Quantification of PEG2000-DMG and TS-202, an ionic lipid, in rabbit plasma by LC-MS/MS: Application to complement C5 siRNA lipid nanoparticles.

A simple and robust assay was developed for the quantification of PEG2000-DMG and TS-202, key components of the lipid nanoparticle (LNP) formulation of E8001-LNP, which encapsulates small-interfering RNA targeting complement C5, in rabbit plasma. The assay utilizes high-performance liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). To prevent degradation of PEG2000-DMG, plasma samples were treated with a serine protease inhibitor prior to protein precipitation extraction. The analytes were separated on a reversed-phase column under gradient elution conditions. Detection wa

Confronted with the real world: how final-year medical students experience interprofessional collaboration in clinical practice.

BACKGROUND: Interprofessional education (IPE) is increasingly embedded in undergraduate medical curricula, yet little is known about how students encounter interprofessional collaboration (IPC) in real-world clinical practice. METHODS: This longitudinal qualitative study analyzed written diaries from eight final-year medical students at one institution (blinded) to explore IPC experiences during clerkships without formal IPE. Deductive coding used an interprofessional learning framework with four domains: "Roles, Hierarchy, and Power Dynamics," "Team Inclusion, Collaboration, and Silos," "Com

Energy landscapes and tunable electronic properties of polar and non-polar SiC nanofilms.

The energy landscapes and electronic properties of non-polar and polar nanofilms of silicon carbide are studied using periodic hybrid density functional calculations. Relative energies and electronic properties are reported as a function of film thickness for a set of structures derived from those of wurtzite, zinc blende and graphite. The energy landscape of the films is complex with pronounced nano-polymorphism. Across polar films different mechanisms in different cases remove or reduce the dipole moment, including charge transfer and phase transitions to the graphitic-like structures which

Mitophagy in Hepatic Ischemia-Reperfusion Injury: From Mitochondrial Dysfunction to Therapeutic Targeting.

Hepatic ischemia-reperfusion injury (HIRI) is a major cause of postoperative liver dysfunction and adverse outcomes in hepatectomy, liver transplantation, and hemorrhagic shock. Among the multiple mechanisms implicated in HIRI, mitochondria are recognized as central organelles that integrate metabolic failure, oxidative stress, inflammation, and cell death. During ischemia, interruption of oxygen and nutrient supply impairs oxidative phosphorylation, depletes ATP, disrupts ionic homeostasis, and renders mitochondria highly vulnerable to subsequent injury. Upon reperfusion, reoxygenation trigge

Synthetic Control of Structure and Magnetism in Low-Dimensional Organic Copper(II) Chloride Hybrids.

Low-dimensional (LD) organic metal halide hybrids (OMHHs) have attracted widespread attention owing to their exceptional structural versatility and emergent photophysical and spin-dependent properties. While hundreds of LD OMHHs have been reported to date, systematic investigations into how organic cations influence structural dimensionality and magnetic behavior remain limited. Here, we report a steric-engineering strategy to control structural dimensionality and magnetism in LD OMHHs. By systematically tuning methyl substitution in 1,3-propanediamine (PDA), we achieve a structural progressio

Oropharyngeal Dysphagia as a Metabolic Emergency: A Comprehensive Review on Nutritional Barriers, Sarcopenia, and Management Strategies.

Oropharyngeal dysphagia (OD) is traditionally managed as a mechanical swallowing impairment. This narrative review proposes a conceptual model that reframes chronic, severe OD as a high-risk clinical condition driving systemic malnutrition and progressive nutritional deterioration. We examine the epidemiological burden of OD-associated malnutrition across geriatric, neurological, and oncological populations, exploring how diagnostic heterogeneity influences reported prevalence ranges. The pathophysiological narrative synthesizes hypotheses regarding the potential disruption of the cephalic pha

Biologic immunomodulators and spinal fusion: evidence, mechanisms, and perioperative considerations.

Purpose Biologic immunomodulators are increasingly used across autoimmune, inflammatory, oncologic, and metabolic diseases, resulting in a growing population of patients undergoing spinal fusion while receiving these agents. Because immune pathways targeted by biologic therapies play essential, time-dependent roles in bone graft incorporation, this review examines how pharmacologic modulation of these pathways may affect fusion outcomes and perioperative risk. Methods We synthesized mechanistic, translational, and clinical evidence examining the impact of major classes of biologic immunomodulat

Amyloid-Like Nanocoatings for Enhanced Hemoperfusion Materials.

Hemoperfusion, a therapeutic technique in which blood is circulated through extracorporeal adsorbents to remove toxins, faces the challenge of simultaneously enhancing the adsorption performance and hemocompatibility of hemoperfusion materials (HPMs) through a facile and eco-friendly surface engineering strategy. Herein, a one-step amyloid-mediated surface modification method is proposed to encapsulate HPMs by immersing them in a phase-transitioned bovine serum albumin (PTB) aqueous solution. The resultant amyloid-like PTB nanocoating modified activated carbon exhibited a nearly four-fold incr

A longitudinal resource for mapping interindividual variation in the aging connectome.

