Molecular profiling of early stage liver fibrosis in patients with chronic hepatitis C virus infection.

The molecular mechanisms of acute hepatitis C virus (HCV) infection, end-stage hepatitis (cirrhosis), and hepatocellular carcinoma have been extensively studied, but little is known of the changes in liver gene expression during the early stages of liver fibrosis associated with chronic HCV infection, that is, the transition from normal liver (NL) of uninfected patients to the first stage of liver fibrosis (F1-CH-C). To obtain insight into the molecular pathogenesis of F1-CH-C, we used real-time quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) to study the mRNA expression

A mammalian severin replaces gelsolin in transformed epithelial cells.

A persisting paradox in cytoskeletal regulation of cell motility is the loss of the actin filament fragmenting protein, gelsolin, in transformed epithelial cells that have gained the ability to migrate. Either actin filament severing does not occur during motility of carcinoma cells or a novel fragmentation protein is expressed during transformation. Using an antibody specific for severin, the Mr 40,000 actin filament severing protein from Dictyostelium discoideum amoebae, we have identified a mammalian form of severin in murine LL/2 carcinoma cells lacking gelsolin. Mammalian severin (M-sever

Comparison of different voice samples for perceptual analysis.

BACKGROUND: Antiphospholipid syndrome (APS) is an acquired autoimmune disorder characterized by recurrent vascular events in large, medium, or small vessels. These events contribute to cardiovascular disease primarily through thrombosis and atherosclerosis (AS). Carotid atherosclerosis (CAS) represents a particularly high-risk manifestation of subclinical AS in patients with APS. However, the shared molecular signatures linking APS and CAS remain unclear. METHODS: Bulk transcriptome datasets from Gene Expression Omnibus (GEO) were analyzed to identify differentially expressed genes (DEGs) in

Tubulin State Determines Proteopathic Fate.

Neurodegenerative diseases have largely been defined by the accumulation of misfolded and aggregated proteins, while cytoskeletal disruption has typically been treated as a secondary consequence of pathology. Here, we argue that tubulin and microtubules are active determinants of whether disease-linked proteins remain functionally engaged or enter pathological assembly pathways. Recent evidence that tubulin redirects Tau:α-synuclein condensates away from oligomerization and amyloid formation toward physiological, microtubule-association competent states supports this view. On this basis, we pr

Determining Proximities of Sarcomeric Proteins in Living Skeletal Muscle Cells Using FRET Imaging.

The technique of fluorescence resonance energy transfer (FRET) has been used in this study to determine the proximities (efficiencies, i.e., E%) of interacting proteins in sarcomeres in living cultured embryonic chicken skeletal muscle cells to draw up a FRETsome of a sarcomere in mature myofibrils. During this study, two known Z-band proteins, FATZ (also called myozenin), and telethonin, were found to be actin binding proteins. The Efficiency of the F-actin and telethonin pair was measured to be 15%, while the F-actin and FATZ pair was 18%. The known F-actin and alpha-actinin interactions res

Double Sided Traction Force Microscopy: A Method to Confine Cells for Physiologically Relevant Force Measurements.

In this paper we describe a technique to make a confined environment of variable stiffness that is suitable for high-resolution live-cell imaging. This versatile and adaptable technique enables cell confinement between soft elastic surfaces made from polyacrylamide gels. The two surfaces retain all their compatibility with multiple approaches to chemically couple adhesion proteins, and additional techniques like micropatterning and traction force microscopy. This method is thus well suited for measuring force production and migration of weakly adherent cells that struggle to migrate in traditi

Transcripts and Protein Expression of Different HMW Non-Myofibrillar Tpm1 Isoforms in Human iPSC-CMs During Differentiation.

