The role of IL-6 -174C/G polymorphism in hepatitis B virus infection: a genetic association study.

The development of the disease after infection with the hepatitis B virus (HBV) is linked to single-nucleotide polymorphisms (SNPs). SNPs' involvement in patients' persistent HBV infection is still unknown, though. Thus, the purpose of the current study was to assess the IL-6174C/G polymorphism in hepatitis B-infected Iraqi patients. The results revealed that serum HBV patients had higher levels of several interleukins, including IL-2, which exhibited a significant (P < 0.05) increase in individuals (237.04 ± 16.29 pg/mL) in comparison to the control group (115.42 ± 12.82 pg/mL). Serum IL-4 le

The bioinformatics approach to identifying pathogenic variants for colorectal cancer (CRC).

Colorectal cancer (CRC) is the third most prevalent cancer globally, accounting for 9.6% of newly diagnosed cases and 9.3% of cancer-related deaths. It develops from the uncontrolled proliferation of glandular cells in the colon and rectum and is categorized into three primary types: sporadic, hereditary, and colitis-associated. While genetic susceptibility is a key factor in CRC pathogenesis, identifying high-impact pathogenic variants remains a significant challenge. This study integrates bioinformatics and population genetics approaches to identify CRC-associated single-nucleotide polymorph

Development a novel in silico mRNA vaccine against Clostridium difficile.

Clostridium difficile, a toxin-producing bacterium, can lead to conditions ranging from mild diarrhea to severe pseudomembranous colitis. Our research introduces an innovative strategy for developing a multi-epitope mRNA vaccine using in-silico methods targeting proteins involved in Clostridium difficile infection (CDI). Through computational models, we identified epitopes for B cells, helper T lymphocytes (HTL), and cytotoxic T lymphocytes (CTL) from protein sequences, assessing their immune response potential, allergenicity, toxicity, and autoimmune risk. The constructed 3D model underwent i

Explainable machine learning and multimodal single cell analysis link NR4A2 to CD8+ T-cell terminal exhaustion and a CD8-adjusted prognostic signature in hepatocellular carcinoma.

BACKGROUND: Terminal exhaustion of CD8+ tumour-infiltrating lymphocytes constrains the benefit of immune-checkpoint blockade in hepatocellular carcinoma (HCC). The orphan nuclear receptor NR4A2 (Nurr1) belongs to a transcription-factor family implicated in murine T-cell exhaustion, yet its behaviour in the human HCC microenvironment has not been characterised across data modalities. We asked whether NR4A2 is specifically coupled to the terminal-exhaustion programme of human HCC CD8+ T cells, and whether that coupling is reflected in chromatin, tissue topography and bulk-tumour outcome data. Evaluating the effect of SNPs located on some biofilm genes on their protein function and structure in Pseudomonas aeruginosa and Klebsiella pneumonia isolates.

To evaluate the impact of biofilm-associated SNPs (previously identified) on protein structure and function, we employed a suite of bioinformatics tools. SIFT, Mupro, INPS-3D, and NCBI Conserved Domain Search were utilized to analyze changes in function, stability, secondary structure, phi/psi angles, and Relative Solvent Accessibility (RSA). Furthermore, SwissDock and molecular dynamics simulations were conducted to compare free energy, RMSF, polarity, flexibility, and molecular contacts between wild-type and mutant proteins. In silico analysis indicated that mutations in quorum-sensing (lasI

A novel adeno-associated viral vector derived from human spleen isolate AAV.hu.S17.

Adeno-associated viruses (AAVs) are widely used vectors for gene therapy owing to their nonpathogenic nature, low immunogenicity, and ability to support long-term transgene expression. However, the tissue tropism and immune response of AAVs are strongly dependent on their capsid serotype. Although hundreds of naturally occurring AAV variants have been isolated, many remain unvectorised and poorly characterized. Therefore, the discovery and development of previously uncharacterised native serotypes retain valuable. Here, we generated a novel recombinant AAV vector rAAV.hu.S17, derived from the

PIP3 antagonist as a molecular regulator in MSC-derived cardiomyocytes: Potential in vitro therapeutic implications for conotruncal heart defects.

Conotruncal heart defects (CTDs) account for approximately one-third of all congenital heart defects. Elevated levels of phosphatidylinositol (3,4,5)-trisphosphate (PIP3) may contribute to CTD pathogenesis. PIP3 plays a pivotal role in mechanotransduction-based biological processes and remodeling of cardiac cytoskeletal proteins. Here, we aimed to evaluate the efficacy of the 322PESB derivative compound as a molecular regulator that antagonizes PIP3 binding pleckstrin homology (PH) domain of the Akt protein using mesenchymal stem cell-derived cardiomyocyte. Human adipose-derived MSCs (Ad-MSCs)

Ancient DNA as a temporal lens: reconstructing evolution, migration, and disease dynamics.

Ancient DNA (aDNA) has transformed evolutionary biology and anthropology by providing direct, chronologically validated genetic evidence over millennia. This review synthesizes significant findings from the paleogenomic era (2010-2025), demonstrating how ancient DNA has resolved persistent debates across four interconnected themes: (i) human migration and admixture, revealing complex population transitions from archaic hominins to Holocene expansions; (ii) adaptation, tracking allele frequency changes during domestication and selection; (iii) pathogen history, clarifying the origins of pandemi
