Pilot study of temporary controllable gastric pseudobezoars for dynamic non-invasive gastric volume reduction.

Invasive surgical procedures for gastric volume reduction or bypass have been considered the most effective approach to sustainable long-term weight reduction. However, non-invasive techniques for dynamic volume reduction from inside the stomach are lacking. The aim of this study was to propose temporary, permeable, controllable pseudobezoars for non-invasive, long-term sustainable gastric volume reduction and to test them in pilot human studies. Permeable sac-like carriers made from biocompatible and biodegradable material were filled with expandable superabsorbent fiber and polymer granules.

Satiety responsiveness following dietary exposure to ultra-processed foods differing in texture-derived eating rate: a secondary analysis of a randomized controlled crossover diet intervention.

PURPOSE: Disrupted satiety signalling has been proposed as a possible mechanism driving high energy intakes associated with the consumption of ultra-processed foods (UPFs). We examined whether transitioning from a diet low in UPFs to a high-UPF diet affects satiety responsiveness. METHODS: In a randomized crossover trial, participants (n = 32; mean ± SD: age 27 ± 2 y; BMI 23.6 ± 2.3 kg/m2) with habitual low-UPF intakes completed two 14-day UPF diets (~ 95%EN from UPFs), separated by a 14-day washout period. Acute satiety responsiveness was assessed using a mixed-meal tolerance test with a fix

The ecological validity of the Intuitive Eating Scale-2 among adults with binge-eating disorder.

OBJECTIVE: This study examined the ecological validity of the Intuitive Eating Scale-2 (IES-2) facets among adults with binge-eating disorder (BED) and subthreshold BED (SBED) by assessing how IES-2 subscales predict positive and negative affect, body satisfaction, hunger and satiety sensitivity, eating self-efficacy, and binge-eating symptoms measured via ecological momentary assessment (EMA). METHOD: The sample included 50 adults with BED/SBED who completed the IES-2 and 14 days of EMA data collection. Generalized linear mixed models examined IES-2 subscales (eating for physical rather than

Glial hypothalamic inhibition of GLUT2 expression alters satiety, impacting eating behavior.

Glucose is a key modulator of feeding behavior. By acting in peripheral tissues and in the central nervous system, it directly controls the secretion of hormones and neuropeptides and modulates the activity of the autonomic nervous system. GLUT2 is required for several glucoregulatory responses in the brain, including feeding behavior, and is localized in the hypothalamus and brainstem, which are the main centers that control this behavior. In the hypothalamus, GLUT2 has been detected in glial cells, known as tanycytes, which line the basal walls of the third ventricle (3V). This study aimed t

Is hunger important to model in fMRI visual food-cue reactivity paradigms in adults with obesity and how should this be done?

We considered 1) influence of self-reported hunger in behavioral and fMRI food-cue reactivity (fMRI-FCR) 2) optimal methods to model this. Adults (N = 32; 19-60 years; F = 21; BMI 30-39.9 kg/m2) participated in an fMRI-FCR task that required rating 240 images of food and matched objects for 'appeal'. Hunger, satiety, thirst, fullness and emptiness were measured pre- and post-scan (visual analogue scales). Hunger, satiety, fullness and emptiness were combined to form a latent factor (appetite). Post-vs. pre-scores were compared using paired t-tests. In mixed-effects models, appeal/fMRI-FCR resp

Deactivation of the left dorsolateral prefrontal cortex in Prader-Willi syndrome after meal consumption.

BACKGROUND/OBJECTIVES: Prader-Willi syndrome (PWS) is a type of human genetic obesity that may give us information regarding the physiology of non-syndromic obesity. The objective of this study was to investigate the functional correlates of hunger and satiety in individuals with PWS in comparison with healthy controls with obesity, hypothesizing that we would see significant differences in activation in the left dorsolateral prefrontal cortex (DLPFC) based on prior findings. SUBJECTS/METHODS: This study compared the central effects of food consumption in nine individuals with PWS (7 men, 2 w

Eight-day consumption of inulin added to a yogurt breakfast lowers postprandial appetite ratings but not energy intakes in young healthy females: a randomised controlled trial.

Increasing feelings of satiety may reduce appetite and energy intake. The role of inulin consumption in impacting satiety is unclear. A randomised double-blind controlled crossover trial aimed to determine the effects of inulin+yogurt on satiety after 1 and 8-d consumption. The preload breakfast included 100 g vanilla yogurt with (yogurt-inulin (YI)) and without (yogurt-control (YC)) 6 g inulin. A total of nineteen healthy females (22·8 (sd 2·7) years) with non-restrained eating behaviour and taking hormonal contraceptives participated in the study. Day 1 and 8 visual analogue scale (VAS) rati

NMDA receptor participation in control of food intake by the stomach.

We previously reported that MK-801 (dizocilpine), an antagonist of N-methyl-D-aspartate (NMDA)-type glutamate receptors, increased meal size and duration in rats. MK-801 did not increase sham feeding or attenuate reduction of sham feeding by intraintestinal nutrient infusions. These results suggested that the MK-801-induced increase in meal size did not depend on antagonism of postgastric satiety signals. Consequently, we hypothesized that the NMDA antagonist might increase food intake by directly antagonizing gastric mechanosensory signals or by accelerating gastric emptying, thereby reducing

A pilot study of laparoscopic gastric plication in adolescent patients with severe obesity.

