Mechanism of action of soybean oil in arthropods: mortality results from tracheal infiltration and obstruction of gas exchange structures.

Oils are widely used in arthropod pest control, yet their modes of action remain poorly understood and often differ across studies. To investigate the mechanism of action of a commercial soybean oil formulation, we evaluated its physical and toxicological effects on three arthropod species across several developmental stages: adults and nymphs of Uroleucon sonchi (L.) (Hemiptera: Aphididae), adults and eggs of Tetranychus urticae Koch (Trombidiformes: Tetranychidae), and larvae and eggs of Plutella xylostella (L.) (Lepidoptera: Plutellidae). Adults, immature stages, and eggs were either topica

Larval character evolution in Holometabola (Insecta).

Larvae, a novel type of immature stage, are a crucial innovation of the Holometabola. Larval features therefore provide key evidence for reconstructing evolutionary transformations in this megadiverse group. Here we present the first formal reconstruction of larval character evolution, using maximum parsimony and Bayesian ancestral-state reconstruction. Our analyses are based on an updated matrix of 88 larval characters and a phylogenetic topology assembled from published transcriptomic and phylogenomic studies. A series of key larval transformations is associated with the origin of Holometabo

An Autolysis-Derived Trypsin Peptide Disrupts Digestive Homeostasis and Reduces Fitness in Spodoptera Frugiperda Caterpillars.

The fall armyworm, Spodoptera frugiperda J.E. Smith (Lepidoptera: Noctuidae), is a significant agricultural pest, and its management is increasingly impeded by resistance to conventional insecticides, underscoring the need for alternative control strategies. This research examined the effects of the synthetic peptide GORE3 on digestive protease homeostasis and larval performance using an integrated approach that combined molecular dynamics simulations, enzymatic assays, physiological analyses, and histopathology. Comparative analyses employed benzamidine and soybean Kunitz trypsin inhibitor (S

Diet, predator, and molecular marker properties influence dietary DNA detection in ladybird beetles (Coleoptera: Coccinellidae) during digestion.

DNA-based gut content analysis is a powerful tool for quantifying predation, yet dietary DNA detectability is influenced by multiple factors. Here, we systematically evaluated the effects of diet, predator traits (species, life stage, and sample type), and molecular marker properties (marker length and type) on dietary DNA detectability in ladybird beetles (Coleoptera: Coccinellidae), using Cryptolaemus montrouzieri Mulsant, 1853 as a representative species alongside other ladybird species. Dietary DNA detectability varied markedly among food types. The 50% detection success (DS50) of Ephestia

Maintenance of Gut Microbial Balance via the Kynurenine Pathway Improves Larval Performance and Resistance to Bacillus thuringiensis in Spodoptera exigua.

The gut microbiota is a key determinant of insect physiology, influencing nutrition, immunity, and interactions with plants and pathogens. In Lepidoptera, larval gut communities are dynamic, but a core microbiota, often dominated by Enterococcus species, persists across instars. In Spodoptera littoralis, the enzyme kynurenine 3-monooxygenase (KMO) regulates gut bacterial composition via 8-hydroxyquinoline-2-carboxylic acid (8-HQA), a secreted iron-chelating compound. To investigate whether this mechanism is conserved in Noctuidae, we generated Spodoptera exigua kmo-/- mutants using CRISPR/Cas9

Unveiling the hidden threat: flupyradifurone soluble liquid's toxic impact on silkworms.

Bombyx mori Linnaeus (Lepidoptera: Bombycidae) is an economically significant insect that consistently contributes to the enhancement of the national economy in all countries with predominant sericulture industries. Flupyradifurone, a butenolide insecticide, is recognized for its minimal toxic effects on bees when applied at standard dosages; however, its potential toxicity or threat to B. mori has not been fully elucidated. This study aims to investigate and evaluate the toxicity of flupyradifurone soluble liquid (SL) on B. mori by administering various concentrations to third instar silkworm

Low and high-density polyethylene and expanded polystyrene biodegradation by the greater wax moth Galleria mellonella L reveals a key role of the gut microbiome.

