The temporal dynamics of coherent motion processing in autism spectrum disorder: evidence for a deficit in the dorsal pathway.

Individuals with autism spectrum disorder (ASD) show impairments in processing coherent motion which have been proposed to be linked to a general deficit in the dorsal visual pathway. However, few studies have investigated the neural mechanisms underlying coherent motion processing in ASD. Thus, the aim of this study was to further test the hypothesis of a dorsal pathway deficit in ASD using visual evoked potentials (VEPs). 16 children and adolescents with ASD and 12 typically developing controls were examined with VEPs elicited by a random dot kinematogram. After an initial experimental seque

Integrative control of the lower urinary tract: preclinical perspective.

Storage and periodic expulsion of urine is regulated by a neural control system in the brain and spinal cord that coordinates the reciprocal activity of two functional units in the lower urinary tract (LUT): (a) a reservoir (the urinary bladder) and (b) an outlet (bladder neck, urethra and striated muscles of the urethral sphincter). Control of the bladder and urethral outlet is dependent on three sets of peripheral nerves: parasympathetic, sympathetic and somatic nerves that contain afferent as well as efferent pathways. Afferent neurons innervating the bladder have A-delta or C-fibre axons.

Surgery for idiopathic retroperitoneal fibrosis by ureterolysis and ureteric protection with a posterior pre-peritoneal fat flap.

Tinnitus is hypothesized to be an auditory phantom phenomenon resulting from spontaneous neuronal activity somewhere along the auditory pathway. The neural abnormalities underlying tinnitus are largely unknown. We evaluated the functional characteristics and the auditory system synchronization using Auditory Brainstem Response (ABR) in normal hearing tinnitus patients. In this observational comparative cross-sectional study, patients with chief complaints of Tinnitus and equal number of age and sex matched controls without hearing loss and tinnitus were enrolled. All patients underwent a full

Simultaneous intrastriatal and intranigral grafting (double grafts) in the rat model of Parkinson's disease.

Experimental and clinical studies of neural transplantation in Parkinson's disease have focused on the placement of fetal dopaminergic grafts not in their ontogenic site (substantia nigra) but in the main nigral target area (striatum). The reason for this is the apparent inability of intranigral nigral grafts to extend axons for long distances reinnervating the ipsilateral striatum. This review presents previous work by our laboratory [I. Mendez, M. Hong, Reconstruction of the striato-nigro-striatal circuitry by simultaneous double dopaminergic grafts: a tracer study using fluorogold and horse

Cholinergic control mechanisms for immunoreactive motilin release and motility in the canine duodenum.

The relationship of immunoreactive (IR) motilin release from the duodenum to duodenal motility changes was investigated in anaesthetized dogs. Stimulation of one or both vagi at 5 or 15 Hz or field stimulation of intrinsic duodenal nerves produced significant increases in duodenal vein IR motilin concentrations and accompanying increases in duodenal motility. However, only stimulation of both vagi at 15 Hz produced significant changes in peripheral venous concentrations of IR motilin. These occurred after a delay at a time when both the duodenum and the antrum were quiescent. Either hexamethon

Brain-Inspired Multi-Pathway Motion Decision-Making for Obstacle Avoidance of Humanoid Arms.

Achieving rapid and accurate obstacle avoidance in complex and dynamic environments remains a significant challenge for robots. To enhance the adaptability and flexibility of humanoid arms for obstacle avoidance, a brain-inspired multi-pathway motion decision-making method is proposed to modulate rational planning and habitual actions of humanoid arms. Firstly, a novel framework integrating both a slow and a fast pathway is designed for motion decision-making tasks. Imitating the rational planning function of the prefrontal cortex, the slow pathway employs an improved planning approach based o

The microbiota-gut-brain axis perspective: mechanisms and intervention strategies for the comorbidity of chronic constipation and depression.

