Disease-associated mosaic variation in clinical exome sequencing: a two-year pediatric tertiary care experience.

Exome sequencing (ES) has become an important tool in pediatric genomic medicine, improving identification of disease-associated variation due to assay breadth. Depth is also afforded by ES, enabling detection of lower-frequency mosaic variation compared to Sanger sequencing in the studied tissue, thus enhancing diagnostic yield. Within a pediatric tertiary-care hospital, we report two years of clinical ES data from probands evaluated for genetic disease to assess diagnostic yield, characteristics of causal variants, and prevalence of mosaicism among disease-causing variants. Machine Learning of Personal Repertoires From Public T Cell Receptors.

The T-cell receptor (TCR) repertoire records an individual's immunological history, but most unique CDR3 sequences in any one person are private and uninformative about anyone else. A small subset, however, recurs predictably across unrelated donors. These public TCRβ sequences are independently generated by multiple mechanisms: enrichment by thymic positive selection on a largely shared self-peptide-MHC ligandome, further amplified by selection on common foreign antigens and convergent recombination, together, they are what makes a personal repertoire computationally legible: they provide the

Multidisciplinary management in panfacial trauma: A clinical study.

Management of panfacial fractures lacks standardized multidisciplinary pathways and variability in coordination, surgical sequencing and airway planning may contribute to delays and postoperative complications. Therefore, it is of interest to assess if a structured multidisciplinary team (MDT) referral system leads to an improvement in process and outcome measures versus usual care where patients are referred sequentially. A comparison of a single-center cohort with two different treatment strategies; primary outcomes were time-to-definitive fixation and major postoperative complications. Seco

Developing a cps-genotyping scheme for the emerging zoonotic pathogen Streptococcus parasuis through genetic analysis of capsular polysaccharide synthesis gene loci.

BACKGROUND: Streptococcus parasuis is an emerging zoonotic pathogen responsible for pneumonia, meningitis, peritonitis, pleuritis, nephritis, cellulitis, cholecystitis, sepsis, and arthritis in humans. The continuous increase in reported human cases of S. parasuis infection indicates its growing threat to public health and highlights the urgent need to monitor its epidemiology. The lack of high-throughput typing tools has seriously impeded the epidemiological surveillance of S. parasuis. Serotyping based on antigenic differences in capsular polysaccharides (CPSs) synthesized by cps loci is wid

Re-Emerging Bacterial Pathogens, Resistance Genes and Promising Bioindicators in Raw and Treated Sewage-Addressing a Known Issue from a Different Angle and Perspective.

Municipal wastewater catchments are significant sources of symbiotic, opportunistic, and drug-resistant bacteria that can acquire or enhance their resistance. This "re-emergence of threats" presents a serious public health issue, especially during crisis conditions (COVID-19). The aim of this study was to propose a sampling algorithm, identify strategically important sites within the wastewater catchment, expand the range of bacterial indicators, and rapidly confirm potential threats using in vitro diagnostics in both raw and treated sewage. A parallel objective was to evaluate the effectivene

Comparative Sequence Analysis of the Envelope Gene of Kyasanur Forest Disease Virus Vaccine Strain With Currently Circulating Field Strains.

The present study was undertaken with the objective of performing comparative sequence analysis of the envelope (E) gene of Kyasanur forest disease virus (KFDV) vaccine strain P9605 with the circulating field strains. The study was taken up as the currently used KFDV vaccine strain, KFDV P9605, was isolated in the 1960s. For this study, we designed two sets of primers targeting the complete amplification of the E gene of KFDV. The sequencing was performed by the Sanger method, and the deduced sequence of the vaccine virus was deposited in GenBank with accession number PX067005. This sequence o

Diversity in Chlamydial plasmids.

BACKGROUND: Evolutionary studies have been conducted that have investigated the chromosomal variance in the genus of Chlamydia. However, no all-encompassing genus-wide comparison has been performed on the plasmid. Therefore, there is a gap in the current knowledge on Chlamydia plasmid diversity. AIMS: This project is aimed to investigate and establish the nature and extent of diversity across the entire genus of Chlamydia, by comparing the sequences of all currently available plasmid carrying strains. METHODS: The PUBMED database was used to identify plasmid sequences from all available stra

Prevalence of Piroplasma in Ticks Collected from Dogs and Cattle in Guangxi, South China Determined by Reverse Line Blot Hybridization Assay.

Piroplasmosis is a disease of domestic and wild animals caused by tick-borne protozoa of the genera Theileria and Babesia. Piroplasmosis leads to substantial economic losses in the livestock industry. This disease has been frequently reported in subtropical and tropical regions worldwide. However, information regarding the prevalence of piroplasma in ticks collected from dogs and cattle is lacking in most areas. To assess the potential threat of piroplasmosis in South China, 671 ticks were collected in Guangxi Province. Deoxyribonucleic acid (DNA) was extracted from these ticks to evaluate the

Complete androgen insensitivity syndrome associated with bilateral sertoli cell adenomas and unilateral paratesticular leiomyoma: A case report.