Polyethylene (PE) is a ubiquitous pollutant that takes long time to degrade naturally. PE breaks down into persistent micro- or nano-plastics that are even more dangerous for the environment and human health. Here we investigated the ability of the wax worm Galleria mellonella (Lepidoptera, Pyralidae) to degrade two different types of PE and expanded polystyrene (EPS). Results showed a reduction of up to 69 %, 73 %, and 50 % in the weight of LDPE, HDPE, and EPS, respectively. Antibiotic treatments indicated that PE consumption is highly dependent upon an intact population of at least eight dif

Bioinsecticide control and enzymatic responses in Spodoptera frugiperda.

Spodoptera frugiperda (Lepidoptera: Noctuidae) is a serious invasive pest, which has attracted concern regarding the effectiveness of environmental bioinsecticide as a substitution for synthetic insecticide in controlling its damage to numerous agricultural crops in recent years. Hence, laboratory and field experiments have investigated insecticidal toxicity in S. frugiperda by 5 insecticides and have determined the activity of acetylcholinesterase (AChE) and detoxifying enzymes, namely mixed function oxidase (MFO), carboxylesterase (CarE), and glutathione S-transferase (GST) on 3rd instar lar

Night and day: Contributions of diurnal and nocturnal visitors to pollen dispersal, paternity diversity, and fruit set in an early-blooming shrub, Daphne jezoensis.

PREMISE: Under uncertain pollinator visit conditions, plants often exhibit long flowering periods and generalized pollination systems. Flowering of the gynodioecious shrub Daphne jezoensis occurs in early spring in cool temperate forests. Pollination by nocturnal moths is expected, given the species' tubular-shaped flowers with sweet fragrance and nectar. However, the effectiveness of nocturnal moths under cool conditions is unknown. We evaluated the relative importance of diurnal and nocturnal visitors as pollinators in early spring. METHODS: We investigated flowering duration, flower visito

Drosophila X virus-like particles as delivery carriers for improved oral insecticidal efficacy of scorpion Androctonus australis peptide against the invasive fruit fly, Drosophila suzukii.

Insect-specific neurotoxic peptides derived from the venoms of scorpions and spiders can cause acute paralysis and death when injected into insects, offering a promising insecticidal component for insect pest control. However, effective delivery systems are required to help neurotoxic peptides pass through the gut barrier into the hemolymph, where they can act. Here, we investigated the potential of a novel nanocarrier, Drosophila X virus-like particle (DXV-VLP), for delivering a neurotoxin from the scorpion Androctonus australis Hector (AaIT) against the invasive pest fruit fly, Drosophila su

Application of transcriptomic analysis to unveil the toxicity mechanisms of fall armyworm response after exposure to sublethal chlorantraniliprole.

The anthranilic diamide insecticide chlorantraniliprole is highly effective against Lepidoptera pests, but the underlying mechanisms of toxic effects of chlorantraniliprole exposures for adapting to the chemical environment are poorly known in fall armyworm (FAW), Spodoptera frugiperda (J.E.Smith). FAW being one of the most pests of maize in Latin America, suddenly appeared in China in 2019 and spread rapidly. In this study, using bioassay and transcriptomic and biochemical analyses, we comprehensively investigated gene expression changes of third instar larvae in response to different subleth

The Corn-Cotton Agroecosystem in the Mid-Southern United States: What Insecticidal Event Pyramids Should be Used in Each Crop to Extend Vip3A Durability.

Recent studies suggest that resistance in Helicoverpa zea (Boddie) (Lepidoptera, Noctuidae) to Cry1A(b/c) and Cry2Ab2 toxins from the bacterium Bacillus thuringiensis Berliner (Bacillales: Bacillaceae) has increased and field efficacy is impacted in transgenic corn and cotton expressing these toxins. A third toxin, Vip3A, is available in pyramids expressing two or more Bt toxins in corn hybrids and cotton varieties, but uncertainty exists regarding deployment strategies. During a growing season, H. zea infests corn and cotton, and debate arises over use of Vip3A toxin in corn where H. zea is n

Do anthropogenic transports facilitate stored-product pest moth dispersal? A molecular approach.