BACKGROUND: Chronic constipation and depression are highly prevalent worldwide. These two conditions frequently co-occur in clinical practice. Accumulating evidence indicates that gut microbiota dysbiosis mediates this comorbidity through the microbiota-gut-brain axis (MGBA). METHODS: This narrative review systematically summarizes current research on MGBA bidirectional communication mechanisms, gut microbiota alterations in comorbid patients, and microbiota-targeted intervention strategies. RESULTS: The MGBA facilitates bidirectional communication through four major pathways: neural pathway

Auditory-cognitive training is associated with selective reorganization of alpha-band connectivity during speech-in-noise comprehension.

Older adults with hearing loss often struggle to understand speech in background noise because degraded auditory input increases demand on attention, working memory, and control. Auditory-cognitive training (ACT) can improve speech-in-noise (SiN) comprehension, but whether such gains are accompanied by changes in large-scale neural connectivity remains poorly understood. We tested whether ACT was associated with pre-to-post changes in source-space alpha-band phase synchrony within an auditory-control network during continuous SiN comprehension, and whether such changes related to ACT-associate

Zebrafish con/disp1 reveals multiple spatiotemporal requirements for Hedgehog-signaling in craniofacial development.

BACKGROUND: The vertebrate head skeleton is derived largely from cranial neural crest cells (CNCC). Genetic studies in zebrafish and mice have established that the Hedgehog (Hh)-signaling pathway plays a critical role in craniofacial development, partly due to the pathway's role in CNCC development. Disruption of the Hh-signaling pathway in humans can lead to the spectral disorder of Holoprosencephaly (HPE), which is often characterized by a variety of craniofacial defects including midline facial clefting and cyclopia 12. Previous work has uncovered a role for Hh-signaling in zebrafish dorsal

LAMP2A-dependent chaperone mediated autophagy regulates Notch signaling and controls neural stem cell differentiation.

Protein degradation systems have profound effects on signaling pathways and molecular networks that regulate organogenesis. Chaperone Mediated Autophagy (CMA) has been linked to neurodegenerative and neurodevelopmental diseases, however, its role in mammalian brain development remains poorly understood. Here, we identify a regulatory role of CMA pathway in neural stem cell (NSC) differentiation. We show that CMA is active in NSCs, while expression of LAMP2A, the principal limiting component of CMA, is associated with the induction of neurogenesis. Most importantly, overexpression and loss-of-f

From the gut to the brain: Potential novel avenues for IBD treatment via serotonergic pathways.

Inflammatory bowel disease (IBD) is an idiopathic condition characterised by chronic relapsing intestinal inflammation, affecting approximately 6.8 million people globally. Crohn's disease and ulcerative colitis are two main subtypes. The microbiota in IBD patients has been extensively researched, and dysbiosis is associated with IBD onset. While current treatments reduce morbidity and improve the quality of life of IBD patients, they have strong anti-inflammatory effects, creating an immunosuppressed environment and increase the risk of comorbidities, highlighting the need for better therapeu

A red nucleus-VTA glutamate pathway underlies exercise reward and the therapeutic effect of exercise on cocaine use.

Physical exercise is rewarding and protective against drug abuse and addiction. However, the neural mechanisms underlying these actions remain unclear. Here, we report that long-term wheel-running produced a more robust increase in c-fos expression in the red nucleus (RN) than in other brain regions. Anatomic and functional assays demonstrated that most RN magnocellular portion (RNm) neurons are glutamatergic. Wheel-running activates a subset of RNm glutamate neurons that project to ventral tegmental area (VTA) dopamine neurons. Optogenetic stimulation of this pathway was rewarding, as assesse

Age-Related Dopaminergic Innervation Augments T Helper 2-Type Allergic Inflammation in the Postnatal Lung.

Young children are more susceptible to developing allergic asthma than adults. As neural innervation of the peripheral tissue continues to develop after birth, neurons may modulate tissue inflammation in an age-related manner. Here we showed that sympathetic nerves underwent a dopaminergic-to-adrenergic transition during post-natal development of the lung in mice and humans. Dopamine signaled through a specific dopamine receptor (DRD4) to promote T helper 2 (Th2) cell differentiation. The dopamine-DRD4 pathway acted synergistically with the cytokine IL-4 by upregulating IL-2-STAT5 signaling an

Celecoxib-induced inhibition of neurogenesis in fetal frontal cortex is attenuated by curcumin via Wnt/β-catenin pathway.