Mammals have four tropomyosin (TPM) genes (TPM1, TPM2, TPM3, and TPM4) that produce various isoforms through alternative splicing. TPM1 generates both myofibrillar (Tpm1.1, Tpm1.2) and non-myofibrillar isoforms, including those with an exon 9d peptide or with an exon 9a peptide. These non-myofibrillar isoforms are involved in regulating actin-based structures and processes in non-muscle cells. This study focused on the expression of the non-myofibrillar HMW Tpm1 isoforms (Tpm1.3, Tpm1.4, Tpm1.5, Tpm1.6, Tpm1.7, and Tpm1.14) during human inducible pluripotent stem cells (hiPSC) differentiation

The I130V and S229F Tropomyosin Mutations Associated With Congenital Heart Disease Disrupt Actin-Myosin Interactions in the Atria.

Tropomyosin is a highly conserved α-helical coiled-coil protein that is involved in maintaining the structural integrity of the sarcomere and regulating contraction of striated muscle. Mutations in the TPM1 gene encoding cardiac α-tropomyosin (Tpm1.1) are associated with left ventricular noncompact cardiomyopathy and congenital heart defects (CHD). We investigated the structural and functional properties of Tpm1.1 with the I130V and S229F mutations associated with CHD. We found that both mutations decreased the Tpm affinity for F-actin and differently affected the sliding velocity of F-actin-T

Hot and Splicey: Kinesin Isoforms Seek Attention.

Kinesins family members (KIFs) are encoded in mammals by approximately 45 genes, which are grouped into 14 families based on structural and functional features. As cells increased in complexity, so too did the demand for specialized kinesin-mediated processes, including intracellular transport, cytoskeletal organization, and cell division. One mechanism driving this diversity is alternative splicing, which can generate multiple protein isoforms with distinct properties. Here, we highlight examples of KIF isoforms that differ in cargo binding and biophysical motor behavior, as well as their rol

Hyperactive KIF5A in Neurodegeneration.

The highly polarised morphology of neurons and the sheer length of their axons make transport of cargoes throughout the cell a formidable task. Decades of evidence obtained from genetic studies on patients and animal models highlight deficits in axonal transport as a recurrent cause, or early contributing factor, in a plethora of neurodegenerative diseases. Axonal transport abnormalities usually manifest as a slowing of cargo trafficked by molecular motors along microtubules; however, hyperactivation of motors can also lead to disease. That is the case for the kinesin-1 protein KIF5A, in which

Harnessing Structure Prediction of Polo-Like Kinase 4 for Drug Repurposing.

Polo-like kinase 4 (PLK4) is a centrosome-specific kinase aberrantly expressed in cancers. Drugs inhibiting its catalytic kinase domain are under clinical phase-1/2 trials in patients with different leukemia types. However, the kinase domain of PLK4 shows structural similarity with other kinases. Therefore, drugs targeting the unique C-terminal polo-box domain (PBD) of PLK4 could provide better specificity. The knowledge of domain orientation in a full-length PLK4 structure is imperative for drug discovery. In this work, we utilized ab initio and threading approaches to predict the full-length

The Cilia-Associated Protein CCDC89 Is Dispensable for Male Fertility in Mice.

Cilia are microtubule-based organelles that protrude from the cell surface and are crucial for cellular sensory and motility functions. Defects in cilia are associated with various diseases, collectively known as ciliopathies. Although single-cell transcriptomics and proteomics have identified many proteins linked to cilia, their physiological roles remain largely unclear. In this study, we identify coiled-coil domain-containing 89 (CCDC89) as a new ciliary protein. Super-resolution imaging reveals that CCDC89 localizes to the axonemal lumen in motile cilia of mouse ependymal multiciliated cel

Isoform Specificity of a Compound Targeting Actin Filaments Containing Tropomyosin Tpm1.8/1.9.

