Selective frontal neurodegeneration of the inferior fronto-occipital fasciculus in progressive supranuclear palsy (PSP) demonstrated by diffusion tensor tractography.

BACKGROUND: The clinical presentation in progressive supranuclear palsy (PSP), an atypical parkinsonian disorder, includes varying degrees of frontal dysexecutive symptoms. Using diffusion tensor imaging (DTI) and tractography (DTT), we investigated whether diffusion changes and atrophy of the inferior fronto-occipital fasciculus (IFO) occurs in PSP and if these changes correlate with disease stage and clinical phenotype. The corticospinal tract (CST), which is often involved in PSP, was investigated for comparison. METHODS: DTI of the whole brain was performed with a 3 T MR scanner using a s

Insula reactivity and connectivity to anterior cingulate cortex when processing threat in generalized social anxiety disorder.

Aberrant subcortical-prefrontal connectivity may contribute to insula hyper-reactivity to threat in generalized social anxiety disorder (gSAD). A novel PsychoPhysiological Interaction (PPI) analysis was used to examine functional 'coupling' between the insula and prefrontal cortex in gSAD patients and healthy controls (HCs). During fMRI, 29 gSAD and 26 HC volunteers performed an Emotional Face Matching Task, involving the processing of fear, angry, and happy expressions. As expected, compared with HCs, gSAD patients exhibited greater bilateral anterior insula (aINS) reactivity for fear vs. hap

Ins and outs of cerebellar modules.

The modular concept of cerebellar connections has been advocated in the lifetime work of Jan Voogd. In this concept, a cerebellar module is defined as the conglomerate of one or multiple and non-adjacent, parasagittally arranged zones of Purkinje cells, their specific projection to a well-defined region of the cerebellar nuclei, and the climbing fiber input to these zones by a well-defined region of the inferior olivary complex. The modular organization of these olivo-cortico-nuclear connections is further exemplified by matching reciprocal connections between inferior olive and cerebellar nuc

Spatial attention, feature-based attention, and saccades: three sides of one coin?

The last three decades has seen a steady growth of neuroscience research aimed at understanding the functions and sources of top-down attentional modulation in the brain. This correlates with recognition that attention may be a necessary component of sensory systems to support natural behaviors in natural environments. Complexity and clutter are two of the most recognizable hallmarks of natural environments, which can simultaneously contain vitally important and completely irrelevant stimuli. Attention serves as an adaptive filter providing each sensory modality preferential processing routes

Laterality of temporoparietal causal connectivity during the prestimulus period correlates with phonological decoding task performance in dyslexic and typical readers.

We examined how effective connectivity into and out of the left and right temporoparietal areas (TPAs) to/from other key cortical areas affected phonological decoding in 7 dyslexic readers (DRs) and 10 typical readers (TRs) who were young adults. Granger causality was used to compute the effective connectivity of the preparatory network 500 ms prior to presentation of nonwords that required phonological decoding. Neuromagnetic activity was analyzed within the low, medium, and high beta and gamma subbands. A mixed-model analysis determined whether connectivity to or from the left and right TPAs

Abnormal language pathway in children with Angelman syndrome.

Angelman syndrome is a genetic disorder characterized by pervasive developmental disability with failure to develop speech. We examined the basis for severe language delay in patients with Angelman syndrome by diffusion tensor imaging. Magnetic resonance imaging/diffusion tensor imaging was performed in 7 children with genetically confirmed Angelman syndrome (age 70 ± 26 months, 5 boys) and 4 age-matched control children to investigate the microstructural integrity of arcuate fasciculus and other major association tracts. Six of 7 children with Angelman syndrome had unidentifiable left arcuate

Temporal and spatial integration of face, object, and scene features in occipito-temporal cortex.

In three neuroimaging experiments, face, novel object, and building stimuli were compared under conditions of restricted (aperture) viewing and normal (whole) viewing. Aperture viewing restricted the view to a single face/object feature at a time, with the subjects able to move the aperture continuously though time to reveal different features. An analysis of the proportion of time spent viewing different features showed stereotypical exploration patterns for face, object, and building stimuli, and suggested that subjects constrained their viewing to the features most relevant for recognition.

