The effects of piracetam on heroin-induced CPP and neuronal apoptosis in rats.

BACKGROUND: Piracetam is a positive allosteric modulator of the AMPA receptor that has been used in the treatment of cognitive disorders for decades. Recent surveys and drug analyses have demonstrated that a heroin mixture adulterated with piracetam has spread rapidly in heroin addicts in China, but its addictive properties and the damage it causes to the central neural system are currently unknown. METHODS: The effect of piracetam on the reward properties of heroin was assessed by conditioned place preference (CPP). Electron microscopy and radioimmunoassay were used to compare the effects of

Voluntary Alcohol Intake following Blast Exposure in a Rat Model of Mild Traumatic Brain Injury.

Alcoholism is a frequent comorbidity following mild traumatic brain injury (mTBI), even in patients without a previous history of alcohol dependence. Despite this correlational relationship, the extent to which the neurological effects of mTBI contribute to the development of alcoholism is unknown. In this study, we used a rodent blast exposure model to investigate the relationship between mTBI and voluntary alcohol drinking in alcohol naïve rats. We have previously demonstrated in Sprague Dawley rats that blast exposure leads to microstructural abnormalities in the medial prefrontal cortex (m

Assessment of individual differences in the rat nucleus accumbens transcriptome following taste-heroin extended access.

Heroin addiction is a disease of chronic relapse that harms the individual through devaluation of personal responsibilities in favor of finding and using drugs. Only some recreational heroin users devolve into addiction but the basis of these individual differences is not known. We have shown in rats that avoidance of a heroin-paired taste cue reliably identifies individual animals with greater addiction-like behavior for heroin. Here rats received 5min access to a 0.15% saccharin solution followed by the opportunity to self-administer either saline or heroin for 6h. Large Suppressors of the h

Reduced salience and default mode network activity in women with anorexia nervosa.

BACKGROUND: The neurobiology of anorexia nervosa is poorly understood. Neuronal networks contributing to action selection, self-regulation and interoception could contribute to pathologic eating and body perception in people with anorexia nervosa. We tested the hypothesis that the salience network (SN) and default mode network (DMN) would show decreased intrinsic activity in women with anorexia nervosa and those who had recovered from the disease compared to controls. The basal ganglia (BGN) and sensorimotor networks (SMN) were also investigated. METHODS: Between January 2008 and January 2012

How the win-lose balance situation affects subsequent decision-making: functional magnetic resonance imaging evidence from a gambling task.

Humans have been consistently shown to be bad at making decisions, especially in disadvantageous situations. In this study, we designed a task that simulates real-life non-strategic gambling to examine the effect of win-lose balance situations (WIN, LOSS, TIE) on decision-making. In behavioral performances, participants showed shorter response time (RT) in LOSS than in WIN and TIE conditions. Imaging results revealed that decisions in WIN are associated with increased brain activations in the posterior cingulate cortex; decisions in LOSS are associated with increased brain activations in the i

Age-related differences in advantageous decision making are associated with distinct differences in functional community structure.

Human decision making is dependent on not only the function of several brain regions but also their synergistic interaction. The specific function of brain areas within the ventromedial prefrontal cortex has long been studied in an effort to understand choice evaluation and decision making. These data specifically focus on whole-brain functional interconnectivity using the principles of network science. The Iowa Gambling Task (IGT) was the first neuropsychological task used to model real-life decisions in a way that factors reward, punishment, and uncertainty. Clinically, it has been used to d

Effect of environmental enrichment devices on behaviors of single- and group-housed squirrel monkeys (Saimiri sciureus).

Squirrel monkeys display an interest in novel places, habituate to new situations, and spend most of their daily activity in the wild in large groups engaging in feeding behaviors over a broad area. Captivity limits these behaviors and consequently may disrupt normal social organizations. In captivity, squirrel monkeys may exhibit stereotypical behaviors that are believed to indicate decreased psychologic well-being. When a monkey's behavior can be made to approach that seen in the wild, and stereotypical behaviors are minimal, it is assumed that psychologic well-being is adequate. Environment

Correlates of bulimia nervosa: early family mealtime experiences.

