Spaced food but not electrical brain stimulation induces polydipsia and air licking.

An attempt was made to induce polydipsia in rats whose lever pressing was reinforced with food pellets or electrical brain stimulation. Nine food-deprived, water-sated rats drank water excessively during sessions in which food pellets were delivered. When brain stimulation was substituted for food, drinking immediately ceased. Delivering brain stimulation according to a variety of schedules, pairing brain stimulation with food reinforcement, and substituting an air stream for water, each failed to produce polydipsic licking. These results show that polydipsia is not induced by all reinforcers.

Nanopore-enabled time-resolved monitoring of catecholamine-related phenylalanine metabolism.

Catecholamines, including dopamine, noradrenaline (also known as norepinephrine) and adrenaline (also known as epinephrine), are essential regulators of neural and endocrine function. Although they are synthesized through a branched phenylalanine metabolic pathway, direct and dynamic monitoring of the enzymatic processes governing their production has remained challenging. Here we reconstitute the complete catecholamine biosynthetic pathway from phenylalanine to adrenaline in vitro using purified enzymes and cofactors, and couple this system with nanopore-based single-molecule sensing. Protecting the heart from ischemia/reperfusion injury: an update on remote ischemic preconditioning and postconditioning.

PURPOSE OF REVIEW: The most effective strategy for reducing acute myocardial ischemic injury is timely and effective reperfusion. However, myocardial reperfusion can induce further cardiomyocyte death (reperfusion injury). Interventions that protect the heart from ischemia/reperfusion injury, reducing infarct size, can involve remote ischemic preconditioning and postconditioning. These interventions have a promising potential clinical application, and have been the focus of recent research. In this review, we provide an update of remote ischemic preconditioning and postconditioning mechanisms.

A Printed Hydrogel Hybrid Electronic System With Thermoresponsive Adhesion for Neurophysiological Monitoring.

Neurological disorders severely impair the daily life of patients, necessitating precise assessments of neural conduction pathway damage to guide effective diagnosis and treatment. However, current clinical neurodiagnostic equipment is expensive, bulky, and restricted to hospital settings. Furthermore, the rigid nature and low adhesion of conventional electrodes lead to the acquisition of low-quality neurophysiological signals. Here, we develop a soft hybrid electronic system assembled with printed responsive hydrogel electrodes. By synthesizing thermoresponsive hydrogel microsphere inks, we a

tubb5 knockout in zebrafish causes neurodevelopmental defects via notch pathways.

Mutations in TUBB5 cause complex cortical dysplasia with other brain malformations. However, the pathogenic mechanisms underlying TUBB5 mutations remain incompletely understood. We generated tubb5 knockout zebrafish using CRISPR/Cas9 genome editing technology. Homozygous mutants were lethality within two weeks post-fertilization along with developmental delay, craniofacial malformations, uncoordinated movement and increased seizure susceptibility. tubb5 knockout larvae also showed reduced locomotor activity and impair photomotor responses. Furthermore, tubb5 knockout larvae exhibited significa

Static and dynamic perspective on brain activity and functional connectivity alternations in the comorbidity of chronic pain and depression: An fMRI study in rats.

BACKGROUND: Clinically, chronic pain and depression often coexist in multiple diseases and reciprocally reinforce each other, which greatly escalates the difficulty of treatment. The alteration in neural networks underlying the chronic pain/depression comorbidity remains unclear. METHODS: Spared nerve injury (SNI) was performed to establish a rat model of comorbidity of chronic pain and depression. Multimodal fMRI was utilized to acquire neuroimaging data of the rat brain, with BOLD and DTI data used to analyze adaptive changes in brain regions and networks associated with comorbidity. RESUL

Biofluid-Permeable and Erosion-Resistant Wireless Neural-Electronic Interfaces for Neurohomeostasis Modulation.

Neural-electronic interfaces through delivering electroceuticals to lesions and modulating pathological endogenous electrical environments offer exciting opportunities to treat drug-refractory neurological disorders. Such an interface should ideally be compatible with the neural tissue and aggressive biofluid environment. Unfortunately, no interface specifically designed for the biofluid environments is available so far; instead, simply stacking an encapsulation layer on silicon-based substrates makes them susceptible to biofluid leakage, device malfunction, and foreign-body reactions. Here, w

Extracellular vesicles: A new therapeutic drug for nerve injury repair.

