Diaphanospondylodysostosis: six new cases and exclusion of the candidate genes, PAX1 and MEOX1.

We report on six cases from four families with the newly described skeletal disorder diaphanospondylodysostosis (DSD). The characteristic radiographic findings included abnormal ossification of vertebral bodies, posterior rib gaps, missing ribs, and a downward tilt of the pubic rami, but normal long bones. The typical facial features of DSD cases were ocular hypertelorism, a short nose, depressed nasal bridge, and low set ears. Other distinctive findings included a short neck with bell-shaped thorax, and nephroblastomatosis. A history of consanguinity and affected siblings with unaffected pare

TATA-binding protein in neurodegenerative disease.

TATA binding protein (TBP) is a general transcription factor that plays an important role in initiation of transcription. In recent years evidence has emerged implicating TPB in the molecular mechanism of a number of neurodegenerative diseases. Wild type TBP in humans contains a long polyglutamine stretch ranging in size from 29 to 42. It has been found associated with aggregated proteins in several of the polyglutamine disorders. Expansion in the CAA/CAG composite repeat beyond 42 has been shown to cause a cerebellar ataxia, SCA17. The involvement of such an important housekeeping protein in

The incidence of deafness is non-randomly distributed among families segregating for Waardenburg syndrome type 1 (WS1).

Waardenburg syndrome (WS) is caused by autosomal dominant mutations, and is characterised by pigmentary anomalies and various defects of neural crest derived tissues. It accounts for over 2% of congenital deafness. WS shows high variability in expressivity within families and differences in penetrance of clinical traits between families. While mutations in the gene PAX3 seem to be responsible for most, if not all, WS type 1, it is still not clear what accounts for the reduced penetrance of deafness. Stochastic events during development may be the factors that determine whether a person with a

Suppressive Effects of Arriheuk Wheat Sprout Extract on Muscle Atrophy in Dexamethasone-Induced C2C12 Myotubes and a Mouse Model.

Skeletal muscle atrophy refers to the loss of muscle strength and mass due to decreased protein synthesis or increased protein degradation. Various conditions can cause muscle atrophy, including aging, heart disease, chronic illness, obstructive pulmonary disease, kidney failure, diabetes, AIDS, cancer, sepsis, and steroid use. Various natural materials have been studied for the prevention of muscle atrophy. In this study, we found that extracts from the sprouts of purple wheat, Arriheuk, prevented muscle atrophy in vitro and in vivo. Arriheuk wheat sprouts extract inhibited the expression of

Dkk3 dependent transcriptional regulation controls age related skeletal muscle atrophy.

Age-related muscle atrophy (sarcopenia) is the leading cause for disability in aged population, but the underlying molecular mechanisms are poorly understood. Here we identify a novel role for the secreted glycoprotein Dickkopf 3 (Dkk3) in sarcopenia. Forced expression of Dkk3 in muscles in young mice leads to muscle atrophy. Conversely, reducing its expression in old muscles restores both muscle size and function. Dkk3 induces nuclear import of β-catenin and enhances its interaction with FoxO3, which in turn activates the transcription of E3 ubiquitin ligase Fbxo32 and Trim63, driving muscle

Genetic profiles of cervical tumors by high-throughput sequencing for personalized medical care.

Cancer treatment is facing major evolution since the advent of targeted therapies. Building genetic profiles could predict sensitivity or resistance to these therapies and highlight disease-specific abnormalities, supporting personalized patient care. In the context of biomedical research and clinical diagnosis, our laboratory has developed an oncogenic panel comprised of 226 genes and a dedicated bioinformatic pipeline to explore somatic mutations in cervical carcinomas, using high-throughput sequencing. Twenty-nine tumors were sequenced for exons within 226 genes. The automated pipeline used

Dynamic epigenetic regulation of the microRNA-200 family mediates epithelial and mesenchymal transitions in human tumorigenesis.

Epithelial-mesenchymal (EMT) and mesenchymal-epithelial (MET) transitions occur in the development of human tumorigenesis and are part of the natural history of the process to adapt to the changing microenvironment. In this setting, the miR-200 family is recognized as a master regulator of the epithelial phenotype by targeting ZEB1 and ZEB2, two important transcriptional repressors of the cell adherence (E-cadherin) and polarity (CRB3 and LGL2) genes. Recently, the putative DNA methylation associated inactivation of various miR-200 members has been described in cancer. Herein, we show that the

Ribonucleoprotein assembly defects correlate with spinal muscular atrophy severity and preferentially affect a subset of spliceosomal snRNPs.

Spinal muscular atrophy (SMA) is a motor neuron disease caused by reduced levels of the survival motor neuron (SMN) protein. SMN together with Gemins2-8 and unrip proteins form a macromolecular complex that functions in the assembly of small nuclear ribonucleoproteins (snRNPs) of both the major and the minor splicing pathways. It is not known whether the levels of spliceosomal snRNPs are decreased in SMA. Here we analyzed the consequence of SMN deficiency on snRNP metabolism in the spinal cord of mouse models of SMA with differing phenotypic severities. We demonstrate that the expression of a

CcWRKY31 Regulates Postharvest Mandarin Fruit Coloration in a Temperature-Dependent Manner.

