Ultrastructural studies and immunolocalization of enamel proteins in rodent secretory stage ameloblasts processed by various cryofixation methods.

BACKGROUND: Cryofixation rapidly immobilizes cell and tissue components in their native state, thereby resulting in an ultrastructural preservation very close to the living situation. We have applied this approach to examine the morphology of secretory stage ameloblasts and the distribution of enamel proteins in these cells. METHODS: Molar and incisor tooth germs from newborn mice and/or rats were quickly dissected and divided into segments. The segments were then rapidly frozen using slam, plunge or pressure freezing, freeze-substituted and embedded in Epon. In addition, incisors from older

Characteristics of Burkholderia pyrrocinia and the biocontrol mechanism by which it affects lily bulb rot caused by F. oxysporum.

BACKGROUND: Lily bulb rot, which is mainly caused by Fusarium oxysporum (F. oxysporum), is a destructive soil-borne disease that significantly reduces lily yield and quality. Biocontrol strategies are preferable for counteracting the threats associated with pathogens. METHODS: Confrontation assays were used to screen antagonistic rhizosphere bacteria against F. oxysporum and evaluate their antifungal activity. The most effective isolate, Burkholderia pyrrocinia, was further characterized using a multi-omics approach to investigate its modulation of lily defense responses. RESULTS: In this st

Inoculation with Stenotrophomonas maltophilia LIMN and Enterobacter roggenkampii LCMG Enhances Maize (Zea mays L.) Yield and Reduces Nitrogen Fertilizer Dependence in Nutrient-Limited Soils of Semi-Arid Regions.

Excessive nitrogen (N) fertilization in maize (Zea mays L.) production causes environmental degradation, particularly in semi-arid regions where high chemical inputs are necessary to maintain crop production. Plant growth-promoting rhizobacteria (PGPR) could reduce the optimal N fertilization doses. This investigation aimed to isolate bacteria, screen them for plant growth-promoting traits, and evaluate their potential effects on maize grain under different chemical N fertilization doses. Isolates Stenotrophomonas maltophilia LIMN and Enterobacter roggenkampii LCMG demonstrated multifunctional

Transient acute kidney injury following femoral fracture in mice : the role of sympathetic activation.

AIMS: Femoral shaft fractures are commonly associated with high-energy trauma, and affect individuals of all ages. Kidney dysfunction frequently complicates fracture healing, and trauma-related acute kidney injury (TRAKI) elevates the risk of adverse outcomes. The mechanisms of post-fracture kidney injury, particularly the involvement of sympathetic activation, remain poorly understood. We aimed to define the impact of femoral fracture on the kidney and investigate mechanistically the potential contribution of sympathetic activation. METHODS: A total of 46 female C57BL/6J mice were subjected

Selection of indigenous bacterial strains having the ability to promote plant growth and control root-knot nematode Meloidogyne spp. on Malabar spinach in Vietnam.

Malabar spinach (Basella alba L.) is a widely consumed leafy vegetable in Vietnamese daily meals. It has a short growth cycle but is susceptible to various diseases, of which the nematode Meloidogyne spp. is a significant factor causing yield reduction. This study focused on finding native plant growth-promoting rhizobacteria that can control root-knot nematodes and stimulate plant growth. The results isolated and selected Bacillus velezensis BHMT4.1, Staphylococcus carnosus CCMT2.1, and Pseudomonas fluorescens HMMT1.1, which showed promise in controlling Meloidogyne spp. in Malabar spinach an

Microbiome matters: how transplantation methods and donor origins shape the successful restoration of the seagrass Posidonia oceanica.

BACKGROUND: Posidonia oceanica forms extensive seagrass meadows in the Mediterranean Sea, providing key ecosystem services. However, these meadows decline due to anthropogenic pressures like anchoring and coastal development. Transplantation-based restoration has been explored for decades, yet the role of the plant-associated microbiome in restoration success remains largely unknown. RESULTS: 16 S rRNA gene amplicon sequencing was used to investigate how different transplantation methods and donor origins influence the bacterial communities of P. oceanica cuttings two years post-transplantati

Biocontrol Potential, Plant Growth-Promotion, and Genomic Insights of Pseudomonas koreensis CHHM-1 Against Bacterial Canker in Actinidia arguta.

In 2019, bacterial canker caused by Pseudomonas syringae pv. actinidiae was first identified in Actinidia arguta. This disease has led to significant yield reduction, plant mortality, and substantial economic losses in A. arguta cultivation. Its emergence poses a novel challenge to the sustainable global production of kiwifruit. Currently available treatments for bacterial canker caused by P. syringae pv. actinidiae are scarce. Moreover, the environmental toxicity of copper-based compounds and emerging antibiotic resistance issues necessitate the development of eco-friendly control strategies.

Enhancement of Tomato Growth Through Rhizobacteria and Biocontrol of Associated Diseases.

The purpose of this study was to investigate the growth-promoting effects of four rhizobacterial isolates (RS60, RS65, RS46, and RP6) isolated from the tomato rhizosphere. These isolates were screened for key plant growth-promoting rhizobacteria (PGPR) mechanisms, including ammonia production, nitrogen fixation, phosphate solubilization, indole-3-acetic acid (IAA) production, and siderophore synthesis. Their potential to enhance seed germination and tomato plant growth was investigated in controlled and greenhouse conditions. Four isolates exhibited multiple PGPR attributes, notably IAA and am

Unraveling the functional characteristics of endophytic bacterial diversity for plant growth promotion and enhanced secondary metabolite production in Pelargonium graveolens.

