Integrated spatially resolved metabolomics and network toxicology to investigate the hepatotoxicity mechanisms of component D of Polygonum multiflorum Thunb.

ETHNOPHARMACOLOGICAL RELEVANCE: The liver toxicity of Reynoutria multiflora (Thunb.) Moldenke. (Polygonaceae) (Polygonum multiflorum Thunb, PM) has always attracted much attention, but the related toxicity materials and mechanisms have not been elucidated due to multi-component and multi-target characteristics. In previous hepatotoxicity screening, different components of PM were first evaluated and the hepatotoxicity of component D [95% ethanol (EtOH) elution] in a 70% EtOH extract of PM (PM-D) showed the highest hepatotoxicity. Furthermore, the main components of PM-D were identified and the

Oxidative Stress Potentiates a STAT1-APOL1 Feedforward Loop and Governs Disease Severity in APOL1-Mediated Kidney Disease.

INTRODUCTION: Apolipoprotein L1 (APOL1) risk variants (G1 and G2) confer immunity against Trypanosoma infection but markedly increase susceptibility to progressive kidney disease. However, only a subset of biallelic risk-variant carriers develops disease progression, distinct from murine models. Here, we evaluated molecular drivers of progressive kidney disease using a model that reflected the low human disease penetrance. METHODS: To investigate the heterogeneity of APOL1-mediated kidney disease, we generated novel APOL1 bacterial artificial chromosome transgenic mice with doxycycline-induci

PFAS contamination in the Western Seto Inland Sea of Japan: Substitution lag and TFA risk alert.

The western Seto Inland Sea (WSIS) is surrounded by major industrial zones in Japan and receives substantial inputs of pollutants. However, per- and polyfluoroalkyl substances (PFAS) contamination in the WSIS remains poorly characterized. In this study, we investigated the occurrence and potential sources of PFAS in seawater samples from the WSIS, and conducted a preliminary ecological risk assessment. A total of 24 PFAS were identified and quantified, with total concentrations (Σ24PFAS) ranging from 519 to 916 ng/L. Trifluoroacetic acid (TFA) was the predominant compound (mean: 680 ± 156 ng/L

MTDANet: lightweight multi-scale temporal hierarchy and distance-regularized linear attention network for motor imagery brain-computer interfaces.

OBJECTIVE: Motor-imagery (MI) electroencephalography (EEG) offers a non-invasive route to brain-computer interaction, yet reliable single-trial decoding remains difficult because task-related patterns are weak, nonstationary, and distributed over multiple temporal scales. This study investigates whether a staged local-to-global model can integrate fine temporal-spatial structure with longer-range context without relying on unrestricted self-attention. APPROACH: We propose MTDANet, an architecture that combines multi-scale temporal-spatial feature extraction, factorized temporal consolidation,

Spatial and Seasonal Dynamics of Microbial and Phytoplankton Communities and Factors Influencing the Surface Water and Sediments of the Ponjavica River in the Ponjavica Nature Park.

Microorganisms are key bioindicators of changes in biogeochemical cycles and the ecological status of aquatic ecosystems. In this paper, seasonal changes in culturable microbial groups, phytoplankton, and physicochemical water characteristics were examined as integrated indicators of environmental status in the Ponjavica River (Ponjavica Nature Park, Serbia). Sampling was conducted at six sites (P2-P7) in 2021, with two campaigns carried out in spring (May) and autumn (November). At all sites, the number of microorganisms was higher in sediment than in water, confirming the importance of sedim

Virtual Reality in Medical Education: A Concept Analysis.

Virtual reality (VR) remains inconsistently defined in medical education, with the term being applied to heterogeneous immersive and nonimmersive technologies. This inconsistency may hinder intervention classification, evidence synthesis, curriculum planning, and technology selection. This study aimed to analyze and clarify the concept of VR in medical education. A concept analysis was conducted using the eight stages of Walker and Avant's framework. A scoping review following the Joanna Briggs Institute methodology provided the original documentary basis. A complementary concept-directed, non

Automated CT-based quantification of knee osteophytes for robotic-assisted total knee arthroplasty: a multi-cohort study.

Preoperative computed tomography (CT) is routinely acquired for planning robotic-assisted total knee arthroplasty (TKA), yet osteophyte burden is not quantitatively characterized. We developed automated CT-based osteophyte quantification and evaluated it against blinded de novo expert-consensus annotations and an intraoperative accessible resected osteophyte volume reference. A human-in-the-loop 3-dimensional U-Net was developed using 387 knee CT scans. The frozen model was evaluated against blinded de novo expert-consensus annotations in two independent external cohorts (100 knees each). Spat

Employing Psychological Distance to Explain Perspective-Specific EQ-5D-Y-3L Health State Valuation: A Bayesian Hierarchical Analysis.

BACKGROUND: The EQ-5D-Y-3L is a widely used instrument for valuing health states in pediatric populations. Previous studies have shown that composite time tradeoff (cTTO) utilities tend to be higher when adults value health states from a 10-year-old child's perspective than from their own. In this study, we applied construal level theory (CLT) to examine whether variations in psychological distance can explain these differences. METHODS: We conducted online cTTO interviews with 150 adults in the United Kingdom, valuing 4 health states using perspectives that experimentally combine high and lo

DCP-TS: A Unified Spatiotemporal Framework for Real-Time Desmoking and Flicker Suppression in Laparoscopic Surgical Videos.

Surgical smoke generated by energy-based instruments during minimally invasive surgery severely degrades intraoperative visibility in laparoscopic procedures, prolonging operation time and elevating surgical risk. Although deep-learning desmoking methods have improved spatial clarity, most operate frame-by-frame and produce temporal artifacts-flicker, brightness drift, and color instability-that hinder clinical adoption. To our knowledge, no prior framework has jointly addressed spatial restoration and temporal consistency within a unified surgical smoke removal pipeline. We proposed DCP-TS, a

VesselVision-Net: Hybrid Global-Local Context Learning for Liver Vessel Segmentation in Hepatic Surgery Preplanning.