A direct projection from the subthalamic nucleus to the ventral thalamus in monkeys.

The current basal ganglia model considers the internal division of the globus pallidus and the substantia nigra pars reticulata as the sole sources of basal ganglia output to the thalamus. However, following the delivery of retrograde tracers into the ventral anterior/ventral lateral thalamic nuclei, a moderate number of labeled neurons were found within the subthalamic nucleus (STN) in control cases, MPTP-treated monkeys and animals with levodopa-induced dyskinesias. Furthermore, dual tracing experiments showed that subthalamo-thalamic and subthalamo-pallidal projections arise from different

Deep brain stimulation of the lateral cerebellar nucleus produces frequency-specific alterations in motor evoked potentials in the rat in vivo.

The cerebral cortex is tightly and reciprocally linked to the cerebellum and the ascending dentato-thalalmo-cortical pathway influences widespread cortical regions. Using a rodent model of middle cerebral artery stroke, we showed previously that chronic, 20 Hz stimulation of the contralateral lateral cerebellar nucleus (LCN) improved motor recovery, while 50 Hz stimulation did not. Using motor evoked potentials (MEP) elicited by intracortical microstimulation, we now show the effect of LCN stimulation on motor cortex excitability as a function of pulse frequency in propofol-anesthetized rats.

Emotional automaticity is a matter of timing.

There has been a long controversy concerning whether the amygdala's response to emotional stimuli is automatic or dependent on attentional load. Using magnoencephalography and an advanced beamformer source localization technique, we found that amygdala automaticity was a function of time: while early amygdala responding to emotional stimuli (40-140 ms) was unaffected by attentional load, later amygdala response (280-410 ms), subsequent to frontoparietal cortex activity, was modulated by attentional load.

Enlargement of thalamic nuclei in Tourette syndrome.

CONTEXT: The basal ganglia and thalamus together connect in parallel closed-loop circuits with the cortex. Previous imaging studies have shown modifications of the basal ganglia and cortical targets in individuals with Tourette syndrome (TS), but less is known regarding the role of the thalamus in TS pathogenesis. OBJECTIVE: To study the morphological features of the thalamus in children and adults with TS. DESIGN: A cross-sectional, case-control study using anatomical magnetic resonance imaging. SETTING: University research center. PARTICIPANTS: The 283 participants included 149 with TS a

Degeneration of the mid-cingulate cortex in amyotrophic lateral sclerosis detected in vivo with MR spectroscopy.

BACKGROUND AND PURPOSE: Various lines of evidence implicate cerebral involvement beyond the motor cortex in ALS, including the cingulate gyrus and the thalamus. The purpose of this study was to assess neurodegeneration in these regions in vivo by using MRSI. MATERIALS AND METHODS: Fourteen patients with ALS and 14 healthy controls underwent MRSI by using a coronal acquisition scheme. The NAA/Cho ratio was quantified in the MCC, thalamus, and motor cortex (PCG). RESULTS: NAA/Cho was reduced in the MCC in patients with ALS compared with the controls (P = .0004). There was no difference in NAA/

Modulation of cortical motor outputs by the symbolic meaning of visual stimuli.

The observation of an action modulates motor cortical outputs in specific ways, in part through mediation of the mirror neuron system. Sometimes we infer a meaning to an observed action based on integration of the actual percept with memories. Here, we conducted a series of experiments in healthy adults to investigate whether such inferred meanings can also modulate motor cortical outputs in specific ways. We show that brief observation of a neutral stimulus mimicking a hand does not significantly modulate motor cortical excitability (Study 1) although, after prolonged exposure, it can lead to

A comparison of the effects of days 1 and 10 unilateral lesions of medial prefrontal cortex on cerebral morphogenesis and behavior.

