Acute COVID-19 is associated with altered CD8 T-cells indicative of impaired ability to control Epstein-Barr virus reactivation.

Increasing evidence suggests that reactivation of latent EBV in patients with COVID-19 may be linked to the development of post-acute sequelae of COVID-19, colloquially known as Long COVID. However, the reason for this co-occurrence of primary infection and reactivation of latent viruses remains elusive. During the first wave of COVID-19, we assessed all major immune cell populations by flow cytometry in a cohort of 61 patients with moderate to critical COVID-19 at the time of hospitalization. Additional blood samples from these patients were biobanked for later analysis. Using these biobanked

Spatial transcriptomic profiling reveals molecular signatures of endocapillary hypercellularity in IgA nephropathy.

BACKGROUND: Immunoglobulin A nephropathy (IgAN) is a leading cause of chronic kidney disease, marked by mesangial IgA1 immune-complex deposition and a multi-hit pathogenesis. The Oxford classification grades histopathological lesions such as endocapillary hypercellularity (E0 and E1), which are associated with disease severity and prognosis. Despite advances in single-cell and spatial transcriptomics, the molecular basis of endocapillary hypercellularity in human IgAN remains unclear. METHODS: Spatial transcriptomic data obtained using the GeoMx DSP platform from kidney biopsy specimens of Ea

Concentration-Dependent Rheological Properties of Atelocollagen Are Associated with Fibroblast Mechanotransduction and Collagen Remodeling in Aged Skin.

Injectable collagen biomaterials are used to modify the dermal extracellular matrix in aged skin; however, whether concentration-dependent rheological differences are associated with distinct fibroblast responses remain unclear. We compared 3% and 6% atelocollagen (AtCOL) and examined three mechanobiological programs: (i) integrin (ITG) α5β1-ERK-cyclin D1 signaling related to proliferation; (ii) ITGβ1-FAK-YAP signaling related to matrix synthesis; and (iii) ITGα11β1/Tensin-1-positive fibrillar adhesion related to collagen assembly. Under the tested oscillatory shear conditions, 6% AtCOL displa

Dual-loaded homotypic exosomes with endogenous IR808 and pH-responsive DOX drive sequenced chemo-PDT and convert "cold" OSCC tumors "hot".

BACKGROUND: Chemotherapy remains central to oral squamous cell carcinoma (OSCC) management but is constrained by poor specificity, systemic toxicity, multidrug resistance, and limited durability. Tumor-derived exosomes (TEX) offer a clinically relevant carrier owing to their biocompatibility, low immunogenicity, and homotypic targeting; however, prevailing high-efficiency loading methods (electroporation/sonication/extrusion, etc.) may disrupt membrane proteins, promote cargo leakage and aggregation, and increase costs-hindering clinical translation. RESULTS: Here we report IR808/Doxorubicin@

Genomic Investigations Unveil the Genetic Underpinnings of Environmental Adaptation in African Goat Populations.

In the expansive African region, goats display a diverse array of breeds that have evolved over time to adapt to varying local climates and environments. This study involved the analysis of 1591 samples, including 63 domestic breeds and 164 individuals from wild related species, focusing on genetic diversity, population structure, gene flow, selective signals, and landscape genomics. Our findings revealed lower genetic diversity in wild related species, particularly in the mountain goat (Oreamnos americanus). Gene flow was observed between wild Yura goats and North African populations. Introgr

Anti-αvβ3 antibodies in FNAIT are associated with disturbed angiogenesis and intracranial hemorrhage in men and mice.

In Caucasians, maternal allo-immunization against human platelet antigen (HPA)-1a is the most common cause for fetal and neonatal alloimmune thrombocytopenia (FNAIT). Intracranial hemorrhage (ICH) is a serious complication. The lack of a consistent correlation between antibody concentration, severity of thrombocytopenia, and ICH makes it difficult to predict thrombocytopenia in subsequent pregnancies. HPA-1a is located on integrin β3, which forms heterodimers with different alpha chains. The αvβ3 heterodimer is present on endothelial cells (ECs), and a specific antibody interaction with fetal

The Heterogeneity of the Neurovascular Unit in the Optic Nerve Head During Glaucomatous Neurodegeneration Based on Single-Cell RNA Sequencing.