Accurate segmentation of liver vessels on contrast-enhanced CT is essential for safe hepatic surgery preplanning. It is still challenging because vessels vary greatly in size and have complex branching, and small branches are difficult to detect due to their low contrast. In this work, we propose VesselVision-Net, a hybrid global-local context learning framework for liver vessel segmentation, which focuses on detecting small vessel branches. The model uses a dilated fusion module (DFM) to capture better small vessel branches and a paired attention module (PAM) to enhance vessel-focused spatial

High-resolution mapping of chromatin conformation in HBeAg-treated macrophage provides insights into pathogenesis of HBV-related liver diseases.

BACKGROUND: Hepatitis B virus (HBV) e antigen (HBeAg) plays a critical role in inducing macrophage activation and subsequent immune tolerance, which facilitates viral immune escape. However, the underlying spatial 3D genomic and epigenetic mechanisms driving this macrophage dysfunction remain largely unknown. METHODS: To map the topological and transcriptional regulatory landscape, we integrated RNA-sequencing (RNA-seq), high-throughput chromosome conformation capture (Hi-C), and chromatin immunoprecipitation-sequencing (ChIP-seq) to comprehensively analyze control and HBeAg-stimulated macrop

Molecular subtyping and prognostic modeling of colon adenocarcinoma based on programmed cell death features: a multi-omics and machine learning study.

BACKGROUND: Programmed cell death (PCD) plays a complex and critical role in the progression of colon adenocarcinoma (COAD). Elucidating PCD-related characteristics is expected to provide new insights for tumor subtyping, prognosis assessment, and personalized therapy. METHODS: This study integrated multi-omics data from COAD and employed 10 clustering algorithms for molecular subtyping. Based on PCD-related genes, a Programmed cell death signature (PCDS) predictive model was constructed using 113 machine learning algorithms. The focus then shifted to the core gene of the model, TERT. Single-

Integrated Time- and Frequency-Domain Reflectometry for Non-Invasive Moisture Monitoring in Historic Masonry Materials.

Moisture monitoring in historic masonry requires non-destructive solutions that can be adapted to the characteristics and value of the asset, the spatial extent of the phenomenon, and the need for repeated measurements over time. This work presents an integrated reflectometric methodology that combines accurate Frequency-Domain Reflectometry (FDR) characterization with the development and validation of a custom-developed, compact, portable, low-cost, and potentially scalable Time-Domain Reflectometry (TDR) prototype. Resonant patch antennas operating at different frequencies were used with a V

Designing a Territorial Composite Vulnerability Index to guide public health action in Cali, Colombia.

BACKGROUND: Social determinants of health are central to explaining health inequalities. In fragmented urban contexts such as Cali, Colombia, territorially focused public health action requires analytical tools to identify priority areas for intervention. OBJECTIVE: To design and apply a Territorial Composite Vulnerability Index for the city of Santiago de Cali to support the micro-planning of public health interventions. METHODS: We conducted an ecological, citywide study with a geospatial, multivariate approach. Seven indicators representing social determinants were selected: multidimensio

Neuroprotective Mechanisms of Ficus deltoidea in an Alzheimer's Disease-Like Rat Model: Targeting Tau Hyperphosphorylation Through Glycogen Synthase Kinase-3 Beta and Protein Phosphatase 2A Regulation.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterised by memory loss, neurodegeneration, amyloid plaque accumulation and tau hyperphosphorylation. Dysregulation of glycogen synthase kinase-3β (GSK-3β) and protein phosphatase 2 A (PP2A) plays a pivotal role in tau pathology, contributing to synaptic dysfunction and memory impairment. Current AD medications offer limited palliative care, underscoring the need for multifaceted therapeutic strategies. Ficus deltoidea (FD), a medicinal plant renowned for its antioxidant and anti-inflammatory properties, has demonstrated

Synthesis of scale-dependent plant biodiversity change in climate warming experiments.

Climate warming is one of the most pressing environmental challenges for plant populations, communities, and ecosystems on earth. Warming experiments are an important tool for establishing causal relationships between climate change and plant diversity responses. However, results from such experiments vary in direction and magnitude. We hypothesize that such variation arises from context-dependent factors, such as the magnitude of the temperature manipulation, the study duration, the spatial grain of the analysis, the background climate in which the study was conducted, and the biodiversity me

Short-term intrinsic connectivity changes induced by cognitive exertion in healthy participants.

INTRODUCTION: Changes in brain intrinsic connectivity on the timescale of minutes, as provoked by a cognitive task, have not been well documented. METHODS: A total of two 7.5-min 7 Tesla functional MRI (fMRI) scans (Run 1 and Run 2), separated by 90 s, were acquired for 23 healthy participants during cognitive exertion induced by the Stroop color-word interference task. Independent component analysis (ICA) of the paired Run 1 and Run 2 fMRI acquisitions identified components with distinct spatial and temporal signatures. RESULTS: The spatial extent of the ICA components coincided with hubs o

Same geometry, different dynamics: what uncontrolled manifold (UCM) analysis misses about motor control.

Uncontrolled Manifold (UCM) analysis separates the variability of multi-joint motion into a part that affects the task goal (task-relevant) and a part that does not (task-irrelevant), using the geometric relationship-the task Jacobian-between joint motion and task outcome. The resulting partition has been interpreted as evidence of neural control strategies that stabilize task goals while permitting flexibility in execution. UCM is conventionally reported as time-averaged variance indices, and this time-averaging step discards the temporal structure of movement variability. Using a three-joint
