Scorpion-Venom-Derived Antimicrobial Peptide Css54 Exerts Potent Antimicrobial Activity by Disrupting Bacterial Membrane of Zoonotic Bacteria.

Antibiotic resistance is an important issue affecting humans and livestock. Antimicrobial peptides are promising alternatives to antibiotics. In this study, the antimicrobial peptide Css54, isolated from the venom of C. suffuses, was found to exhibit antimicrobial activity against bacteria such as Listeria monocytogenes, Streptococcus suis, Campylobacter jejuni, and Salmonella typhimurium that cause zoonotic diseases. Moreover, the cytotoxicity and hemolytic activity of Css54 was lower than that of melittin isolated from bee venom. Circular dichroism assays showed that Css54 has an α-helix str

Bacteraemias in tropical Australia: changing trends over a 10-year period.

Bacteraemia is an important cause of morbidity and mortality worldwide. This is the largest reported study of bacteraemias in Australia. The presence of organisms endemic to the tropical region and the changing trends described have significant implications for empirical antibiotic therapy. This retrospective study examined 8976 blood cultures from Townsville Hospital, a regional Australian hospital located in the tropics over a 10-year period. The rate of bacteraemic episodes during the study period was 10.12 per 1000 admissions. Intravenous devices (18.7%), immunosuppressive therapy (16.1%),

Isolation of Arcobacter species and other neglected opportunistic agents from aborted bovine and caprine fetuses.

BACKGROUND: Infectious abortion in ruminants is a problem in animal husbandry worldwide. It is important to obtain a diagnosis, to make sure that proper control measures can be instituted, but most abortion cases remain without an etiologic diagnosis. This report describes the presence of Arcobacter species and several neglected opportunistic abortifacient agents in ruminant abortion cases showing or not co-infections among at least one of the major recognized protozoal, fungal, bacterial and viral abortifacient agents. RESULTS: A total of 67 fetuses (55 cattle and 12 goats) and just one plac

The cholesterol-dependent cytolysin promotes Streptococcus systemic spread and induces arachidonic acid accumulation-mediated lethality in a murine intraperitoneal infection model.

Streptococcal peritonitis is associated with a high mortality risk and sometimes accompanied by streptococcal toxic shock-like syndrome (STSLS). Here, we used a murine intraperitoneal model of Streptococcus suis serotype 2 (SS2) infection and applied bacterial lineage tracing to investigate the pathogen's population dynamics during streptococcal intraperitoneal infection. We found that the cholesterol-dependent cytolysin suilysin (SLY) was essential for acute lethality and facilitated bacterial systemic spread by mitigating infection bottlenecks and enabling vascular leakage in the peritoneal

A Single-Copy Sensitive and Field-Deployable One-Pot RT-RPA CRISPR/Cas12a Assay for the Specific Visual Detection of the Nipah Virus.

Nipah virus (NiV) is an emerging bat-borne zoonotic virus that can be transmitted to humans and other animals through infected bats or contaminated foods. The disease is highly lethal in humans (40%-75%) and has the potential for human-to-human transmission. Currently, there are no approved treatments or vaccines for NiV infection in humans or animals. Consequently, there is a pressing need for a highly sensitive, precise, and visually detectable assay to enable early intervention and mitigate the transmission of NiV infection. Here, we report a single-copy sensitive, field-deployable, one-pot

Advances of Caspase-11 for veterinary respiratory infections.

Respiratory diseases represent a major health burden in global livestock production and are driven by bacterial and viral pathogens that trigger severe inflammatory responses and substantial economic losses. Caspase-11 serves as a core component of the non-canonical inflammasome, functioning as an intracellular lipopolysaccharide (LPS) sensor that mediates pyroptosis and inflammatory cytokine maturation. Although extensively studied in human and murine systems, the role of Caspase-11 in veterinary respiratory infections remains poorly characterized. This review synthesizes current knowledge on

Development of Novel Antimicrobial Peptides and Their Engineering into Nanoparticles to Improve Performance.

Antimicrobial peptides (AMPs) are promising alternatives to antibiotics, but discovering potent, low-toxicity candidates and improving their delivery remain challenging. In this study, novel AMPs were identified by constructing and screening a synthetic random peptide library using a bacterial surface display system. Rational design generated derivative peptides, among which WP-4 and WP-6 showed high antimicrobial activity, good biocompatibility, rapid bactericidal effects, and low propensity for resistance development. WP-6 also showed good in vivo therapeutic potency in a murine Escherichia

Biogenic Selenium Nanoparticles from Food-Grade Pediococcus acidilactici JD-21: Selenite Bioreduction, Enhanced Probiotic Traits, and Antioxidant Protection.

Selenite bioreduction by food-grade lactic acid bacteria enables mild production of selenium nanoparticles (SeNPs) together with selenium-enriched biomass. Here, a highly Se(IV)-tolerant isolate from Enshi soil was identified as Pediococcus acidilactici JD-21, which efficiently reduced 5 mmol/L Se(IV) and accumulated SeNPs with an average diameter of 46.4 ± 7.7 nm, potentially associated with protein- and polysaccharide-related biomolecules. Selenium enrichment markedly enhanced the antibacterial activity of JD-21 against Escherichia coli, Staphylococcus aureus and Salmonella enteritidis and i

Association of Common Zoonotic Pathogens With Concentrated Animal Feeding Operations.

Animal farming has intensified significantly in recent decades, with the emergence of concentrated animal feeding operations (CAFOs) in industrialized nations. The congregation of susceptible animals in CAFOs can lead to heavy environmental contamination with pathogens, promoting the emergence of hyper-transmissible, and virulent pathogens. As a result, CAFOs have been associated with emergence of highly pathogenic avian influenza viruses, hepatitis E virus, Escherichia coli O157:H7, Streptococcus suis, livestock-associated methicillin-resistant Staphylococcus aureus, and Cryptosporidium parvu

The site of the lesion causing hearing loss in bacterial meningitis: a study of experimental streptococcal meningitis in guinea-pigs.

The pathological basis of hearing loss in bacterial meningitis was investigated using an animal model of Streptococcus suis meningitis. Forty guinea-pigs were infected after their hearing had been assessed by brain stem auditory evoked potentials. In 17 animals, it was possible to repeat the procedure at the onset of meningitis; this included one animal with subclinical disease. Fifteen animals showed evidence of hearing loss, which on subsequent histological examination was found to be associated invariably with suppurative labyrinthitis. The remaining two animals without hearing loss had nor

Pathogenicity and molecular characterization of emerging porcine reproductive and respiratory syndrome virus in Vietnam in 2007.

In 2007, Vietnam experienced swine disease outbreaks causing clinical signs similar to the 'porcine high fever disease' that occurred in China during 2006. Analysis of diagnostic samples from the disease outbreaks in Vietnam identified porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV-2). Additionally, Escherichia coli and Streptococcus equi subspecies zooepidemicus were cultured from lung and spleen, and Streptococcus suis from one spleen sample. Genetic characterization of the Vietnamese PRRSV isolates revealed that this virus belongs to the North
