Current cell-based therapies administered after myocardial infarction (MI) show limited efficacy due to subpar cell retention in a dynamically beating heart. In particular, cardiac patches generally provide a cursory level of cell attachment due to the lack of an adequate microenvironment. From this perspective, decellularized cell-derived ECM (CDM) is attractive in its recapitulation of a natural biophysical environment for cells. Unfortunately, its weak physical property renders it difficult to retain in its original form, limiting its full potential. Here, a novel strategy to peel CDM off f

BACKGROUND: The reconstruction of heart valves provides substantial benefits, particularly in the pediatric population. We present our experience using decellularized extracellular matrix (dECM, CorMatrix® ) for aortic valve procedures.
METHODS: We retrospectively reviewed the case histories of 6 patients (aged from 2 months - 14 years) who underwent surgery for severe aortic valve stenosis (n = 4) or regurgitation (n = 2). Aortic valve repair was performed on all patients using dECM as a leaflet replacement or leaflet extension. Follow-ups were performed using echocardiography. Reoperation w

BACKGROUND: The extracellular matrix (ECM) of articular cartilage has an inhibitory effect on vascularization, yet clinical utilization has been technically challenging. In this study, we aimed to fabricate a biologically functional ECM powder suspension from porcine articular cartilage that inhibits neovascularization (NV).
METHODS: The digested-cartilage acellular matrix (dg-CAM) was prepared by sequential processes of decellularization, enzymatic digestion and pulverization. Physicochemical properties of dg-CAM were compared with that of native cartilage tissue (NCT). Cellular interactions

UNLABELLED: Extracellular matrix (ECM) scaffolds are being used for the clinical repair of soft tissue injuries. Although improved functional outcomes have been reported, ECM scaffolds show limited tissue specific remodeling response with concomitant deposition of fibrotic tissue. One plausible explanation is the regression of blood vessels which may be limiting the diffusion of oxygen and nutrients across the scaffold. Herein we develop a composite scaffold as a vasculo-inductive platform by integrating PEGylated platelet free plasma (PFP) hydrogel with a muscle derived ECM scaffold (m-ECM).

Severe peripheral nerve injury (PNI) requiring nerve graft remains a clinical challenge due to limitations associated with currently available grafts. While decellularized nerve grafts (DNGs) are commonly used, their efficacy is largely restricted to short-gap repairs due to their acellular and dense structure, which poses a persistent challenge in the treatment of critical long-gap nerve defects. It is hypothesized that making porous DNGs (PDNGs) can create a suitable microenvironment that would facilitate the cell infiltration, recellularization, and further axonal growth to enhance nerve re

BACKGROUND: The current standard of care to treat a ruptured anterior cruciate ligament (ACL) is ACL reconstruction (ACLR), which involves replacing the torn ligament with a tendon graft. The implanted tendon graft undergoes a graft-maturation process known as ligamentization. However, the synthesis of type III collagen, which is typically found in weaker scar tissue, continues at higher concentrations, and the heterogeneous composition of collagen fibers of varying diameters of native ACL is never restored. Culture-expanded ACL-derived cells reseeded to tendon graft have shown the potential f

Bone related diseases such as osteoporosis, osteoarthritis, metastatic bone cancer, osteogenesis imperfecta, and Paget's disease, are primarily treated with pharmacologic therapies that often exhibit limited efficacy and substantial side effects. Bone injuries or fractures are primarily repaired with biocompatible materials that produce mixed results in sufficiently regenerating healthy and homogenous bone tissue. Each of these bone conditions, both localized and systemic, use different strategies with the same goal of achieving a healthy and homeostatic bone environment. In this study, we dev

Partial or whole regeneration of the uterine endometrium using extracellular matrix (ECM)-based scaffolds is a therapeutic strategy for uterine infertility due to functional and/or structural endometrial defects. Here, we examined whether the entire endometrium can be regenerated circumferentially using an acellular ECM scaffold (decellularized endometrial scaffold, DES) prepared from rat endometrium. We placed a silicone tube alone to prevent adhesions or a DES loaded with a silicone tube into a recipient uterus in which the endometrium had been surgically removed circumferentially. Histologi

Defects of articular joints are becoming an increasing societal burden due to a persistent increase in obesity and aging. For some patients suffering from cartilage erosion, joint replacement is the final option to regain proper motion and limit pain. Extensive research has been undertaken to identify novel strategies enabling earlier intervention to promote regeneration and cartilage healing. With the introduction of decellularized extracellular matrix (dECM), researchers have tapped into the potential for increased tissue regeneration by designing biomaterials with inherent biochemical and i

PURPOSE OF REVIEW: Urethrotomy remains the first-line therapy in the treatment of a urethral stricture despite data showing no real chance of a cure after repeated urethrotomies. An anastomotic or an augmentation urethroplasty using oral mucosa can be offered to patients following failed urethrotomy. The potential for a tissue engineered solution as an alternative to native tissue has been explored in recent years and is reviewed in this article.
RECENT FINDINGS: More than 80 preclinical studies have investigated a tissue-engineered approach for urethral reconstruction mostly using decellular

