Accelerated Glofitamab as Urgent Bridging to CAR T-Cell Therapy in High-Risk Relapsed/Refractory Large B-Cell Lymphoma.

Conventional glofitamab step-up dosing reaches the 30-mg target dose approximately 3 weeks after obinutuzumab. This interval may be difficult to accommodate in patients with relapsed or refractory large B-cell lymphoma (R/R LBCL) who require disease control within days before cellular therapy. We retrospectively evaluated an accelerated bridging schedule of obinutuzumab 1000 mg on day 1 followed by glofitamab 2.5 mg on day 4, 10 mg on day 8, and 30 mg on day 11. Forty-one consecutive adults with R/R LBCL were included. All patients had a documented plan to proceed to chimeric antigen receptor

Gradient-accelerated nutation-based fast alignment for transmitter-receiver misalignment in inter-satellite free-space optical communication systems.

Free-space optical (FSO) communication has been widely adopted for inter-satellite links. To address the challenge of transmitter-receiver misalignment in FSO systems, this study proposes a gradient-accelerated nutation algorithm. The proposed method retains the inherent advantages of conventional nutation approaches while leveraging the optical field distribution of the FSO link to significantly enhance convergence speed. Specifically, an analytical relationship between the step size and the optical power gradient is derived, enabling the algorithm to automatically switch between rapid conver

CD73 overexpression in ADSCs accelerates bladder repair by regulating the NFκB/NLRP3/caspase-1 signaling axis in neurogenic bladder rats.

Neurogenic bladder (NB) is a disabling condition lacking effective therapies. This study investigated whether CD73-expressing adipose-derived stem cells (ADSCs) promote bladder repair in a rat model of NB and explored the underlying mechanisms. ADSCs were sorted into CD73⁺ and CD73⁻ populations, and CD73⁺ cells were further modified to generate CD73⁺/ev ADSCs and CD73-overexpressing CD73⁺/⁺ADSCs, while CD73 inhibition was achieved using APCP. Conditioned media were applied to rat bladder smooth muscle cells in vitro, and ADSCs were injected into the bladder wall of rats subjected to bilateral

Synergistic Modulation of Triplet Density and Heavy-Atom Effect Accelerates Reverse Intersystem Crossing for Narrowband Multi-Resonance TADF Emitters.

Combining rapid triplet-to-singlet spin conversion with BT.2020-relevant color purity in a single purely organic emitter remains a major challenge for OLED development. Here, we report a synergistic strategy that couples increased triplet density with a modest heavy-atom effect, in which an energetically matched sulfur-containing fragment is fused into a multi-resonance thermally activated delayed fluorescence (MR-TADF) skeleton to accelerate reverse intersystem crossing (RISC) while preserving narrowband emission. The resulting emitter exhibits pure-green emission at 514 nm with a full width

Bifidobacterium Infantis Promotes FOSL1-Mediated Transcriptional Repression of VDR to Accelerate Autoimmune Pancreatitis Progression Through Activation of Oxidative Stress.

Even though colonization by Bifidobacterium spp. is a prominent feature of the gut microbial community in autoimmune pancreatitis (AIP), the detailed mechanism remains to be explored. The purpose of this study is to determine whether Bifidobacterium infantis (B. infantis) accelerates the progression of AIP through the activation of plasmacytoid dendritic cells (pDCs) and the molecular mechanism. Mild and severe AIP mice were developed. B. infantis was gavaged into mild AIP mice, and transcriptome sequencing was conducted to analyze transcriptomic differences of pDCs in the pancreas after B. in

Thermoneutrality Accelerates a Distinctive Metabolic Dysfunction-Associated Steatohepatitis Phenotype in a Diet-Induced Male Mouse Model With Similarities to Human Disease.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatohepatitis is a prevalent chronic liver disease characterized by steatosis, inflammation, and hepatocyte damage, with or without fibrosis. Robust preclinical models that represent the clinical features of metabolic dysfunction-associated steatohepatitis are critical for drug development. We aimed to examine the effects of 2 widely used Western-type diets and thermoneutral housing on the induction of metabolic dysfunction-associated steatohepatitis and liver fibrosis in mice. METHODS: Male mice were fed either a Gubra-Amylin metabolic dy

Task-guided accelerated cTBS simultaneously treats depression and social dysfunction in patients with major depressive disorder: a randomized clinical trial.

Current antidepressant therapies predominately target symptom remission in major depressive disorder (MDD) but often overlook social dysfunction, which is a critical determinant of functional recovery. Individualized, accelerated continuous theta burst stimulation (cTBS) targeting the right dorsolateral prefrontal cortex (R.DLPFC) may offer a potential therapeutic approach for concurrently improving clinical and psychosocial outcomes in MDD. In this randomized, double-blind, sham-controlled trial, 70 patients with MDD were randomly assigned to receive either active (n = 37) or sham (n = 33) ac

Fine particulate matter accelerates the senescence of bone marrow-conserved stem cells in a type 2 diabetes mellitus mouse model.

