Transferrin receptors (TfRs) play an essential role in iron-withholding strategy, and are involved in immune response against bacterial infection. In this study, the transferrin receptor 1 (OnTfR1) and transferrin receptor 2 (OnTfR2) genes are identified and characterized in Nile tilapia (Oreochromis niloticus). The open reading frames of OnTfR1 and OnTfR2 are 2220 and 2343 bp of nucleotide sequence, encoding 739 and 780 amino acids, respectively. The deduced proteins of OnTfR1 and OnTfR2 are highly homologous to those of other species, containing three conserved TfR superfamily domains (PA Tf

Growth hormone (GH) and its receptors are critical regulators of somatic growth and metabolism. It has been shown in mammals that the methylation of cytosines within the GH promoter plays a key role in regulating transcripts expression. In the present study, the GH, GHR1 and GHR2 proximal promoters were identified and the methylation levels of these genes in corresponding tissues were assayed. The results suggested that significant arising of GH putative promoter methylation levels in pituitary was observed in females compared with males. However, no such sex-specific changes were found in GHR

Functional feed additives are increasingly incorporated into aquafeeds to improve growth performance, metabolic status, and overall health of farmed fish. However, their regulatory effects on adaptive immunity, particularly T cell-mediated immune responses, remain poorly characterized. In this study, we evaluated the effects of dietary ferulic acid supplementation on T cell immunity and antioxidant capacity in Nile tilapia (Oreochromis niloticus). Fish were fed diets supplemented with 0, 200, or 400 mg/kg ferulic acid and subsequently challenged with Aeromonas hydrophila. Compared with the bas

Complement receptor 3 (CR3), also known as integrin αMβ2, CD11b/CD18, or Mac-1, is a heterodimeric leukocyte-specific integrin composed of the αM (CD11b) and β2 (CD18) subunits. To elucidate the role of CR3 in immunity in Nile tilapia (Oreochromis niloticus), we cloned and characterized the αM (OnCD11b) and β2 (OnCD18) subunits and investigated their functions both in vivo and in vitro. Sequence analysis revealed that OnCD11b contains a 3378-bp open reading frame (ORF) encoding a protein of 1128 amino acids (124.7 kDa), while OnCD18 comprises a 2337-bp ORF encoding 778 amino acids (85.7 kDa).

Transferrin (TF), an iron-binding glycoprotein, plays an important role in host defense against pathogenic infection, which inhibits the growth and proliferation of pathogens, deprives iron from invading pathogens, and activates anti-microbial responses in macrophages. In this study, a TF homologue (OnTF) was identified from Nile tilapia (Oreochromis niloticus) and characterized at expression pattern against bacterial infection and capability binding bacterial pathogens. The open reading frame of OnTF is 2118 bp of nucleotide sequence encoding polypeptides of 705 amino acids. The deduced prote

Interferon-stimulated gene 15 (Isg15), a ubiquitin-like protein induced by type I interferon (IFN), plays a pivotal role in antiviral innate immunity; however, its antibacterial functions in teleosts remain less clear. In this study, we identified and characterized an isg15 gene (Onisg15) from Nile tilapia (Oreochromis niloticus) and explored its regulatory role in antibacterial immune responses. Onisg15 encodes a 156-amino-acid protein containing two conserved ubiquitin-like (UBL) domains and a canonical C-terminal LRLRGG conjugation motif, which are characteristic structural features of Isg1

BACKGROUND: Disease outbreaks caused by Aeromonas veronii pose a significant threat to Nile tilapia (Oreochromis niloticus) aquaculture. Lectin-based adjuvants, such as concanavalin A (ConA), can potentially enhance vaccine efficacy by stimulating both innate and adaptive immunity. This study evaluated the immunoprotective potential of a ConA-adjuvanted, formalin-inactivated A. veronii vaccine administered intraperitoneally.
METHODS: Nile tilapia were vaccinated intraperitoneally with the inactivated A. veronii vaccine formulated with ConA. Safety was assessed by monitoring fish behavior and

Candida albicans is a pathogenic yeast recently associated with diverse diseases in aquaculture. The present study investigated the efficacy of chitosan nanogel (CNG) in ameliorating effects of C. albicans on Nile tilapia (Oreochromis niloticus). Fish were randomly distributed into four groups (control, waterborne CNG at 75 μg/L, intraperitoneally challenged with C. albicans (1.8 × 107 CFU/mL), and waterborne CNG at 75 μg/L + C. albicans at 1.8 × 107 CFU/mL). Results showed that C. albicans infection reduced survival rate (57.5%) and caused marked clinical symptoms in fish among all infected g