The rich diversity of microbial endophytic communities associated with plants, often referred to as the second genome, serves as a compelling illustration of efficient co-evolution. This noteworthy partnership plays a pivotal role in sustaining plant well-being and enhancing plant adaptability across diverse habitats. Therefore, examining the diversity of endophytic microbes associated with their particular host plant is valuable for gaining insights into the vast spectrum of plant-microbe interactions. The present experiments aimed at investigating the bacterial endophytic diversity in both r

Comparative Proteomic Analysis Reveals the Regulatory Effects of H2S on Salt Tolerance of Mangrove Plant Kandelia obovata.

As a dominant mangrove species, Kandelia obovata is distributed in an intertidal marsh with an active H2S release. Whether H2S participates in the salt tolerance of mangrove plants is still ambiguous, although increasing evidence has demonstrated that H2S functions in plant responses to multiple abiotic stresses. In this study, NaHS was used as an H2S donor to investigate the regulatory mechanism of H2S on the salt tolerance of K. obovata seedlings by using a combined physiological and proteomic analysis. The results showed that the reduction in photosynthesis (Pn) caused by 400 mM of NaCl was

Soil microbial diversity, stability, and function are enhanced by cover cropping: A machine learning-based pooled analysis of Mississippi agroecosystems.

Cover cropping has emerged as a pivotal strategy to enhance soil health and microbiome functionality across diverse agroecosystems. However, the extent to which cover crops reshape microbial diversity, composition, functional capacity, and ecological stability remains insufficiently understood, particularly within heterogenous environments like Mississippi. In this study, we conducted a comprehensive pooled analysis of 473 soil samples collected from multiple independent cover crop trials (2020-2024) across diverse cropping systems and edaphic contexts in Mississippi. Amplicon sequencing (16S

Continuous Cropping Is Associated with Shifts in Strawberry Root-Associated Bacterial Communities and Predicted Carbon-Nitrogen Functional Profiles.

Long-term cultivation of strawberries can alter plant-associated microbial communities; however, the responses of root-associated bacteria within the combined rhizoplane and endophytic community remain insufficiently characterized. This study compared bacterial community composition, co-occurrence patterns, and predicted ecological functions in root-associated bacteria from short-term (4 months; 4 mon) and long-term (16 months; 16 mon) cultivation of Dandong 99 strawberry plants. Because the two groups differed simultaneously in soil cultivation history, plant age, and root developmental stage

Insights into remediation effects and bacterial diversity of different remediation measures in rare earth mine soil with SO4 2- and heavy metals.

The increased demand for rare earth resources has led to an increase in the development of rare earth mines (REMs). However, the production of high-concentration leaching agents (SO4 2-) and heavy metals as a result of rare earth mining has increased, necessitating the removal of contaminants. Here, a series of experiments with different remediation measures, including control (CK), sulfate-reducing bacteria (SRB) alone (M), chemicals (Ca(OH)2, 1.5 g/kg) plus SRB (CM-L), chemicals (Ca(OH)2, 3.0 g/kg) plus SRB (CM-M), and chemicals (Ca(OH)2, 4.5 g/kg) plus SRB (CM-H), were conducted to investig

Excessive nitrogen addition accelerates N assimilation and P utilization by enhancing organic carbon decomposition in a Tibetan alpine steppe.

High amounts of deposited nitrogen (N) dramatically influence the stability and functions of alpine ecosystems by changing soil microbial community functions, but the mechanism is still unclear. To investigate the impacts of increased N deposition on microbial community functions, a 2-year multilevel N addition (0, 10, 20, 40, 80 and 160 kg N ha-1 year-1) field experiment was set up in an alpine steppe on the Tibetan Plateau. Soil microbial functional genes (GeoChip 4.6), together with soil enzyme activity, soil organic compounds and environmental variables, were used to explore the response o

Global analysis of gene expression in response to L-Cysteine deprivation in the anaerobic protozoan parasite Entamoeba histolytica.

BACKGROUND: Entamoeba histolytica, an enteric protozoan parasite, causes amebic colitis and extra intestinal abscesses in millions of inhabitants of endemic areas. E. histolytica completely lacks glutathione metabolism but possesses L-cysteine as the principle low molecular weight thiol. L-Cysteine is essential for the structure, stability, and various protein functions, including catalysis, electron transfer, redox regulation, nitrogen fixation, and sensing for regulatory processes. Recently, we demonstrated that in E. histolytica, L-cysteine regulates various metabolic pathways including ene

Mesorhizobium bavaricum sp. nov. and Mesorhizobium monacense sp. nov., two novel Lotus-associated species harbouring symbiotic plasmids.

Legumes establish a mutualistic interaction with nitrogen-fixing rhizobia. Lotus japonicus is a model for studying this symbiosis; however, only a limited number of rhizobial species nodulating this host have been taxonomically described. Here, we characterise four Mesorhizobium strains (DC-1.1T, Qj1B1, DC-1.5T, and Qj2B2) isolated from root nodules of Lotus japonicus and Lotus burttii. Multi-locus phylogeny and phylogenomic analyses resolved these isolates into two well-supported monophyletic clades. Genome-based comparisons supported their classification as distinct taxa, with strains DC-1.1