Fine particulate matter (PM, diameter < 2.5 μm) is a significant public health concern because it can disrupt the bone marrow (BM) microenvironment through oxidative stress and systemic inflammation. Patients with type 2 diabetes mellitus (T2DM) exhibit heightened vulnerability to environmental stressors such as PM2.5, which exacerbates metabolic dysfunction. However, the effects of PM2.5 on BM resident stem cells in the context of diabetes have not been explored. In this study, we investigated the impact of atmospherically relevant PM2.5 exposure on BM-resident stem cells in T2DM mice. Diabet

CD4 + cell depletion accelerates dendritic cell migration and enhances resident dendritic cell proliferation in tumor-draining lymph nodes.

Depletion of CD4+ cells during an anti-tumor immune response promotes tumor regression and accelerates proliferation of tumor-antigen-specific CD8+ T cells in draining lymph nodes (dLNs). However, the effect of the depletion on inter-organ kinetics of antigen presentation and qualitative changes in dendritic cells (DCs) are not yet understood. Here, we established a novel approach for simultaneous detection of cellular movement and cell cycle phase in vivo by KikGR/Fucci mice and used it to examine migratory and LN resident DC (LNDC) dynamics after CD4+ cell depletion. We found that CD4+ cell

A multifunctional Bletilla Striata polysaccharide-based Bsp/GA-rGO electroactive composite sponge accelerates wound healing by modulating the microenvironment.

Wound healing involves four overlapping stages, yet traditional single-function dressings often fail to address the multiple challenges of repair simultaneously. Although electroactive dressings can regulate cellular behavior, natural alternatives to synthetic conductive polymers are needed. This study aimed to develop a multifunctional composite sponge based on Bletilla striata polysaccharide (Bsp) and gallic acid-reduced graphene oxide (GA-rGO). The sponge was fabricated by a mixing and freeze-drying process, yielding a highly porous structure with adequate mechanical properties. Combining t

A GPU-accelerated brachytherapy planning framework for template-based applications: Evaluated on three-dimensional printed individual templates in cervical cancer.

PURPOSE: 3D-printed individualized templates improve high-dose-rate (HDR) brachytherapy, but planning remains limited by slow optimization and geometry-driven needle design. This study proposes a GPU-accelerated framework with a fast dose engine and adaptive inverse optimizer to enhance efficiency and plan quality in template-based workflows. MATERIALS AND METHODS: The PyTorch-implemented framework was validated against a commercial TPS via single-source tests and full-plan recalculations. Retrospective re-optimization of 13 cervical cancer patients treated with 3D-printed templates evaluated

Electroactive bacteria-driven electron transfer accelerates molecular transformation of tetracycline in iron-carbon coupled wetland microbial fuel cells.

Conventional constructed wetlands (CWs) often suffer from adsorption saturation and incomplete degradation when treating antibiotic wastewater, posing risks of pollutant re-release. An iron-carbon substrate was coupled with a microbial fuel cell in a vertical subsurface flow CW (FeC-MFC) based on the hypothesis that electroactive microbial enrichment and strengthened electron transfer would promote TC transformation and treatment stability under chronic TC stress (800 μg/L). FeC-MFC achieved 99.97% TC removal, reducing residual parent TC to 0.28 μg/L, while maintaining stable COD (86.95%) and

Genomic-epidemiological analysis of 15 million SARS-CoV-2 genomes reveals accelerated fitness gain of JN.1 lineage.

The evolution of SARS-CoV-2 has been driven by successive globally circulating waves, including the Alpha and Delta lineages, early Omicron (BA.1-BA.5), XBB, and the recently dominant JN.1 lineages. Although the marked advantage in fitness of early Omicron over Delta lineages has been recognized, there is a lack of systematic evaluation of SARS-CoV-2 fitness across 2020 to 2025. Here, we analyzed 15.23 million SARS-CoV-2 genomes available through May 2025. The accumulation of mutations in the spike protein of the virus has continued to accelerate over time, whereas the trend slowed in the othe

Harnessing design of experiments and automation significantly improves transient protein production in Chinese hamster ovary cells and accelerates drug discovery.

Chinese Hamster Ovary (CHO) cells produce the majority of approved therapeutic proteins and are the de facto platform for rapid, milligram-scale production via transient expression during early biologics discovery. However, performance is highly sensitive to transfection composition, feed strategy, and execution details; and vendor protocols often underperform without platform-specific optimization. We integrated Design of Experiments (DOE) with liquid-handling automation to develop a rapid, data-driven optimization workflow for CHO transient expression. Key factors and ranges were defined for
