A novel three-dimensional method for detailed analysis of RGC central projections under acute ocular hypertension.

Normal perception of visual information relies not only on the quantity and quality of retinal ganglion cells (RGCs), but also on the integrity of the visual pathway, within which RGC central projection predominates. However, the exact changes of RGC central projection under particular pathological conditions remain to be elucidated. Here, we report a whole-brain clearing method modified from iDISCO for 3D visualization of RGC central projection. The CTB-labeled RGC central projection was visualized three-dimensionally with minimized both fluorescence quenching and the time taken. For observat

Persistent Thalamic Sound Processing Despite Profound Cochlear Denervation.

Neurons at higher stages of sensory processing can partially compensate for a sudden drop in peripheral input through a homeostatic plasticity process that increases the gain on weak afferent inputs. Even after a profound unilateral auditory neuropathy where >95% of afferent synapses between auditory nerve fibers and inner hair cells have been eliminated with ouabain, central gain can restore cortical processing and perceptual detection of basic sounds delivered to the denervated ear. In this model of profound auditory neuropathy, auditory cortex (ACtx) processing and perception recover despit

Effect of retinal ischemia on the non-image forming visual system.

Retinal ischemic injury is an important cause of visual impairment. The loss of retinal ganglion cells (RGCs) is a key sign of retinal ischemic damage. A subset of RGCs expressing the photopigment melanopsin (mRGCs) regulates non-image-forming visual functions such as the pupillary light reflex (PLR), and circadian rhythms. We studied the effect of retinal ischemia on mRGCs and the non-image-forming visual system function. For this purpose, transient ischemia was induced by raising intraocular pressure to 120 mm Hg for 40 min followed by retinal reperfusion by restoring normal pressure. At 4 w

Age-related changes in calbindin and calretinin immunoreactivity in the central auditory system of the rat.

Age-related changes in the levels of major intracellular calcium buffers are known to occur in different parts of the mammalian brain, including the central auditory pathway. In the present study, we evaluate with immunohistochemistry and the western blot technique the effect that aging has on the calbindin- and calretinin-expressing system of neurons in the higher structures of the central auditory pathway, in the inferior colliculus (IC), medial geniculate body (MGB) and auditory cortex (AC) of two rat strains, the slowly aging Long-Evans and the fast aging Fischer 344. Interestingly, the ag

Experience-dependent retinogeniculate synapse remodeling is abnormal in MeCP2-deficient mice.

Mutations in MECP2 underlie the neurodevelopmental disorder Rett syndrome (RTT). One hallmark of RTT is relatively normal development followed by a later onset of symptoms. Growing evidence suggests an etiology of disrupted synaptic function, yet it is unclear how these abnormalities explain the clinical presentation of RTT. Here we investigate synapse maturation in Mecp2-deficient mice at a circuit with distinct developmental phases: the retinogeniculate synapse. We find that synapse development in mutants is comparable to that of wild-type littermates between postnatal days 9 and 21, indicat

Paired pulse facilitation of corticogeniculate EPSCs in the dorsal lateral geniculate nucleus of the rat investigated in vitro.

To investigate paired pulse facilitation of corticogeniculate EPSCs, whole-cell patch-clamp recordings were made from principal cells in the rat dorsal lateral geniculate nucleus (dLGN) in vitro. Thalamic slices, oriented so that both corticogeniculate and retinogeniculate axons could be stimulated, were cut from young (16- to 37-day-old) DA-HAN rats. Corticogeniculate EPSCs displayed pronounced paired pulse facilitation at stimulus intervals up to 400 ms. The facilitation had a fast and a slow component of decay with time constants of 12 +/- 7 and 164 +/- 47 ms (means +/- S.D.), respectively.

Cerebral microgyria, thalamic cell size and auditory temporal processing in male and female rats.

Induction of microgyria by freezing injury to the developing somatosensory cortex of neonatal rats causes a defect in fast auditory processing in males, but not in females. It was speculated that early damage to the cortex has sexually dimorphic cascading effects on other brain regions mediating auditory processing, which can lead to the observed behavioral deficits. In the current series of experiments, bilateral microgyri were induced by placement of a freezing probe on the skulls of newborn male and female rats, and these animals were tested in adulthood for auditory temporal processing. [Hemianopic visual field defects--methods of study and localization problems].

In a survey of methods for determining hemianopic visual field defects, a distinction is made between gross visual field screening and actual visual field examination with kinetic and automatic static perimetry. These methods may be arranged according to increasing the concentration required as well as to ability to cooperate, as follows: hand-movement stimulation of shifts of gaze, finger-counting with repetition by the patient, brightness and color comparison among the respective halves and quadrants of the visual field, then kinetic perimetry with the Goldmann perimeter, and, most exacting

Geniculotectal neurons of the lateral part of the ventral lateral geniculate nucleus in the hereditary microphthalmic rat: a retrograde WGA-HRP study.

The distribution and morphological characteristics of the geniculotectal neurons from the lateral part of the ventral lateral geniculate nucleus in the hereditary microphthalmic rat were examined by the WGA-HRP method. Unilateral injection of the tracer into the microphthalmic superior colliculus showed that WGA-HRP-positive neurons were present in the ipsilateral lateral part of the ventral lateral geniculate nucleus. However, the number of labeled geniculotectal neurons of the microphthalmia was about 50% that of the normal rats. Furthermore, the size of labeled geniculotectal neurons of the

Cortical and subcortical projections of the middle temporal area (MT) and adjacent cortex in galagos.

