Study on the spatiotemporal regulation of interferon-stimulated genes during Zika virus infection.

BACKGROUND: Zika virus (ZIKV) can cause severe neurological disorders such as congenital microcephaly, and there are currently no approved prevention or treatment measures. Elucidating the antiviral mechanism of the host interferon-stimulated genes (ISGs) is crucial for the development of therapeutic strategies. METHODS: This study integrated multisystem transcriptomic data [human brain organoids, monocyte-derived dendritic cells (moDCs), and peripheral blood mononuclear cells (PBMCs) from infected patients], combined with cellular experiments and bioinformatics analysis, to systematically in

Antiseizure Prescription for Children With Severe Congenital Heart Defects and Children With Gastrointestinal Anomalies.

BACKGROUND: Children with congenital anomalies (CAs) are at an increased risk of developing epilepsy, but the relative risk (RR) for specific anomaly subtypes remains underexplored. This study aims to estimate the risk of epilepsy, as indicated by antiseizure medication (ASM) prescriptions, among children with various CAs compared to children without anomalies. METHODS: We utilized data from six European regions participating in the European network for surveillance of congenital anomalies registries, covering births from 2000 to 2015. Children with major CAs, classified by International Clas

Severe Progression of Scoliosis Beyond Skeletal Maturity in Partial Trisomy 13: A Case Report Expanding the Phenotypic Spectrum.

Partial trisomy 13 (Patau syndrome) is a rare chromosomal disorder with variable survival and a wide spectrum of congenital anomalies. While skeletal abnormalities have been described, scoliosis remains infrequently reported, particularly in adolescents with partial trisomy 13 who survive beyond early childhood. We report the case of an 18-year-old male with partial trisomy 13 due to an unbalanced 13;21 translocation who developed progressive thoracolumbar scoliosis during adolescence. The patient was diagnosed with scoliosis at age 13, measuring 33°, and was initially managed with Boston brac

KIF23 in disease pathogenesis and its therapeutic and diagnostic potential.

BACKGROUND: Kinesin family member 23 (KIF23) is a microtubule-dependent motor protein essential for cytokinesis, organelle transport, and signaling pathway regulation. Its dysregulation contributes to both tumorigenesis and non-malignant disorders; however, a comprehensive review integrating recent mechanistic and translational insights is currently lacking. METHODS: A literature search across PubMed, Web of Science, Embase, and public databases (such as TCGA), using keywords including "KIF23," "MKLP1," and "cytokinesis" was performed. Published bioinformatic findings, including pan cancer sc

Distinct developmental outcomes in DNA repair-deficient FANCC c.67delG mutant and FANCC-/- Mice.

Fanconi Anemia (FA) is an autosomal recessive disorder characterized by diverse clinical manifestations such as aplastic anemia, cancer predisposition, and developmental defects including hypogonadism, microcephaly, organ dysfunction, infertility, hyperpigmentation, microphthalmia, and skeletal defects. In addition to the well-described defects in DNA repair, mitochondrial dysfunction due to defects in mitochondrial autophagy (mitophagy) is also associated with FA, although its contribution to FA phenotypes is unknown. This study focused on the FANCC gene, which, alongside other FA genes, is i

Characterizing trends in clinical genetic testing: A single-center analysis of EHR data from 1.8 million patients over two decades.

A lack of structural data in electronic health records (EHRs) makes assessing the impact of genetic testing on clinical practice challenging. We extracted clinical genetic tests from the EHRs of more than 1.8 million patients seen at Vanderbilt University Medical Center from 2002 to 2022. With these data, we quantified the use of clinical genetic testing in healthcare and described how testing patterns and results changed over time. We assessed trends in types of genetic tests, tracked usage across medical specialties, and introduced a new measure, the genetically attributable fraction (GAF),

Novel SSR4 gene splice variant leads to congenital disorder of glycosylation, type Iy.

BACKGROUND: Congenital disorders of glycosylation (CDG) are a group of multi-systemic genetic disorders. Over 100 monogenic human diseases were known related with defects in glycosylation process. Defects of SSR4 gene lead to a rare X linked pattern of CDG which has been rarely reported. METHOD: We reported a Chinese boy with developmental delay, microcephaly, and epileptic seizures. Whole exome sequencing and Sanger sequencing were performed in the family. RESULT: A novel maternal splice variant c.351+1del in SSR4 gene was identified by trio-exome sequencing, and confirmed by Sanger sequenc

An ultrapotent human antibody neutralizes all maturation states of Zika virus.

