Spinal cord injury (SCI), representing a devastating trauma to the central nervous system (CNS), is often accompanied by complex immune responses and metabolic dysregulation, significantly affecting patients' functional recovery and quality of life. In recent years, the gut-spinal axis, as an important pathway connecting the gut microbiome and the CNS, has emerged as a pivotal area of investigation regarding the pathological mechanisms of SCI. Changes in the gut microbiome modulate inflammatory responses and tissue repair processes after SCI by regulating immune system activation and metabolic

Therapeutic plasma exchange (TPE) is a cornerstone extracorporeal therapy with broad immunomodulatory effects across diverse immune-mediated and inflammatory diseases. By removing circulating pathogenic substances such as autoantibodies, immune complexes, complement components, and pro-inflammatory cytokines, TPE rapidly interrupts humoral pathways driving tissue injury. This review summarizes the mechanisms through which plasma exchange modulates immune activity, including both depletion of harmful plasma factors and restoration of normal homeostatic proteins when donor plasma is used. Glioblastoma multiforme (GBM) is the most aggressive form of brain cancer with the very poor survival and high recurrence rate. Tumor-associated neutrophils (TANs) play a pivotal role in regulation of the tumor microenvironment. In this study, we developed a new mathematical model of the critical GBM-TAN interaction in the heterogeneous brain tissue. The model reveals that the dual and complex role of TANs (either anti-tumorigenic N1 and the pro-tumorigenic N2 type) regulates the phenotypic trajectory of the evolution of tumor growth and the invasive patterns in white and gray matter via media

Patients with impaired tumour-specific major histocompatibility complex class I (tsMHC-Iimpaired) often fail to respond to immune checkpoint blockade (ICB), presenting a major clinical challenge. However, through our multicentre investigation, we observed that a subset of patients with tsMHC-Iimpaired remains responsive to ICB, a phenomenon that has not been fully explained. Here we identify a COTL1high natural killer (NK) subset that mediates ICB responsiveness in these patients. Mechanistically, PD-L1+ macrophages coexpress GITRL and engage GITR on COTL1high NK cells, whereas PD-L1 blockade

Microglia maintain brain homeostasis via iC3b-mediated synaptic pruning. The Arp2/3 complex has been implicated in iC3b-mediated macrophage phagocytosis, but it is unclear whether it is similarly required in microglia in the CNS. We examined the question of CR3-dependent clearance of iC3b in microglia using a combination of in vitro and in situ physical confinement studies. Arp2/3 inhibition decreased iC3b phagocytosis and cell motility in vitro. Furthermore, microglia-like cells remove immobilized iC3b from the substrate in an Arp2/3-dependent fashion, in a process reminiscent of trogocytic s

Hypoxia is a hallmark of solid tumors, and hypoxia-inducible factors (HIFs) are the master transcription factors that allow cancer cells to sense and cope with a hypoxic microenvironment. However, the mechanism by which the HIF-mediated transcription process is precisely regulated remains poorly understood. Here, we demonstrate that the RNA helicase DDX21 facilitates breast tumor growth and metastasis by triggering HIF-mediated transcription elongation. Mechanistically, DDX21 induces the release of CDK9 from the 7SK complex, enhancing the interaction between CDK9 and HIF-1α. This interaction s

Acute kidney injury (AKI) is a severe clinical syndrome with limited therapeutic options. Here, we identified a previously unrecognized role of the nuclear pore complex protein POM121 in AKI. Analysis of the public transcriptomic dataset GSE153625, together with qPCR, Western blot, and immunohistochemical validation, demonstrated that POM121 expression was markedly increased in ischemia-reperfusion injury (IRI)-induced AKI. Due to embryonic lethality of homozygous POM121 knockout mice, heterozygous POM121⁺/⁻ mice were used for functional studies. POM121 haploinsufficiency significantly aggrava

Volatile organic compounds (VOCs) emitted by certain fungi have previously been suggested to modulate plant development and their response to stresses. While some fungal species, such as Trichoderma harzianum, are known to produce VOCs, the production of VOCs in other phylogenetically divergent species, such as Mucor mucedo, was previously unknown. This study aimed to analyse the VOCs profiles emitted by these fungal species and examine their effects on the growth and defensive responses of Arabidopsis thaliana. Volatilome analysis revealed that the most abundant VOCs for both species were alc

With a lack of new antimicrobials recently entering the clinic, another tactic to combat antimicrobial resistance is to inhibit the resistance mechanisms to existing antibiotics, prolonging the usefulness of these important drugs. One key clinical antibiotic is fusidic acid which inhibits bacterial protein synthesis by binding to EF-G on the ribosome and preventing its release, stalling protein synthesis. The FusB-family of proteins bind to EF-G and confer fusidic acid resistance by promoting the dissociation of these stalled complexes. This resistance mechanism could be inhibited by preventin