Extraembryonic tissues play a crucial role in embryonic development and fetal growth and exhibit remarkable biological and biomechanical properties including, pro-angiogenic, anti-inflammatory, immunomodulatory, anti-fibrotic, antimicrobial, and viscoelastic. These characteristics have driven the development of innovative therapies for regenerative medicine applications. Unlike other biological tissues, the mammalian placenta is considered waste material, making it a highly accessible and easily obtainable resource that poses no additional risks to the health of the mother or fetus. BACKGROUND: Tendon injuries cause pain and functional limitations, posing a clinical burden due to poor healing and recurrence. Combining mesenchymal stem cells (MSCs), tissue-specific extracellular matrix (ECM) scaffolds, and bioactive factors may enhance tendon regeneration. This study investigated integrating adipose-derived MSCs (AD-MSCs) seeded on a decellularized tendon scaffold (DTS) with G-CSF injection for tendon repair.
METHODS: DTS was prepared from rat Achilles tendons through freeze-thaw cycles and sodium dodecyl sulfate decellularization, preserving ECM structure. In vitro, AD-M

Autologous chondrocyte implantation (ACI) is a promising approach to repair cartilage defects; however, the cartilage trauma-induced inflammatory environment compromises its clinical outcomes. Cell-derived decellularized extracellular matrix (DECM) has been used as a culture substrate for mesenchymal stem cells (MSCs) to improve the cell proliferation and lineage-specific differentiation. In this study, DECM deposited by synovium-derived MSCs was used as an in vitro expansion system for rabbit articular chondrocytes and the response of DECM-expanded chondrocytes to pro-inflammatory cytokines s

OBJECTIVES: Although adipose-derived stem cells (ADSCs) have shown promise in cardiac regeneration, stable engraftment is still challenging. Acellular bioengineered cardiac patches have shown promise in positively altering ventricular remodeling in ischemic cardiomyopathy. We hypothesized that combining an ADSC sheet approach with a bioengineered patch would enhance ADSC engraftment and positively promote cardiac function compared with either therapy alone in a rat ischemic cardiomyopathy model.
METHODS: Cardiac patches were generated from poly(ester carbonate urethane) urea and porcine decel

BACKGROUND: Metastasis is the leading cause of cancer-related mortality, yet experimental models often fail to recapitulate the tissue-specific microenvironments shaping metastatic dissemination. While in vivo systems provide physiological relevance, they remain challenging for mechanistic studies. Conversely, conventional in vitro assays lack the organ-specific extracellular matrix (ECM) that regulates invasive behavior. Accessible models that balance biological relevance with experimental feasibility are thus needed.
RESULTS: We developed an ex vivo invasion platform based on mild detergent

Premature ovarian insufficiency (POI) means menopause before 40 years of age affecting about 1 % of women. Approaches based on cell therapy and the paracrine effects of stem cells or bioproducts such as platelet-rich plasma have been proposed, but concerns remain about undesired systemic effects, as well as the need to optimize delivery methods through bioengineering methods. This study explores the efficacy of decellularized bovine ovarian cortex extracellular matrix (OvaECM) hydrogels alone and as a growth factor (GF) carrier (OvaECM+GF) in a chemotherapy-induced POI murine model. In vitro a

BACKGROUND: Induced pluripotent stem cell-derived cardiomyocytes (iPSC-CM) are regarded as promising cell type for cardiac cell replacement therapy, but it is not known whether the developmental stage influences their persistence and functional integration in the host tissue, which are crucial for a long-term therapeutic benefit. To investigate this, we first tested the cell adhesion capability of murine iPSC-CM in vitro at three different time points during the differentiation process and then examined cell persistence and quality of electrical integration in the infarcted myocardium in vivo.

Chronic diabetic wounds remain a major clinical challenge due to impaired angiogenesis, persistent inflammation, and delayed extracellular matrix remodeling. Stabilization of hypoxia-inducible factor-1 alpha (HIF-1α) represents a promising therapeutic strategy to restore pro-healing signaling in diabetic tissues. In the present study, a bioactive decellularized dermal matrix scaffold (DDMS) loaded with the hypoxia-mimetic agent dimethyloxalylglycine (DMOG) was developed and evaluated for its regenerative potential in a type 2 diabetic wound model. Type 2 diabetes was induced in Wistar rats usi

Traditional two-dimensional (2D) cell cultures fail to capture the structural, mechanical, and biochemical complexity of living tissues, prompting a major paradigm shift toward three-dimensional (3D) cell culture models. 3D systems faithfully replicate in vivo physiological architectures, dynamic cell-to-cell contacts, and extracellular matrix (ECM) interactions, bridging a critical gap in preclinical studies by predicting human therapeutic responses more accurately. This review explores the technological evolution from simple multicellular tumor spheroids to sophisticated microphysiological n
