Pyroptosis-Related Gene Signatures Can Robustly Diagnose Skin Cutaneous Melanoma and Predict the Prognosis.

Skin cutaneous melanoma (SKCM) is a chronically malignant tumor with a high mortality rate. Pyroptosis, a kind of pro-inflammatory programmed cell death, has been linked to cancer in recent studies. However, the value of pyroptosis in the diagnosis and prognosis of SKCM is not clear. In this study, it was discovered that 20 pyroptosis-related genes (PRGs) differed in expression between SKCM and normal tissues, which were related to diagnosis and prognosis. Firstly, based on these genes, nine machine-learning algorithms were shown to perform well in constructing diagnostic classifiers, includin

Forecasting short-term peak concentrations from a network of air quality instruments measuring PM2.5 using boosted gradient machine models.

Machine learning algorithms are used successfully in this paper to forecast reliably upcoming short-term high concentration episodes, or peaks (<60-min) of fine particulate air pollution (PM2.5) 1 h in advance. Results are from a network around Christchurch, New Zealand, with an objective to forecast the occurrence of short-term peaks using a gradient boosted machine with a binary classifier as the response (1 = peak, 0 = no peak). Results are successful, with 80-90% accurate forecasting of whether a peak in PM2.5 would occur within the next 60-min period. Elevated and variable nitrogen monoxi

Integration of Nitrogen Oxides Into a Triage-Based Index for Predicting Adverse Outcomes in ST-Segment Elevation Myocardial Infarction Patients.

BACKGROUND: Early risk stratification in ST-segment elevation myocardial infarction (STEMI) remains challenging. The Hypoxia-Age-Shock Index (HASI), incorporating SpO₂, age, heart rate, and systolic blood pressure, offers improved prediction over traditional indices but may benefit from machine learning (ML) and environmental data. OBJECTIVES: To compare HASI with established shock indices for mortality prediction in STEMI triage and assess whether adding machine learning and ambient NOx data improves early risk assessment. METHODS: We retrospectively analyzed 711 STEMI patients. HASI was co

XGBoost-based urinary microbial signatures enable non-invasive diagnosis and prognosis for urothelial carcinoma.

BACKGROUND: Urothelial carcinoma (UC) is the most common malignant tumor of the urinary system, characterized by high incidence and recurrence rates, posing a serious threat to human health. While previous studies have linked urinary microbiota alterations to bladder cancer, little is known about the broader spectrum of UC subtypes or their clinical implications. To address this gap, we analyzed microbiota profiles across multiple UC subtypes, incorporating microbial network analyses and machine learning to establish diagnostic models and identify prognostic biomarkers. METHOD: A total of 112

Fully connected network with multi-scale dilation convolution module in evaluating atrial septal defect based on MRI segmentation.

BACKGROUND AND OBJECTIVE: Atrial septal defect (ASD) is a common congenital heart disease. During embryonic development, abnormal atrial septal development leads to pores between the left and right atria. ASD accounts for the largest proportion of congenital heart disease. Therefore, the design and implementation of an ASD intelligent auxiliary segmentation system based on deep learning segmentation of the atria has very important practical significance, which we aim to achieve in this paper. METHODS: This study proposes a multi-scale dilated convolution module, which is composed of three par

Computer-Assisted Pterygium Screening System: A Review.

Pterygium is an eye condition that causes the fibrovascular tissues to grow towards the corneal region. At the early stage, it is not a harmful condition, except for slight discomfort for the patients. However, it will start to affect the eyesight of the patient once the tissues encroach towards the corneal region, with a more serious impact if it has grown into the pupil region. Therefore, this condition needs to be identified as early as possible to halt its growth, with the use of simple eye drops and sunglasses. One of the associated risk factors for this condition is a low educational lev

Employing network toxicology, molecular docking, machine learning, and single-cell analysis to analyze BPA exposure-induced ccRCC.

This study investigates the molecular mechanisms underlying bisphenol A (BPA)-induced clear cell renal cell carcinoma (ccRCC). We integrated transcriptomic data from multiple GEO datasets and performed differential expression analysis and WGCNA to identify BPA-associated candidate genes. Enrichment analyses implicated pathways including cell adhesion, lipid metabolism, arachidonic acid signaling, and innate immune response. Using twelve machine learning algorithms, we identified four core genes (ITGB2, TBXAS1, LIPA, and CLEC7A), all upregulated in ccRCC. Molecular docking suggested stable BPA-

Screening Biomarkers Related to Circadian Rhythm of Gastric Cancer Through Bioinformatics.