Extracellular vesicles are membranous structures actively released by cells. They efficiently mediate intercellular communication and play a role in reshaping the microenvironment. Their diverse bioactive cargo, including miRNAs, proteins, and lipids, along with their ability to cross the blood-brain barrier, holds broad therapeutic promise in the field of neuroregeneration. Current research focuses on exploring the mechanisms of action and clinical translational potential of extracellular vesicles from different sources, including exosomes derived from mesenchymal stem cells, neural stem cell

Increased listening effort and cochlear neural degeneration underlie speech-in-noise deficits in normal-hearing middle-aged adults.

Middle age represents a critical period of accelerated brain changes and provides a window for early detection and intervention in age-related neurological decline. Hearing loss is a key early marker of such decline and is linked to numerous comorbidities in older adults. Yet, ~10% of middle-aged individuals who report hearing difficulties show normal audiograms. Cochlear neural degeneration (CND) could contribute to these hidden hearing deficits, though its role remains unclear due to a lack of objective diagnostics and uncertainty regarding its perceptual outcomes. Here, we employed a cross-

Aberrant auditory prediction patterns robustly characterize tinnitus.

Phantom perceptions like tinnitus occur without any identifiable environmental or bodily source. The mechanisms and key drivers behind tinnitus are poorly understood. The dominant framework, suggesting that tinnitus results from neural hyperactivity in the auditory pathway following hearing damage, has been difficult to investigate in humans and has reached explanatory limits. As a result, researchers have tried to explain perceptual and potential neural aberrations in tinnitus within a more parsimonious predictive-coding framework. In two independent magnetoencephalography studies, participan

LASP to the Future of Atomic Simulation: Intelligence and Automation.

Atomic simulations aim to understand and predict complex physical phenomena, the success of which relies largely on the accuracy of the potential energy surface description and the efficiency to capture important rare events. LASP software (large-scale atomic simulation with a Neural Network Potential), released in 2018, incorporates the key ingredients to fulfill the ultimate goal of atomic simulations by combining advanced neural network potentials with efficient global optimization methods. This review introduces the recent development of the software along two main streams, namely, higher

Prenatal drug exposure affects neonatal brain functional connectivity.

Prenatal drug exposure, particularly prenatal cocaine exposure (PCE), incurs great public and scientific interest because of its associated neurodevelopmental consequences. However, the neural underpinnings of PCE remain essentially uncharted, and existing studies in school-aged children and adolescents are confounded greatly by postnatal environmental factors. In this study, leveraging a large neonate sample (N = 152) and non-invasive resting-state functional magnetic resonance imaging, we compared human infants with PCE comorbid with other drugs (such as nicotine, alcohol, marijuana, and ant

Oxygen-glucose-deprived rat primary neural cells exhibit DJ-1 translocation into healthy mitochondria: a potent stroke therapeutic target.

AIMS: DJ-1 is a key redox-reactive neuroprotective protein implicated in regulation of oxidative stress after stroke. However, the molecular mechanism, especially the role of mitochondrial function, by which DJ-1 protects neural cells in stroke remains to be elucidated. The aim of this study was to reveal whether DJ-1 translocates into the mitochondria in exerting neuroprotection against oxidative stress. In particular, we examined DJ-1 secretion from primary rat neural cells (PRNCs) exposed to experimental stroke. METHODS: Primary rat neural cells were exposed to the oxygen-glucose deprivati

A zebrafish model of axenfeld-rieger syndrome reveals that pitx2 regulation by retinoic acid is essential for ocular and craniofacial development.

PURPOSE: The homeobox transcription factor PITX2 is a known regulator of mammalian ocular development, and human PITX2 mutations are associated with Axenfeld-Rieger syndrome (ARS). However, the treatment of patients with ARS remains mostly supportive and palliative. METHODS: The authors used molecular genetic, pharmacologic, and embryologic techniques to study the biology of ARS in a zebrafish model that uses transgenes to mark neural crest and muscle cells in the head. RESULTS: The authors demonstrated in vivo that pitx2 is a key downstream target of retinoic acid (RA) in craniofacial devel
