Dysfunctions of leptin, ghrelin, BDNF and endocannabinoids in eating disorders: beyond the homeostatic control of food intake.

A large body of literature documents the occurrence of alterations in the physiology of both central and peripheral modulators of appetite in acute patients with anorexia nervosa (AN) and bulimia nervosa (BN). Until more recently the role of most of the appetite modulators in the control of eating behavior was conceptualized solely in terms of their influence on homeostatic control of energy balance. However, it is becoming more and more evident that appetite modulators also affect the non-homeostatic cognitive, emotional and rewarding component of food intake as well as non food-related rewar

[Circadian regulation of sleep-wake cycles and food anticipation].

The circadian clock is crucial for efficient physiological function and drives the temporal regulation of the sleep-wake state, metabolism, and behavior. The timing of food intake and the accompanying behavior are both controlled by the internal clock, which is located in the suprachiasmatic nucleus (SCN) of the anterior hypothalamus. The SCN is considered as the master clock because the circadian rhythms for most physiological and behavioral processes are terminated after SCN ablation. The molecular framework of circadian oscillations can be best studied in the SCN. A "core" set of circadian

Operative Procedure for Laparoscopy-Assisted Vagus Nerve and Pylorus-Preserving Gastrectomy (LAVNPPG) for Early Gastric Cancer.

Laparosocpy-assisted pylorus-preserving gastrectomy (LAPPG) is a widely accepted surgical procedure for the treatment of early gastric cancer in the middle third of the stomach. We have been performing this operation since 2007. Compared with traditional distal gastrectomy, LAPPG has postoperative nutritional benefits for patients. However, this procedure preserves only the pyloric branch of the vagus nerve and not the celiac branch. We found that patients retain a large amount of residual food in the gastric remnant, which interferes with the detection of secondary cancer on endoscopic follow

Hypolipidemic effect of n-butanol Extract from Asparagus officinalis L. in mice fed a high-fat diet.

During industrial processing of Asparagus (Asparagus officinalis L.), around half of each spear is discarded. However, these discarded asparagus (by-products) might be used as food supplements for their potential therapeutic effects. This study evaluated the hypolipidemic effect of n-butanol extract (BEA) from asparagus by-products in mice fed a high-fat diet (HFD). Continuous HFD feeding caused hyperlipidemia, oxidative stress and liver damage in mice. Interestingly, while BEA significantly decreased the levels of body weight gain, serum total cholesterol and low density lipoprotein cholester

Vagal control of satiety and hormonal regulation of appetite.

The paradigm for the control of feeding behavior has changed significantly. In this review, we present evidence that the separation of function in which cholecystokinin (CCK) controls short-term food intake and leptin regulate long-term eating behavior and body weight become less clear. In addition to the hypothalamus, the vagus nerve is critically involved in the control of feeding by transmitting signals arising from the upper gut to the nucleus of the solitary tract. Among the peripheral mediators, CCK is the key peptide involved in generating the satiety signal via the vagus. Leptin recept

Acrolein: sources, metabolism, and biomolecular interactions relevant to human health and disease.

Acrolein (2-propenal) is ubiquitously present in (cooked) foods and in the environment. It is formed from carbohydrates, vegetable oils and animal fats, amino acids during heating of foods, and by combustion of petroleum fuels and biodiesel. Chemical reactions responsible for release of acrolein include heat-induced dehydration of glycerol, retro-aldol cleavage of dehydrated carbohydrates, lipid peroxidation of polyunsaturated fatty acids, and Strecker degradation of methionine and threonine. Smoking of tobacco products equals or exceeds the total human exposure to acrolein from all other sour

Finite element simulation of food transport through the esophageal body.

The peristaltic transport of swallowed material in the esophagus is a neuro-muscular function involving the nerve control, bolus-structure interaction, and structure-mechanics relationship of the tissue. In this study, a finite element model (FEM) was developed to simulate food transport through the esophagus. The FEM consists of three components, i.e., tissue, food bolus and peristaltic wave, as well as the interactions between them. The transport process was simulated as three stages, i.e., the filling of fluid, contraction of circular muscle and traveling of peristaltic wave. It was found t

Effects of central infusion of ghrelin on food intake and plasma levels of growth hormone, luteinizing hormone, prolactin, and cortisol secretion in sheep.

Ghrelin is an endogenous ligand for the GH secretagogue/ghrelin receptor (GHS-R) and stimulates feeding behavior and GH levels in rodents and humans. A preprandial increase in plasma ghrelin levels is seen in sheep on programmed feeding, followed by a postprandial rise in plasma GH levels, but effects on food intake and endocrine function are not defined in this ruminant species. We administered ghrelin to female sheep in various modes and measured effects on voluntary food intake (VFI) and plasma levels of GH, LH, prolactin, and cortisol. Whether administered intracerebroventricularly or iv,

Multi-level mechanistic insights into phytone bioactivity: the healthy aging characteristics of C. elegans, molecular interactions and enzyme inhibition.

Phytone, an important terpene in fermented foods, has anti-aging potential. This study explored its interactions with α-amylase (αAmy), pepsin (PEP), trypsin (TPS), human serum albumin (HSA), and anti-aging effects via Caenorhabditis elegans (C. elegans). In nematodes, it prolongs lifespan, increases pharyngeal pump rate (14.98%), reduces heat stress mortality (10.10%), and preserves reproductive capacity. At the same time, it can significantly enhance the activity of SOD, CAT, and GSH-Px. Multispectral analysis and molecular simulation confirmed phytone forms stable complexes with these prote

Preparation and Characterization of Theasinensin A-Enriched Instant Black Tea.

