No alteration in the primary structure of the mineralocorticoid receptor in a family with pseudohypoaldosteronism.

We have studied the molecular structure of the mineralocorticoid receptor (MR) complementary DNA (cDNA) in a kindred affected by pseudohypoaldosteronism (PHA). In this family, the clinical symptoms included salt wasting and failure to thrive, accompanied by high urinary levels of sodium despite hyponatremia, hyperkalemia and metabolic acidosis, elevation of PRA, and high plasma aldosterone levels. The patients were resistant to mineralocorticoid administration, but their symptoms ameliorated after a period of sodium supplementation, which was discontinued in older patients. Binding studies per

[Epidemiologic markers of Neisseria (author's transl)].

The world-wide increase of gonococcal infections and meningococcal infections should incite to face up all necessary means of checking which can be actually envisaged against these diseases. The major phenomenon regarding gonococcal infections is the appearance, since 1976, of plasmidic resistance by a beta-lactamase secretion. Two different types of strains have been described: one originating from West-Africa, the other one from South-East Asia (that latter presenting a conjugating plasmid to totalizing 24,5 M-daltons involving coding plasmid for beta-lactamase). Among approximately thirty s

Characterization of COVID-19 vaccine clinical trial discussions on the social question-and-answer site Quora.

INTRODUCTION: Throughout the COVID-19 pandemic, underserved populations, such as racial and ethnic minorities, were disproportionately impacted by illness, hospitalization, and death. Equity in clinical trials means that the participants in clinical trials represent the people who are most likely to have the health condition and need the treatment that the trial is testing. Infodemiology approaches examining user conversations on social media platforms have the potential to elucidate specific barriers and challenges related to clinical trial participation, including among racial and ethnic min

'It's a can of worms': understanding primary care practitioners' behaviours in relation to HPV using the Theoretical Domains Framework.

BACKGROUND: The relationship between infection with high-risk human papillomavirus (HPV) and cervical cancer is transforming cervical cancer prevention. HPV tests and vaccinations have recently become available. In Ireland, as elsewhere, primary care practitioners play a key role in prevention. ATHENS (A Trial of HPV Education and Support) aims to develop a theory-based intervention to support primary care practitioners in their HPV-related practice. This study, the first step in the intervention development process, aimed to: identify HPV-related clinical behaviours that the intervention will

Epigenetic, chromatin-associated, and epitranscriptomic regulation in chronic pain: mechanisms and translational prospects.

Chronic pain is sustained by durable yet dynamic changes in gene regulation across sensory neurons, spinal circuits, glial and immune cells, and higher brain regions. This review critically evaluates three related but conceptually distinct regulatory layers: chromatin-associated epigenetic mechanisms, including DNA methylation and hydroxymethylation, histone modifications, and chromatin accessibility; non-coding RNAs that directly interface with chromatin-regulatory machinery; and RNA N6-methyladenosine (m6A), an epitranscriptomic mechanism that acts primarily at the RNA level. Across pain mod

Comparison of Natural Language Processing and Traditional Qualitative Analyses of Caregiver Feedback From a Community-Based Intervention for Children With Autism.

BACKGROUND: Satisfaction data is a commonly used construct to evaluate programs, products, and experiences. This data is collected through forced choice (scales, surveys) and/or open-ended measures. While open-ended questions have the greatest potential for more useful and candid feedback, analysis of this data can be time-consuming. OBJECTIVE: This paper compares two evaluation methods, traditional qualitative analysis (TQA) and Natural Language Processing (NLP), to analyze themes from open-ended satisfaction data of a caregiver-mediated intervention for families of autistic children, called

From Tumor Suppressor to Tumor Promoter - The Dual Role of LIMCH1 in Cancer and Its Clinical Implications.

