Ureolytic Biomineralization Reduces Proteus mirabilis Biofilm Susceptibility to Ciprofloxacin.

Ureolytic biomineralization induced by urease-producing bacteria, particularly Proteus mirabilis, is responsible for the formation of urinary tract calculi and the encrustation of indwelling urinary catheters. Such microbial biofilms are challenging to eradicate and contribute to the persistence of catheter-associated urinary tract infections, but the mechanisms responsible for this recalcitrance remain obscure. In this study, we characterized the susceptibility of wild-type (ure+) and urease-negative (ure-) P. mirabilis biofilms to killing by ciprofloxacin. Ure+ biofilms produced fine biomine

Ecabet sodium eradicates Helicobacter pylori infection in gastric ulcer patients.

Ecabet sodium (ecabet), a new agent that has protective effects on the gastric mucosa has anti-Helicobacter pylori effects, binding with urease to inhibit H. pylori activity, and causing the bacterial to become non-viable. Ecabet monotherapy eradicates H. pylori infection in Japanese monkeys. We investigated a new regimen that included ecabet to eradicate H. pylori infection in gastric ulcer patients. Fifty-five H. pylori-positive patients with gastric ulcer were randomly assigned to one of two groups: group 1 received dual therapy with lansoprazole (30 mg o.d.) for 8 weeks plus clarithromycin

[Helicobacter pylori and molecular biology--virulence, diagnosis and vaccine development].

The discovery of H. pylori as the etiologic agent of chronic antral type B gastritis and the finding that H.pylori is involved in the pathogenesis of gastroduodenal ulcer disease and gastric carcinoma have triggered intensive research about this organism. After overcoming considerable initial difficulties, researchers succeeded in adapting the instruments of molecular biology to the study of this highly fastidious organism. This has led to a rapid increase of knowledge concerning the basis of H.pylori virulence (as well as that of the related animal pathogens H. mustelae and H. felis) and to t

Targeting bacterial nickel transport with aspergillomarasmine A suppresses virulence-associated Ni-dependent enzymes.

Microbial Ni2+ homeostasis underpins the virulence of several clinical pathogens. Ni2+ is an essential cofactor in urease and [NiFe]-hydrogenases involved in colonization and persistence. Many microbes produce metallophores to sequester metals necessary for their metabolism and starve competing neighboring organisms. The fungal metallophore aspergillomarasmine A (AMA) shows narrow specificity for Zn2+, Ni2+, and Co2+. Here, we show that this specificity allows AMA to block the uptake of Ni2+ and attenuate bacterial Ni-dependent enzymes, offering a potential strategy for reducing virulence. Bac

Evaluation of Novel Isatin-Schiff Base Derivatives: Monoamine Oxidase Inhibition and Antimicrobial Assessment.

Five novel series of isatin-Schiff base derivatives (1a-5e) were synthesized, and their neuroprotective and anti-infective potential was evaluated. Their half-maximal inhibitory concentration (IC50) was determined against both monoamine oxidase A (MAO-A) and MAO-B targets for the treatment of depression and Parkinson's disease, respectively. The anti-infective potential of the synthesized derivatives was evaluated by measuring their IC50 values against urease, an enzyme utilized by specific pathogenic bacteria as a virulence factor. The minimum inhibitory concentration for each compound was as

Hyperammonemia syndrome due to Pluralibacter gergoviae bacteremia in an immunocompromised patient: case report.

BACKGROUND: Pluralibacter gergoviae is a rare, opportunistic, urease-producing gram-negative organism infrequently implicated in invasive infections. Reports in organ transplant or extracorporeal membrane oxygenation (ECMO) settings are exceedingly uncommon. This case is distinctive for describing P. gergoviae bacteremia potentially associated with severe hyperammonemia and encephalopathy in a lung transplant recipient on prolonged ECMO support, expanding the clinical spectrum of infection-related hyperammonemia beyond the traditionally recognized Ureaplasma and Mycoplasma species. Multi-Targeting Carnosic Acid Kills Drug-Resistant Helicobacter pylori With Narrow-Spectrum Activity.

Helicobacter pylori (H. pylori) is a significant global human pathogen intricately linked to gastritis, peptic ulcers, and gastric cancer. The escalating challenge of antimicrobial resistance and the adverse effects of conventional antibiotics on the gut microbiome necessitate the development of novel, targeted therapeutics. In this study, we demonstrate that carnosic acid (CA), a natural compound derived from traditional Chinese medicine, exhibits potent and specific anti-H. pylori activity in vitro, with no detectable resistance observed after prolonged serial passaging. CA also displayed en

Blue light emitting diodes enhance the antivirulence effects of Curcumin against Helicobacter pylori.

Introduction. Growing concern about the increasing frequency of resistance of Helicobacter pylori to the available antimicrobial agents worldwide has encouraged the search for new strategies in treating and eradicating H. pylori infections. Endoscopic blue-light therapy has been used in patients with H. pylori gastritis with limited success due to subsequent repopulation with H. pylori. Clinical trials using Curcumin could not eradicate infection either.Aim. We studied the effect of blue light emitting diodes (LEDs) in conjunction with Curcumin on H. pylori, since this has not been previously
