Immune checkpoint inhibitors (ICIs) are immunomodulatory antibodies that intensify the host immune response, thereby leading to cytotoxicity. The primary targets for checkpoint inhibition have included cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), programmed cell death receptor-1 (PD-1) or programmed cell death ligand-1 (PD-L1). ICIs have resulted in a change in treatment landscape of various neoplasms. Among hematologic malignancies, ICIs have been most successful in certain subtypes of lymphomas such as classic Hodgkin lymphoma (cHL) and primary mediastinal B-cell lymphoma (PMBCL). H

INTRODUCTION: A 2016 assessment of frontline health care workers (HCWs) in Ghana identified knowledge, skill, and attitude gaps related to immunization during the second year of life (2YL). The U.S. Centers for Disease Control and Prevention subsequently supported the Ghana Health Service Immunization Program to apply best practices of adult learning and training of trainers (TOT) for a cascade training program for 2YL.
METHODS: Five districts from each of the 3 regions (Greater Accra, Northern, and Volta) were selected for the TOT based on key measles and rubella vaccination coverage indicat

Introduction: South Africa is yet to introduce rubella-containing vaccines (RCV) into its routine immunization schedule. Selecting the target population when introducing RCV should take into account the ages of susceptible individuals in the population. We aimed to determine the seroprevalence of antibodies to rubella and characterize immunity gaps among individuals of all ages in South Africa. Methods: We tested for rubella immunoglobulin G (IgG) antibodies with a commercial enzyme-linked immunosorbent assay. We used residual samples collected from 2016 through 2018 as part of the national me

AIMS: Salivary duct carcinoma (SDC) is an aggressive salivary malignancy that results in high mortality rates and is often resistant to chemotherapy. Anti-programmed death-1 (PD-1)/programmed death ligand-1 (PD-L1) checkpoint inhibitors have led to dramatic improvements in patients with various cancers. Other immunotherapeutic approaches, e.g. cancer vaccines, have shown promising results. Cancer testis antigens, e.g. preferentially expressed antigen in melanoma (PRAME), are regarded as promising vaccine targets because of their tumour-specific expression pattern.
METHODS AND RESULTS: We anal

Ibrutinib, a first-generation Bruton's tyrosine kinase (BTK) inhibitor, could improve immunity of relapsed or refractory (R/R) chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL) patients. Whether zanubrutinib, a second-generation selective BTK inhibitor, has similar effects as ibrutinib remains to be determined. Dynamics of number and immunophenotype of immune cells during zanubrutinib treatment in 25 R/R CLL/SLL patients were examined by flow cytometry and blood routine tests. The expression intensity of programmed death-1 (PD-1) on total CD4+ (P < .01), total CD8+ (P < .01), a

Emerging knowledge shows the importance of early life events in programming the intestinal mucosal immune system and development of the intestinal barrier function. These processes depend heavily on close interactions between gut microbiota and host cells in the intestinal mucosa. In turn, development of the intestinal microbiota is largely dependent on available nutrients required for the specific microbial community structures to expand. It is currently not known what the specificities are of intestinal microbial community structures in relation to the programming of the intestinal mucosal i

BACKGROUND: A desperate need for novel therapies in pediatric ependymoma (EPN) exists, as chemotherapy remains ineffective and radiotherapy often fails. EPN have significant infiltration of immune cells, which correlates with outcome. Immune checkpoint inhibitors provide an avenue for new treatments. This study characterizes tumor-infiltrating immune cells in EPN and aims at predicting candidates for clinical trials using checkpoint inhibitors targeting PD-L1/PD-1 (programmed death ligand 1/programmed death 1).
METHODS: The transcriptomic profiles of the primary study cohort of EPN and other

Hepatocellular carcinoma (HCC) is the fourth most prevalent and the second most lethal cancer in China. Immune checkpoint inhibitors (ICIs) have dramatically improved treatment outcomes for advanced HCC. However, ICIs can also cause immune-related adverse events (irAEs), including the rare occurrence of ICI-related pancreatitis (ICIPI). After successful treatment of ICI-related pancreatitis, further research is needed to determine whether continuation of ICI therapy is appropriate for the patient. Currently, the clinical feasibility of resuming ICI therapy following complete resolution of ICIP

Melanoma progression is characterized by complex molecular mechanisms and immune dysregulation. In this study, we investigated the role of pyroptosis, a caspase-dependent and pro-inflammatory form of programmed cell death, as a potential modulator of anti-tumor immunity. Analysis of datasets from The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) revealed that B lymphocyte tyrosine kinase (BLK), a member of the Src family kinases, is significantly downregulated in melanoma samples. Notably, elevated BLK expression was positively associated with improved overall survival. Func

Disulfidptosis is a new form of programmed cell death. However, there is limited information available regarding the impact of disulfidptosis on septic acute respiratory distress syndrome (ARDS). The 16 disulfidptosis-related genes (DRGs) were collected from a previous study. Gene expression data of sepsis and septic ARDS samples were downloaded from the Gene Expression Omnibus database. The risk score model in septic ARDS was constructed based on the DRGs, followed by the investigation of immune microenvironment in septic ARDS patients. Furthermore, septic ARDS patients were divided into diff