LIM and Calponin Homology Domain 1 (LIMCH1) is a multi-domain protein-coding gene located on human chromosome 4p13. Its encoded protein participates in the dynamic regulation of the actin cytoskeleton through an N-terminal calponin homology domain and a C-terminal LIM domain. LIMCH1 specifically binds to non-muscle myosin IIA (NM-IIA). This binding enhances the dual phosphorylation of myosin regulatory light chains. As a result, it increases the adenosine triphosphatase (ATPase) activity and assembly efficiency of NM-IIA, stabilizes stress fibers, promotes focal adhesion maturation, and inhibi

Genome-wide association studies of osteoarthritis: from genetic discovery to precision medicine.

Osteoarthritis (OA) affects more than 500 million people worldwide, yet disease-modifying therapies remain unavailable. Genome-wide association studies (GWAS) have transformed understanding of OA genetic susceptibility, progressing from early single-cohort analyses to multi-ancestry meta-analyses involving 2 million individuals and prioritizing hundreds of loci and 700 effector genes. This review summarizes the development of OA GWAS and examines how genetic discoveries are being translated into biological mechanisms and precision medicine. We discuss how integrating GWAS with fine-mapping, mo

Non-Coding RNA Biomarkers in Prostate Cancer: Evidence Mapping and In Silico Characterization.

Non-coding RNAs (ncRNAs) have emerged as promising biomarkers for prostate cancer (PCa), yet evidence remains dispersed across heterogeneous studies and their regulatory context is seldom analyzed in an integrated manner. This study systematically maps ncRNAs reported as diagnostic biomarkers for PCa and characterizes their molecular interactions through in silico analyses. A comprehensive evidence-mapping strategy across major bibliographic databases identified 693 studies, of which 58 met eligibility criteria. Differentially expressed ncRNAs were extracted and classified by RNA type. Subsequ

Prostate cancer associated 3 (PCA3) is a potential indicator of the pathological grades of prostate cancer.

Prostate cancer (PCa) is a prevalent malignancy of the male reproductive system and ranks as the second most common cancer in men worldwide. The prognosis and clinical management of PCa are closely linked to its pathological grade; however, the high heterogeneity of the disease poses significant challenges in these assessments. Prostate cancer associated 3 (PCA3), an upregulated long non-coding RNA (lncRNA) in PCa, has been established as a urinary biomarker for PCa detection. Nevertheless, its potential role in distinguishing pathological grades remains unclear. In this study, we investigated

Iron homeostasis and ferroptosis: a converging axis in cancer therapy.

Cancer comprises a diverse group of complex diseases driven by genetic and epigenetic alterations that disrupt cellular signaling, metabolism, and cell death mechanisms. Despite significant advances in therapy, challenges such as tumor heterogeneity, treatment resistance, and escape from regulated cell death continue to impede curative outcomes. Among the various modes of regulated cell death, ferroptosis, an iron-dependent mechanism characterized by excessive lipid peroxidation and oxidative stress, has emerged as a promising therapeutic avenue in oncology. Notably, ferroptosis is intricately

Comparative roles between Metastasis Associated Lung Adenocarcinoma Transcript 1 (MALAT1) in lung and colorectal cancer: insights into tumorigenesis and metastasis.

Long non-coding RNA Metastasis Associated Lung Adenocarcinoma Transcript 1 (MALAT1) has emerged as a key regulator in lung and colorectal cancers, two leading causes of cancer-related mortality worldwide. This review adopts a comparative lens to examine MALAT1’s shared and cancer-specific molecular roles and prognostic implications in lung and colorectal malignancies. MALAT1 is consistently overexpressed in both lung and colorectal cancers, correlating with advanced tumor stage, lymph node metastasis, and poor overall survival (OS). MALAT1 is able to promote cancer progression through the PI3K

Muscle transcriptome profiling reveals novel molecular pathways and biomarkers in laminin-α2 deficient patients.

Merosin-deficient congenital muscular dystrophy (LAMA2-RD) is a neuromuscular disorder caused by mutations in the LAMA2 gene, coding for the α2 subunit of laminin-211 (merosin). LAMA2 mutations leading to complete laminin-211 absence result in a severe clinical phenotype with profound muscle weakness and respiratory insufficiency, whereas mutations allowing the production of a partially functional protein are often associated with milder phenotypes. While several dysregulated pathways linked to LAMA2-RD have been reported, an in-depth characterization of muscle gene expression in patients with

Teratoid Wilms Tumor: A Patient-Level Systematic Review of the Published Literature.