Auxin, a key plant hormone, regulates growth and development through two distinct receptor systems. The well-established transport inhibitor response 1 (TIR1)/auxin-signaling F-box protein (AFBs) pathway mediates transcriptional responses in the nucleus, whereas a distinct non-transcriptional pathway involving the plasma membrane-associated co-receptors auxin binding protein 1 (ABP1)/ABP1-like protein 1 and 2 (ABL1/2) and transmembrane kinases (TMKs) mediates rapid cellular responses. Recent breakthroughs have resolved long-standing debates regarding the apoplastic auxin signaling, highlightin

Macrophages are crucial for host defense against the pathogen. However, pathogens such as Mycobacterium tuberculosis (Mtb) have evolved mechanisms to alter macrophage physiology and exploit these cells as their primary niche. Mtb-infected macrophages upregulate several metabolic pathways including glutamine metabolism. We previously showed that inhibiting glutamine metabolism with the pleiotropic glutamine metabolism antagonist prodrug JHU083 has dual antibacterial and immunomodulatory effects in a mouse model of tuberculosis. In the present study, using single-cell RNA sequencing and LS-MS/MS

Osteoarthritis (OA) is a whole-joint disease characterized by progressive structural degeneration and chronic low-grade inflammation affecting the cartilage, synovium, subchondral bone, and immune compartments. It is a complex degenerative disorder associated with substantial morbidity, heterogeneous clinical trajectories, and limited disease-modifying treatment options. Extracellular vesicles (EVs) have emerged as important mediators of intercellular communication within the OA joint microenvironment, and are implicated in pathophysiological responses to mechanical stress, inflammatory cues,

Cardiovascular-kidney-metabolic (CKM) syndrome is not merely the simple superimposition of isolated target organ lesions, but rather a systemic disorder sustained by a complex network of inter-organ communication. The circadian clock functions as the central spatiotemporal regulator of physiological homeostasis, and the circadian disruption is increasingly recognized as an important contributor to CKM progression. In this review, we systematically elucidate the core mechanisms and biological functions of the circadian clock in mediating inter-organ crosstalk within CKM syndrome. We first delin

Tumor metastasis is the leading cause of mortality in patients with malignant cancers, and the establishment of the pre-metastatic niche (PMN) is recognized as a pivotal step in this process. With growing insight into the tumor microenvironment, the roles of immune cells and stromal mediators in PMN formation have gained increasing attention. This review systematically summarizes the core functions of stromal components and immune cells in PMN development and their coordinated regulatory mechanisms within the metastatic cascade, encompassing macrophages, tumor-associated fibroblasts, neutrophi

Hypertrophic scars (HS) represent a significant clinical challenge due to their complex pathophysiology and resistance to conventional therapies, often resulting in persistent symptoms such as itching, pain, and impaired joint mobility that compromise patients' quality of life. Current treatment modalities, including compression therapy, pharmacological agents, radiation, silicone gel, and laser therapies, have faced limitations primarily due to inadequate drug penetration into the dense fibrotic scar tissue. In this context, microneedle-mediated controlled delivery systems have emerged as a p

Inflammatory bowel diseases (IBD) represent a spectrum of chronic gastrointestinal disorders fueled by a complex multifactorial interplay, where mitochondrial dysfunction has emerged as a critical contributor. The SLC25 family of mitochondrial transporters mediates the exchange of essential metabolites across the mitochondrial inner membrane, serving as pivotal regulators of cellular energy homeostasis in both the intestinal epithelium and immune cells. Accumulating evidence indicates that dysregulation of SLC25-dependent metabolic transport disrupts the crosstalk between cellular metabolism a

The mammalian brain fundamentally relies on precise lipid homeostasis to maintain structural integrity and complex neural signaling. Emerging evidence positions lipid metabolism reprogramming not merely as a secondary pathological byproduct but as a core initiating driver of age-related neurodegenerative diseases. This review systematically evaluates the mechanisms of cerebral lipid dyshomeostasis during brain aging, highlighting glial cells as the central mediators of this pathological cascade. We comprehensively dissect the age-associated "lipid drift", emphasizing apolipoprotein E (APOE)-in

Despite extensive characterisation of immune responses across peripheral organs, the immunological landscape of the central nervous system (CNS) remains incompletely defined. Among the resident immune cell populations in the brain, mast cells and microglia have emerged as key modulators of neuroinflammatory processes. Research has increasingly also shown that these cells contribute to fundamental homeostatic functions, including regulation of sleep, affective states, and energy balance. These cells engage in complex bidirectional communication, mediated in part by the biogenic amine histamine.

Invasive candidiasis, predominantly caused by Candida albicans, threatens immunocompromised individuals with high mortality. Dendritic cell-derived interleukin-1β, orchestrated via Dectin-1-mediated β-glucan recognition, is central to antifungal defense. While the Dectin-1/Syk/CARD9 axis governing Il1b transcription is defined, the epigenetic regulatory mechanisms remain elusive. Here, we identify Complement C1q binding protein (C1QBP) as a critical negative regulator of Dectin-1-mediated interleukin-1β production. C1QBP sequesters phosphorylated protein kinase C δ in the cytoplasm, preventing