Trajectories of age-related neurocognitive decline are nonuniform, and are impacted by numerous environmental and physiological factors. Earlier life phases set the stage for later life neurocognitive function, with midlife marking a critical transition characterized by increasing variability in cognitive, affective, and physiological functioning. Despite its importance, this turbulent period remains underrepresented in open neuroimaging data resources. To address this gap, the Nathan Kline Institute - Rockland Sample (NKI-RS) created 'Mapping Interindividual Variation in the Aging Connectome'

Early Local Treatment Delays Multiple Metastases in Non-Small Cell Lung Cancer With Solitary Distant Metastasis: A Multicenter Retrospective Cohort Study.

Solitary distant metastasis (SDM) is a distinct oligometastatic subtype associated with a relatively favorable prognosis. This multicenter retrospective study including 139 non-small cell lung cancer (NSCLC) patients with SDM (2019-2024) was conducted to investigate prognostic factors for multiple metastases and identify the critical time window for local treatment. Statistical analyses included Cox regression, Kaplan-Meier survival analysis, and restricted mean survival time (RMST). Results showed that driver gene status, tumor staging, systemic therapy, and local treatment were significantly

Calm after the storm; transition from crisis: Iranian nurses' experiences of the COVID-19 pandemic - a qualitative study.

OBJECTIVES: The COVID-19 pandemic had significant physical and psychosocial impacts on nurses. While the initial phases of the pandemic presented considerable challenges, these appear to have diminished over time. This study aimed to explore the lived experiences of Iranian nurses working in clinical settings during the post-pandemic period of COVID-19. DESIGN: This is a qualitative study conducted using the conventional content analysis approach proposed by Granheim and Lundman (2004). SETTING: This study was conducted at secondary care hospitals in northwest Iran that served as referral ce

Bridge-Disease Curriculum Transfer Enables Smartphone-Based Balance Assessment in Rare Neurologic Disorders.

Video-based motor assessment could enable scalable assessment of neurological function, but rare-disease applications are constrained by extreme data scarcity and poor generalization from small cohorts. We develop and evaluate a smartphone-based balance-assessment framework in NF2-related schwannomatosis (NF2-SWN), an ultra-rare disorder for which objective balance assessment is clinically valuable yet logistically challenging. At its core, a staged bridge-disease curriculum adapts a skeleton-based spatiotemporal graph model from large-scale action recognition through an intermediate Parkinson

Microbial Survival, Sublethal Injury, and Recovery in Frozen Food Systems: Implications for Safety, Quality Deterioration, and Cold-Chain Control.

Freezing is widely regarded as a microbiologically protective process that suppresses growth and extends shelf life but rarely eliminates microorganisms. Microbial risk in frozen foods is, therefore, determined not only by survival during storage at -18°C but by post-thaw repair and growth during thawing and handling. This review synthesizes freeze-induced injury mechanisms, including membrane damage, osmotic stress, oxidative injury, and macromolecular disruption, and links them to physiological state transitions, lag phase behavior, and recovery potential. Rather than treating sublethal inju

Adaptive Network Stabilization Despite Profound Structural Reorganization in Human Organotypic Cultures.

Human organotypic slice cultures provide experimental access to adult human neuronal circuits ex vivo, yet it remains unclear whether these networks preserve function or undergo fundamental reorganization following the profound perturbation of slice preparation. Here, a multimodal approach combines extracellular electrophysiology, longitudinal calcium imaging, and quantitative histology to track the changes of human cortical slice cultures over several weeks in vitro. Early phases are marked by pronounced variability and instability, with reduced firing rates, increased burst propensity in pri

The KRAS targeting revolution in metastatic pancreatic cancer: insights from the landmark RASolute-302 trial and emerging allele-specific strategies at ASCO 2026.

Survival in metastatic pancreatic ductal adenocarcinoma (mPDAC) has long been limited by a dismal second-line therapeutic ceiling dictated by conventional chemotherapy. However, breakthrough data from the ASCO 2026 Annual Meeting and the publication of the phase III RASolute-302 trial mark a definitive shift toward targeted KRAS inhibition. This correspondence highlights how the first-in-class pan-RAS (ON) inhibitor daraxonrasib (RMC-6236) virtually doubled median overall survival (13.2 vs. 6.6 months) and progression-free survival compared to chemotherapy in second-line mPDAC, establishing a

Therapeutic potentials of mesenchymal stem cell-derived exosomes for major solid malignancies: A narrative systematic review.

Treatments for solid tumors, the most common malignant neoplasms, are often confounded by tumor microenvironments that impede the achievement of uniform anti-tumor effects throughout the entire malignant mass, which contributes to recurrence and progression, negatively impacting clinical outcomes. Improved treatment methods for solid malignancies are therefore needed. Mesenchymal stromal cells (MSCs) have been investigated for treatments for various types of solid tumor cancers due to their ability to target tumor cells with similar cell surface protein profiles. MSC-derived exosomes (MSC-Exos

CTNND1 affects trophoblast proliferation and specification during human embryo implantation.

The placenta, serving as the crucial link between maternal and infant, plays a pivotal role in maintaining a healthy pregnancy. Placental dysplasia can lead to various complications, underscoring the importance of understanding trophoblast lineage development. During peri-implantation, the trophectoderm undergoes differentiation into cytotrophoblast, syncytiotrophoblast, and extravillous trophoblast. However, the specification and regulation of human trophoblast lineage during embryo implantation, particularly in the peri-implantation phase, remain to be explored. In this study, we employed a