Glaucoma is a multifactorial neurodegenerative disease, and the dysfunction of the neurovascular unit (NVU) in optic nerve head (ONH) has been gradually recognized to contribute to the neurodegeneration progression, yet its heterogeneity features remain poorly understood. Here, we established rat models of high intraocular pressure (HIOP) and lipopolysaccharide-induced ocular inflammation (LPS), and performed single-cell RNA sequencing (scRNA-seq) of ONH-centered tissues to characterize the cellular and molecular characteristics of NVU and its main cell components. Distinct alterations in the

Tumor budding in colorectal cancer: partial EMT, microenvironmental remodeling, and metastatic competence.

Tumor budding (TB) is a pathological hallmark of malignant invasion at the invasive front of colorectal cancer (CRC) and provides a morphologic window into early dissemination. In the International Tumor Budding Consensus Conference (ITBCC) framework, buds are single tumor cells or clusters of up to four cells, graded by hotspot counting to support standardized evaluation. Evidence from histopathology, single-cell profiling, spatially resolved analyses, and functional models links TB to invasion-competent tumor states. TB often tracks partial epithelial-mesenchymal transition, with weakened ce

NG2-ITGA4 axis regulates Rho GTPases and leukemic aggressiveness in KMT2A-r B-ALL and is targetable with natalizumab.

KMT2A-rearranged B-cell acute lymphoblastic leukemia (KMT2A-r B-ALL) is an aggressive subtype of leukemia, characterized by high relapse rates, therapy resistance, and poor prognosis. Although CD19-targeted immunotherapies have significantly benefited patients with relapsed/refractory (R/R) disease, relapses remain common and long-term survival is especially poor in KMT2A-r B-ALL patients. We recently identified the membrane-bound proteoglycan NG2 (CSPG4) as direct transcriptional target of KMT2A fusions, with its expression associated with poor prognosis, early relapse, and glucocorticoid res

αvβ3 CAR-T Cells Simultaneously Targeting Tumor and Metastases Produce Highly Effective Control in Preclinical Models of Melanoma and Triple-Negative Breast Cancer.

One of the many barriers to successful chimeric antigen receptor (CAR) T-cell immunotherapy against solid tumors is the scarcity of targetable molecules common to multiple types of cancer. The purpose of this study was to determine the efficacy of αvβ3 CAR-T cells against solid tumors, particularly melanoma and triple-negative breast cancer, two malignancies recognized for dependence on the αvβ3 pathway for tumor progression and metastases. A novel αvβ3 CAR-T cell developed in our lab and demonstrated to be effective and safe in preclinical models of glioblastoma and pediatric diffuse intrinsi

Beyond attenuation: a translational review of curative-intent pharmacological targets in idiopathic pulmonary fibrosis.

Idiopathic Pulmonary Fibrosis (IPF) is a progressive, fatal lung disease with a median survival of 3-5 years, driven by a profound unmet medical need. The current standard-of-care agents, pirfenidone and nintedanib, merely slow disease progression and are burdened by significant toxicity. This review synthesizes the basic, clinical, and translational evolution of novel pharmacological approaches for IPF, analyzing the critical lessons from recent high-profile clinical trial failures and successes. This review followed a narrative, analytical methodology, examining key Phase 2 and 3 clinical tr

Bioactive hydroxyl-terminated phosphorus dendrimers mediate protein/drug co-delivery for enhanced multi-target ischemic stroke therapy.

The treatment of ischemic stroke (IS) faces significant challenges due to the complex pathophysiology, which encompasses oxidative stress, neuroinflammation, and blood-brain barrier (BBB) dysfunction. Here, we report the development of a bioactive per se hydroxyl-terminated phosphorus dendrimer-based nanoplatform for protein/drug co-delivery to the ischemic brain. We show that through sequential physical complexation and loading, nanocomplexes (NCs) composed of phosphorus dendrimers, a protein drug of fibronectin (FN) with anti-inflammatory/antioxidant/angiogenic properties and a small molecul

Human Cell-Derived Extracellular Matrix Modulates Endothelial Cell Morphology and Metabolism in Response to Fluid Shear Stress.

