Memory-T-cell-derived interferon-γ instructs potent innate cell activation for protective immunity.

Cells of the innate immune system are essential for host defenses against primary microbial pathogen infections, yet their involvement in effective memory responses of vaccinated individuals has been poorly investigated. Here we show that memory T cells instruct innate cells to become potent effector cells in a systemic and a mucosal model of infection. Memory T cells controlled phagocyte, dendritic cell, and NK or NK T cell mobilization and induction of a strong program of differentiation, which included their expression of effector cytokines and microbicidal pathways, all of which were delay

An unusual clinical presentation of gingival melanoacanthoma.

Gingival melanoacanthoma is a rare, benign pigmented lesion characterized clinically by sudden onset and rapid growth of a macular brown black lesion and histologically by acanthosis of superficial epithelium and proliferation of dendritic melanocytes. This article reports a previously undescribed case of pigmented unilateral diffuse gingival enlargement, which on histopathological examination proved to be melanoacanthoma. Intraoral examination revealed pigmented unilateral diffuse gingival enlargement in relation to second and third quadrants buccally, palatally/lingually. Based on these clin

CDKL5 ensures excitatory synapse stability by reinforcing NGL-1-PSD95 interaction in the postsynaptic compartment and is impaired in patient iPSC-derived neurons.

Mutations of the cyclin-dependent kinase-like 5 (CDKL5) and netrin-G1 (NTNG1) genes cause a severe neurodevelopmental disorder with clinical features that are closely related to Rett syndrome, including intellectual disability, early-onset intractable epilepsy and autism. We report here that CDKL5 is localized at excitatory synapses and contributes to correct dendritic spine structure and synapse activity. To exert this role, CDKL5 binds and phosphorylates the cell adhesion molecule NGL-1. This phosphorylation event ensures a stable association between NGL-1 and PSD95. Accordingly, phospho-mut

Inhibition of DC-SIGN-mediated HIV infection by a linear trimannoside mimic in a tetravalent presentation.

HIV infection is pandemic in humans and is responsible for millions of deaths every year. The discovery of new cellular targets that can be used to prevent the infection process represents a new opportunity for developing more effective antiviral drugs. In this context, dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN), a lectin expressed at the surface of immature dendritic cells and involved in the initial stages of HIV infection, is a promising therapeutic target. Herein we show the ability of a new tetravalent dendron containing four copies of a linear trimannoside mimic to in

Immunotherapeutic strategies for the treatment of malignant melanoma.

The incidence of cutaneous malignant melanoma is increasing at a faster rate than any other cancer worldwide. Despite new advances in surgical management of melanoma, this malignancy remains one of the most aggressive and intractable to treat among other solid tumors. Continuous search for better therapeutics led to the development of various immunological approaches applicable to the treatment of this melanocytic malignancy. Multiple peptide, dendritic cell, adjuvant, lymphocyte, and virus-based strategies were established and tested in preclinical and clinical studies with varying degrees of

[CD8+ cytotoxic lymphocytes in cutaneous leishmaniasis].

Review of the literature on the role of CD8+ T cell in the immune response against Leishmania species that cause cutaneous leishmaniasis. The role of macrophages, dendritic cells, CD4 T cells and NK cells has been extensively analyzed in leishmaniasis, yet very little knowledge has been gained on CD8+ T cells in this disease. Murine models of leishmaniasis suggest that CD8+ T cells participate through IFNg production, yet their cytotoxic capacity may also play a crucial role, as has been found in human disease. It is an enigma what mechanisms underlie the CD8+ T cell activation. It is possible

Expression of cochlin in the vestibular organ of rats.

The COCH gene mutated in autosomal dominant sensorineural deafness (DFNA9) encodes cochlin, a major constituent of the inner ear extracellular matrix. Cochlin constitutes 70% of the inner ear protein and cochlin isoforms can be classified into three subgroups, p63s, p44s and p40s. Symptoms of some DFNA9 patients are consistent with those of Ménière's disease. Here, we report the expression of cochlin in the vestibular organ of rats using isoform-specific antibodies that recognize all three isoforms. Cochlin is highly expressed in the stromata of the maculae of otolithic organs and cristae of s

The effects of thymostimulin on immunological function in patients with head and neck cancer.