Storing mandarin fruit at 25 °C accelerates postharvest coloration of mandarin fruit, while 32 °C inhibits it. From the mandarin fruit stored at these two temperatures, we identified the temperature-responsive transcription factor CcWRKY31 as a key regulator of pigment metabolism. CcWRKY31 localizes to the nucleus and exhibits transcriptional activation activity in tobacco cells but not in yeast. Its transient overexpression in tobacco leaves caused visible yellowing and significantly reduced chlorophyll and its metabolites, with faster yellowing at 25 °C than at 32 °C. In mandarin fruit peel,

Evaluating eye-tracking as a method for deception detection against the polygraph as deployed in practice.

Can credibility assessment be based on methods more reliable than those currently used? Traditional polygraph examinations, although widely employed, have well-documented limitations, motivating the search for alternative, noninvasive approaches. In this study, we developed an eye-tracking-based method to detect concealed memory traces by analyzing patterns of eye movements during a Concealed Information Test. Unlike previous research that examined eye movements in isolation, we directly compared the performance of the eye-tracking method with polygraph measurements within the same participant

Targeting XBP1 in Cancer: A Review of Therapeutic Approaches and Strategies.

X-box binding protein 1 (XBP1) is an essential unfolded protein response (UPR) transcription factor that has important roles in cancer biology. Malignant XBP1 signaling promotes tumor survival, drug resistance, and immune evasion and thus represents a potential therapeutic target and biomarker. A structured literature search was conducted using PubMed, Embase, Springer, Elsevier, ISI Web of Knowledge, and Google Scholar. The studies that had investigated the expression, role, and therapeutic targeting of XBP1 in various cancers were identified and critically assessed. XBP1 overexpression is as

Chemosensitizer Effects of Coencapsulation of Curcumin and Cabazitaxel in Nanostructured Lipid Carriers in Glioblastoma Cells.

Gliomas are the most common primary tumors of the central nervous system and are associated with poor prognosis. Current therapy with Temozolomide shows limited efficacy, highlighting the need for new treatment options. Cabazitaxel (CBZ), a chemotherapeutic agent approved for prostate cancer, has shown efficacy in preclinical glioblastoma models. However, there is a need for strategies to reduce toxicity and enhance delivery. Curcumin (CUR), known for its antitumor, anti-inflammatory, and antioxidant properties, has been investigated as a potential chemosensitizer. Here, we developed and chara

Hansen-guided green recovery of non-extractable hesperidin from lemon by-products with antioxidant and cholinesterase inhibitory potential.

Non-extractable phenolics (NEPs), including high-value flavonoids such as hesperidin, remain trapped in residues after conventional and advanced extraction processes, despite their recognized antioxidant and neuroprotective potential. Their sustainable recovery is key to fully valorizing citrus by-products. For the first time, a Box-Behnken experimental design was applied to optimize the recovery of extractable phenolics (EPPs) from lemon by-products using aqueous-organic pressurized liquid extraction (PLE), followed by the recovery of non-extractable hesperidin from the residual matrix using

Melatonin as a daily rhythm control signal for glucose homeostasis and insulin secretion in horses.

An involvement of melatonin in the regulation of glucose homeostasis and insulin secretion in pancreatic β-cells has been proposed. To evaluate the features of the daily rhythms of plasma melatonin, blood glucose, and insulin in horses, ten clinically healthy Italian Saddle geldings, 9-10 years old with a mean body weight of 490 ± 25 kg, were enrolled. The animals were housed in individual boxes under natural environmental conditions (sunrise 06:10; sunset 18:10; 12-15°C, 77% RH). All horses were cannulated the day before the start of the study for sample collection, which was performed every

Oral Consumption of Cannabidiol During Pregnancy Alters Behavior in Mouse Offspring.

BACKGROUND: Pregnant people use cannabidiol (CBD) to treat nausea, insomnia, anxiety, and pain. However, CBD crosses the placenta and enters the fetal brain, where it can affect several targets important for brain development. While consumption of high doses of CBD during pregnancy has been shown to disrupt offspring neurodevelopment, pain sensitivity, and cognitive behavior in mice, lower doses have not been assessed. METHODOLOGY: We administered 10 mg/kg/day CBD by oral gavage to pregnant C57Bl6 mice from embryonic day 5 through birth. We used the puzzle box, the forced swim test, and Hargr

Tumor-SAM: Segment Anything Model for Semi-automatic Lung Tumor Segmentation in CT.

Lung tumor segmentation in thoracic CT scans is vital for radiomics analysis and treatment assessment, but is hindered by heterogeneous tumor morphology, ambiguous boundaries, and inaccuracy and labor-intensiveness of manual segmentation. Existing methods, including traditional machine learning and deep learning methods, often suffer from over-/under-segmentation or poor robustness. In this study, we propose an improved Segment Anything Model (called Tumor-SAM) for semi-automatic lung tumor segmentation, integrating U-Net for multi-scale feature extraction and a novel ellipse prompt.