Zika virus (ZIKV), a flavivirus, causes a range of clinical complications including microcephaly in human fetuses. Currently, there is no treatment or vaccine. Different maturation states (mature and immature forms) of flavivirus particles have been observed to be released from infected cells and are infectious. To understand how an ultrapotent human antibody (HMAb) A9E can neutralize these Zika particles, we determined the cryoEM structures of the A9E Fab fragment complexed with mature (mZIKV) and immature (immZIKV) ZIKV to 2.8Å and 7.5Å, respectively. A9E binds to an epitope spanning Domain

In vivo and in vitro susceptibility and inflammatory response of postnatal mouse cortical neurons and glial cells to zika virus infection.

Zika virus (ZIKV) poses a significant threat to neural tissue, causing substantial damage to unborn children exposed to the virus in utero, with consequences that can manifest even after birth, despite being born with a normal head circumference. Regardless of the extensive research, the interactions between ZIKV and the nervous system cells remain insufficiently understood, particularly regarding how neuronal responses influence broader inflammatory and viral dynamics especially in postnatal stages of development. This study evaluated the susceptibility to ZIKV infection, viral replication, i

Oropouche Virus (OROV) and Breastfeeding Safety: Analysis of Related Orthobunyaviruses for Mother-Infant Vertical Transmission in Breast Milk.

The discovery that the Oropouche virus (OROV) can be transmitted vertically from an infected pregnant mother to the fetus, resulting in fetal and placental OROV infection, miscarriage, stillbirth, and congenital malformations including microcephaly, has emphasized its public health significance. Because of the importance of breastfeeding in those areas affected by the Oropouche fever outbreak, public health agencies have continued to encourage nursing among mothers who have had OROV infection or who reside or travel in endemic regions. However, the basis for this recommendation has not been st

A New Threat on the Rise: Oropouche Viral Infection.

The Oropouche virus (OROV), a member of the Simbu group's Orthobunyaviruses, is a vector-borne virus with three single-stranded negative-sense RNA segments and a spherical lipid sheath. It is mainly spread by Culicoides paraensis midges and, to a lesser extent, Culex quinquefasciatus mosquitoes. OROV, first reported in Trinidad and Tobago in 1955, has since spread to Brazil, Colombia, and Peru, causing serological epidemics and recently invading nonendemic regions. The severity of the disease can vary from cold symptoms to severe illness, such as meningitis. Recent outbreaks in Acre and Brazil

Ibuprofen-induced multiorgan malformation during embryogenesis in Xenopus laevis (FETAX).

Ibuprofen, one of the most commonly prescribed nonsteroidal anti-inflammatory drugs, has not been fully assessed for embryonic toxicity in vertebrates. Here, we systematically assessed the embryotoxicity of ibuprofen in Xenopus laevis at various concentrations during embryogenesis. Embryos were treated with different concentrations of ibuprofen, ranging from 8 to 64 mg/L, at 23 °C for 96 h, and examined daily and evaluated at 72 hpf. Lethal or teratogenic effects were documented. For histological analysis, paraffin embedded embryos were transversely sectioned at a thickness of 10-μm and staine

Antiviral drug discovery: Pyrimidine entry inhibitors for Zika and dengue viruses.

Vector-borne diseases, constituting over 17 % of infectious diseases, are caused by parasites, viruses, and bacteria, and their prevalence is shaped by environmental and social factors. Dengue virus (DENV) and Zika virus (ZIKV), some of the most prevalent infectious agents of this type of diseases, are transmitted by mosquitoes belonging to the genus Aedes. The highest prevalence is observed in tropical regions, inhabited by around 3 billion people. DENV infects millions of people annually and constitutes an additional sanitary challenge due to the circulation of four serotypes, which has comp

Chromosomal 1p Duplication in a Pediatric Patient: A Case Report.

Chromosomal 1p duplications are a rarity, with minimal literature on the topic. As a result, it is useful to document patient presentations with this defect to help guide the management and treatment of future patients with this genetic abnormality. We present a successful case report of a patient with a chromosome 1p31.3p31.1 duplication, including her initial presentation, the path to genetic testing, and patient outcome. Chromosomal duplication was found on genetic testing performed for failure to thrive and inability to meet her developmental milestones. The patient was significantly under