Celecoxib is widely used in pregnant women but its influence on fetal brain neurogenesis is largely unknown. The objective of the present study was to examine the influence of celecoxib to fetal brain development and to investigate whether curcumin could ameliorate celecoxib-induced neurotoxicity. Pregnant mice, cultured neurons and cultured neural progenitor cells were all treated with celecoxib with or without curcumin. The change in proliferation, differentiation and the activity of Wnt/β-catenin signaling pathway were then assessed. Here, we report that prenatal celecoxib exposure inhibite

Medullary mediation of the laryngeal adductor reflex: A possible role in sudden infant death syndrome.

The laryngeal adductor reflex (LAR) is a laryngeal protective reflex. Vagal afferent polymodal sensory fibres that have cell bodies in the nodose ganglion, originate in the sub-glottal area of the larynx and upper trachea. These polymodal sensory fibres respond to mechanical or chemical stimuli. The central axons of these sensory vagal neurons terminate in the dorsolateral subnuclei of the tractus solitarius in the medulla oblongata. The LAR is a critical, reflex in the pathways that play a protective role in the process of ventilation, and the sychronisation of ventilation with other activiti

Zika Virus NS4A and NS4B Proteins Deregulate Akt-mTOR Signaling in Human Fetal Neural Stem Cells to Inhibit Neurogenesis and Induce Autophagy.

The current widespread outbreak of Zika virus (ZIKV) infection has been linked to severe clinical birth defects, particularly microcephaly, warranting urgent study of the molecular mechanisms underlying ZIKV pathogenesis. Akt-mTOR signaling is one of the key cellular pathways essential for brain development and autophagy regulation. Here, we show that ZIKV infection of human fetal neural stem cells (fNSCs) causes inhibition of the Akt-mTOR pathway, leading to defective neurogenesis and aberrant activation of autophagy. By screening the three structural proteins and seven nonstructural proteins

The vagal innervation of the gut and immune homeostasis.

The central nervous system interacts dynamically with the immune system to modulate inflammation through humoral and neural pathways. Recently, in animal models of sepsis, the vagus nerve (VN) has been proposed to play a crucial role in the regulation of the immune response, also referred to as the cholinergic anti-inflammatory pathway. The VN, through release of acetylcholine, dampens immune cell activation by interacting with α-7 nicotinic acetylcholine receptors. Recent evidence suggests that the vagal innervation of the gastrointestinal tract also plays a major role controlling intestinal

Noses and neurons: induction, morphogenesis, and neuronal differentiation in the peripheral olfactory pathway.

Non-axial mesenchymal/epithelial (M/E) induction guides peripheral olfactory pathway differentiation using cellular and molecular mechanisms similar to those in the developing limbs, aortic arches, and branchial arches. At each of these bilaterally symmetric sites off the midline axis, a thickened ectodermal epithelium is apposed to a specialized mesenchyme derived largely, but not exclusively, from the neural crest. The capacity of M/E interaction in the olfactory primordia (the combined olfactory placodal epithelium and adjacent mesenchyme) to induce a distinct class of sensory receptor neur

A corticohypothalamic circuit that mediates maternal awakening to pup cries.

Maternal caregiving requires sensory vigilance to ensure offspring survival, particularly during sleep. Infant cries are salient sensory cues that reliably trigger maternal arousal and caregiving responses in both humans and dams, but the underlying neural circuitry remains poorly defined. Here we identify a corticohypothalamic circuit in mice that mediates maternal awakening in response to pup calls. We show that layer 6 (L6) neurons in the temporal association cortex and ectorhinal cortex (TeA/Ect) projecting to the preoptic area (POA) exhibit enhanced responsiveness to pup calls after the t
