Effects of microgravity on osteoblast growth.

Studies from space flights over the past two decades have demonstrated that basic physiological changes occur in humans during space flight. These changes include cephalic fluid shifts, loss of fluid and electrolytes, loss of muscle mass, space motion sickness, anemia, reduced immune response, and loss of calcium and mineralized bone. The cause of most of these manifestations is not known and until recently, the general approach was to investigate general systemic changes, not basic cellular responses to microgravity. Recently analyzed data from the 1973-1974 Skylabs disclose that there is a r

[Qualification of human body reactions to microgravity].

The purpose of this paper is to qualify developments in the human body during long-term exposure to microgravity in the light of current physiological and pathophysiological postulates. The work is grounded on the analysis of the data about human body reactions to extended stay in microgravity and adopted from the literature. The syndrome of space motion sickness (SMS) evolving at the onset of space flight can be qualified as a temporary reaction to inadequate ambient conditions. In long-term microgravity after mitigation of SMS by dint of appropriate countermeasures, no subjective disturbing

Comparison of gradual and rapid onset runs in a short-arm centrifugation.

A gradual onset run (GOR) in a short-arm centrifugation was performed on ten healthy students. The centrifuge had a 1.8 m radius, and the subjects sat on a chair in a cabin. The Gz force increased to 2.2 Gz at 0.1 degree/sec2 for 32 min. and the same Gz-level was maintained for 20 min. Three out of ten subjects completed the whole protocol; the load on the others was terminated because of symptoms or increased heart rate. There were few symptoms such as vertigo, that was a common problem with a rapid onset run (ROR) in former experiments, due to the short-arm centrifugation. The changes of the

Effects of artificial gravity on the vascular system during 60 days of strict head-down tilt bedrest.

During spaceflight, the lack of gravity-induced hydrostatic pressure gradients in the body results in a headward fluid shift hypothesized to play a role in pathophysiological changes in the cardiovascular, ocular, and central nervous systems. Here, 24 subjects (n = 8 control) underwent 60 days of strict 6° head-down tilt (HDT) bedrest, an analog of spaceflight where we tested the ability of daily 30-min artificial gravity (AG) exposure via short-arm human centrifugation (intermittent AG, n = 8, and continuous AG, n = 8) to reverse the HDT-induced headward fluid shift. Right internal jugular ve

Relevance analysis of N-glycan variations in C2C12 cells and mouse serum under simulated microgravity using a quaternary phosphonium hydrazide labeling strategy-based mass spectrometry quantitation approach.

Mass spectrometry combined with stable isotope labeling is a powerful technique for detecting disease-related changes in glycosylation patterns and identifying potential biomarkers. However, stable isotope labeling reagents that simultaneously offer high sensitivity, low cost, and stable sialic acid modifications remain scarce. In this study, we developed a convenient and cost-effective microwave-assisted method for synthesizing a stable isotopic quaternary phosphonium hydrazide labeling reagent pair, 14N/15N-P4HZD, for the quantitation difference analysis of N-glycans using HPLC-ESI-HRMS with

Integrative cardiovascular dose-response to graded lower-body negative pressure.

Lower-body negative pressure (LBNP) has been posited as a potential spaceflight countermeasure to counteract the physiological deconditioning related to fluid shifts in microgravity. However, open questions remain regarding the magnitude of LBNP that should be applied. We systematically characterized the cardiovascular effects of LBNP and quantified the effect size of varied LBNP doses across different parts of the cardiovascular system. Twenty-four subjects (12 male and 12 female) were exposed to graded LBNP, increasing from 0 to -50 mmHg in 10 mmHg increments, in both supine (0°) and 15° hea

Impact of muscle atrophy on bone metabolism and bone strength: implications for muscle-bone crosstalk with aging and disuse.

Bone fractures in older adults are often preceded by a loss of muscle mass and strength. Likewise, bone loss with prolonged bed rest, spinal cord injury, or with exposure to microgravity is also preceded by a rapid loss of muscle mass. Recent studies using animal models in the setting of hindlimb unloading or botulinum toxin (Botox) injection also reveal that muscle loss can induce bone loss. Moreover, muscle-derived factors such as irisin and leptin can inhibit bone loss with unloading, and knockout of catabolic factors in muscle such as the ubiquitin ligase Murf1 or the myokine myostatin can

Disruption of NF-κB1 prevents bone loss caused by mechanical unloading.

Mechanical unloading, such as in a microgravity environment in space or during bed rest (for patients who require prolonged bed rest), leads to a decrease in bone mass because of the suppression of bone formation and the stimulation of bone resorption. To address the challenges presented by a prolonged stay in space and the forthcoming era of a super-aged society, it will be important to prevent the bone loss caused by prolonged mechanical unloading. Nuclear factor κB (NF-κB) transcription factors are activated by mechanical loading and inflammatory cytokines. Our objective was to elucidate th

Histology of the utricle in kinetotically swimming fish: a parabolic aircraft flight study.