The unbranched actin filaments in mammalian cells are usually composed of co-polymers of a specific tropomyosin isoform with actin. Genetic manipulation has revealed that the tropomyosins largely define the functional properties of actin filaments in an isoform-specific, non-redundant manner. Tropomyosin isoforms play a role in human diseases including cancers, thrombocytopenia and thrombocythaemia, endometrial decidualisation resistance, and ulcerative colitis. Hence, the development of compounds that target different tropomyosins are potentially valuable tools for cell biology as well as pot

Dissecting the Structural Organization, Recruitment and Activation Mechanisms of Centrosomal γ-TuRCs.

Visualizing human centrosomes using cryo-electron tomography revealed the native structure and molecular organization of γ-tubulin ring complexes (γ-TuRCs). γ-TuRCs localized to two distinct centrosomal pools, one in the pericentriolar material (PCM) and another in the centriole lumen, which is released during mitosis. All detected γ-TuRCs were associated with the tetrameric adaptor protein NEDD1. Within the PCM, binding to the centrosomin (CM1) motif of the microcephaly protein CDK5RAP2 in different patterns correlates with conformational changes of γ-TuRCs. In the centriole lumen, the augmin

Astral Microtubules Are Dispensable for Pavarotti Localization During Drosophila Spermatogonial Mitoses.

We analysed here the dynamic of the kinesin-like Pavarotti (Pav) during male gametogenesis of wild-type and Sas4 mutant flies. Pav localizes to the equatorial region and the inner central spindle of late anaphase wild-type spermatogonia and displays a strong concentration at the midbody during late telophase. At metaphase of the first meiotic division, Pav shows widespread localization on the equatorial region of the spermatocytes. This unusual distribution restricts and enhances during anaphase where antiparallel cortical microtubules overlap. Additional Pav staining is also found in the inne

The Interplay Between Early Chondrocyte Spreading and Inflammatory Responsivity.

Joint injuries are increasingly common and initiate a degenerative cascade in the cartilage extracellular matrix. Chondrocytes experience both intra- and extra-cellular changes during the initial phases of this process, including inflammatory activation and morphological change, initiating a catabolic feedback cycle that progresses toward osteoarthritis (OA). However, the link between this early morphological spreading and susceptibility to future inflammatory events is unclear. Thus, the objective of this study was to explore the implications of cellular spreading on early inflammatory activa

Discovery of an Atypical Arp2/3 Complex in Malaria Parasites Sheds New Light on Nuclear Actin.

The Arp2/3 complex is a key actin nucleator essential for cytoskeletal dynamics in eukaryotes. Previously believed absent in apicomplexan parasites, we recently identified an atypical Arp2/3 complex in malaria parasites consisting of five divergent subunits and a putative kinetochore-associated factor. This complex ensures proper kinetochore-spindle attachment during male gametogenesis, likely by nucleating actin at the mitotic spindle. Disruption of Arp2/3 function or actin polymerization leads to defective DNA segregation into gametes and developmental arrest of the parasite in the mosquito.

Myosin VI Is Associated With the Endoplasmic Reticulum in Regions of Sertoli Cells Containing Tubulobulbar Complexes.

Myosin VI has been reported by others to localize in association with various regions of apical tubulobulbar complexes (TBCs) at sites of attachment between Sertoli cells and late spermatids in the mouse. Tubulobulbar complexes internalize "intact" intercellular junctions during sperm release and during spermatocyte translocation through the blood-testis barrier. Here, we use super-resolution (STED-stimulated emission depletion) and electron microscopy of immunolabeled sections of rat testis to clearly define the localization of anti-myosin VI reactivity both at apical and basal sites in the e

Duchenne and Becker muscular dystrophy: Cellular mechanisms, image analysis, and computational models: A review.

The muscle is the principal tissue that is capable to transform potential energy into kinetic energy. This process is due to the transformation of chemical energy into mechanical energy to enhance the movements and all the daily activities. However, muscular tissues can be affected by some pathologies associated with genetic alterations that affect the expression of proteins. As the muscle is a highly organized structure in which most of the signaling pathways and proteins are related to one another, pathologies may overlap. Duchenne muscular dystrophy (DMD) is one of the most severe muscle pa
