Effects of fibrate compounds on expression of plasminogen activator inhibitor-1 by cultured endothelial cells.

The consistent positive correlation between triglyceride and plasminogen activator inhibitor-1 (PAI-1) levels in plasma and the fact that very low density lipoprotein (VLDL) induces secretion of PAI-1 from cultured human umbilical vein endothelial cells (HUVECs) and human hepatoblastoma cells have raised the question of whether fibrate treatment, the main effect of which is a profound lowering of plasma concentrations of VLDL, might improve fibrinolytic function by reducing the plasma levels of PAI-1. However, the findings of controlled clinical trials using various fibrate compounds have been

Heparanase Blockade as a Novel Dual-Targeting Therapy for COVID-19.

The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has caused over 5 million deaths worldwide. Pneumonia and systemic inflammation contribute to its high mortality. Many viruses use heparan sulfate proteoglycans as coreceptors for viral entry, and heparanase (HPSE) is a known regulator of both viral entry and inflammatory cytokines. We evaluated the heparanase inhibitor Roneparstat, a modified heparin with minimum anticoagulant activity, in pathophysiology and therapy for COVID-19. We found that Roneparstat significantly de

Control of the overall lipid profile.

The importance of overall lipid control in cardiovascular prevention is reviewed. Several studies and meta-analyses show that the control of LDL cholesterol (LDL-C) still maintains a high cardiovascular risk, which is related to the presence of triglyceride-rich lipoproteins, and therefore with an increase in plasma triglycerides and the values of apolipoprotein B (apoB) containing these lipoproteins. The importance of this relationship is due to the change in the lipid profile of our population in recent years. This is related to the increase in obesity and insulin resistance, and is called a

In vitro and in vivo performance of monoacyl phospholipid-based self-emulsifying drug delivery systems.

This study investigates the effect of monoacyl phospholipid incorporation on the in vitro and in vivo performance of self-emulsifying drug delivery systems (SEDDS). Monoacyl phosphatidylcholine (Lipoid S LPC 80 (LPC)) was incorporated into four different fenofibrate (FF)-loaded long-chain SEDDS to investigate the impact of LPC on the emulsion droplet size, extent of digestion, colloidal structure evolution and drug precipitation during in vitro lipolysis simulating human conditions and drug bioavailability in a rat model. The four investigated SEDDS containing long-chain glycerides, polyoxyl 3

LC-MS/MS-based metabolomics and PK/PD correlation analysis reveal the mechanism of hyperlipidemia treatment with Gastrodia elata fine powder.

Gastrodia elata Blume is used as both a food and a traditional medicine in East Asia. This study evaluated the lipid-lowering effects of G. elata fine powder (GFP) and the associated metabolic responses in Sprague-Dawley rats with high-fat diet (HFD)-induced hyperlipidemia. Thirty-five rats were assigned to seven groups: a normal-diet control, an untreated HFD model, three GFP groups (150, 300, or 500 mg kg-1 per administration, three times daily), and two positive-control groups receiving simvastatin or fenofibrate. Following a 6 week modeling period and a 2 week intervention, serum biochemic

Microemulsion Eyedrop for Delivery of a PPARα Agonist To Improve Diabetic Keratopathy Corneal Wound Healing.

Diabetic keratopathy (DK) is characterized by delayed corneal wound healing in diabetes, and recent studies have linked this defect to impaired mitochondrial respiration and reduced peroxisome proliferator-activated receptor-α (PPARα) signaling in the corneal epithelium. The PPARα agonist fenofibrate (Feno) has shown therapeutic potential in diabetic corneal disease, but prior topical and oral approaches still face formulation and translational limitations. In this study, we developed a Feno-loaded microemulsion (Feno microemulsion) eyedrop to improve drug solubility, precorneal residence, and

Bezafibrate, a Pan PPAR agonist, attenuates experimental rheumatoid arthritis via PPAR dependent modulation of inflammatory pathways with emphasis on PPAR γ activity.

Peroxisome proliferator-activated receptor gamma (PPAR-γ) plays a key role in immunoregulatory functions, making it a promising target for distinct inflammatory diseases, including rheumatoid arthritis. In the present study, we aimed to identify and evaluate a clinically viable PPAR-γ agonist with anti-arthritic potential using a structure-based drug repurposing approach. Initially, molecular docking was performed using a panel of fibrate-class compounds (including pemafibrate, bezafibrate, fenofibrate, gemfibrozil, and clofibrate) against the ligand-binding domain of PPAR-γ (PDB ID: 7WGO). The serum acylcarnitines profile in epileptic children treated with valproic acid and the protective roles of peroxisome proliferator-activated receptor a activation in valproic acid-induced liver injury.

Valproic acid (VPA) is widely used as a major drug in the treatment of epilepsy. Despite the undisputed pharmacological importance and effectiveness of VPA, its potential hepatotoxicity is still a major concern. Being a simple fatty acid, the hepatotoxicity induced by VPA has long been considered to be due primarily to its interference with fatty acid β-oxidation (β-FAO). The aim of this study was to investigate the biomarkers for VPA-induced abnormal liver function in epileptic children and to determine potential mechanisms of its liver injury. Targeted metabolomics analysis of acylcarnitines

Targeted disruption of the Kcnj5 gene in the female mouse lowers aldosterone levels.

Aldosterone is released from adrenal zona glomerulosa (ZG) cells and plays an important role in Na and K homoeostasis. Mutations in the human inwardly rectifying K channel CNJ type (KCNJ) 5 (KCNJ5) gene encoding the G-coupled inwardly rectifying K channel 4 (GIRK4) cause abnormal aldosterone secretion and hypertension. To better understand the role of wild-type (WT) GIRK4 in regulating aldosterone release, we have looked at aldosterone secretion in a Kcnj5 knockout (KO) mouse. We found that female but not male KO mice have reduced aldosterone levels compared with WT female controls, but higher

Vision Loss in Older Adults.

Vision loss affects 37 million Americans older than 50 years and one in four who are older than 80 years. The U.S. Preventive Services Task Force concludes that current evidence is insufficient to assess the balance of benefits and harms of screening for impaired visual acuity in adults older than 65 years. However, family physicians play a critical role in identifying persons who are at risk of vision loss, counseling patients, and referring patients for disease-specific treatment. The conditions that cause most cases of vision loss in older patients are age-related macular degeneration, glau

Stage-dependent signaling networks and therapeutic strategies in diabetic retinopathy: from neurovascular dysfunction to angiogenesis and fibrosis.

Diabetic retinopathy (DR) is a consequence of multiple metabolic, neuronal, inflammatory, vascular and fibrotic changes, not a single cause. Early hyperglycaemia, oxidative stress, lipid disturbance, and mitochondrial dysfunction weaken the retinal neurovascular unit before clinically apparent vascular changes develop. With the accumulation of damage, nuclear factor kappa B, high-mobility group box 1, Toll-like receptor 4, receptor for advanced glycation end products, cyclic GMP-AMP synthase-stimulator of interferon genes, and the NOD-like receptor protein 3 inflammasome enhance inflammation,
