"Root phenomics for harnessing hidden-half dynamic plasticity under water stress".

Adapting crops to climate-driven water extremes is a cornerstone of agricultural sustainability. Historically overlooked due to phenotyping constraints, root phenes have now emerged as a primary frontier for enhancing crop resilience. We review root architectural and anatomical responses to contrasting water extremes -drought and waterlogging- across major crop species. We examine key root phenes and their associated functions as modulated by developmental and environmental cues. Non-destructive longitudinal root phenomics offers an unprecedented view of the adaptive plastic responses that und

Rainfed sugarcane under water limitation: agronomic challenges and breeding perspectives.

Sugarcane is a highly productive crop due to its high photosynthetic efficiency. Such productivity is associated with high water requirements, making sugarcane sensitive to water limitation. With a predominance of rainfed cultivation worldwide, sugarcane is increasingly exposed to drought under changing climate conditions. We highlight how sugarcane's response to water limitation is driven by plant developmental stage, canopy architecture and dynamics, root system functioning, and how these factors influence the identification of traits related to drought tolerance. We synthesize current knowl

PELOTA at the Crossroads of Ribosome-associated quality control and Silencing: Coordinating Development, Stress, and Immunity in Plants.

Ribosome-associated quality control (RaQC) pathways, including no-go decay (NGD) and non-stop decay (NSD), are essential for maintaining translational fidelity and regulating gene expression in eukaryotes. Central to these pathways is the conserved ribosome rescue factor PELOTA, which resolves stalled ribosomes and promotes the clearance of aberrant mRNAs and nascent polypeptides. While NGD and NSD have been extensively characterized in yeast and animals, our understanding of these processes in plants remains limited. Nevertheless, emerging evidence indicates that PELOTA plays a pivotal role i

A specific 296 bp insertion in the DFR1 promoter activates ethylene-mediated anthocyanin biosynthesis during autumn leaf senescence in Quercus aliena.

Red-leaf ornamental trees are commonly characterized by the enrichment of anthocyanins. Quercus aliena, a dominant native oak species in China, displays striking autumn foliage with yellow to red colors. However, the molecular regulation of anthocyanin biosynthesis during leaf senescence remains largely unknown. In this study, we identified a 296 bp insertion in the promoter of Dihydroflavonol 4-Reductase (DFR1) from the red-leaf Q. aliena cultivar 'Qiuyun', which was significantly associated with high anthocyanin accumulation. We established a highly efficient method for genetic transformatio

Phylogenomic insights into the genus Tulipa (Liliaceae): Taxonomy, evolution, and biogeography.

PREMISE: Tulips are one of the best-known geophytes, but their taxonomy remains convoluted and their evolutionary history poorly understood. Here, we used plastid genomes to identify some issues with current classification, understand the diversification history of the genus, and identify potential species linked to historical cultivation. METHODS: We gathered a large number of tulip specimens from living collections, herbaria, and online sources and through extensive fieldwork to infer maximum likelihood phylogenies of the plastid genomes of Tulipa (Liliaceae), representing ~86% of accepted

EXPANSIN A6 and GRAIN WIDTH 2 genes differentially affect grain weight, grain number and yield of wheat through their physiological determinants.

The physiological basis of the trade-off between grain number (GN) and thousand grain weight (TGW) is key to wheat yield improvement. To that end, three wheat line groups were assessed at conventional (300-350 pl m-2) and low (44 pl m-2) planting rates in field experiments: TaExpA6 (EXPANSIN A6 overexpression), TaGW2 (GRAIN WIDTH 2 triple knockout), and TaP1xGW2A (VEGETATIVE TO REPRODUCTIVE TRANSITION 2 ectopic expression, and TaGW2-A knockout), together with their wild types (WT). Lines TaExpA6 and TaGW2 increased TGW over their respective WTs, whereas line TaP1xGW2A showed no effect on this

Transcription factors SlBBX20 and SlMYB9 in tomato modulate plant height through gibberellin metabolic and response pathways.

