Functional connectivity associated with hand shape generation: Imitating novel hand postures and pantomiming tool grips challenge different nodes of a shared neural network.

Clinical research suggests that imitating meaningless hand postures and pantomiming tool-related hand shapes rely on different neuroanatomical substrates. We investigated the BOLD responses to different tasks of hand posture generation in 14 right handed volunteers. Conjunction and contrast analyses were applied to select regions that were either common or sensitive to imitation and/or pantomime tasks. The selection included bilateral areas of medial and lateral extrastriate cortex, superior and inferior regions of the lateral and medial parietal lobe, primary motor and somatosensory cortex, a

Cortical abnormalities and association with symptom dimensions across the depressive spectrum.

BACKGROUND: Few studies have investigated the relationship between structural brain abnormalities and dimensions of depressive symptomatology. METHODS: In the current study, we examined the relationship between cortical structural abnormalities and specific behavioral dimensions relevant to depression in a sample of unmedicated patients with major depressive disorder (MDD, n=57) and demographically similar healthy control volunteers (HC, n=29). All subjects underwent diagnostic assessment with the SCID, MRI at 3T, and dimensional assessments using the visual analog scales (VAS). Cortical regi

Substance abuse risk in emerging adults associated with smaller frontal gray matter volumes and higher externalizing behaviors.

BACKGROUND: During emerging adulthood, alcohol and substance use peak. Previous research has suggested that prefrontal and subcortical brain volumes may relate to risk for development of substance abuse. Epidemiological studies indicate that early initiation of alcohol or drug use significantly increases the likelihood of later substance use disorder diagnoses. We hypothesized that frontal regions would be smaller in young adults with early substance use and related problems (early-risk, ER), compared with a control group without early use/problems (C). We further hypothesized that these volum

Zazen meditation and no-task resting EEG compared with LORETA intracortical source localization.

Meditation is a self-induced and willfully initiated practice that alters the state of consciousness. The meditation practice of Zazen, like many other meditation practices, aims at disregarding intrusive thoughts while controlling body posture. It is an open monitoring meditation characterized by detached moment-to-moment awareness and reduced conceptual thinking and self-reference. Which brain areas differ in electric activity during Zazen compared to task-free resting? Since scalp electroencephalography (EEG) waveforms are reference-dependent, conclusions about the localization of active br

A computational hypothesis for allostasis: delineation of substance dependence, conventional therapies, and alternative treatments.

The allostatic theory of drug abuse describes the brain's reward system alterations as substance misuse progresses. Neural adaptations arising from the reward system itself and from the antireward system provide the subject with functional stability, while affecting the person's mood. We propose a computational hypothesis describing how a virtual subject's drug consumption, cognitive substrate, and mood interface with reward and antireward systems. Reward system adaptations are assumed interrelated with the ongoing neural activity defining behavior toward drug intake, including activity in the

Altered fronto-striatal and fronto-cerebellar circuits in heroin-dependent individuals: a resting-state FMRI study.

BACKGROUND: The formation of compulsive pattern of drug use is related to abnormal regional neural activity and functional reorganization in the heroin addicts' brain, but the relationship between heroin-use-induced disrupted local neural activity and its functional organization pattern in resting-state is unknown. METHODOLOGY/PRINCIPAL FINDINGS: With fMRI data acquired during resting state from 17 male heroin dependent individuals (HD) and 15 matched normal controls (NC), we analyzed the changes of amplitude of low frequency fluctuation (ALFF) in brain areas, and its relationship with histor

Reduced dorsal anterior cingulate cortical activity during emotional regulation and top-down attentional control in generalized social phobia, generalized anxiety disorder, and comorbid generalized social phobia/generalized anxiety disorder.

BACKGROUND: Generalized social phobia (GSP) and generalized anxiety disorder (GAD) are both associated with emotion dysregulation. Research implicates dorsal anterior cingulate cortex in both explicit emotion regulation (EER) and top-down attentional control (TAC). Although studies have examined these processes in GSP or GAD, no work compares findings across the two disorders or examines functioning in cases comorbid for both disorders (GSP/GAD). Here we compare the neural correlates of EER and TAC in GSP, GAD, and GSP/GAD. METHODS: Medication-free adults with GSP (EER n = 19; TAC n = 18), GA

Gray matter reduction associated with psychopathology and cognitive dysfunction in unipolar depression: a voxel-based morphometry study.

BACKGROUND: Functional neuroimaging studies on both cognitive processing and psychopathology in patients with major depression have reported several functionally aberrant brain areas within limbic-cortical circuits. However, less is known about the relationship between psychopathology, cognitive deficits and regional volume alterations in this patient population. METHODS: By means of voxel-based morphometry (VBM) and a standardized neuropsychological test battery, we examined 15 patients meeting DSM-IV criteria for major depression disorder and 14 healthy controls in order to investigate the

Shared and distinct neurophysiological components of the digits forward and backward tasks as revealed by functional neuroimaging.