BACKGROUND: Metabolic and bariatric surgery in adolescents with severe obesity has been demonstrated to be safe and have long-term treatment efficacy. However, less than 0.1% of adolescents meeting criteria undergo surgery. METHODS: A prospective pilot study of adolescents who underwent laparoscopic gastric plication (LGP) was conducted to evaluate safety and effects on anthropometrics and weight-related medical and psychological comorbidities. RESULTS: Four adolescents enrolled and underwent LGP; two withdrew 90 days postoperatively and two were followed through 36 months. Preprocedure body

Confirmatory factor analysis of the Baby Eating Behaviour Questionnaire and associations with infant weight, gender and feeding mode in an Australian sample.

The aim of this study was to evaluate the factor structure of the Baby Eating Behaviour Questionnaire (BEBQ) in an Australian community sample of mother-infant dyads. A secondary aim was to explore the relationship between the BEBQ subscales and infant gender, weight and current feeding mode. Confirmatory factor analysis (CFA) utilising structural equation modelling examined the hypothesised four-factor model of the BEBQ. Only mothers (N=467) who completed all items on the BEBQ (infant age: M=17 weeks, SD= weeks) were included in the analysis. The original four-factor model did not provide an

Importance of reward and prefrontal circuitry in hunger and satiety: Prader-Willi syndrome vs simple obesity.

BACKGROUND: The majority of research on obesity (OB) has focused primarily on clinical features (eating behavior, adiposity measures) or peripheral appetite-regulatory peptides (leptin, ghrelin). However, recent functional neuroimaging studies have demonstrated that some reward circuitry regions that are associated with appetite-regulatory hormones are also involved in the development and maintenance of OB. Prader-Willi syndrome (PWS), characterized by hyperphagia and hyperghrelinemia reflecting multi-system dysfunction in inhibitory and satiety mechanisms, serves as an extreme model of geneti

Melanocortin and leptin signaling systems: central regulation of catabolic energy balance.

The recent cloning of the ob gene (leptin) has revolutionized our understanding of obesity and the underlying factors that govern weight homeostasis. There is growing evidence that long term food intake regulation is controlled by the central nervous system by a number of peptide hormones in response to changes in leptin levels. Studies of these hormones, using both genetic and pharmacological approaches, have provided a foundation for decoding the molecular logic of the neuronal circuits which regulate food intake control and energy balance. A review of the current progress in the melanocorti

Glucagon-like peptide 1 promotes satiety and suppresses energy intake in humans.

We examined the effect of intravenously infused glucagon-like peptide 1 (GLP-1) on subjective appetite sensations after an energy-fixed breakfast, and on spontaneous energy intake at an ad libitum lunch. 20 young, healthy, normal-weight men participated in a placebo-controlled, randomized, blinded, crossover study. Infusion (GLP-1, 50 pmol/ kg.h or saline) was started simultaneously with initiation of the test meals. Visual analogue scales were used to assess appetite sensations throughout the experiment and the palatability of the test meals. Blood was sampled throughout the day for analysis

Early Life Added Sugars and Associated Appetite, Satiety, Growth and Adiposity in the First 2 Years of Life.

Introduction: Added sugar (AS) intake has been linked to chronic diseases, yet evidence in children under 2 years remains limited. Aim: Characterise AS intake in children ≤ 2 years with associated appetite, satiety, growth, adiposity, and breastfeeding duration. Methods: We analysed data from 248 mother-child pairs from the MAS-Lactancia birth cohort. Intake of AS and energy was estimated using data from 24 h dietary recalls. AS intake was classified in tertiles as low (0 g), medium (0.01-6.96 g), and high (>6.96 g). Major food group contributors to AS intake were identified. Appetite and sati

Hippocampal atrophy and altered brain responses to pleasant tastes among obese compared with healthy weight children.

OBJECTIVE: The hippocampus is a key structure implicated in food motivation and intake. Research has shown that the hippocampus is vulnerable to the consumption of a western diet (i.e., high saturated fat and simple carbohydrates). Studies of patients with obesity (OB), compared with healthy weight (HW), show changes in hippocampal volume and response to food cues. Moreover, evidence suggests that OB children, relative to HW, have greater hippocampal response to taste. However, no study has examined the association of hippocampal volume with taste functioning in children. We hypothesized that

Dissociation of epinephrine's hyperglycemic and anorectic effect.

The contributions of alpha- and beta-adrenergic receptor mechanisms to epinephrine-induced hyperglycemia, to prandial hepatic glycogenolysis, and to epinephrine-induced inhibition of feeding were investigated in rats, because epinephrine's hyperglycemic and anorectic effect might be related to each other. Intraperitoneal injection of the selective alpha-adrenergic receptor antagonist phentolamine (0.5 mg/kg body weight) but not of the selective beta-adrenergic receptor antagonist propranolol (0.5 mg/kg body weight) reduced the hyperglycemic effect of a subsequent epinephrine injection (0.05 mg

Gastrointestinal factors in hunger and satiety.

The evidence for the participation of signals from the gastrointestinal tract in hunger and satiety is reviewed. Various methods of subjecting only certain portions of the gastrointestinal tract to food are described including sham feeding, crossing of the intestines of two animals, and intragastric, intraduodenal and intravenous feeding. These methods have revealed that animals eat more when only the mouth is exposed to food, but consume less when food enters the stomach directly and still less when feeding themselves intravenously. The role of the stomach in satiety is discussed in relation