Background. Highly stable, evolutionarily conserved, small, non-integrative plasmids are commonly found in members of the Chlamydiaceae and, in some species, these plasmids have been strongly linked to virulence. To date, evidence for such a plasmid in Chlamydia pecorum has been ambiguous. In a recent comparative genomic study of porcine, ovine, bovine, and koala C. pecorum isolates, we identified plasmids (pCpec) in a pig and three koala strains, respectively. Screening of further porcine, ovine, bovine, and koala C. pecorum isolates for pCpec showed that pCpec is common, but not ubiquitous i

Natatory-stage cymothoid isopods: description, molecular identification and evolution of attachment.

Cymothoid isopods are parasites that have a biphasic life cycle comprised of free-swimming micropredatory stages that eventually attach permanently to fishes, on which they change sex and morphology. Thus, matching free-swimming and permanently attached life-history stages is difficult. We attempted to identify natatory-stage cymothoids by seeking matches of partial 16S rRNA gene sequences from these stages and described adult cymothoids. Natatory cymothoids were collected from light traps at Lizard Island, Great Barrier Reef and adult females were collected from fishes from several sites alon

MOLECULAR CHARACTERIZATION OF SARCOCYSTIS SPOROCYSTS IN INTESTINES OF BOBCAT (LYNX RUFUS) FROM MINNESOTA.

Sarcocystis species are heteroxenous protozoan parasites of major veterinary and public health relevance, yet the role of wild felids in their natural transmission cycles remains poorly understood. In this study, we investigated Sarcocystis infections in 189 bobcats (Lynx rufus) harvested from Minnesota during 2024-2025. Intestinal homogenates of 179 were examined microscopically, and Sarcocystis sporocysts were found in 38 bobcats (21.2%). DNA was extracted from all 38 positive samples, of which 19 were PCR-positive, and 14 yielded high-quality sequences for molecular characterization using p

MORPHOLOGICAL OBSERVATION AND DETAILED MOLECULAR CHARACTERIZATION OF FISCHOEDERIUS ELONGATUS (DIGENEA: GASTROTHYLACIDAE) FROM THE RUMEN OF DOMESTIC CATTLE IN CAMBODIA.

Fischoederius elongatus (Poirier, 1883) is a common rumen fluke found in Asia and Africa. Nucleotide sequence data for F. elongatus have been reported but are limited to certain ribosomal and mitochondrial DNA regions. High diversity in the nucleotide sequences of the mitochondrial cytochrome c oxidase subunit 1 (COI) haplotypes has been reported in some Asian countries. Some researchers have advocated the possibility of cryptic species within F. elongatus based on the high genetic diversity within the species as well as the genetic distance within and between sister species. However, the numb

Genetic evolution analysis of novel recombinant pseudorabies virus strain in Sichuan, China.

Pseudorabies is a disease that seriously endangers the pig industry in China. Recently, we successfully isolated a pseudorabies virus from the brain tissue of piglets at a farm in Sichuan, China, and named it the FJ62 strain. In order to understand the molecular biological characteristics of the strain, primers were designed for glycoproteins gB, gC, gD and gE, which were amplified by a polymerase chain reaction (PCR) and sequenced. After comparing the sequence with the GenBank 22 pseudorabies virus reference strains and establishing the genetic evolutionary tree, it was found that the gB gene

Characterisation and functional analysis of canine TLR5.

In this study, we characterised the single exon TLR5 gene of the Chinese rural dog. Sequence analysis revealed a 2577 nucleotide-long open reading frame of canine TLR5, encoding an 858 amino acid-long protein. The putative amino acid sequence of canine TLR5 consisted of a signal peptide sequence, 15 LRR domains, a LRR C-terminal domain, a transmembrane domain and an intracellular Toll-IL-1 receptor domain. The amino acid sequence of the canine TLR5 protein shared 95.4% identity with vulpine, 72.2% with feline and 64.7% with human TLR5. Plasmids expressing canine TLR5 and NF-κB-luciferase were

Molecular analyses reveal high species diversity of trematodes in a sub-Arctic lake.

To identify trematode diversity and life-cycles in the sub-Arctic Lake Takvatn, Norway, we characterised 120 trematode isolates from mollusc first intermediate hosts, metacercariae from second intermediate host fishes and invertebrates, and adults from fish and invertebrate definitive hosts, using molecular techniques. Phylogenies based on nuclear and/or mtDNA revealed high species richness (24 species or species-level genetic lineages) and uncovered trematode diversity (16 putative new species) from five families typical in lake ecosystems (Allocreadiidae, Diplostomidae, Plagiorchiidae, Schis

Changes in the neuropeptide content of Biomphalaria ganglia nervous system following Schistosoma infection.

BACKGROUND: Molluscs, including snails, are prone to parasite infection, which can lead to massive physiological and behavioural changes, yet many of the molecular components involved remain unresolved. Central to this point is the neural system that in snails consists of several ganglia that regulate the animals' physiology and behaviour patterns. The availability of a genomic resource for the freshwater snail Biomphalaria glabrata provides a mean towards the high throughput analysis of changes in the central nervous system (CNS) following infection with Schistosoma miracidia.
