Ontogeny of hypothalamo-adenohypophyseal-gonadal (HAG) interrelationships in the chick embryo.

In the male chick embryo the components of the hypothalamo-adenohypophyseal-testicular axis initially function independently of each other. It is not until days 12.5-13.5 that the adenohypophysis begins to regulate testosterone synthesis and secretion; on day 13.5 plasma testosterone reaches a maximum embryonic level. This feed forward regulation of the pituitary-testicular unit appears to involve a cause and effect relation between a statistically significant increase in the number of testicular interstitial cell LH receptors on days 12.5 and 13.0 and an increase in plasma LH levels on day 13

B lymphocyte precursors in embryonic and adult W anemic mice.

Mice homozygous for mutations at the dominant spotting or W locus on chromosome 5 have been extensively used as models of severe macrocytic anemia caused by defective hemopoietic stem cells. We examined cells of the developing B lineage in adult and embryonic W anemic mice both by phenotypic analyses and by three distinctly different functional assays for B lymphocyte precursors. Adult W/Wv mice had normal numbers of B cells in the spleen and bone marrow, and normal numbers of pre-B cells and cells identified by a monoclonal antibody directed to a B lineage cell surface antigen (14.8) in the b

Placental transfer of thiamphenicol in the rat.

Measurement of thiamphenicol transfer to the rat embryo during organogenesis was performed as one step of the determination of possible drug embryotoxicity in man. Extrapolation of data on animal embryotoxicity to man will only become possible when data on the toxicokinetic properties of the substance under investigation are available for both animal and man. 1) Thiamphenicol, given between day 11.5 and 14 of rat gestation, rapidly reached the embryo; 4--6 h after single i.v. or s.c. injection, embryonic and maternal thiamphenicol levels became equal and decreased from that time on at the same

Embryonic endocrine pancreas and mature beta cells acquire alpha and PP cell phenotypes upon Arx misexpression.

Aristaless-related homeobox (Arx) was recently demonstrated to be involved in pancreatic alpha cell fate specification while simultaneously repressing the beta and delta cell lineages. To establish whether Arx is not only necessary, but also sufficient to instruct the alpha cell fate in endocrine progenitors, we used a gain-of-function approach to generate mice conditionally misexpressing this factor. Mice with forced Arx expression in the embryonic pancreas or in developing islet cells developed a dramatic hyperglycemia and eventually died. Further analysis demonstrated a drastic loss of beta

Variabilities in prenatal development of orofacial system.

Reliable information on embryonic and fetal development of the human oro-facial system is meager. Much of the data available at present is not entirely reliable, because it was derived from a small number of specimens. An embryological approach with human materials is important for establishing a normal standard of development including individual variabilities as well as clarifying the embryogenesis and etiology of defective development (Nishimura et al. 1977). It is important in human craniofacial embryology to know the variabilities, that is, individual differences in developmental phenomen

Embryonic origin of two ASD subtypes of social symptom severity: the larger the brain cortical organoid size, the more severe the social symptoms.

BACKGROUND: Social affective and communication symptoms are central to autism spectrum disorder (ASD), yet their severity differs across toddlers: Some toddlers with ASD display improving abilities across early ages and develop good social and language skills, while others with "profound" autism have persistently low social, language and cognitive skills and require lifelong care. The biological origins of these opposite ASD social severity subtypes and developmental trajectories are not known. METHODS: Because ASD involves early brain overgrowth and excess neurons, we measured size and growt

Subcellular distribution of glucose transporter (GLUT-1) during development of the blood-brain barrier in rats.

Electron microscopy was used to quantify the subcellular distribution of the GLUT-1 isoform of the glucose transporter in developing microvessels of the brain of embryonic rats from E (embryonic stage) 13 to E19 and in adult rats. Gold-conjugated secondary antibodies were used to localize, on ultrathin sections of brain, a rabbit polyclonal antiserum (anti-GLUT-1) raised against a synthetic peptide encoding 13 amino acids of the C-terminus of the human glucose transporter. Staining was weak at E13 but increased in density during development into adulthood. The increase represented an increase

Characterizing the effects of in utero exposure to valproic acid on murine fetal heart development.