Gibberellins (GAs) are plant hormones that regulate growth and development. We investigated how a transcription factor, SlBBX20, modulates plant height by affecting GA metabolism and response in tomato (Solanum lycopersicum L.). We found that overexpression of SlBBX20 caused dwarfism, which was rescued by GA treatment, while knockout of SlBBX20 increased plant height. SlBBX20 directly activated the expression of GA metabolic oxidase genes (SlGA2ox1, SlGA2ox4, and SlGA2ox5) and repressed the expression of a GA-responsive gene, SlMYB9. Plants overexpressing SlMYB9 grew taller than wild type and

Fine-mapping, candidate gene identification, and marker development for the apple scab resistance gene Rvi2.

Breeding elite apple cultivars with scab resistance is a key global goal, as reliance on fungicides is unsustainable. The causal fungus, Venturia inaequalis, evolves rapidly, threatening cultivars with single-gene resistance. Since the 1980s, breeding programmes have introduced novel resistance sources via backcrossing. Here, we generated a haplotype-phased genome assembly of Russian apple R12740-7A and an Oxford Nanopore assembly of the Rvi2-resistance accession TSR34T15, enabling detailed dissection of the Rvi2 resistance locus. Fine-mapping using a 'Royal Gala' × TSR34T15 segregating family

A viral protein promotes host L-asparaginase activity and weakens the asparagine-induced resistance pathway for the benefit of virus infection.

Amino acids are not only essential for plant nutrition but also serve as critical immune signaling molecules, particularly during pathogen invasion. Pathogens can manipulate amino acid metabolic pathways to counteract host immune defenses, yet the underlying mechanisms remain poorly understood. Here, we demonstrate that the pepper mild mottle virus (PMMoV) 126 kDa protein interacts with the host L-asparaginase (LA), identified as a negative regulator of antiviral defense. LA converts asparagine (Asn) to aspartic acid (Asp). Exogenous application of Asn markedly enhanced resistance to PMMoV, wh

Sulfur in dialogue with phosphorus, nitrogen, and iron: regulatory networks in plant nutrient homeostasis.

Sulfur (S) is an essential macronutrient for plant growth and resilience. The S-amino acids cysteine (Cys) and methionine (Met) are indispensable for protein synthesis and structural integrity, as well as redox homeostasis and cofactor assembly. Over the past several decades, biochemical and molecular genetic studies demonstrated the core steps in sulfate uptake and assimilation pathways, while it has become increasingly evident that S homeostasis in plants cannot be understood in isolation. Robust and reciprocal regulatory interactions link S with phosphorus (P), nitrogen (N), and iron (Fe).

Immature fruit abscission is triggered by hormonal changes in the seed coat of avocado.

Avocado (Persea americana) is an economically important tree crop that exhibits a high rate of immature fruit abscission (IFA), reducing yield. As the seed coat derived from recently abscised fruitlets displays a senescence phenotype, we hypothesized that this maternal organ plays a critical role in initiating IFA. Here, we show that fruitlets fated to abscise undergo growth arrest before shrinking and detaching from the tree. Comparative RNA-seq analysis indicated that growth arrest is associated with a transcriptome reprogramming that is first initiated in the seed coat and then transmitted

Taxonomic reassessment of fossil Sequoia and Protosequoia from the Upper Miocene of Central Honshu, Japan, with implications for leaf morphological variation in extant S. sempervirens.

PREMISE: Since its emergence in the Mesozoic, Sequoia (Cupressaceae) has been considered to possess conserved leaf morphology. However, recent studies have shown that the leaves of extant S. sempervirens become smaller, with a scale form, with increasing tree height. The fossil-genus Protosequoia has a similar scale leaf morphology, but its relationship to co-occurring Sequoia fossils, which bear linear leaves, remains unresolved. METHODS: To resolve this relationship, we re-examined specimens of two fossil species of Sequoia and Protosequoia from the Upper Miocene of Japan, focusing on leaf

Placing very long branch taxa in the plant tree of life: A case study with Dioscoreales mycoheterotrophs.

