Copromicroscopic study of gastrointestinal parasites in captive mammals at Central Zoo, Lalitpur, Nepal.

BACKGROUND: Zoological gardens (Zoo) provide abode to various threatened animals or trafficked animals seized by the authorities, and injured and orphaned animals. Captive animals are more susceptible to infection as they are under significant stress due to diet and space which further dwindle their resistance to parasitic infections. OBJECTIVES: This study was conducted to determine the prevalence and burden of gastrointestinal parasites in captive mammals housed at Central Zoo. METHODS: Fresh faecal samples from three orders of mammals including carnivora (n = 24), rodentia (n = 28) and ar

The effect of plant bioactive compounds on lamb performance, intake, gastrointestinal parasite burdens, and lipid peroxidation in muscle.

Plant extracts have been widely used in animal production systems due to the benefits promoted by their bioactive components, mainly through their antioxidant effects and positive effects on animal growth, health, and meat quality. We determined whether bioactive compounds (BC) from grape (Vitis vinifera), olive (Olea europaea), and pomegranate (Punica granatum) have beneficial effects on lamb growth while controlling gastrointestinal parasitism and reducing skeletal lipid peroxidation in muscle. Fourteen lambs (4 to 5 mo of age) were housed in individual pens and assigned to two treatment gro

Tuft Cells Increase Following Ovine Intestinal Parasite Infections and Define Evolutionarily Conserved and Divergent Responses.

Helminth parasite infections of humans and livestock are a global health and economic problem. Resistance of helminths to current drug treatment is an increasing problem and alternative control approaches, including vaccines, are needed. Effective vaccine design requires knowledge of host immune mechanisms and how these are stimulated. Mouse models of helminth infection indicate that tuft cells, an unusual type of epithelial cell, may 'sense' infection in the small intestine and trigger a type 2 immune response. Currently nothing is known of tuft cells in immunity in other host species and in

Influence of the physiological stage of Blackbelly sheep on immunological behaviour against gastrointestinal nematodes.

The influence of the physiological stage of a Blackbelly flock against natural infection by gastrointestinal parasitic nematodes (GIN) was investigated. Data on serum and saliva IgA levels and peripheral cellular immune response were recorded in 51 sheep for 6 months. The flock was divided based on their physiological stage as follows: a) pregnant ewes, b) lactating ewes, c) non-pregnant ewes, d) pre-weaning lambs, e) growing lambs and f) replacement sheep. Additionally, sheep were classified based on the parasite infection as stabled or grazing management system as follows: g) non-infected st

The use of nemabiome metabarcoding to explore gastro-intestinal nematode species diversity and anthelmintic treatment effectiveness in beef calves.

Next-generation deep amplicon sequencing, or metabarcoding, has revolutionized the study of microbial communities in humans, animals and the environment. However, such approaches have yet to be applied to parasitic helminth communities. We recently described the first example of such a method - nemabiome sequencing - based on deep-amplicon sequencing of internal transcribed spacer 2 (ITS-2) rDNA, and validated its ability to quantitatively assess the species composition of cattle gastro-intestinal nematode (GIN) communities. Here, we present the first application of this approach to explore GI

Anthelmintic resistance in gastrointestinal nematodes of beef cattle in the state of Rio Grande do Sul, Brazil.

Gastrointestinal nematodes resistant to anthelmintics have been reported in several regions of Brazil, and they may be associated with economic losses for the cattle industry. This study aimed to evaluate the resistance status of gastrointestinal nematodes from naturally infected beef cattle to several commercially available anthelmintics, as well as to test the efficacy of combinations of anthelmintics against multi-resistant gastrointestinal nematodes. Ten farms located in Rio Grande do Sul state were selected by: farmers' consent; extensive raising system; availability of calves aged from 7

Discovery of anthelmintic drug targets and drugs using chokepoints in nematode metabolic pathways.