BACKGROUND: Recently, the use of the antiepileptic drug valproic acid (VPA) for the treatment of psychiatric conditions has been on the rise. However, studies have shown that in utero VPA exposure can affect embryonic development, including being associated with congenital heart defects. One proposed mechanism of VPA-initiated teratogenicity is the inhibition of histone deacetylase, which is involved in the regulation of transcription factors that regulate cardiogenesis. Myocyte enhancing factor 2C (Mef2c), a transcription factor involved in the development of cardiac structure and cardiomyocy

Safety profile of Hoodia gordonii extract: rabbit prenatal developmental toxicity study.

Hoodia gordonii extract was orally administered by gavage to groups of 22 female New Zealand white rabbits from day 3-28 after mating at doses of 0 (control), 3, 6 or 12 mg/kg bodyweight/day. These doses were reached by a dose escalation phase between days 3 and 7 after mating. As well as a vehicle control group, a control group pair-fed to the high dose was also included. On day 29 after mating the females were euthanized and examined. Treatment at 6 or 12 mg/kg/day was associated with a dose-related reduction in feed intake and bodyweight gain. Feed consumption and bodyweight gain was unaffe

Fifteen years after "Wingspread"--environmental endocrine disrupters and human and wildlife health: where we are today and where we need to go.

In 1991, a group of expert scientists at a Wingspread work session on endocrine-disrupting chemicals (EDCs) concluded that "Many compounds introduced into the environment by human activity are capable of disrupting the endocrine system of animals, including fish, wildlife, and humans. Endocrine disruption can be profound because of the crucial role hormones play in controlling development." Since that time, there have been numerous documented examples of adverse effects of EDCs in invertebrates, fish, wildlife, domestic animals, and humans. Hormonal systems can be disrupted by numerous differe

Maternal oral intake mouse model for fetal alcohol spectrum disorders: ocular defects as a measure of effect.

BACKGROUND: This work was conducted in an effort to establish an oral intake model system in which the effects of ethanol insult that occur during early stages of embryogenesis can be easily examined and in which agents that may modulate ethanol's teratogenicity can be readily tested in vivo. The model system described utilizes the alcohol deprivation effect to obtain teratogenic levels of maternal ethanol intake on days 7 and 8 of pregnancy in C57Bl/6J mice. Ocular defects including microphthalmia and uveal coloboma, which have previously been shown to result from ethanol administered by gava

Integrated genomic and transcriptomic analysis of polydactyly in chickens.

INTRODUCTION: Polydactyly-the presence of extra digits-is a heritable limb anomaly observed in several chicken breeds. The Puan Panjiang black-bone chicken uniquely exhibits both four- and five-toed phenotypes, yet the genetic and transcriptional bases of this trait remain unclear. This study aimed to elucidate the genomic variants and gene expression changes underlying polydactyly in this breed. METHODS: We performed whole-genome resequencing (WGS) on 43 Puan Panjiang chickens (22 four-toed, 21 five-toed) and integrated publicly available data from 17 red junglefowl (RJF). After stringent qu

Generation of an mESC model with a human hemophilia B nonsense mutation via CRISPR/Cas9 technology.

BACKGROUND: Hemophilia B is a rare inherited genetic bleeding disorder caused by a deficiency or lack of coagulation factor IX, the gene for which (F9) is located on the X chromosome. Hemophilia B is currently incurable and the standard treatment is coagulation factor replacement therapy. Although gene therapy has the potential to cure hemophilia, significant barriers are still needed to be overcome, e.g., off-target effects and immunoreactivity, so new approaches must be explored. Nonsense mutations account for 8% of all the hemophilia B mutation types and can result in the development of coa

Study on the preparation and activity of intelligent response poly(lactic-co-glycolic acid)-ss-polyethylene glycol copolymer micelles.