BACKGROUND: Teratoid Wilms tumor (TWT) is a rare morphological variant of Wilms tumor characterized by extensive heterologous differentiation. Its clinical significance remains uncertain because the available evidence consists predominantly of isolated case reports and small case series. We present a patient-level systematic review of published definite TWT cases. METHODS: A systematic review of original reports was performed using PubMed/MEDLINE, Google Scholar, citation tracking, and manual reference screening. Patient-level data were extracted exclusively from original publications and har

What Makes an Emergency Visit 'Avoidable'? A Qualitative Systematic Review of Multidisciplinary Clinician Perspectives on Acute Oncology Care Delivery.

BackgroundEmergency department (ED) presentations by patients with cancer are increasing globally. Policy frameworks often classify these as "potentially preventable emergency department visits" (PPEDVs) using administrative coding, yet the validity of this approach is contested. No qualitative systematic review of clinician perspectives on avoidable oncology ED use currently exists.ObjectiveTo synthesise multidisciplinary clinician perspectives on avoidable ED use in oncology and identify the barriers and facilitators that must be addressed to support alternatives to emergency attendance.Meth

Meaning-centered psychotherapy for Chinese caregivers of patients with advanced cancer: Cultural and linguistic adaptation.

PURPOSE: Cancer incidence is rising in Hong Kong, with over half of common cancers diagnosed at advanced stages, placing substantial demands on families and driving high caregiver distress. Meaning-centered psychotherapy for cancer caregivers (MCP-C) shows promise in Western settings but requires cultural tailoring for Chinese caregivers whose values are shaped by Confucian ethics. This study aimed to culturally and linguistically adapt MCP-C for Chinese caregivers of patients with advanced cancer in Hong Kong to enhance acceptability, relevance and conceptual equivalence. METHODS: Guided by

LMSCDA: A Secondary Structure Enhanced Language Model for Predicting CircRNA and Disease Associations.

Circular RNA (circRNA) is a kind of non-coding RNA widely present in cells. CircRNA plays a critical role in the occurrence and treatment of diseases. Unraveling the relationships between circRNAs and diseases has become a focus for diagnosis. While computational methods for predicting circRNA-disease associations (CDA) exist, they often oversimplify the representation of circRNA structures. To address this gap, we propose a novel method LMSCDA, which focuses on enhancing circRNA and disease representation by language model to predict CDAs. Specifically, we first calculate circRNA secondary st

Development and evaluation of a virus like particle based vaccine candidate against Indian Isolate of porcine circovirus 2d in pig.

Porcine circovirus type 2d (PCV2d) is the predominant genotype etiological agent of porcine circovirus-associated disease (PCVAD), causes substantial economic losses to the swine industry worldwide, including in India. Since currently available vaccines are mainly based on the PCV2a genotype, which is unable to offer cross protective immunity against emerging PCV2d strains, development of PCV2d specific vaccine is necessary. In this study, an indigenous virus-like particle (VLP)-based vaccine candidate was developed using the capsid coding gene (orf2) of an Indian PCV2d isolate expressed in a

The Impact of Clinical Variables and Dialysis Modality on Kynurenine Pathway Enzymes Expression.

Chronic kidney disease (CKD) and kidney failure significantly reduce patients' quality of life and markedly increase cardiovascular risk and overall mortality. Disturbed metabolism of tryptophan (Trp) through kynurenine (KYN) pathway was implicated as an important factor in kidney damage and its complications. However, the expression of genes coding crucial enzymes of KYN pathway was not examined so far. The goal of the present study was to analyze the expression of Ido (indoleamine-2,3-dioxygenase), Kat1 (kynurenine aminotransferase 1), Kat2 (kynurenine aminotransferase 2), and Kmo (kynurenin
