Nitric oxide interacts with brassinosteroid signalling to regulate plant growth and development in an organ-specific manner.

Nitric oxide (NO) is a plant gasotransmitter that regulates plant growth and development by interacting with different regulatory pathways. Among these, brassinosteroids (BRs) have become candidate phytohormones that interact antagonistically and closely with NO to regulate plant growth. Seedling growth in NO-deficient mutants is enhanced and hyper-responds to BR treatments, while NO overaccumulator mutants show some developmental defects. Interestingly, this hyper-response was also observed in mutants that had both constitutively activated BR signalling and reduced NO levels. BR signalling mu

Transcriptional and phytohormonal regulation of positional ear development reveals yield strategies in maize.

Maize (Zea mays L.) is a vital global crop, contributing approximately 37% of annual grain production. Enhancing yield per unit area is crucial for food security, yet research has primarily focused on single-ear traits, overlooking the regulation of double ears-a key determinant of prolificacy. While secondary ears drive yield variability under prolificacy-favoring conditions, the mechanisms governing ear formation across shoot positions remain poorly understood. Here, we performed high-resolution transcriptomic analysis of 66 samples from 3 ear types (primary, secondary, and tertiary) in maiz

Modeling Complex Developmental Disease: The Case of Polycystic Kidney Disease.

Both genetics and the environment affect the phenotypes of polycystic kidney diseases (PKD), such as autosomal dominant (AD) PKD, autosomal recessive (AR) PKD and nephronophthisis (NPH). Variable phenotypes, pleiotropy, divergent severity and progression, even in family members who inherited the same disease-causing mutation(s), signal the involvement of networked genes and modifiers. Several PKD-linked genes function in development and renal tubule morphogenesis. Cystic renal tissues feature metabolic remodeling, functional reprogramming and dysregulation of several shared factors and pathway

Octopamine Signaling Regulates Phototactic Behavior via a β-Adrenergic-like Receptor in Diaphorina citri.

The Asian citrus psyllid (Diaphorina citri, ACP) is the primary vector of citrus huanglongbing and exhibits positive phototactic behavior. Octopamine (OA) is a critical neuromodulator of diverse insect behaviors, yet its regulatory mechanism in ACP phototactic behavior remains largely uncharacterized. In this study, three core genes of the OA signaling pathway in ACP were identified and characterized: the biosynthetic enzyme DcTβH and two β-adrenergic-like OA receptors (DcOctβ1R, DcOctβ2R). All three genes exhibited a U-shaped developmental expression pattern and high transcript abundance in t

Harnessing cutting-edge techniques to identify novel gene expression signatures in acute myeloid leukemia patients.

INTRODUCTION: Acute Myeloid Leukemia (AML) is a heterogeneous hematological malignancy with poor prognosis, despite therapeutic advances. Gene expression analysis has emerged as a powerful tool for identifying novel biological markers that can aid in predicting patient outcomes. This study is aimed to identify prognostic gene expression signatures from RNA-seq data of 457 AML patients. METHODS: Filtering methods were applied to reduce the set of genes, resulting in a subset of 685. Lasso-Cox, Bayesian Model Averaging (BMA), Random Survival Forest (RSF), and a Cox-time Neural Network (CtNN) we

Single-cell lineage tracing unveils sex differences in VSMCs derived from the cardiac neural crest and second heart field.

Stanford type A thoracic aortic aneurysm and dissection (TAAD) affects the ascending aorta and aortic arch and can cause sudden death due to rupture. Epidemiological studies have shown that TAAD occurs more frequently in men than women. Vascular smooth muscle cells (VSMCs) are one of the major cell types in the vascular wall and play a critical role in the pathogenesis of TAAD. Recent studies have highlighted the developmental heterogeneity of VSMCs in the ascending aorta and aortic arch, which are derived primarily from the second heart field (SHF) and cardiac neural crest (CNC), respectively

Gateway: patient olfactory neurons for large-scale discovery in neurodegenerative disease.