Instant black tea is widely used in both the food and pharmaceutical industries. However, traditional water-mediated preparation leads to the loss of theasinensin A (TSA), a characteristic compound of black tea. This study aimed to optimize the preparation of instant black tea using TSA content as the primary quality indicator. Through response surface methodology, the ideal extraction parameters were identified as 49% ethanol (v/v), extraction at 70 °C for 41 min, and a material-to-liquid ratio of 1:60 g/mL. Under these optimized conditions, the TSA content reached a maximum of 456.05 ± 4.45

Structural Characterization and Antioxidant Activity of a Crude Polysaccharide from Cannabis sativa Leaves.

As a traditional Chinese herbal medicine, Cannabis sativa holds broad prospects for application in the development of functional foods, pharmaceutical formulations, dietary supplements, and cosmetic products. However, the bioactivity of polysaccharides in C. sativa has been largely overlooked. In this study, crude C. sativa leaf polysaccharide (CSLP) was extracted using the hot-water extraction and ethanol-precipitation method. CSLP contains 64.15 ± 1.96% carbohydrates and 2.13 ± 0.47% protein, with a yield of 6.71 ± 0.84% (w/w). Preliminary structural characterization showed that CSLP was mai

Engineering starch-based matrices: comparative impacts of conventional hydrocolloids versus novel algal biomass on potato purée.

Hydrocolloids are traditionally used to structure starch-based foods, yet demand is rising for sustainable, nutrient-dense matrices with enhanced bioactivity. To the best of our knowledge, this study is among the first to characterize algal fortification effect on potato purée, providing a novel route to develop starch-based foods with enhanced nutritional and functional properties. Light microscopy, colorimetry, texturometry and rheology were performed. Chlorella vulgaris at 3% exhibited the strongest water immobilization (aw = 0.27 ± 0.01), suggesting a possible mechanism of physical cellula

A novel fluorescent probe based on isolongifolanone-coupled acridine derivative for ultrafast and ultrasensitive detection of glyphosate via Cu2+ displacement and its application in environmental, food samples and biological imaging.

The extensive use of glyphosate in agriculture and the accumulation of heavy metal ions in the environment pose serious threats to food safety and human health. Herein, a dual-function fluorescent probe BO-BABP-OH based on an isolongifolanone scaffold was reported, and it realized the continuous detection of Cu2+ and glyphosate by in situ complex and selective replacement. Probe BO-BABP-OH displayed a fluorescence quenching response induced by Cu2+, and the fluorescence color of the probe was changed from bright yellow to colorless. However, the bright yellow emission of the probe was immediat

Water Activation on Sulfur Vacancies Enables Pyrite-Mediated Methylmercury Demethylation and Immobilization.

The accumulation of neurotoxic methylmercury (MeHg) in rice paddies presents a major global food safety challenge, yet effective in situ remediation strategies for flooded, anoxic soils are lacking. Here, we report that sulfur vacancy (SV)-rich pyrite efficiently degrades MeHg under dark, anoxic conditions via an innovative pathway involving vacancy-activated water (H2O). Experimental and theoretical evidence reveals that SVs serve a dual function: they enhance MeHg adsorption, weakening the carbon-mercury bond, while simultaneously activating ambient H2O to generate surface-bound hydroxyl rad

DOCK8 and STAT3 cooperate to restrain IgE-inducing T follicular helper cells.

Patients with loss-of-function DOCK8 or dominant-negative STAT3 variants have hyper-IgE syndrome, although only DOCK8 deficiency consistently presents with elevated food-specific IgE and symptomatic allergy. We previously found in mice that DOCK8 restricts the differentiation of IL-13+ T follicular helper (Tfh13) cells that drive anaphylactic IgE, although the mechanisms were unclear. Here, we show that DOCK8 promotes STAT3 activity, which inhibits GATA3 in T cells. However, only patients with DOCK8, but not STAT3, deficiency had elevated Tfh13 cells. Cell-specific deletion of either Dock8 (T-

Dietary Sulfur Compounds and Halitosis: Bridging Food Science, Microbial Metabolism, and Oral Health: A Comprehensive Review.

Intraoral halitosis is predominantly caused by anaerobic microbes within the tongue biofilm that break down sulfur-containing amino acids, especially cysteine and methionine, to volatile sulfur compounds (VSCs). In addition to microbial activity, there is growing evidence to suggest that dietary factors are able to influence VSC formation by affecting substrate availability, redox equilibrium, and oral ecological stability and that some of these effects are due to extraoral metabolic activity. This review integrates food chemistry, microbial ecology, and oral health to explain how dietary expo

Comparative study on the mechanisms of citrus pectin and polygalacturonic acid in alleviating food allergies.

Pectin has a complex structure, and different structures lead to different biological functions. Citrus pectin (PC) has been widely proven to possess immunomodulatory activity. This study compared and analyzed the anti-allergic activities of PC and polygalacturonic acid (PGA) based on intestinal microbiota, metabolomics, and molecular docking. The results showed that dietary interventions with PC and PGA significantly relieved allergic symptoms, intestinal damage, and the contents of specific antibodies IgE and IgG1. Among them, PGA exhibited a significant suppressive effect on histamine and a

Temporal dynamics of bioactive compounds in sweet tea (Lithocarpus litseifolius (Hance) Chun): Linking harvest stages to flavor and health benefits.

Sweet tea, the leaves of Lithocarpus litseifolius (Hance) Chun, received widespread attention for its edible and healthy values. This study employed non-targeted (UPLC-Q-Orbitrap-MS/MS) and quantitative (UPLC-QqQ-MS/MS) analyses to investigate the influence of harvest timing and geographic origin on its metabolite profile, flavor, and bioactivity. Among 505 identified metabolites, flavonoids and dihydrochalcones exhibited significant variation across developmental stages and regions. Electronic tongue analysis revealed that young leaves harvested in March, containing over 23 % trilobatin, disp