Nile tilapia (Oreochromis niloticus) is an important food fish species that is mainly cultivated in tropical and subtropical countries. However, microbial diseases have created various difficulties for this industry. The fundamental prerequisite for tackling disease outbreak prevention and disease resistance is to know how hosts' immune responses against invading microbes are initiated. Toll-like receptors (TLRs) are vital pattern recognition receptors and play pivotal roles in the cellular innate immunity defense that is able to recognize pathogen-associated molecular patterns (PAMPs). The species name Cryptosporidium bollandi n. sp. is proposed for Cryptosporidium piscine genotype 2 based on morphological, biological and molecular characterisation. Phylogenetic analyses of 18S rRNA (18S) sequences revealed that C. bollandi n. sp. was most closely related to piscine genotype 4 (5.1% genetic distance) and exhibited genetic distances of 10.0%, 12.2% and 25.2% from Cryptosporidium molnari, Cryptosporidium huwi and Cryptosporidium scophthtalmi, respectively. At the actin locus, C. bollandi n. sp. was again most closely related to piscine genotype 4 (6.8% genetic distance) and ex

Nile tilapia (Oreochromis niloticus) is a pivotal economic fish that has been plagued by Streptococcus infections. Tumor necrosis factor receptor-associated factor 5 (TRAF5) is a crucial adaptor molecule, which can trigger downstream signaling cascades involved in immune pathway. In this study, Nile tilapia TRAF5 coding sequence (named OnTRAF5) was obtained, which contained typical functional domains, such as RING, zinc finger, coiled-coil and MATH domain. Different from other TRAF molecules, OnTRAF5 had shown relatively low identify with its homolog, and it was clustered into other teleost TR

Acid-base disturbances caused by environmental factors and physiological events including feeding have been well documented in several fish species, but little is known about the impact of dietary electrolyte balance (dEB). In the present study, we investigated the effect of feeding diets differing in dEB (-100, 200, 500 or 800 mEq/kg diet) on the growth, nutrient digestibility and energy balance of Nile tilapia. After 5 weeks on the test diet, the growth of the fish was linearly affected by the dEB levels (P< 0·001), with the lowest growth being observed in the fish fed the 800 dEB diet. The

ZAP70 is essential for initiating the early events of T-cell antigen receptor (TCR) signaling cascade to ensure proper T cell activation and function. However, whether this molecule takes part in the T cell immune response of early vertebrates remains unclear. In the present study, using a teleost model Nile tilapia (Oreochromis niloticus), we investigated the potential involvement of ZAP70 in the T cell activation and adaptive immunity of fish species. Both primary and tertiary structures of O. niloticus ZAP70 (On-ZAP70) are highly conserved with those from other vertebrates. On-ZAP70 protein

Nile tilapia (Oreochromis niloticus) is the most common aquaculture fish, but is exposed to various pollutants and may be susceptible to infectious diseases due to reduction in their antioxidant status and immune defense. Therefore, researchers have tried to find feed supplements of natural origin to increase the health status of fish and decrease the incidence of drug resistance. The current study was conducted to investigate the effect of dietary supplementation of Miswak (Salvadora persica, SP) on Nile tilapia. Fish were randomly allocated into four experimental groups (30 fish each); the c

Despite the potential antimicrobial and water purification benefits of chitosan-based nanocomposites, there are growing concerns regarding the hazards of leached nanoparticles (NPs) to the in-contact circumference. The antibacterial performance of the nanocomposites of chitosan with silver and copper NPs and carbon nanotubes was assessed with an emphasis on their impact on fish health. The minimal inhibitory concentrations of each preparation and the growth curves of Aeromonas hydrophila exposed to different nanocomposites were measured. Five groups of Oreochromis niloticus were exposed to chi

Hydroferrate fluid, MRN-100, an iron-based compound derived from bivalent and trivalent ferrates, is a potent antioxidant compound. Therefore, we examined the protective effect of MRN-100 against γ-radiation-induced lethality and damage to hematopoietic tissues in fish. A total of 216 Nile tilapia fish (Oreochromis niloticus) were randomly divided into four groups. Group 1 served as a control that was administered no radiation and no MRN-100 treatment. Group 2 was exposed only to γ-radiation (15 Gy). Groups 3 and 4 were pre-treated with MRN-100 at doses of either 1 ml/l or 3 ml/l in water for

Aquatic habitats are increasingly endangered by oxyfluorfen (OXY), a diphenyl ether herbicide that induces hepatotoxicity and oxidative stress in aquatic organisms. Banana (Musa acuminata) peel, a valuable byproduct, is known for its antioxidant properties; however, its potential to counteract OXY-induced liver toxicity in Oreochromis niloticus remains unexplored. Therefore, this study investigated the hepatoprotective effects of banana peel powder (BPP) against OXY-induced liver damage and the underlying pathways. A hepatotoxicity model was implemented by subjecting O. niloticus to OXY (0.33

Chemokines play crucial roles in the regulation of immune cell migration and development. The CXCL12/CXCR4 axis has been extensively studied in mammals, but its regulatory mechanism in teleost fish remains unclear. In this study, we used Nile tilapia (Oreochromis niloticus) as a teleost model to investigate the mediation of the CXCL12/CXCR4 axis in IgM+ B cells. Our findings demonstrate that the CXCL12/CXCR4 axis exhibits chemotactic activity on IgM+ B cells and promotes the phagocytosis of IgM+ B cells. Blocking CXCR4 severely impairs the chemotaxis and phagocytosis of IgM+ B cells in vitro a