Circadian rhythm (CR) dysregulation is increasingly recognized as a contributor to gastric cancer (GC) development, yet the biological and immunological consequences of CR-related alterations remain unclear. In this study, integrative transcriptomic analyses identified 147 CR-related differentially expressed genes in GC. Functional enrichment revealed significant involvement in circadian processes, cytokine activity, and immune-related signaling pathways. By applying three complementary machine learning algorithms, five robust hub genes (INHBA, DEPTOR, PDCD4, SELENBP1, and SIRT4) were identifi

Predictive modeling of corticosteroids sensitivity in sepsis using a supervised learning approach.

Dealing with Sepsis poses a critical challenge in healthcare and necessitates rapid and well-adapted treatment responses. Corticosteroids have been used as a treatment but individual-level effects vary widely. This study aims at improving treatment efficacy by leveraging machine learning techniques to predict patients' sensitivity to corticosteroids. We use two comprehensive datasets of sepsis patients to evaluate two distinct model configurations. These configurations employ Logistic Regression and Random Forest algorithms, both with and without class balancing using the Synthetic Minority Ov

Multi-model machine learning framework for prediction of greenhouse gas emissions during composting of organic solid waste.

Accurate prediction and regulation of greenhouse gas (GHG) emissions during composting are essential for mitigating global warming. Herein, two multi-model machine learning frameworks were developed based on Gradient Boosting Regression, Extreme Gradient Boosting, and Extra Trees Regression algorithms. In particular, the modular multi-model demonstrated superior performance in predicting CO2, CH4, and N2O emissions, achieving R2 values of 0.9662, 0.9729, and 0.9051, respectively. The stacking ensemble model also exhibited high accuracy in cumulative emission prediction (R2 = 0.9278). SHapley A

Oral microbiome-derived biomarkers for non-invasive diagnosis of head and neck squamous cell carcinoma.

Mounting evidence suggests that sustained microbial dysbiosis is associated with the development of multiple cancers, while the species-level bacterial taxa and metabolic dysfunction of oral microbiome in patients with head and neck squamous cell carcinoma (HNSCC) remains unclear. In this cross-sectional study, comprehensive metagenomic and 16S rRNA amplicon sequencing analyses of oral swab samples from 172 patients were performed. Unsupervised clustering algorithms of relative microbial abundance profiles revealed three distinctive microbiome clusters. Based on the metagenomic and 16S rRNA am

Identification of CCR7 as a potential biomarker in polycystic ovary syndrome through transcriptome sequencing and integrated bioinformatics.

Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder, yet its mechanisms remain elusive. This study employed transcriptome sequencing on granulosa cells from 5 PCOS women and 5 controls, followed by bioinformatic analyses. We identified 684 mRNAs and 167 lncRNAs with significant differential expression. Gene Ontology and KEGG analyses highlighted enrichment in immune and inflammatory responses among these genes. Through CytoHubba plug-in and three machine learning algorithms, CCR7 was identified as the hub gene of PCOS, further validated through analysis of GSE65746, GSE34526 and

Logic gate-driven dual-index balanced visualization strategy for tumor metastasis diagnosis.

Exfoliated tumor cells are integral to malignant tumors diagnosis. The process of clinical cytology of exfoliation involves several complex steps that require at least two days of preparation. Here, we develop a balanced-etching visual kit based on concentration differences of Glutathione/Glucose (GSH/Glu) to distinguish normal from exfoliated tumor cells rapidly and accurately. The balanced-etching visualization kit can be used to obtain color cards and screen exfoliated tumor cells initially (within 10 min). Furthermore, by utilizing logic gates and machine learning algorithms for RGB extrac

Machine learning-based prediction of cognitive outcomes in de novo Parkinson's disease.

Cognitive impairment is a debilitating symptom in Parkinson's disease (PD). We aimed to establish an accurate multivariate machine learning (ML) model to predict cognitive outcome in newly diagnosed PD cases from the Parkinson's Progression Markers Initiative (PPMI). Annual cognitive assessments over an 8-year time span were used to define two cognitive outcomes of (i) cognitive impairment, and (ii) dementia conversion. Selected baseline variables were organized into three subsets of clinical, biofluid and genetic/epigenetic measures and tested using four different ML algorithms. Irrespective

A Multichannel Fluorescent Tongue for Amyloid-β Aggregates Detection.

Attention has been paid to the early diagnosis of Alzheimer's disease, due to the maximum benefit acquired from the early-stage intervention and treatment. However, the sensing techniques primarily depended upon for neuroimaging and immunological assays for the detection of AD biomarkers are expensive, time-consuming and instrument dependent. Here, we developed a multichannel fluorescent tongue consisting of four fluorescent dyes and GO through electrostatic and π-π interaction. The array distinguished multiple aggregation states of 1 µM Aβ40/Aβ42 with 100% prediction accuracy via 10-channel s