Projections of the middle temporal visual area, MT, and of visual cortex adjoining MT were investigated with autoradiographic methods in the prosimian primate, Galago senegalensis. Ipsilateral cortical targets of MT included area 17, area 18, cortex caudal to MT, cortex ventral to MT, and parietal-occipital cortex dorsal to MT. This pattern of projections suggests that extrastriate cortex contains a number of visual subdivisions in addition to MT. Contralateral projections were to MT and parietal-occipital cortex. Projections from MT to areas 17 and 18 connected regions representing similar pa

Laminar segregation of excitatory and inhibitory projections from the inferior colliculus to the superior colliculus.

The superior colliculus (SC) integrates multimodal spatial information to guide orientation behaviors. Visual inputs to superficial layers drive these behaviors through circuits in deeper layers, which also receive input from other sensory modalities. The inferior colliculus (IC), a central midbrain hub of auditory system, provides both VGluT2-expressing excitatory and VGAT/GAD67-expressing inhibitory projections to the SC, but the organization of these projections remains incompletely understood. In this study, we systematically mapped excitatory and inhibitory IC projections to the SC. Cre-d

Deep Brain Stimulation for Primary Refractory Tinnitus: A Systematic Review.

BACKGROUND: tinnitus is a common and often debilitating condition with limited evidence-based treatment options. Deep brain stimulation (DBS) is an approved treatment modality for certain neurological conditions; its experimental use as a treatment modality for severe tinnitus is novel and beginning to show promise. This systematic review focuses on the current evidence for the safety and efficacy of DBS for treatment of refractory tinnitus. METHODS: a systematic search in PubMed and EMBASE was performed to identify peer-reviewed studies on DBS of non-cortical structures for the primary indic

Neural correlates of sensory gating in the rat: decreased Fos induction in the lateral septum.

In the P(50) gating or conditioning-testing paradigm in the rat, two identical click stimuli are presented with an inter-click interval of 500 ms. The reaction towards the second click, as measured with evoked potentials, is reduced in respect to that towards the first click; this phenomenon is called sensory gating. In the present experiments, the inter-click interval was varied systematically and auditory evoked potentials were measured. Sensory gating was found to occur only at intervals between 500 and 1000 ms, but not at longer intervals. Fos immunohistochemistry was then performed using

Chemotherapeutic treatment of extensive optic pathway tumors in infants.

Two infants, ages 14 and 4 months, with extensive optic pathway tumors were treated with intensive chemotherapy called MADDOC: nitrogen mustard, doxorubicin, cis-platinum, dacarbazine, vincristine, and cyclophosphamide. The first child had hydrocephalus with an enhancing mass at the hypothalamus which followed the optic radiation to include the lateral geniculate body and medial temporal lobe. A v-p shunt was placed, and biopsy revealed a Grade II astrocytoma. One month later, the child developed malignant ascites. Intensive induction chemotherapy was then begun with cis-platinum 100 mg/m2 and

Subcortical projections to the pontine nuclei in the cat.

In a systematic attempt to trace all projections from the brainstem and diencephalon to the pontine nuclei of the cat, implantations and injections of horseradish peroxidase-wheat germ agglutinin (HRP-WGA) or Fluoro-Gold were placed in the pontine nuclei of 21 cats. In most of the cases there was no evidence of spread of tracer outside the pontine nuclei. Retrogradely labeled cells in the brainstem and diencephalon were carefully mapped and counted. The number labeled cells in the brainstem and diencephalon ranged from 24 in cases with very small implantations to 3,490 in cases with large inje

Circuitry Underlying Experience-Dependent Plasticity in the Mouse Visual System.

Since the discovery of ocular dominance plasticity, neuroscientists have understood that changes in visual experience during a discrete developmental time, the critical period, trigger robust changes in the visual cortex. State-of-the-art tools used to probe connectivity with cell-type-specific resolution have expanded the understanding of circuit changes underlying experience-dependent plasticity. Here, we review the visual circuitry of the mouse, describing projections from retina to thalamus, between thalamus and cortex, and within cortex. We discuss how visual circuit development leads to

Neuronal degeneration in the dorsal lateral geniculate nucleus following lesions of primary visual cortex: comparison of young adult and geriatric marmoset monkeys.

Neuronal loss in the lateral geniculate nucleus (LGN) is a consequence of lesions of the primary visual cortex (V1). Despite the importance of this phenomenon in understanding the residual capacities of the primate visual system following V1 damage, few quantitative studies are available, and the effect of age at the time of lesion remains unknown. We compared the volume, neuronal number, and neuronal density in the LGN, 6-21 months after unilateral V1 lesions in marmoset monkeys. Stereological sampling techniques and neuronal nuclei (NeuN) staining were used to assess the effects of similar-s

Changes in visual fields and lateral geniculate nucleus in monkey laser-induced high intraocular pressure model.

Monkey eyes are useful for ophthalmologic research into eye diseases because their histological and functional properties are very similar to those of humans. The monkey laser-induced high intraocular pressure (IOP) model is a common model for ophthalmologic research, especially into glaucoma. Although several studies using this model have focused on changes in visual field, retinal ganglion cells (RGC), and lateral geniculate nucleus (LGN), clear relationships among these changes in one and the same monkey have not been established. We therefore examined visual field changes, RGC and LGN numb

Parallel thalamocortical pathways for echolocation and passive sound localization in a gleaning bat, Antrozous pallidus.

We present evidence for parallel auditory thalamocortical pathways that serve two different behaviors. The pallid bat listens for prey-generated noise (5-35 kHz) to localize prey, while reserving echolocation [downward frequency-modulated (FM) sweeps, 60-30 kHz] for obstacle avoidance. Its auditory cortex contains a tonotopic map representing frequencies from 6 to 70 kHz. The high-frequency (BF > 30 kHz) representation is dominated by FM sweep-selective neurons, whereas most neurons tuned to lower frequencies prefer noise. Retrograde tracer injections into these physiologically distinct cortic
