Essential roles of Meltrin beta (ADAM19) in heart development.

Morphogenesis of the heart requires development of the endocardial cushion tissue that gives rise to the membranous septa and valves. Here we show that Meltrin beta/ADAM19, a novel metalloprotease-disintegrin, participates in the development of the endocardial cushion. Mice lacking Meltrin beta exhibit ventricular septal defect (VSD) and immature valves, and most of the animals die soon after birth. During development of the endocardial cushion, epithelial-mesenchymal transformation (EMT) of endocardial epithelial cells generates most of the cushion mesenchymes that constitute the main compone

Anti-retinoic acid monoclonal antibody localizes all-trans-retinoic acid in target cells and blocks normal development in early quail embryo.

Avian cardiovascular development is vitamin A-dependent, and retinoic acid has been suggested to be active in this important developmental event. We report here that a monoclonal antibody against all-trans-retinoic acid blocks normal embryonic development in the quail causing cardiovascular abnormalities typical of avian vitamin A deficiency. In whole-mount preparations of stage 5 normal quail embryos the fluorescence associated with the antiretinoic acid monoclonal antibody localizes in Hensen's node and in caudal area. In stage 7-8 embryos fluorescence localizes in heart-forming areas as wel

Retinoic acid and craniofacial development: molecules and morphogenesis.

Retinoic acid (RA), a derivative of vitamin A, is essential for normal mammalian development. Developmental abnormalities induced by RA excess and vitamin A deficiency are different even though they affect the same organ systems, and it is clear that there are intraembryonic tissue differences in the requirement for RA. The developmental functions of RA are mediated by its effects on gene expression. In the nucleus, two different forms of RA bind to and activate two families of nuclear receptors, which themselves co-operate in initiating the transcription of target genes. In this article I pro

Retinoic acid causes an anteroposterior transformation in the developing central nervous system.

All-trans retinoic acid (RA) is well known as a biologically active form of vitamin A and a teratogen. The identification of nuclear receptors for this ligand suggests strongly that it is an endogenous signal molecule, and measurements of RA and teratogenic manipulations suggest further that RA is a morphogen specifying the anteroposterior axis during limb development. Besides the limb, RA and other retinoids affect development of other organs, including the central nervous system (CNS). None of these other effects has been investigated in detail. Our purpose here was to begin analysing the ef

Biallelic expression of the H19 and IGF2 genes in hepatocellular carcinoma.

The imprinted genes, H19 and insulin-like growth factor II (IGF2), have been demonstrated to be necessary for embryonal development in humans. Both genes are reciprocally imprinted, with expression of the maternal H19 and paternal IGF2 alleles, and are normally characterized by monoallelic expression. Recently, loss of imprinting of these genes producing biallelic expression has been observed in childhood tumors including Wilms' tumors (WT), embryonal rhabdomyosarcoma, and adulthood tumors such as lung cancer. To test the existence of loss of imprinting in hepatocellular carcinoma (HCC), we an

Effect of prenatal ethanol exposure on fetal calcium metabolism.

Alcohol consumption has adverse effects on both adult and developing bone. The mechanisms by which alcohol affects bone, however, are unknown. This study examined the possibility that maternal alcohol consumption may affect fetal bone development by altering fetal levels of parathyroid hormone (PTH), 1,25(OH)2D, or calcitonin (hormones that regulate calcium (Ca) and bone metabolism in the adult animal). Female Sprague-Dawley rats were bred and divided into three groups: 1 group was fed lab chow ad libitum (Control; C) and the other 2 groups received a liquid diet with (Ethanol; E) or without (

Research progress on RNA m6A methylation modification mediating placental disorders induced by maternal stress†.

Maternal stress caused by the environmental factors varied in intrauterine and extrauterine during pregnancy may significantly affect placental and fetal development, as well as offspring health in adulthood. Epigenetic mechanisms are frequently invoked to elucidate these effects. RNA N6-methyladenosine (m6A) modification, one of the most prevalent and abundant post-transcriptional epigenetic modifications in eukaryotic mRNA, has recently garnered widespread attention in life sciences. RNA m6A modification plays critical roles in RNA splicing, translation, localization, stability, and has been

Arginine methyltransferase PRMT1 equipoises trophoblast development to prevent early pregnancy loss.

Idiopathic recurrent pregnancy loss (RPL) affects 1-2% of human pregnancies, yet the underlying molecular causes are poorly understood. Here we identify defective Protein Arginine Methyltransferase 1 (PRMT1) function in trophoblast progenitors as a cause of early pregnancy failure. PRMT1 is highly conserved in mammalian trophoblast progenitors, and conditional deletion of Prmt1 in mouse trophoblast progenitors arrests placental and embryonic development, resulting in lethality at ~E7.5. In humans, a subset of idiopathic RPL cases exhibits loss of PRMT1 in cytotrophoblast progenitors (CTBs). Hu

Development-related gene expression disruption in the human midcingulate cortex in Huntington's disease.

Huntington's disease (HD) patients with anterior cingulate cortex atrophy typically exhibit mood symptomatology. However, the midcingulate cortex's (MCC) role in HD is poorly understood. mRNA sequencing was utilized to examine the MCC transcriptome in HD, and differentially expressed transcripts were validated by NanoString analysis. Transcriptomic analysis of 14 HD patients exhibiting mood, motor, or mixed symptoms revealed differential expression of 223 genes, including several homeo-domain, developmental, and noncoding genes in the MCC. Protein-protein interaction, gene ontology, and cell-t

Elucidating cardiomyocyte state transitions in zebrafish heart development using transcriptome dynamics.