The digits forward (DF) and backward (DB) tasks are widely used neuropsychological measures believed to tap overlapping systems of phonological processing and working memory. Studies of focal brain lesions have partially elucidated the brain regions essential for these tasks; however relatively little information exists on the underlying functional neuroanatomy in the intact brain. We therefore examined the shared and separate neural systems of these tasks in two positron emission tomography (PET) experiments. In Experiment 1, eight healthy participants performed verbal DF, DB, and a sensorimo

Changes in neuronal activation in patients with bipolar disorder during performance of a working memory task.

OBJECTIVES: Several lines of evidence suggest that deficits in cognition persist in bipolar patients during periods of euthymia. Working memory impairment has been observed in euthymic bipolar patients and noted to be a significant source of functional deficits in psychiatric disorders. Functional changes associated with these cognitive deficits however, remain poorly understood. We hypothesized that patients with bipolar disorder would demonstrate changes in neuronal activation in specific regions forming part of the working memory network. METHODS: Fifteen euthymic bipolar patients and fift

Exercise-induced changes in local cerebral glucose utilization in the rat.

In exercise, little is known about local cerebral glucose utilization (LCGU), which is an index of functional neurogenic activity. We measured LCGU in resting and running (approximately 85% of maximum O2 uptake) rats (n = 7 in both groups) previously equipped with a tail artery catheter. LCGU was measured quantitatively from 2-deoxy-D-[1-14C]glucose autoradiographs. During exercise, total cerebral glucose utilization (TCGU) increased by 38% (p < 0.005). LCGU increased (p < 0.05) in areas involved in motor function (motor cortex 39%, cerebellum approximately 110%, basal ganglia approximately 30

Comparative Efficacy of Non-invasive Brain Stimulation for Pain, Anxiety, and Depression: A Systematic Review and Network Meta-analysis.

INTRODUCTION: Although non-invasive brain stimulation (NIBS) has been used to manage chronic musculoskeletal pain (CMP) and related emotional symptoms, the comparative efficacy of different NIBS techniques remains unclear. Therefore, this network meta-analysis (NMA) compared the effects of NIBS interventions on pain intensity, anxiety, and depressive symptoms in CMP. METHODS: We searched seven databases from inception to 7 October 2025, including PubMed, the Cochrane Library, Web of Science, Embase, MEDLINE, PsycINFO, and CINAHL, and included only English-language randomized controlled trials

Neural Substrates of Emotion Processing and Cognitive Control Over Emotion in Youth Anxiety: An RDoC-Informed Study Across the Clinical to Nonclinical Continuum of Severity.

OBJECTIVE: Clinically anxious youth are hypervigilant to emotional stimuli and display difficulty shifting attention from emotional to nonemotional stimuli, suggesting impairments in cognitive control over emotion. However, it is unknown whether the neural substrates of such biases vary across the clinical-to-nonclinical range of anxiety or by age. METHOD: Youth aged 7 to 17 years with clinical anxiety (n = 119) or without an anxiety diagnosis (n = 41) matched emotional faces or matched shapes flanked by emotional face distractors during magnetic resonance imaging, probing emotion processing

Top-down influence of the inferior frontal gyrus on motor cortex excitability during pain empathy.

Empathy for pain can reduce activity in the observer's motor system, but the neural processes that shape this response remain unclear. Here we show that observing another person in pain reduces corticospinal excitability and increases perceived pain intensity when viewed from a first-person perspective, but not from a third-person perspective. This motor suppression is absent when noxious events are applied to a non-biological object. Using single-pulse, paired-pulse, and dual-site transcranial magnetic stimulation, we find no detectable change in local motor cortical inhibitory or facilitator

Children With Idiopathic Toe Walking Display Different Cortical Activation Patterns When Interpreting Tactile Sensation.

Idiopathic toe walking (ITW) is a diagnosis given to children who walk with an absence of heel strike in the contact phase of the gait cycle, who are otherwise typically developing. This study aimed to determine if children with ITW had cortical activation differences when completing a tactile sensation test compared to typically developing children. The study included 15 typically developing children and 15 children with ITW. Participants were required to perform a two-point discrimination test during an fMRI using a standard block design.Four clusters were identified, where the controls exhi

The temporal onset of associations of cortical proteins with cognitive resilience vary during late life.

BACKGROUND: Cortical proteins associated with cognitive resilience have been identified but their temporal onset in older adults is unknown. We present a multistage approach to first identify cortical proteins associated with cognitive resilience and then examine their associated temporal onset. METHODS: We used data from a subset of 1088 decedents from two cohort-studies who had selected reaction monitoring proteomics from the dorsolateral prefrontal cortex, and at least 3 cognitive assessments. Cognition was assessed using a composite derived from 19 tests. We first used linear mixed-effect

Cross-spatial scale processing of hierarchical auditory sequences in human brains revealed using 7 T magnetic resonance imaging.

Human auditory cognition involves multiregional hierarchical processing, yet the neural mechanisms integrating cortical layers and large-scale networks remain unclear. Using high-resolution fMRI, we investigate cross-spatial scale dynamics during hierarchical auditory processing. Distinct activations across layers of superior temporal gyrus (STG) and inferior frontal gyrus (IFG) are detected during standard stimuli and violation. We identify the layer-specific effective connectivity among key nodes in the auditory hierarchy using dynamic causal modelling. The results are consistent with hierar