Over 80% of colorectal cancer (CRC) cases are classified as microsatellite stable (MSS), a subtype characterized by an immunosuppressive tumor microenvironment that poses a significant challenge for chemotherapy. Emerging evidence suggests that inflammatory programmed cell death pathways can effectively activate anti-cancer immunity across various cancer types. However, studies exploring the role of these death pathways in reversing the immune desertification and providing unique therapeutic advantages in MSS-CRC remain scarce. Herein, we present a strategy using iridium(III)-based inducers to

BACKGROUND/AIM: Pulmonary arterial hypertension (PAH) is a significant and challenging complication for patients with systemic lupus erythematosus (SLE). It is thought to arise from immune dysregulation and vascular remodeling, ultimately leading to progressive right heart failure. Molecular hydrogen therapy, a selective antioxidant and anti-inflammatory agent, may modulate the immune responses seen in autoimmune diseases. This case report details the use of adjuvant hydrogen capsule therapy in a patient with SLE with PAH, suggesting its potential as a novel approach for this difficult clinica

B cells contribute to humoral immunity by producing antibodies, presenting antigens, and secreting cytokines. Traditionally, B cells have been viewed as predominantly pro-immunogenic effectors that promote adaptive immune responses. However, accumulating evidence has identified a distinct B cell subset with immunosuppressive properties, termed regulatory B cells (Bregs), which restrain pathogenic T-cell responses by secreting anti-inflammatory cytokines including interleukin-10 (IL-10), IL-35, and transforming growth factor β (TGF-β), and by expressing immunoregulatory surface molecules such a

PANoptosis, an integrated programmed cell death modality encompassing apoptosis, necroptosis, and pyroptosis, exerts pleiotropic effects on tumor immune surveillance and immune escape. However, the molecular mechanisms underlying PANoptosis in pediatric acute myeloid leukemia (AML) remain poorly elucidated. Here, we performed bioinformatic analyses of AML bone marrow transcriptomic data from the TARGET database, aiming to define PANoptosis-associated molecular subtypes, characterize tumor immune microenvironment (TIME) heterogeneity, and identify candidate regulatory genes and small-molecule b

Nivolumab, a programmed death-1 (PD-1) immune checkpoint inhibitor (ICI), improves outcomes in advanced renal cell carcinoma (RCC) by enhancing antitumor T-cell activity. Immune activation may result in immune-related adverse events, including endocrine dysfunction. Thyroid abnormalities are commonly observed, whereas adrenal insufficiency is less frequent but clinically significant. We report a 59-year-old man with metastatic chromophobe RCC who developed autoimmune thyroiditis followed by secondary adrenal insufficiency after treatment with nivolumab. Thyroid dysfunction occurred approximate

Staphylococcus aureus is a leading cause of skin and soft tissue infections (SSTIs), yet the cellular mediators of protective memory remain incompletely defined. Using a self-resolving murine SSTI model, we combined longitudinal single-cell RNA sequencing with flow cytometry to characterize immune memory within the skin following infection. We identified a persistent population of CD4+ T cells that remained in the skin after bacterial clearance and acquired a transcriptional program consistent with tissue residency, including expression of genes associated with tissue retention and long-term r

Prostate cancer is common in older men, yet mechanisms linking aging to tumor progression remain incompletely defined. Beyond genetic alterations, aging reshapes the prostate microenvironment through cellular senescence, chronic low-grade inflammation, immune dysfunction, stromal remodeling, metabolic stress, and impaired tissue repair, collectively promoting inflammaging. This persistent inflammatory state may create a permissive niche for tumor initiation, progression, immune evasion, and treatment resistance. Senescent epithelial and stromal cells release cytokines, chemokines, growth facto

Lung cancer is a deadly disease with the highest rates of mortality. Over recent decades, a better understanding of the biological mechanisms implicated in its pathogenesis has led to the development of targeted therapies and immunotherapy, resulting in improvements in patient outcomes. To better understand lung cancer tumor biology and advance towards precision oncology, a comprehensive tumor profile is necessary. In recent years, novel in situ spatial multiomics approaches have emerged offering a more detailed view of the spatial location of tumor and tumor microenvironment cells, identifyin

It is newly revealed that collagen works as a physical barrier to tumor immune infiltration, oxygen perfusion, and immune depressor in solid tumors. Meanwhile, after radiotherapy (RT), the programmed death ligand-1 (PD-L1) overexpression and transforming growth factor-β (TGF-β) excessive secretion would accelerate DNA damage repair and trigger T cell exclusion to limit RT efficacy. However, existing drugs or nanoparticles can hardly address these obstacles of highly effective RT simultaneously, effectively, and easily. In this study, it is revealed that inducing mitochondria dysfunction by usi