Congenital heart disease (CHD), the most prevalent congenital anomaly, remains genetically incompletely understood. To address this knowledge gap, we employed a zebrafish model to investigate transcriptional dynamics during cardiac morphogenesis. Bulk RNA sequencing (Bulk RNA-seq) analysis of embryonic hearts at 24, 30, 36, and 48 h post-fertilization (hpf) linked the gene expression dynamics to four morphogenetic events: heart tube formation, chamber formation, heart tube looping, and valve formation. Weighted gene co-expression network analysis identified the M5 module, enriched for cardiac

Primitive Hepatoblasts Driving Early Liver Development.

The embryonic development of the liver is initiated by the emergence of hepatoblasts, originating from the ventral foregut endoderm adjacent to the heart. Here, we identify and characterize a previously unrecognized population of early hepatoblasts at the ventroposterior part of the emerging liver bud, traced from Cdx2-positive endoderm progenitors, which we term primitive hepatoblasts. Mouse and human single-cell transcriptomics reveals the expression of both canonical hepatoblast markers TBX3, FGB, and KRT8/18 and primitive-specific mesenchymal markers ID3, VIM, and GATA4. Overexpression of Kdm6b induces testicular differentiation in a temperature-dependent sex determination system.

In reptiles, such as the red-eared slider turtle ( Trachemys scripta elegans), gonadal sex determination is highly dependent on the environmental temperature during embryonic stages. This complex process, which leads to differentiation into either testes or ovaries, is governed by the finely tuned expression of upstream genes, notably the testis-promoting gene Dmrt1 and the ovary-promoting gene Foxl2. Recent studies have identified epigenetic regulation as a crucial factor in testis development, with the H3K27me3 demethylase KDM6B being essential for Dmrt1 expression in T. s. elegans. However,

Towards a "Testis in a Dish": Generation of Mouse Testicular Organoids that Recapitulate Testis Structure and Expression Profiles.

The testis is responsible for sperm production and androgen synthesis. Abnormalities in testis development and function lead to disorders of sex development and male infertility. Currently, no in vitro system exists for modelling the testis. Here, we generated testis organoids from neonatal mouse primary testicular cells using transwell inserts and show that these organoids generate tubule-like structures and cellular organization resembling that of the in vivo testis. Gene expression analysis of organoids demonstrates a profile that recapitulates that observed in in vivo testis. Embryonic tes

Togaram1 is expressed in the neural tube and its absence causes neural tube closure defects.

Neural tube closure defect pathomechanisms in human embryonic development are poorly understood. Here we identified spina bifida patients expressing novel variants of the TOGARAM gene family. TOGARAM1 has been associated with the ciliopathy Joubert syndrome, but its connection to spina bifida and role in neural development is unknown. We show that Togaram1 is expressed in the neural tube and Togaram1 knockout mice have abnormal cilia, reduced sonic hedgehog (Shh) signaling, abnormal neural tube patterning, and display neural tube closure defects. Neural stem cells from Togaram1 knockout embryo

Development of the foregut and the formation of the trachea and esophagus in rat embryos. A symphony of confusion.

Introduction: During embryonic development, the trachea emerges from an area of the foregut, which is often referred to as "anterior" or "common" foregut tube or simply foregut. To explain this process of differentiation, four competing models exist to date. The outgrowth and watershed models propose a foregut that remains constant in length. In the outgrowth model, the trachea buds off and elongates from the foregut, while in the watershed model, a mesenchymal wedge splits the growing foregut into the trachea and esophagus. In contrast, the septation model proposes a cranial splitting and thu

Ontogeny of the American alligator (Alligator mississippiensis) prenatal head: A morphometric approach.

Variation in vertebrate cranial morphology is both extensive and functionally significant. Morphometric analysis attempts to explain such variation in form in evolutionary and functional contexts. Developmental morphometric analyses of vertebrate crania are less common, and many taxa are underrepresented. For example, the published morphometric studies of crocodilian cranial development focus mainly on posthatchling head development, with few, incomplete morphometric analyses of prehatchling heads. To further explore ontogenetic changes in the alligator head, we recorded and analyzed six linea

Lymphatic endothelial-cell expressed ACKR3 is dispensable for postnatal lymphangiogenesis and lymphatic drainage function in mice.

Atypical chemokine receptor ACKR3 (formerly CXCR7) is a scavenging receptor that has recently been implicated in murine lymphatic development. Specifically, ACKR3-deficiency was shown to result in lymphatic hyperplasia and lymphedema, in addition to cardiac hyperplasia and cardiac valve defects leading to embryonic lethality. The lymphatic phenotype was attributed to a lymphatic endothelial cell (LEC)-intrinsic scavenging function of ACKR3 for the vascular peptide hormone adrenomedullin (AM), which is also important during postnatal lymphangiogenesis. In this study, we investigated the express

Identification of CXCL11 as part of chemokine network controlling skeletal muscle development.

The chemokine, CXCL12, and its receptors, CXCR4 and CXCR7, play pivotal roles during development and maintenance of limb muscles. CXCR7 additionally binds CXCL11, which uses CXCR3 as its prime receptor. Based on this cross-talk, we investigate whether CXCL11 would likewise affect development and/or function of skeletal muscles. Western blotting and immunolabelling demonstrated the developmentally restricted expression of CXCL11 in rat limb muscles, which was contrasted by the continuous expression of its receptors in proliferating and differentiating C2C12 cells as well as in late embryonic to