Patients with head and neck carcinoma show deficits of cellular immunity, probably due to low molecular mass factors (LMMFs) released by the tumour. Thymostimulin (TP1) restores the defective monocyte chemotaxis and dendritic cell clustering capability in the presence of the tumour when administered pre-operatively. In the present study we investigated these immune parameters in 39 patients treated with TP1 for 10 days pre- and 6 months post-operatively and in 22 patients who were not treated with TP1 for 6 months after operation. Removal of the tumour in non-TP1-treated patients also resulted

Can megadoses of thiamine prevent ethanol-induced damages of rat hippocampal CA1 pyramidal neurones?

The specific aim of this study was to evaluate whether high doses of thiamine can compensate or prevent alcohol-induced damages of rat hippocampus CA1 pyramids. Twenty weeks of ethanol consumption together with a dose of thiamine in the range of 1.19 mg/100 mg food induced significant enlargement (parameters measured were length of the whole spine and diameter of the end-bulb) of dendritic spines. Hypertrophy can be interpreted as a compensation process due to alcohol-induced cell death because viable spines are in search of new synaptic contacts. In contrast, dendritic spines of the alcohol g

The induction of melanogenesis by ultraviolet light in the pigmentary system of Rhesus monkeys.

Except for the face, eyelids, friction surfaces, and lips, the epidermis of the rhesus monkey contains no discernible melanocytes. After ultraviolet irradiation, however, dopa-positive dendritic cells appeared. With daily sequential irradiation, the number of histochemically demonstrable dopa-positive dendritic cells peaked after 30 exposures, then declined to a basal level which was maintained for the duration of the experiment (216 exposures or 43 weeks). Pigment cells can be restimulated by shading part of the irradiated area and then reirradiating after 3 months. While shaded, dopa-positiv

Single-cell and spatial transcriptomics unveil myeloid-lymphoid cross talk and the dermal immune niche underlying palmoplantar pustulosis.

BACKGROUND: Palmoplantar pustulosis (PPP) is a chronic, recurrent inflammatory dermatosis characterized by sterile pustules on the palms and/or soles, and its pathogenesis remains incompletely understood. OBJECTIVE: We sought to investigate the immune landscape underlying PPP progression using a multiomics approach. METHODS: We conducted single-cell RNA sequencing on pustular and nonpustular sole lesions from patients with PPP. Key findings were further validated at the tissue level using high-resolution spatial transcriptomics (ST). RESULTS: We identified elevated JAK-STAT signaling in ker

Allergen-specific immunotherapy with Der p 1/2 mRNA vaccines relieves house-dust-mite-induced allergic airway inflammation in a mouse model.

Allergen-specific immunotherapy (AIT) is the only disease-modifying treatment for allergic diseases, but current crude-extract-based approaches suffer from poor standardization and variable safety and efficacy. This study developed lipid nanoparticle (LNP)-encapsulated mRNA vaccines encoding Der p 1 and Der p 2, the major house dust mite (HDM) allergens, and evaluated them in a murine model of HDM-induced airway inflammation. LNP-mRNA vaccines triggered potent humoral immunity and exhibited good tolerability. Subcutaneous immunotherapy significantly reduced early allergic responses, airway hyp

Convergence-divergence circuits for multimodal integration of innate and learned opponent valences.

Valence detection in complex environment is critical for natural behaviors like foraging. Previous studies have explored valence processing in brain regions like lateral horn (LH) and mushroom body (MB) using simple synthetic stimuli in Drosophila. However, the neural basis for valence detection of natural objects in complex contexts remains unclear. Here, by brain-wide connectome analysis, we identified the evolutionarily conserved superior protocerebrum (SP) that integrates brain-wide multimodal inputs mainly via LH and MB, and sends widespread outputs particularly to the central complex (CX

Shared autonomous HERV loci transcription identifies a unique circulating CD14+-xCR1+ mononuclear cell phenotype in a patient group with post-acute sequelae of COVID-19.

