Inter-plant communication in broccoli (Brassica oleracea var. italica) through root colonization with the endophytic fungus Trichoderma hamatum: New findings in a "wired communication".

Inter-plant communication has emerged as a critical yet poorly understood component of plant defense strategies, particularly belowground and beyond mycorrhizal systems. Our work demonstrates that the endophytic fungus Trichoderma hamatum mediates effective "wired communication" between neighboring broccoli (Brassica oleracea var. italica) plants, enhancing resistance against the necrotrophic pathogen Sclerotinia sclerotiorum. Using a novel axenic culture system that allows controlled hyphal connections between plant roots, we show that foliar infection of one plant triggers systemic defense p

Bio-preservative potential of marjoram and fennel essential oil nano-emulsions against toxigenic fungi in citrus: integrating in-vitro, in-vivo, and in-silico approaches.

Citrus fruits, known for their vibrant flavours and health benefits, are susceptible to fungal attacks, particularly from toxigenic Penicillium fungi, which pose a significant pre- and post-harvest hazard. However, aromatic oils and their nanoparticles may effectively address this issue. Marjoram and fennel oils, alongside their nanoparticles, were extracted, and their aromatic constituents and antimicrobial activities were evaluated. A simulated medium with fungal spores was used to assess anti-toxigenic activity, and a simulated infection experiment was conducted with orange and lemon fruits

Biocontrol potential of the active substance isolated from the endophytic fungus Aa-Lcht against apple Valsa canker.

Apple Valsa canker (AVC), primarily caused by the pathogenic fungus Valsa mali, is a devastating disease of apple. The development and application of high efficiency and low toxicity fungicides are of great significance for disease control. Natural active substances serve as a vital foundation for the development of novel green fungicides. In previous studies, the endophytic fungus Aa-Lcht was confirmed to contain specific inhibitory effect against V. mali. This study confirmed that Aa-Lcht fermentation broth (FB)'s suppression of V. mali growth and conidial germination, exhibiting AVC prevent

Growth and differentiation properties of pikromycin-producing Streptomyces venezuelae ATCC15439.

Streptomycetes naturally produce a variety of secondary metabolites, in the process of physiological differentiation. Streptomyces venezuelae differentiates into spores in liquid media, serving as a good model system for differentiation and a host for exogenous gene expression. Here, we report the growth and differentiation properties of S. venezuelae ATCC-15439 in liquid medium, which produces pikromycin, along with genome-wide gene expression profile. Comparison of growth properties on two media (SPA, MYM) revealed that the stationary phase cell viability rapidly decreased in SPA. Submerged

Enzymatic System of Antioxidant Protection of Erythrocytes in Diabetic Rats Treated with Medicinal Mushrooms Agaricus brasiliensis and Ganoderma lucidum (Agaricomycetes).

Excessive glucose concentrations in blood and cells promote the intensification of auto-oxidation. This is one of the mechanisms through which free radicals form in hyperglycemia. As a result of hyperglycemia, oxidative stress develops and lipid peroxidation (LPO) is enhanced. Erythrocytes are particularly susceptible to reactive oxygen species and LPO, which can violate cell functions. This article describes the analysis of the influence of mycelia from the medicinal mushrooms Agaricus brasiliensis and Ganoderma lucidum on the enzymatic link of the antioxidant system in rat erythrocytes under

Constitutive overexpression of the sucrose transporter SoSUT1 in potato plants increases arbuscular mycorrhiza fungal root colonization under high, but not under low, soil phosphorus availability.

The sucrose transporter SUT1 functions in phloem loading of photoassimilates in solanaceous plant species. In the present study, wildtype and transgenic potato plants with either constitutive overexpression or antisense inhibition of SUT1 were grown under high or low phosphorus (P) fertilization levels in the presence or absence of the arbuscular mycorrhizal (AM) fungus Glomus intraradices. At a low soil P fertilization level, the extent of AM fungal root colonization was not different among the genotypes. In all plants, the AM symbiosis contributed significantly to P uptake under these condit

Cross-Resistance Between Pseudomonas chlororaphis Strain AFS009 Metabolites (Howler EVO) and Fludioxonil in Botrytis cinerea.