An estimated 42% of Americans over age 55 will develop dementia, but the molecular understanding of dementia and neurodegenerative disease is constrained because the living human brain cannot be routinely sampled during disease progression. Olfactory sensory neurons provide a clinically accessible neuronal tissue source with developmental, transcriptional, and disease-relevant links to the central nervous system. Here we describe Gateway, a platform that combines device guided olfactory epithelium biopsy, onsite fixation, and 10x Genomics FLEX RNA profiling to generate single-cell transcriptom

Prenatal APPV infection delays cerebellar myelination and alters gene expression dynamics during recovery from congenital tremor in pigs.

Atypical porcine pestivirus (APPV) causes congenital tremor (CT) type A-II in piglets, a globally distributed neurological condition, yet its impact on central nervous system (CNS) development and recovery remains poorly understood. This longitudinal case-control study investigated APPV-induced alterations in cerebellar and cerebral cellular processes during clinical disease and recovery by integrating transcriptomic profiling with immunohistochemistry (IHC) in CT pigs at three ages (newborn, 3-week-old, 4-5 months) compared to controls. Group-level transcriptional patterns were assessed using

Comparative Genomics Analysis Provides Insights into Pregnancy Maintenance Mechanisms in the Ovoviviparous Teleost Black Rockfish (Sebastes schlegelii).

The black rockfish (Sebastes schlegelii), an economically important ovoviviparous teleost in Chinese aquaculture, exhibits a distinctive reproductive strategy characterized by internal fertilization and prolonged embryonic development within females until live birth. However, this reproductive mode poses substantial challenges in artificial breeding practices due to a poor understanding of pregnancy (hereafter referring to intraovarian gestation in S. schlegelii) maintenance mechanisms. Here, we conducted a comparative genomic analysis between S. schlegelii and representative oviparous and ovo

Functional verification of a landmark gene EVM713 involved in spermatogenesis in the marine bivalve Chlamys nobilis.

The formation of broodstock gametes is closely linked to the yield and quality in aquaculture production, yet molecular mechanisms underlying this process remain insufficiently understood. The noble scallop Chlamys nobilis, an economically significant dioecious bivalve species, serves as an excellent model for studying gametogenesis. In this study, the adult scallops with testis at different developmental stage were chosen for histological examination and transcriptome analysis to dig genes related gonad development. Totally, 2663 DEGs and their set modules significantly related to spermatogen

Typical Thermochromic Dye Crystal Violet Lactone Acts as an Endocrine Disruptor: Nontargeted Screening, Prioritization, and In Vivo Experimental Verification.

Identification and characterization of endocrine disruptors and their associated ecological and health risks are critical areas of environmental toxicology. In this study, we aimed to identify previously unrecognized endocrine disruptors in thermal paper by employing a nontargeted screening coupled ToxPi Ranking (NSTR) approach. Among the substances identified with high confidence in thermal paper, Crystal Violet Lactone (CVL), a typical thermochromic dye, was prioritized with a high endocrine disruptive potential. CVL can be absorbed through the skin and excreted in the urine. In addition, we

Age-dependent differential iron deficiency responses of rosette leaves during reproductive stages in Arabidopsis thaliana.

Iron (Fe) is essential for plant development throughout the life cycle. Rosette leaves are responsive to Fe supply in Arabidopsis thaliana. Little is known about the dynamics of Fe deficiency (-Fe) responses of rosette leaves during the reproductive stages. We studied the dynamics of Fe-dependent responses at four consecutive reproductive stages (rosette, bolting, flowering, and mature silique stages, herein named RS, BS, FS, and MS). We examined the growth of rosette leaves, elemental contents, and gene expression patterns of Fe homeostasis genes belonging to differently regulated groups. We

RSPO2 Coordinates with GDF9:BMP15 Heterodimers to Promote Granulosa Cell and Oocyte Development in Mice.

The generation of mature oocytes, a cornerstone of reproduction, relies on the coordinated interactions between oocytes and surrounding follicular somatic cells. Central to this process is the bidirectional communication between the oocyte and granulosa cells, mediated by oocyte-secreted factors (OSFs), including GDF9 and BMP15. While GDF9 and BMP15 are well-established regulators of oocyte and follicle development, the role of additional OSFs and their coordination with GDF9 and BMP15 remains largely unclear. Here, RSPO2 is identified as a key OSF that coordinates with the GDF9:BMP15 heterodi

Cell-specific expression biases in human cortex of genes associated with neurodevelopmental disorders.