The relationship of early mealtime experiences to later bulimia was investigated in a sample of 128 female college students. Significant group differences were found between bulimics, nonbulimics, and repeat dieters on an early meal experience questionnaire, with the bulimic group reporting the most negative and unusual experiences. Significant differences were also found between groups on depression but not on family adaptability or cohesion. These findings suggest that bulimic, repeat dieter, and nonbulimic females have differentiating histories regarding eating-related issues in the family

Sugar consumption in Aboriginal children explored from a social practice perspective.

Australian Aboriginal children have higher sugar intake and are disproportionately affected by sugar-related diseases such as obesity, diabetes and dental caries than non-Aboriginal children. The intention of this paper was to highlight the need for approaches that can capture the complexity of health practices without reducing them to either individual choice or structural determination. We explore sugar consumption in Aboriginal children from a social practice perspective. Here, the unit of analysis is the practice, not the individual, where the elements of a social practice (competence, mat

Semaglutide and Alcohol use Disorder: Breaking Free of Addiction?

Alcohol use disorder (AUD) is a chronically relapsing disorder characterized by a compulsive alcohol drinking pattern, loss of control over intake, and the emergence of negative emotional states promoting alcohol dependence. It is unquestionably a major public health issue worldwide and a highly stigmatized condition. Unfortunately, AUD remains untreated in a significant number of adults. Since the approval of disulfiram by the FDA in 1951, only two medications (naltrexone and acamprosate) have gained subsequent FDA approval for this disorder. However, even when these medications are administe

Task affordances shape hornbills' problem-solving strategies.

Innovative problem-solving in birds has traditionally focused on corvids and parrots, but growing evidence shows that many other bird species are also capable of such feats. The vertical string-pulling paradigm, a hallmark test of avian cognition, requires birds to retrieve suspended food by incrementally pulling the string with their bill while securing it with their foot, demonstrating sensorimotor flexibility and the capacity to perceive some of the task's properties. However, Oriental pied hornbills (Anthracoceros albirostris) present an interesting case due to their distinctive foot morph

Meta-Reinforced-Model-Based Planning and Fault-Tolerant Control for a Saturation Diving Decontamination Decompression Chamber.

Saturation diving is the only viable method that enables divers to withstand prolonged exposure to high-pressure environments, and it is increasingly used in underwater rescue and marine resource development. This study presents the control system design for a specialized saturation diving decontamination decompression chamber. As a multi-compartment structure, the system requires precise inter-cabin pressure differentials to ensure safe decontamination and ventilation control under dynamic conditions, particularly in the presence of potential faults, such as valve offset, actuator malfunction

Cocaine-context memories are transcriptionally encoded in nucleus accumbens Arc ensembles.

Learned associations between the rewarding effects of drugs and the context in which they are experienced are critical for context-induced relapse. While context re-exposure triggers the recall of such drug-related associative memories it is unclear whether this relies on the reactivation of and plasticity in neuronal populations previously engaged in their acquisition. Here, using the immediate early gene Arc, we captured a discrete population of nucleus accumbens (NAc) cells activated during the encoding of cocaine-context memory in mice and showed that this neuronal ensemble is later reacti

Pharmacological Perspective on the Neurobiology of PI3K-Akt-mTOR Signalling in Opioid Dependence.

Opioid addiction is a condition of the central nervous system that occurs as a result of using opiate-based substances, which can be either natural or synthetic chemicals. These have effects identical to those of morphine and work by interacting with opioid receptors such as morphine, heroin, opium, buprenorphine, and Oxycontin. Dopamine has been suggested to play a role in the mechanisms linked to opioid addiction. Additionally, neurotransmitters such as serotonin, norepinephrine, glutamate, and GABA may also have a significant impact. These processes play a critical role in the formation of

Associations between intraindividual reaction time variability, baseline respiratory sinus arrhythmia, and trait impulsivity in males with substance use disorders.

Trait impulsivity, characterized by the tendency to act without adequate consideration of potential consequences in pursuit of immediate rewards, is a recognized vulnerability marker for substance use disorders (SUDs). Cognitive control deficits are implicated in the manifestation of high trait impulsivity, and intraindividual reaction time variability (IIRTV), an indicator of cognitive control, may influence trait impulsivity. Baseline respiratory sinus arrhythmia (RSA) reflects vagal tone, serves as an index of physiological self-regulation, which has been demonstrated to be related to trait