Amphiphilic polymer micellar carriers are the most commonly used nanocarriers for oral delivery of hydrophobic drugs because their hydrophilic shell can avoid the recognition of the reticuloendothelial system (RES), has excellent drug-carrying capacity, and protect the drug from inactivation in the gastrointestinal fluid. The polymer micelle shell can enter cancer cells by endocytosis, and autophagy in cells, degradation by lysosomal pathway, so as to release drugs, prolong the circulation time of drugs in vivo, and then achieve the effect of drug sustained release. In this study, the glutathi

Volume and vascularity of the yolk sac assessed by three-dimensional and power doppler ultrasound.

The yolk sac is an organ of increasingly recognized importance in the initial mechanisms of pregnancy maintenance and the early growth and welfare of the embryo. The aim of our study was to assess the vascularity of the yolk sac and vitelline duct in 150 patients between the 6th and 10th weeks of normal, and uncomplicated pregnancies who were scheduled for termination of pregnancy for psychosocial reasons and 130 complicated pregnancies. In same patients volume of the yolk sac was assessed using Combison 530 3D Voluson, Medison-Kretz Company. Overall visualization rate for yolk sac vessels was

[A study of the UV-sensitivity in Drosophila strains with different repair abilities].

Sensitivity of Drosophila embryos to lethal effect of UV rays was studied in mutants rad202G1 and mei-9a (a homologue of the gene for xeroderma pigmentosum) that are deficient in excision repair, the mutant mei-41D5 (a homologue of the gene for AT) with distorted check-point function in the cell cycle, and wild-type line Oregon R. The mortality of embryos, which were exposed to radiation within the 0.5-16-h interval of embryonic life, served as a criterion of sensitivity. During this interval of embryogenesis, the multicellular system of Drosophila embryo sequentially consecutively passes thro

Ectomesenchyme contributes to epidermal stem cell formation through mesenchymal-to-epithelial transition.

Cutaneous development is initiated by crosstalk between non-neural ectodermal epithelium and underlying mesenchymal cells. Recent studies have shown that some mesenchymal cells contribute to keratinocyte regeneration in injured skin. However, whether mesenchymal cells contribute to keratinocyte formation in physiologically normal skin remains unclear. Here we show, using lineage tracing, single-cell transcriptome and epigenome analyses of mouse skin, that the interfollicular epidermis consists largely of mesenchymal-lineage cells. Further lineage-tracing and live-imaging analysis of mouse embr

Lens tissue transcriptome analysis in patients with total congenital cataract.

BACKGROUND: Total congenital cataract (TCC), a leading cause of childhood visual impairment. We analyzed transcriptome profiles of lens epithelial cells (LECs) and lens fiber cells (LFCs) in pediatric patients with TCC to identify differentially expressed genes (DEGs) and potential pathogenic pathways. RESULTS: In LECs, 1,645 DEGs were identified, enriched in structural constituents of the eye lens extracellular matrix component and extracellular matrix organization. LFCs exhibited 2,708 DEGs, enriched in SRP-dependent cotranslational protein targeting to membrane, protein targeting to ER, an

From Stone to Sponge: Hippo/YAP Mechanotransduction Empowers Cell Fates and Functions in Bone and Liver.

Cells experience and respond to both biochemical and mechanical signals throughout embryonic development and adult homeostasis. Bone and liver represent hard and soft tissues, respectively, and they are critically controlled by their mechanoenvironment. Although the significance of mechanotransduction, the process by which mechanical forces are converted into biological cues, has been acknowledged since the nineteenth century, only recent advances have begun to reveal the molecular underpinnings bridging biophysical stimuli and gene regulation. This review synthesizes the latest insights into

Non-coding RNAs in congenital heart disease and placental development: Bridging molecular mechanisms to clinical biomarkers and therapies.

Non-coding RNAs (ncRNAs) have emerged as pivotal regulators of gene expression, orchestrating embryonic development and disease pathogenesis. This review synthesizes current knowledge on the origin, biogenesis, and functional diversity of ncRNAs, with a focus on their regulatory crosstalk in congenital heart disease (CHD) and placental development. The fetal heart-placenta axis, a bidirectional signaling network essential for cardiogenesis and placental morphogenesis, is spatiotemporally modulated by ncRNAs through epigenetic and post-transcriptional mechanisms. Through precise regulation of c