The purpose of this study was to compare the behavioral and anatomical effects of unilateral medial prefrontal cortex (mPFC) ablation on newborn (day of birth, P1), neonatal (10 days old, P10), and sham-operated rats. The infant operates were impaired bilaterally at skilled reaching and forelimb inhibition in swimming, and contralaterally at traversing a rung ladder. There was no effect of age-at-injury. They also had mild deficits on spatial navigation tasks. Anatomically, the cortex of the injured hemisphere was generally thinner throughout, with the effect being larger in the rostral half o

Injury of the spino-thalamo-cortical pathway is necessary for central post-stroke pain.

OBJECTIVES: We investigated the relationship between injury of the spino-thalamo-cortical pathway (STP) and central post-stroke pain (CPSP) in patients with intracerebral hemorrhage, using diffusion tensor tractography (DTT). METHODS: 30 consecutive chronic patients, in whom integrity of the STP and the medial lemnisco-thalamo-cortical pathway (MLP) were spared in both hemispheres, were recruited. We classified the patients into two groups according to the presence of the CPSP. DTTs were obtained using the FMRIB Software Library. Fractional anisotropy (FA), mean diffusivity (MD), and tract vo

Intraurethral stimulation for reflex bladder activation depends on stimulation pattern and location.

AIMS: Reflex bladder excitation has been demonstrated by stimulation of the pudendal nerve and several of its distal branches. However, excitation parameters have not been consistent and the relationship to anatomical locations within the urethra has not been fully investigated. An improved understanding of the lower urinary tract neurophysiology will improve human studies and neuroprosthetic device development. METHODS: Intraurethral stimulation was performed in nine cats at near isovolumetric bladder volumes before and/or after spinalization. Bladder excitability profiles were obtained for

Looming responses of telencephalic neurons in the pigeon are modulated by optic flow.

The movement of animals through space filled with various objects requires the interaction between neuronal mechanisms specialized for processing local object motion and those specialized for processing optic flow generated by self-motion of the animal. In the avian brain, visual nuclei in the tectofugal pathway are primarily involved in the detection of object motion. By contrast, the nucleus of the basal optic root (nBOR) and the pretectal nucleus lentiformis mesencephali (nLM) are dedicated to the analysis of optic flow. But little is known about how these two systems interact. Using single

Occipital nerve stimulator lead pathway length changes with volunteer movement: an in vitro study.

BACKGROUND: Occipital nerve stimulation is a modality reserved for refractory headache disorders. Leads (wires) are inserted subcutaneously in the occipital region to stimulate the distal C1-3 nerves; lead migration may result from repeated mechanical forces on the lead associated with patient movement. The primary aim of this study was to determine implantation pathways associated with the least pathway length change secondary to body movement in an in vitro model of an occipital stimulator system. METHODS: After institutional review board approval, 10 volunteers were recruited. The expected

Drug-induced and genetic alterations in stress-responsive systems: Implications for specific addictive diseases.

From the earliest work in our laboratory, we hypothesized, and with studies conducted in both clinical research and animal models, we have shown that drugs of abuse, administered or self-administered, on a chronic basis, profoundly alter stress-responsive systems. Alterations of expression of specific genes involved in stress responsivity, with increases or decreases in mRNA levels, receptor, and neuropeptide levels, and resultant changes in hormone levels, have been documented to occur after chronic intermittent exposure to heroin, morphine, other opiates, cocaine, other stimulants, and alcoh

A dual DTI approach to analyzing white matter in children with dyslexia.

Using voxel-based (VBA) and region-of-interest (ROI) diffusion tensor imaging (DTI) analyses, we examined white matter (WM) organization in seven children with dyslexia and six age-matched controls. Both methods demonstrated reduced fractional anisotropy (FA) in the left superior longitudinal fasciculus (SLF) and abnormal orientation in the right SLF in dyslexics. Application of this complementary dual DTI approach to dyslexia, which included novel analyses of fiber orientation, demonstrates its usefulness for analyzing mild and complex WM abnormalities.