Howler EVO is a biological fungicide based on metabolites of the bacterium Pseudomonas chlororaphis strain AFS009. One of the metabolites, pyrrolnitrin, is a chemical analogue of the phenylpyrrole fludioxonil used to manage gray mold of fruit crops caused by Botrytis cinerea. Resistance to fludioxonil in B. cinerea is well documented and linked to mutations in the transcription factor mrr1, leading to overexpression of the ATP-binding cassette transporter gene BcatrB. Moderately resistant isolates are designated MDR1 and MDR1h based on the specific variation of mutations in mrr1 and the level

Erinacine S, a small active component derived from Hericium erinaceus, protects oligodendrocytes and alleviates mood abnormalities in cuprizone-exposed rodents.

Hericium erinaceus mycelium extract (HEM), containing erinacine A (HeA) and erinacine S (HeS), has shown promise in promoting the differentiation of oligodendrocyte precursor cells (OPCs) into mature oligodendrocytes (OLs), crucial for myelin production in the central nervous system (CNS). The main aim of this study was to characterize the protective effects of HEM and its components on OLs and myelin in demyelinating rodents by exposure to cuprizone (CPZ), a copper chelating agent commonly used to induce demyelination in the corpus callosum of the brain. Rats were fed by CPZ-containing diet a

Comparison of antifungal activity of selected essential oils against Fusarium graminearum in vitro.

INTRODUCTION: Fusaria are microscopic filamentous fungi which are spread in soil, in various organic substrates, and include more than 80 phytopathogenic species which are predominantly hosted by cereals, fruits and vegetables. Many of these species, under certain conditions, are capable of synthesizing secondary metabolites, mycotoxins. At present, various substances are used for their elimination and one of the solutions appears to be essential oils. In the presented study, the antifungal activity of essential oils was researched in vitro. MATERIAL AND METHODS: In this study, two standard f

Gα-cAMP/PKA pathway positively regulates pigmentation, chaetoglobosin A biosynthesis and sexual development in Chaetomium globosum.

Sensing the environmental signals, the canonical Gα-cAMP/PKA pathway modulates mycelial growth and development, and negatively regulates some secondary metabolism in filamentous fungi, e.g. aflatoxin in Aspergillus nidulans. Here we report the characterization of this signaling pathway in Chaetomium globosum, a widely spread fungus known for synthesizing abundant secondary metabolites, e.g. chaetoglobosin A (ChA). RNAi-mediated knockdown of a putative Gα-encoding gene gna-1, led to plural changes in phenotype, e.g. albino mycelium, significant restriction on perithecium development and decreas

The extremely high Al resistance of Penicillium janthineleum F-13 is not caused by internal or external sequestration of Al.

Penicillium janthinellum F-13 has been isolated in previous work as a fungus tolerating the presence of high concentrations of Al (as high as 100 mM AlCl3). Here its growth rate and yield in three acidic (pH 3.0) media of different composition with varying concentrations of Al are reported. The presence of Al did not affect these parameters. except that the growth yield was somewhat lower in GM (a glucose/peptone/yeast extract-containing medium) with the highest concentration tested (100 mM AlCl3). The amount of Al found in the mycelium was so low that it cannot lead to a significant decrease

Identification of a Coccidioides immitis antigen 2 domain that expresses B-cell-reactive epitopes.

Antigen 2 (Ag2), a major immunoreactive component of Coccidioides immitis mycelium- and spherule-phase cell walls, was recently cloned in our laboratory and was shown to elicit T-cell responses in Coccidioides-immune mice. In this investigation, we evaluated recombinant Ag2 (rAg2) and PCR-generated Ag2 truncations for expression of B-cell-reactive epitopes in enzyme-linked immunosorbent and immunoblot assays with sera from patients with active coccidioidomycosis, a hyperimmune goat anti-Ag2 serum, and a murine anti-Ag2 monoclonal antibody that recognizes a conformational epitope.
