Identification of a divalent metal transporter required for cellular iron metabolism in malaria parasites.

Plasmodium falciparum malaria parasites invade and multiply inside red blood cells (RBCs), the most iron-rich compartment in humans. Like all cells, P. falciparum requires nutritional iron to support essential metabolic pathways, but the critical mechanisms of iron acquisition and trafficking during RBC infection have remained obscure. Parasites internalize and liberate massive amounts of heme during large-scale digestion of RBC hemoglobin within an acidic food vacuole (FV) but lack a heme oxygenase to release porphyrin-bound iron. Although most FV heme is sequestered into inert hemozoin cryst

Responses of Rat Mesenchymal Stromal Cells to Nanocellulose with Different Functional Groups.

Cellulose nanofibrils (CNFs) are multiscale hydrophilic biocompatible polysaccharide materials derived from wood and plants. TEMPO-mediated oxidation of CNFs (TO-CNF) turns some of the primary hydroxyl groups to carboxylate and aldehyde groups. Unlike carboxylic functional groups, there is little or no information about the biological role of the aldehyde groups on the surface of wood-based CNFs. In this work, we replaced the aldehyde groups in the TO-CNF samples with carboxyl groups by another oxidation treatment (TO-O-CNF) or with primary alcohols with terminal hydroxyl groups by a reduction

Vitamin D Supplementation for Benign Paroxysmal Positional Vertigo: A Systematic Review.

OBJECTIVE: Benign paroxysmal positional vertigo (BPPV) is commonly attributed to displaced otoconia. These have been shown to have biomineralization close to that of bone, and vitamin D deficiency has been associated with BPPV. We aim to systematically review the available literature on vitamin D supplementation and BPPV intensity and recurrence in adults. DATABASES REVIEWED: PubMed, MEDLINE, EMBASE, Cochrane Central Register of Controlled Trials (CENTRAL), Current Controlled Trials, and ClinicalTrials.gov. METHODS: We systematically reviewed the available literature from 1947 to April 2020.

Nonrenal Systemic Arterial Calcification Predicts the Formation of Kidney Stones.

Background: Recent data indicate that kidney stone formers (KSFs) may have increased biomineralization at anatomic sites throughout the body compared with nonstone formers (NSFs). The objective of this study is to compare the volume of nonrenal systemic calcifications between KSF and NSF by using a standardized system to analyze calcifications in the abdominal aorta (AA) and splenic artery (SA). Methods: The NSF cohort was obtained from a kidney donor's prospectively maintained database. One hundred ninety-seven NSF were matched to 197 KSF based on age, gender, and body mass index. Noncontrast

Bioglass implant-coating interactions in synthetic physiological fluids with varying degrees of biomimicry.

Synthetic physiological fluids are currently used as a first in vitro bioactivity assessment for bone grafts. Our understanding about the interactions taking place at the fluid-implant interface has evolved remarkably during the last decade, and does not comply with the traditional International Organization for Standardization/final draft International Standard 23317 protocol in purely inorganic simulated body fluid. The advances in our knowledge point to the need of a true paradigm shift toward testing physiological fluids with enhanced biomimicry and a better understanding of the materials'

Novel Reduced Graphene Oxide/Zinc Silicate/Calcium Silicate Electroconductive Biocomposite for Stimulating Osteoporotic Bone Regeneration.

In the absence of external assistance, autogenous healing of bone fracture is difficult due to impaired regeneration ability under osteoporosis pathological conditions. In this study, a reduced graphene oxide/zinc silicate/calcium silicate (RGO/ZS/CS) conductive biocomposite with an optimal surface electroconductivity of 5625 S/m was prepared by a two-step spin-coating method. The presence of lamellar apatite nanocrystals on the surfaces of the biocomposite suggests that it has good in vitro biomineralization ability. The silicon and zinc released from the biocomposite induced a significant in

Trichomes of tobacco excrete zinc as zinc-substituted calcium carbonate and other zinc-containing compounds.

Tobacco (Nicotiana tabacum L. cv Xanthi) plants were exposed to toxic levels of zinc (Zn). Zn exposure resulted in toxicity signs in plants, and these damages were partly reduced by a calcium (Ca) supplement. Confocal imaging of intracellular Zn using Zinquin showed that Zn was preferentially accumulated in trichomes. Exposure to Zn and Zn + Ca increased the trichome density and induced the production of Ca/Zn mineral grains on the head cells of trichomes. These grains were aggregates of submicrometer-sized crystals and poorly crystalline material and contained Ca as major element, along with

Formation of the sclerites in Solenogastres and Caudofoveata (Mollusca, Aplacophora).

Solenogastres and Caudofoveata are vermiform, shell-less molluscs, with the mantle covered by calcareous scales or spines (sclerites) embedded in a cuticle secreted by the mantle epithelium. Molecular studies recover them as a single clade (Aplacophora), sister of Polyplacophora, and together, they form the sister group to all other molluscs. Within this phylogenetic framework, sclerites emerge as potentially informative characters for reconstructing early molluscan evolution. However, the cellular mechanisms underlying sclerite formation in Aplacophora remain poorly resolved. Previous work ha

Dual mechanisms of Microbacterium algeriense C14 in alleviating cadmium toxicity during Zinnia elegans seed germination and early seedling growth.