PREMISE: The inclusion of heterotrophic plants in broader systematic studies is challenging due to substantial morphological modification, gene loss, and accelerated substitution rates following photosynthesis loss. In particular, inclusion of very long branch (VLB) taxa with highly elevated rates may lead to phylogenomic misinference. METHODS: We explored how rapidly evolving plastomes behave in model-based phylogenetic inferences in the yam order Dioscoreales, which experienced convergent photosynthesis losses. Our taxon sampling included mycoheterotrophic lineages displaying a broad range

Plant bioacoustics: decoding ultrasonic emissions.

For decades, plant-environment interactions were understood to be primarily mediated by chemical and visual stimuli. However, recent evidence has unveiled an underexplored dimension of plant physiology: airborne ultrasonic emissions. This review evaluates current research in plant bioacoustics, focusing on ultrasound generation, characteristic features, and specificity across different stress conditions. We discuss proposed mechanisms for plant ultrasound production, with a primary emphasis on xylem cavitation. Advancements in non-contact monitoring have enabled the characterization of acousti

Positive pressure does not efficiently repair xylem embolism after drought.

Numerous plant species can actively pressurize water in their xylem. It has been hypothesized that regular peaks in positive pressure help plants to remove xylem embolism and to replenish water reservoirs. This may happen after rewatering when plants have suffered from soil water deficit (WD) and/or following diurnal cycles at well-watered (WW) conditions. Using X-ray microcomputed tomography (µCT), we tested these hypotheses in tomato plants by quantifying xylem functionality and internal water pools in stems of 35 plants subjected to WD intensities before and after rewatering and 14 WW plant

Mechanisms of intrusive growth of secondary xylem fibers in flax (Linum usitatissimum L.).

This study explores the mechanisms of intrusive growth in secondary xylem fibers, a process determining their length and future mechanical performance. The flax variety CDC Bethune and the derived from it rdf mutant with reduced fiber length were analyzed to identify factors influencing intrusive growth. The structure and development of secondary xylem, as well as the composition, architecture, and mechanical properties of cell walls in xylem fibers, were studied using microscopy. Hypotheses of intrusive growth initiation were tested using computer modeling. Biomechanical differences between c

A wheat cultivar with reduced root ethylene sensitivity shows improved root penetration, leaf water relations and yield in dry compact soil.

Soil compaction is an increasing problem in global agriculture, causing more severe effects when soil dries. Since insensitivity to the plant hormone ethylene maintains root elongation in compact soil, wheat cultivars with reduced sensitivity to ethylene were hypothesised to have greater root and shoot growth in compact soil, and higher plant water status when this soil dried. Screening wheat cultivars revealed root-applied ethephon solution or exposure to ethylene gas inhibited root elongation of the cultivar Chewink#1 more than Kutz. These cultivars were glasshouse-grown in soil columns with

Incomplete climate-driven peripatric speciation in Moehringia sect. Moehringia (Caryophyllaceae) in the European Alps.

PREMISE: The origin of endemic species in the European Alps is commonly attributed to the climatic oscillations of the Quaternary. Moehringia sect. Moehringia, with 12 of 15 species endemic to the Alps and mostly restricted to well-known glacial refugia, is a prime system for investigating the diversification of a lineage originating in the Alps. METHODS: We used all species in this lineage, most of which were represented by multiple accessions, and a target enrichment approach with baits specifically designed for Moehringia to obtain a phylogenetic tree from 829 nuclear loci. We also dated t

Phytohormone-microbiome interactions and epigenetic regulation of plant stress responses and priming.

Plants continuously face fluctuating environmental conditions, requiring tightly coordinated regulatory systems to balance growth and stress responses. This review synthesizes current knowledge on phytohormones as central integrators of plant-microbiome interactions, highlighting their dual role as internal regulators and ecological gatekeepers that shape microbiome assembly and function. Phytohormones, such as auxins, cytokinins, gibberellins, abscisic acid, ethylene, salicylic acid, and jasmonates, dynamically regulate plant development, immunity, and rhizosphere chemistry, thereby influenci