Parasitic roundworm infections plague more than 2 billion people (1/3 of humanity) and cause drastic losses in crops and livestock. New anthelmintic drugs are urgently needed as new drug resistance and environmental concerns arise. A "chokepoint reaction" is defined as a reaction that either consumes a unique substrate or produces a unique product. A chokepoint analysis provides a systematic method of identifying novel potential drug targets. Chokepoint enzymes were identified in the genomes of 10 nematode species, and the intersection and union of all chokepoint enzymes were found. By studyin

Multiscale epidemiology of gastrointestinal nematode infections in small ruminants: field evidence from Inner Mongolia and a national meta-analysis in China.

BACKGROUND: Gastrointestinal nematodes (GINs) are among the most important parasites constraining small-ruminant production. However, epidemiological evidence in China remains fragmented because existing studies differ substantially in geographic coverage, host species, sampling periods, diagnostic approaches, and ecological settings. METHODS: We applied a multiscale framework integrating a regional survey in Inner Mongolia with a national systematic review and meta-analysis. In the regional survey, 2,084 fecal samples were collected from sheep aged 6-12 months in Ulanqab, Alxa, and Ordos fro

Assessment of Anthelmintic Resistance in Major Gastrointestinal Nematodes in Sheep and Goat Farms in Mymensingh, Bangladesh.

Anthelmintic resistance (AR) is a well concern in livestock production. Hence, the study was designed to assess the status of AR in sheep and goat farms at Mymensingh sadar using fecal egg count reduction test (FECRT) and egg hatch assay (EHA). To do this, 10 farms were selected. For FECRT, 40 animals of each farm having more than 200 EPG of feces were selected randomly by employing the McMaster technique and treated with albendazole (ABZ), levamisole (LEV), and ivermectin (IVM). Fecal samples from animals of each treated and control group were collected directly from the rectum on Day 0 (pret

Hookworm genes encoding intestinal excreted-secreted proteins are transcriptionally upregulated in response to the host's immune system.

Hookworms are intestinal parasitic nematodes that chronically infect ~500 million people. How hookworms successfully overcome host protective mechanisms is unclear, but it may involve hookworm proteins that digest host tissues, or counteract the host's immune system, or both. To find such proteins in the zoonotic hookworm Ancylostoma ceylanicum, we used mass spectrometry to identify 565 genes encoding excreted-secreted (ES) proteins from adults, and used RNA-seq to identify genes expressed both in young adults (12 days post-infection) and in intestinal and non-intestinal tissues dissected from

Declining effectiveness of major anthelmintic groups against gastrointestinal nematodes of farmed ruminants in Africa based on faecal egg count reduction: systematic review and first meta-analysis.

OBJECTIVES: Anthelmintic resistance (AR) in gastrointestinal nematode (GIN) parasites of ruminant livestock has been widely reported in Africa but analysis of variation between regions and over time is lacking. The objective of this paper was to collate data on AR and, through meta-analyses, identify patterns in its occurrence in order to guide future research, practice and policy. RESEARCH DESIGN AND METHODS: A systematic literature review following PRISMA guidelines retrieved publications on AR, from which comparable data on treatment effectiveness using the faecal egg count reduction (FECR

Ethnobotanical knowledge on native Brazilian medicinal plants traditionally used as anthelmintic agents - A review.

Intestinal helminthiasis is a neglected disease that affects a significant portion of the global population, specifically in developing countries, where medicinal plants are widely used for therapeutic purposes. With the purpose to identify the native species used in traditional Brazilian medicine for the management of helminthiasis, ethnopharmacological books edited in Brazil documenting the folk use of medicinal plants were analyzed. The native species cited in at least three studies were selected, and bibliographic research was performed using electronic databases to identify their scientif

The transcription factor NHR-8: A new target to increase ivermectin efficacy in nematodes.

Resistance to the anthelmintic macrocyclic lactone ivermectin (IVM) has a great impact on the control of parasitic nematodes. The mechanisms by which nematodes adapt to IVM remain to be deciphered. We have identified NHR-8, a nuclear hormone receptor involved in the xenobiotic response in Caenorhabditis elegans, as a new regulator of tolerance to IVM. Loss-of-function nhr-8(ok186) C. elegans mutants subjected to larval development assays and electropharyngeogram measurements, displayed hypersensitivity to IVM, and silencing of nhr-8 in IVM-resistant worms increased IVM efficacy. In addition, c

Heterologous Expression in Remodeled C. elegans: A Platform for Monoaminergic Agonist Identification and Anthelmintic Screening.