Cadmium (Cd) contamination, a critical type of heavy metal pollution, exerts severe adverse effects on plant growth and poses substantial risks to food safety. Although plant growth-promoting rhizobacteria (PGPR) have demonstrated great potential in mitigating metal-induced stress in plants, a critical limitation remains: most PGPR strains only provide protective effects to plants, failing to reduce the bioavailability of toxic metals in the environment. We herein report the isolation and characterization of Microbacterium algeriense strain C14, which exhibits dual functionality: indole-3-acet

Investigating the effects of disposable face masks leaching compounds on marine biota: a toxicogenomic approach using stress indices and transcriptomics in tissues of mussel Mytilus galloprovincialis.

The present study provides integrated evidence that Disposable Face Masks (DFMs) leaching process could adversely affect marine bivalves, such as the mussel species Mytilus galloprovincialis. Specifically, cellular (through lysosomal membrane stability, using the Neutral Red Retention Time/NRRT), oxidative (by measuring superoxide anions/O2.-, nitric oxides/NO, and lipid peroxidation, via malondialdehyde/MDA content), and genotoxic (by measuring micronucleus/MN formation) effects were determined in hemolymph/hemocytes of mussels exposed for 96 h to artificial seawater containing DFM leaching c

Effect of Hemodialysis With Citrate on Vascular Calcification.

INTRODUCTION: Vascular calcification, specifically medial calcification, is prevalent in patients undergoing hemodialysis (HD) and leads to poor outcomes. Citrate is present in some HD solutions and could potentially inhibit vascular calcification. This was examined by comparing the progression of medial arterial calcification, as measured using mammography, in female patients with end-stage renal disease (ESRD) undergoing HD with and without citrate. METHODS: In a cross-over study, 28 female chronic in-center HD patients with preexisting breast arterial calcification (BAC) underwent HD using

Accurate informatic modeling of tooth enamel pellicle interactions by training substitution matrices with Mat4Pep.

Extracellular matrices direct the formation of mineral constituents into self-assembled mineralized tissues. We investigate the protein and mineral constituents to better understand the underlying mechanisms that lead to mineralized tissue formation. Specifically, we study the protein-hydroxyapatite interactions that govern the development and homeostasis of teeth and bone in the oral cavity. Characterization would enable improvements in the design of peptides to regenerate mineralized tissues and control attachments such as ligaments and dental plaque. Progress has been limited because no ava

Identifying magnetosome-associated genes in the extended CtrA regulon in Magnetospirillum magneticum AMB-1 using a combinational approach.

Magnetotactic bacteria (MTB) are worth studying because of magnetosome biomineralization. Magnetosome biogenesis in MTB is controlled by multiple genes known as magnetosome-associated genes. Recent advances in bioinformatics provide a unique opportunity for studying functions of magnetosome-associated genes and networks that they are involved in. Furthermore, various types of bioinformatics analyses can also help identify genes associated with magnetosome biogenesis. To predict novel magnetosome-associated genes in the extended CtrA regulon, we analyzed expression data of Magnetospirillum magn

Counter-regulatory phosphatases TNAP and NPP1 temporally regulate tooth root cementogenesis.

Cementum is critical for anchoring the insertion of periodontal ligament fibers to the tooth root. Several aspects of cementogenesis remain unclear, including differences between acellular cementum and cellular cementum, and between cementum and bone. Biomineralization is regulated by the ratio of inorganic phosphate (Pi) to mineral inhibitor pyrophosphate (PPi), where local Pi and PPi concentrations are controlled by phosphatases including tissue-nonspecific alkaline phosphatase (TNAP) and ectonucleotide pyrophosphatase/phosphodiesterase 1 (NPP1). The focus of this study was to define the rol

Removing Pb2+ ions from aqueous solution with freshwater cyanobacteria: Combined adsorption and intracellular precipitation.

This study investigated the removal of lead ions (Pb2+) from aqueous solutions by two freshwater cyanobacteria with distinct cellular inclusions: Synechococcus elongatus FACHB-410, which is rich in polyphosphate (polyP), and Cyanothece sp. PCC 7425, containing both polyP and amorphous calcium carbonate (iACC). Both strains were exposed to Pb2+ concentrations ranging from 1 to 50 mg/L and demonstrated high removal efficiencies exceeding 92%. The removal mechanism involved not only adsorption onto cell surfaces but also significant intracellular precipitation. The proportion of intracellular pre

A comprehensive evaluation on the bioactivity, antibacterial efficacy and cell viability of PVA-PVP-chitosan electrospun scaffolds reinforced with calcium/zinc silicate.

Various silicate compounds have been extensively used for scaffold fabrication via electrospinning. However, harnessing the bioactive properties of Calcium silicate (CS) and Zinc silicate (ZS) are less explored in electrospinning. This work discusses the fabrication of PVA-PVP-Chitosan polymer-matrix scaffolds via electrospinning incorporating various percentages of Calcium Silicate/Zinc silicate and the scaffolds are named as PPCC1, PPCC2, and PPCC3 for CS incorporated scaffolds and PPCZ1, PPCZ2 and PPCZ3 for ZS incorporated scaffolds. Leveraging Spatial Transcriptomics to Decode Craniofacial Development.

Understanding how intricate cellular networks and signaling pathways communicate during the formation of craniofacial tissues like the palate and tooth has been the subject of intense investigation for several decades. Both organ systems undergo patterning morphogenesis and the subsequent terminal differentiation of matrix-producing cells that form biomineralized matrices like bone, enamel, dentin, and cementum. Until recently, gene expression profiles could only be assessed for a select number of cells without the context of the entire milieu of genes expressed by neighboring cells and tissue