The human genome contains approximately 3,200 near full-length autonomous human endogenous retroviral (HERV) loci capable of transcriptional activation through long terminal repeats. In our previous study, we developed the Window-based HERV Alignment (WHA) method to detect locus-specific HERV transcription from single-cell RNA sequencing (scRNA-seq) data. Using WHA, we previously identified patterns of autonomous HERV loci expression in monocytes from 12 patients with post-acute sequelae of COVID-19 (PASC). In the current study, we extended the analysis to peripheral blood mononuclear cells (P

Platelet-neutrophil niches associate with epidermal immune activation and systemic inflammation in psoriatic disease.

Platelets are increasingly recognised as inflammatory mediators that influence leukocyte behaviour, yet their spatial organisation and contribution to psoriatic skin inflammation remain incompletely understood. Here, spatial transcriptomics was used to map platelet-leukocyte niches (PLNi) across inflammatory skin diseases, contrasting psoriasis (PsO) with atopic dermatitis (AD). PLNi were selectively expanded in PsO lesions, where platelet-neutrophil co-localisation defined transcriptionally active regions enriched for stress and inflammatory mediators. PsO and psoriatic arthritis (PsA) shared

Virus-specific resident memory T cell networks sustain immunity in human oral mucosa.

BACKGROUND: Durable protection against viral infections relies on tissue-resident memory T (TRM) cells at pathogen entry sites. The oral cavity is a critical mucosal barrier frequently exposed to viral pathogens; however, the phenotypic characteristics and spatial organization of virus-specific T cells in the human oral mucosa remain poorly understood. OBJECTIVE: This study aimed to define how virus-specific T cells maintain long-term residency in the oral mucosa and to identify cellular interactions supporting their persistence. METHODS: We profiled predicted virus-specific T cells in the b

Esketamine mitigates endotoxin-induced hippocampal injury by regulating calcium transient and synaptic plasticity via the NF-α1/CREB pathway.

Esketamine (ES) has been shown to confer neuroprotection partly by exerting anti-inflammation, alleviating oxidative stress, enhancing neuronal vitality, and promoting synaptic remodeling. Nonetheless, its precise function in SAE and the associated mechanisms are not understood. In this study, we investigated the neuroprotective potential of ES at behavioral, structural, and functional levels in vivo and in vitro. C57BL/6J mice administered with lipopolysaccharide (LPS) served as the research model and were injected with 10 mg/kg ES intraperitoneally. Fiber photometry was performed to record C

Diverse Lenabasum pathway activation in dermatomyositis patients' blood.

Lenabasum, a non-psychoactive cannabinoid type-2 receptor (CB2R) agonist, has shown promise in reducing cutaneous disease in Dermatomyositis (DM) patients. Lenabasum activates two distinct receptors: CB2R and the nuclear peroxisome proliferator-activated receptor-γ (PPARγ). Our goal was to investigate the dominant mechanism of action leading to pathogenic IFNβ reduction by lenabasum (through CB2R or PPARγ) across leukocytes. We utilized whole blood leukocytes from 14 DM patients and grouped patients as in vitro responders or non-responders. We stimulated leukocytes in vitro in the presence of

Uncovering the mechanism of troglitazone-mediated idiosyncratic drug-induced liver injury with individual-centric models.

Idiosyncratic drug-induced liver injury is a rare and unpredictable event. Deciphering its initiating-mechanism is a hard task as its occurrence is individual dependent. Thus, studies that utilize models that are not individual-centric might drive to a general mechanistic conclusion that is not necessarily true. Here, we use the individual-centric spheroid model to analyze the initiating-mechanism of troglitazone-mediated iDILI risk. Individual-centric spheroid models were generated using a proprietary cell educating technology. These educated spheroids contain hepatocytes, hepatic stellate ce

Aberrant monocytopoiesis drives granuloma development in sarcoidosis.

In sarcoidosis, granulomas develop in multiple organs including the liver and lungs. Although mechanistic target of rapamycin complex 1 (mTORC1) activation in macrophages drives granuloma development in sarcoidosis by enhancing macrophage proliferation, little is known about the macrophage subsets that proliferate and mature into granuloma macrophages. Here, we show that aberrantly increased monocytopoiesis gives rise to granulomas in a sarcoidosis model, in which Tsc2, a negative regulator of mTORC1, is conditionally deleted in CSF1R-expressing macrophages (Tsc2csf1rΔ mice). In Tsc2csf1rΔ mic