Monoamines, such as 5-HT and tyramine (TA), paralyze both free-living and parasitic nematodes when applied exogenously and serotonergic agonists have been used to clear Haemonchus contortus infections in vivo. Since nematode cell lines are not available and animal screening options are limited, we have developed a screening platform to identify monoamine receptor agonists. Key receptors were expressed heterologously in chimeric, genetically-engineered Caenorhabditis elegans, at sites likely to yield robust phenotypes upon agonist stimulation. This approach potentially preserves the unique phar

Field efficacy of four anthelmintics and confirmation of drug-resistant nematodes by controlled efficacy test and pyrosequencing on a sheep and goat farm in Denmark.

We describe a case of anthelmintic resistance on one of the largest organic small ruminant farms in Denmark. The flock was established in 2007 by purchase of animals from other Danish farms and had history of clinical parasitism, high mortality of young stock and anthelmintic treatment failure. In October 2011, 40 lambs and 40 kids were selected for a faecal egg count reduction test (FECRT) with fenbendazole (FBZ), ivermectin (IVM), moxidectin (MOX) and levamisole (LEV). Lambs were treated with the recommended sheep dose of each product while kids received the sheep dose of IVM, 1.5× sheep dos

Assessing anthelmintic resistance risk in the post-genomic era: a proof-of-concept study assessing the potential for widespread benzimidazole-resistant gastrointestinal nematodes in North American cattle and bison.

As genomic research continues to improve our understanding of the genetics of anthelmintic drug resistance, the revolution in DNA sequencing technologies will provide increasing opportunities for large-scale surveillance for the emergence of drug resistance. In most countries, parasite control in cattle and bison has mainly depended on pour-on macrocyclic lactone formulations resulting in widespread ivermectin resistance. Consequently, there is an increased interest in using benzimidazole drugs which have been used comparatively little in cattle and bison in recent years. This situation, toget

Synthesis, characterization and antiparasitic activity of organometallic derivatives of the anthelmintic drug albendazole.

Helminthiases, a group of neglected tropical diseases, affect more than one billion people mainly in tropical and subtropical regions. Moreover, major intestinal protozoa have a significant impact on global public health. Albendazole (ABZ) is a broad-spectrum anthelmintic recommended by the World Health Organisation (WHO). However, drug resistance is emerging due to its widespread use. In order to tackle this problem, taking into account the spectacular results obtained with ferroquine, an organometallic derivatization of the antimalarial drug chloroquine, we have prepared, in this study, a se

Sensitivity and specificity of the FAMACHA(©) system in Suffolk sheep and crossbred Boer goats.

Sheep and goats are the species of farm animal with the highest growth rate in Paraná State. The main problems facing Paraná State flocks are gastrointestinal parasites and anthelmintic resistance. One of the newest resources used to slow down the development of anthelmintic resistance is the FAMACHA(©) system, a selective method useful for controlling gastrointestinal verminosis in small ruminants. The purpose of the present research was to evaluate the sensitivity and specificity of the FAMACHA(©) system in sheep and goats and to compare the results for both species. The conjunctivae of 83 S

Relative abilities of young sheep and goats to self-medicate with tannin-rich sainfoin when infected with gastrointestinal nematodes.

Plant secondary metabolites (PSM) are one of the promising options to control gastrointestinal nematodes in sheep and goats. The objective of this study was to assess the abilities of sheep and goats to self-medicate with tannin-rich sainfoin (Onobrychis viciifolia) (SF) when infected with gastrointestinal nematodes, using a cafeteria and an operant conditioning trial. Hypotheses were that parasitized (P) lambs and goat kids would show greater intake and preference for SF than their non-parasitized (NP) counterparts, that kids would eat more SF than lambs (due to their lower resistance against