Endothelial cells integrate biochemical cues from the extracellular matrix (ECM) with mechanical cues from blood flow to regulate vascular function, yet the combined influence of native human ECM complexity and physiologic fluid shear stress remains poorly understood. Here, we developed a custom 3D-printed cone-and-plate rheometer compatible with compliant, heterogeneous, and cell-derived substrates, enabling the application of controlled laminar shear stress to the human cell-derived matrix (hCDM) and hydrogels without structural disruption. Using this platform, we investigated endothelial ce

Tissue-Engineered Brain-Mimetic Niches to Model Braintropic Triple-Negative Breast Cancer Metastasis.

Brain metastases of triple-negative breast cancer (TNBC) rapidly progress, causing severe neurological decline with a median survival of less than 6 months. This tragic disease is often difficult to identify with sufficient time for treatment and is exacerbated by a lack of effective pharmacological intervention. Replicating the biochemical and mechanical properties of the premetastatic niche in vitro is a critical step in expediting the development of new therapeutics. However, a high-fidelity and reproducible model system is needed. To quantify the influence of a brain-mimetic microenvironme

Tanyu Tongzhi Decoction Improves Cardiac Function by Inhibiting Platelet Activation and Alleviating Coronary Microthrombosis for Coronary Heart Disease Mice.

Background: Coronary heart disease (CHD) has a high global disease burden. According to traditional Chinese medicine theory, the main syndrome type of CHD is the syndrome of intermingled phlegm and blood stasis (SI-GPBS). Tanyu Tongzhi Decoction (TYTZD) exerts clear cardioprotective effects on CHD patients with SI-GPBS, while its specific regulatory mechanism remains unclear. Methods: Clinical serum proteomics and network pharmacology were used to screen key targets and pathways for CHD with SI-GPBS. An APOE-/- mouse model of CHD complicated with SI-GPBS was established and treated with TYTZD.

Astaxanthin suppresses thrombosis by inhibiting both platelet activation and blood coagulation.

BACKGROUND AND PURPOSE: Thrombosis, the leading cause of death worldwide, involves both platelet activation and coagulation reaction. Astaxanthin (ASX) is a carotenoid with potent antioxidant activity and beneficial effects to the cardiovascular system. This study aims to systematically elucidate the antithrombotic potential of ASX and the underlying mechanisms. EXPERIMENTAL APPROACH: Rats and mice were treated with ASX by gavage to examine the in vivo effects of ASX. Human platelets and liver cells were directly treated with ASX to evaluate its in vitro effects. Network pharmacology and immu

Altered cytoskeletal integrity underlies impaired platelet shape change and defective thrombus formation in ETV6-related thrombocytopenia.

ETV6-related thrombocytopenia (ETV6-RT) is an inherited platelet disorder caused by germline ETV6 variants. Despite recent progress, the mechanisms underlying platelet dysfunction in ETV6-RT remain unclear. We investigated 12 patients from six families using functional assays, electron microscopy, quantitative proteomics and cytoskeletal imaging. Most patients exhibited mild-to-moderate thrombocytopenia with variable paediatric bleeding symptoms (median International Society on Thrombosis and Haemostasis Bleeding Assessment Tool 3, range 1-9) but consistently mild bleeding in adulthood (median

Taxifolin Inhibits Invasion and Endovascular Differentiation of Extravillous Trophoblast HTR-8/SVneo Cells.

Adequate placental development and function, prerequisites for the development of a healthy fetus, rely on controlled trophoblast invasion into the decidua and remodeling of the spiral arteries. These tightly regulated processes involve epithelial-mesenchymal transition (EMT) and endovascular differentiation of trophoblast cells. Taxifolin (dihydroquercetin), a natural flavonoid with various pharmacological effects, previously showed cytoprotective, antioxidant, and anti-inflammatory activity on trophoblast cells. Given that the literature indicates that this flavonoid suppresses EMT and can a