OBJECTIVES: Humans taking part in parabolic aircraft flights (PAFs) may suffer from space motion sickness, which is a form of kinetosis. As it has been repeatedly shown that some fish in a given batch also reveal kinetotic behaviour (especially so-called spinning movements and looping responses) during PAFs, and as a result of the homology of the vestibular apparatus of all vertebrates, fish can be used as model systems to investigate the origin of susceptibility to motion sickness. Therefore. we were prompted to examine the utricular maculae, which are responsible for the internalization of g

Carp experiment in space microgravity--a visual-vestibular sensory conflict model.

In the 8-d flight mission of Spacelab-J (STS-47) conducted in 1992, behavior of the dorsal light response (DLR) and EEG activity of the cerebellum were intermittently examined for two carp, normal and otolith-removed. The latter carp had immobilization trouble caused by twisting of the EEG cable on day 2 inflight. The problem continued for the remainder of the experiment. Analyses made on the normal carp provided additional evidence in fish for sensory-motor disorder and readjustment during early phase of microgravity, thus supporting the sensory conflict hypothesis for space motion sickness.

Biomedical results of the Space Shuttle orbital flight test program.

On July 4, 1982, the Space Shuttle Columbia landed at Edwards Air Force Base, CA, thus successfully completing the fourth and last in a series of Orbital Flight Tests (OFT) of the Space Transportation System (STS). The primary goal of medical operations support for the OFT was to assure the health and well-being of flight personnel during all phases of the mission. To this end, crew health status was evaluated preflight, inflight, and postflight. Biomedical flight test requirements were completed in the following areas: physiological adaptation to microgravity, cabin acoustical noise, cabin at

Coping with space motion sickness in Spacelab missions.

A substantial number of persons, around 75%, making their first transition into orbital flight will need to adapt to this unique environment. The two most powerful instruments in the prevention of space motion sickness reside in the selection process and in acquiring adaptation-prelaunch. Today, neither of these means is practical. One logical alternative is to administer preventative medication to all or none. One candidate drug is a high-potency transdermal therapeutic system (TTS)-scopolamine. This is marketed in the nature of a patch that is affixed to the skin behind the ear 12 hr before

Ankle Somatosensation and Lower-Limb Neuromuscular Function on a Lunar Gravity Analogue.

Background/Objectives: The adverse effects of low gravity on human physiology are well documented; however, much of the literature is directed at changes which occur in microgravity (µg: weightlessness) with relatively less documented on changes in hypogravity (<1 g; >µg: gravity less than Earth's but more than microgravity). Somatosensation and neuromuscular control may be of particular importance for astronauts as they prepare for future missions to walk on the Moon. This study aimed to explore the effect of reduced weight bearing (to simulate conditions of hypogravity) on ankle somatosensat

Immersed false vertical room. A new motion sickness model.

We evaluated a new model of motion sickness--an enclosure decorated with visual cues to upright which was immersed either inverted or "front"-wall down, in Johnson Space Center's Weightless Environment Training Facility (WETF) pool. This "WETF False Vertical Room" (WFVR) was tested with 19 male and 3 female SCUBA diver subjects, aged 23 to 57, who alternately set clocks mounted near the room's 8 corners and made exaggerated pitch head movements. We found that (1) the WFVR test runs produced motion sickness symptoms in 56% and 36% of subjects in the room-inverted and room-front-down positions,

Reduced Mechanical Tactile Stimulation Under Space Microgravity Affects Synaptic Signaling and Contributes to Neuromuscular Aging in Caenorhabditis elegans.

Although space travel is becoming more accessible, our understanding of how the space environment and microgravity (μG) affect biology, physiology, and human health remains incomplete. This study examined the effects of μG on synaptic signaling and neuromuscular aging in Caenorhabditis elegans. The D01 cohort, consisting of L4 larvae to young adults raised in μG, exhibited a downregulation of genes linked to synaptic signaling, dopamine response, locomotion, cuticle development, and mitochondrial metabolism. This was accompanied by altered synapse dynamics, reduced motility, and shorter body l

Effect of short-term gravitational changes on the human minor salivary gland stem cell characteristics.

OBJECTIVES: Human minor salivary gland stem cells (huMSGSCs) are promising in regenerative medicine. Their multipotent capabilities enable tissue regeneration and offer treatment potential for various diseases. The effects of hypergravity (HyperG) and microgravity (MicroG) on stemness and therapeutic potential are not well explored. Therefore, this study investigated the effects of short-term HyperG and MicroG exposure on huMSGSC stemness and differentiation potential for treating salivary gland dysfunction. METHODS: huMSGSCs were exposed to 1G, MicroG, and HyperG. Cell morphology, proliferat

Effect of Nightly Lower Body Negative Pressure on Choroid Engorgement in a Model of Spaceflight-Associated Neuro-ocular Syndrome: A Randomized Crossover Trial.

IMPORTANCE: Astronauts returning from long-duration spaceflight experience ocular remodeling related to cephalad fluid shifts induced by microgravity. It is hypothesized that the absence of diurnal reductions in intracranial pressure in microgravity creates a low but persistent pressure gradient at the posterior aspect of the eye, which results in ocular remodeling and space-associated neuro-ocular syndrome (SANS) over many months. OBJECTIVE: To determine whether partial reintroduction of footward fluid shifts during simulated microgravity via lower body negative pressure (LBNP) during sleep