Stored-product moths cause large economic damage in food processing industries and storage facilities. Control of indoor pests is currently dealt with locally, and control strategies seldom include different mills or cooperative industries in joint efforts to reduce infestations. In colder climates where conditions hinder flight dispersal of stored-product moths, we hypothesize that human transport between mills will facilitate dispersal. Albeit considered intuitive, this hypothesis has so far never been tested. Male moths from three mills (populations) in southern Sweden and Denmark were coll

Trypsinogen Pro-Region as a Molecular Scaffold for the Design of Competitive Peptide Inhibitors of Trypsin-Like Proteases.

Soybean (Glycine max L. Merrill) productivity is severely impacted by the velvetbean caterpillar Anticarsia gemmatalis Hübner, 1818 (Lepidoptera: Erebidae), a major defoliating pest in soybean-producing regions. Current management strategies rely heavily on chemical insecticides, which are associated with environmental risks and the development of resistance. Protease inhibitors have emerged as a promising alternative, as protein digestion in lepidopteran larvae is primarily mediated by trypsin-like serine proteases. In this context, the rational design of short inhibitory peptides represents

Disruption of Spodoptera exigua serine protease 2 (Ser2) results in male sterility by CRISPR/Cas9 technology.

BACKGROUND: Sperm development and behavior present promising targets for environmentally safer, target-specific biorational control strategies. Serine protease in seminal fluid proteins plays a crucial role in the post-mating reproductive processes of lepidopteran pest insects. The serine protease 2 has been identified as the initiatorin of the seminal fluid protein in Lepidoptera, and its loss of function leads to male sterility. Nevertheless, the genetic pattern of this gene mutation and the impacts of various mutant genotypes on the hatchability of the eggs of pests remain unclear. RESULTS

Effects of iron on the growth, biofilm formation and virulence of Klebsiella pneumoniae causing liver abscess.

BACKGROUND: Klebsiella pneumoniae is considered the most clinically relevant species of Enterobacteriaceae, known to cause severe infections including liver abscesses. To the best of our knowledge, a large proportion of iron in the human body is accumulated and stored in the liver. We hypothesize that increased iron availability is an important factor driving liver abscess formation and we therefore aim to understand the effects of iron on K. pneumoniae causing liver abscesses. RESULTS: All tested K. pneumoniae clinical isolates, including those isolated from liver abscesses and other abdomin

Galleria mellonella as a model system to study virulence potential of mucormycetes and evaluation of antifungal treatment.

Mucorales can cause cutaneous to deep-seated infections, mainly in the immunocompromised host, resulting in high mortality rates due to late and inefficient treatment. In this study, Galleria mellonella larvae were evaluated as a heterologous invertebrate host to study pathogenicity of clinically relevant mucormycetes (Rhizopus spp., Rhizomucor spp., Lichtheimia spp., Mucor spp.). All tested species were able to infect G. mellonella larvae. Virulence potential was species-specific and correlated to clinical relevance. Survival of infected larvae was dependent on (a) the species (growth speed a

Virulence of Candida haemulonii complex in Galleria mellonella and efficacy of classical antifungal drugs: a comparative study with other clinically relevant non-albicans Candida species.

Candida haemulonii complex has emerged as notorious yeasts causing invasive infections with high rates of treatment failures. Since there is a particular interest in the development of non-mammalian host models to study microbial virulence, with the aim to evade the ethical impact of animal tests, herein we compared the virulence of C. haemulonii, C. duobushaemulonii and C. haemulonii var. vulnera with non-albicans Candida species (C. tropicalis, C. krusei and C. lusitaniae) on Galleria mellonella and the efficacy of antifungal drugs. Up-Regulation of Antimicrobial Peptides Gallerimycin and Galiomicin in Galleria mellonella Infected with Candida Yeasts Displaying Different Virulence Traits.

Galleria mellonella has been described as a cheap and an easy-to-reproduce model for the study of fungal infections. We hypothesized that yeasts with higher virulence potential decrease survival and significantly trigger an immune response in G. mellonella through the regulation of innate immunity-related genes encoding antimicrobial peptides (AMPs) such as gallerimycin and galiomicin. Candida albicans SC5314 and Candida dubliniensis CBS 7987, selected because of their different virulence potential, were used for a killing assay followed by the determination of gene expression using qPCR.