Up to one third of congenital brain malformations and neurodevelopmental disorders are attributable to single-gene pathogenic variants, and yet we have little understanding of the cellular pathophysiology in the nervous system that arises from these variants. To investigate cortical cell type-specific biases in gene expression associated with distinct neurodevelopmental phenotypes, we integrated phenotypic information from two cohorts of subjects with monogenic neurodevelopmental diagnoses with two human cortical single-nucleus RNA-sequencing (snRNAseq) datasets. Phenotype data was gathered fr

Synostosis of joints caused by mutant FBN2 is linked to the abnormalities and misdifferentiation of articular surface cells.

PURPOSE: FBN2, a high-confidence effector gene for osteoarthritis (OA), was investigated for its potential role in synostosis of joints (SJ) because several OA-related genes are known to cause SJ. METHODS: We analyzed variants in OA-related genes using exome sequencing data from Chinese-Han participants with radioulnar synostosis (RUS). Variants were classified following American College of Medical Genetics and Genomics/Association for Molecular Pathology guidelines. Fbn2 knockout mice were generated via CRISPR/Cas9 and evaluated through radiological and histopathological analyses at multiple

Integrated single-nuclei and spatial transcriptomic profiling of human sacrococcygeal teratomas reveals heterogeneity in cellular composition and X-chromosome inactivation.

Sacrococcygeal teratomas (SCTs) are the most common neonatal tumors, yet their cellular origins, clinical stratification, and sex bias-occurring three times more in XX than XY individuals-remain poorly understood. To address these gaps, we examined six postnatal (one male and five female) and two prenatal (both female) SCTs by single nuclei RNA-seq and spatial transcriptomics. We identified five broad cellular lineages in SCTs: stroma, epithelia, endothelia, neuroectoderm, and immune. The transcriptomes and lineage compositions showed significant heterogeneity, which offer a framework for futu

A B-Box (BBX) Transcription Factor from Cucumber, CsCOL9 Positively Regulates Resistance of Host Plant to Bemisia tabaci.

B-box (BBX) transcription factors play crucial roles in plant growth, development, and defense responses to biotic and abiotic stresses. In this study, we cloned a BBX transcription factor gene, CsCOL9I, from cucumber and analyzed its role in the plant's defense against the feeding of Bemisia tabaci. CsCOL9 is expressed throughout all developmental stages in cucumber, with the highest expression in the leaves. CsCOL9 is induced by B. tabaci feeding, salicylic acid (SA), methyl jasmonate (MeJA), and hydrogen peroxide (H2O2). Cucumber plants with CsCOL9 silence (TRV2-CsCOL9) and overexpression (

Hypohidrotic Ectodermal Dysplasias: Phenotypic and Genotypic Findings in 32 Cases.

Hypohidrotic ectodermal dysplasias are a genetic condition affecting ectoderm-derived structures such as hair, teeth, nails, and sweat glands, resulting from variations in the EDA, EDAR, EDARADD, and WNT10A genes. This study examined 32 cases from 25 unrelated families from Türkiye, identifying seven novel variants in the EDA, EDAR, and WNT10A genes. The distribution of genetic alterations across the cohort revealed that 44% of the families (11/25) harbored variants in EDA, whereas EDAR and WNT10A variants were identified in 32% (8/25) and 24% (6/25) of families, respectively. Clinical evaluat

Genetic analysis of a Chinese family with non-syndromic tooth agenesis may reveal a potential multi-locus etiology.

Tooth agenesis (TA), one of the most common craniofacial developmental anomalies, is characterized by the congenital absence of one or more teeth. While numerous genes have been implicated in non-syndromic tooth agenesis (NSTA), its genetic architecture often remains complex. In this study, we investigated the genetic basis of NSTA in a two-generation Chinese family utilizing whole-exome sequencing (WES) complemented by Sanger sequencing. Our analysis revealed a complex segregation pattern of multiple variants. After systematic filtering based on pathogenicity predictions and minor allele freq
