Early adipose tissue wasting in a preclinical model of human lung cancer cachexia.

Cancer cachexia (CC), a syndrome of skeletal muscle and adipose wasting, reduces responsiveness to therapies and increases mortality. There are no approved treatments for CC, which may relate to discordance between preclinical models and human CC. To address the need for clinically relevant models of lung CC, we generated inducible, lung epithelial cell-specific KrasG12D/+ (G12D) mice. G12D mice develop CC over a protracted time course and phenocopy tissue and tumor, cellular, mutational, transcriptomic, and metabolic characteristics of human lung CC. G12D mice demonstrate early loss of adipos

The fibroblast-driven melanoma/Treg vitiligo mouse model is effectively suppressed by IFNγ blocking antibody.

The use of immunosuppressive drugs for vitiligo treatment carries a potential risk of increased infection, whereas IFNγ bispecific antibodies represent an alternative therapeutic strategy. Researchers employing the melanoma/Treg vitiligo mouse model identified a critical role of dermal fibroblasts during the progressive phase of vitiligo. These fibroblasts respond to IFNγ and recruit CD8 + T cells, a mechanism that also contributes to the bilateral symmetrical depigmentation commonly observed in vitiligo patients. To evaluate the suitability of this model as a platform for developing fibroblas

Animal models of SARS-CoV-2 and COVID-19 for the development of prophylactic and therapeutic interventions.

Infections of the Coronavirus SARS-CoV-2 continue to spread around the globe, causing Coronavirus Disease (COVID)-19. Infected people are at risk of developing acute interstitial pneumonia, which can result in lethal complications, particularly in patients with pre-existing co-morbidities. Novel prophylactic and therapeutic interventions are urgently needed to limit the infection-associated health risk for the population and to contain the pandemic. Animal models are indispensable to assessing the efficacy and safety of potential new antivirals, vaccines, and other innovative therapies, such a

New finite element study protocol: Clinical simulation of orthodontic tooth movement.

The aim of this work was to model tooth movement in a more clinically-exact fashion, thanks to the use of new IT tools and imaging systems (cone-beam). Image segmentation and 3D reconstruction now enable us to model the anatomy realistically, while finite element (FE) analysis makes it possible to evaluate stresses and their distribution on the level of the tooth, the periodontal ligament (PDL) and the alveolar bone when a force is applied. The principle is to monitor tooth movement by obtaining optical impressions at each stage of treatment. The model corresponds to a genuine clinical situati

Sub-clustering based recommendation system for stroke patient: Identification of a specific drug class for a given patient.

Stroke is one of the leading causes of death worldwide. Previous studies have explored machine learning techniques for early detection of stroke patients using content-based recommendation systems. However, these models often struggle with timely detection of medications, which can be critical for patient management and decision-making regarding the prescription of new drugs. In this study, we developed a content-based recommendation model using three machine learning algorithms: Gaussian Mixture Model (GMM), Affinity Propagation (AP), and K-Nearest Neighbors (KNN), to aid Healthcare Professio

Distal Radius Fracture Management: Surgeon Factors Markedly Influence Decision Making.

INTRODUCTION: It is our hypothesis that physician-specific variables affect the management of distal radius (DR) fractures in addition to patient-specific factors. METHODS: A prospective cohort study was conducted evaluating treatment differences between Certificate of Additional Qualification hand surgeons (CAQh) and board-certified orthopaedic surgeons who treat patients at level 1 or level 2 trauma centers (non-CAQh). After institutional review board approval, 30 DR fractures were selected and classified (15 AO/OTA type A and B and 15 AO/OTA type C) to create a standardized patient data se

Testing patients with non-specific symptoms for antibodies against Borrelia burgdorferi sensu lato does not provide useful clinical information about their aetiology.

The aim of this study was to determine whether patients with antibodies against Borrelia burgdorferi sensu lato or who report a history of erythema migrans (EM) or tick bite are more likely to have non-specific symptoms such as musculoskeletal pain, fatigue, sensory disorder, and headache. The study group comprised 423 subjects with non-specific symptoms tested for antibodies against B. burgdorferi sensu lato between July 2012 and December 2014 because of suspicion of Lyme borreliosis (LB). Of these, 285 were females (67%) and 138 were males (33%); the median age was 53 years (range, 7-89 year

From regression to machine learning: improving prediction of rectal cancer recurrence.

PURPOSE: Recurrence after curative-intent treatment of rectal cancer remains a major clinical challenge. Conventional regression models may not adequately capture complex clinical interactions, whereas machine learning algorithms may improve predictive accuracy. This study compared regression-based and machine learning models for predicting recurrence using routinely collected clinical data. METHODS: A retrospective cohort of 581 patients with rectal adenocarcinoma treated between 2013 and 2022 at 2 university hospitals in Thailand was analyzed. Seventeen demographic, pathological, and treatm

Effects of inspiratory waveform, inspiratory time, and pause on compartmental energy partitioning in a heterogeneous two-compartment respiratory model.

In mechanically heterogeneous respiratory systems, the temporal pattern of inspiratory flow may influence how airway-opening work is partitioned between mechanically distinct compartments, independently of global mechanical power (MP). Using a simplified time-resolved two-compartment model with common airway-opening pressure, we investigated whether inspiratory waveform, inspiratory time, and end-inspiratory pause affect model-derived compartmental airway-level energy routing, including cases matched for global MP. Mechanical heterogeneity was introduced by varying compliance and resistance ra

Automatic diagnosis of pneumothorax with M-mode ultrasound images based on D-MPL.

PURPOSE: To address the difficulties of M-mode ultrasound images classification in pneumothorax diagnosis and the shortcomings of existing neural network algorithms in this field, we proposed an M-mode ultrasound images classification model based on Disturbed Meta-Pseudo-Labels (D-MPL). METHODS: An M-mode ultrasound image augmentation system was designed to make the model more robust and generalizable. In D-MPL, teacher-generated pseudo-labeling was first taught to students through a soft mask, and additional disturbance data were added to the teacher network. As the loss of the teacher netwo

Machine learning-based prediction of the necessity for the surgical treatment of distal radius fractures.

BACKGROUND: Treatments for distal radius fractures (DRFs) are determined by various factors. Therefore, quantitative or qualitative tools have been introduced to assist in deciding the treatment approach. This study aimed to develop a machine learning (ML) model that determines the need for surgical treatment in patients with DRFs using a ML model that incorporates various clinical data concatenated with plain radiographs in the anteroposterior and lateral views. METHODS: Radiographic and clinical data from 1,139 patients were collected and used to train the ML models. To analyze and integrat

Angioarchitectural features of brain arteriovenous malformation presented with seizures.

Seizures are the second most common manifestations of brain arteriovenous malformations (bAVMs). This study was conducted to investigate the clinical and angioarchitectural features of bAVMs with seizures and provide guidelines for the clinical management of these patients. We collected clinical and radiological data on patients with bAVMs diagnosed by digital subtraction angiography between January 2013 and December 2020 and dichotomized the patients into the seizures and non-seizures groups. We identified differences in demographic and angiographic features. Logistic regression and random fo

Endoscopic third ventriculostomy for adults with hydrocephalus: creating a prognostic model for success: protocol for a retrospective multicentre study (Nordic ETV).

INTRODUCTION: Endoscopic third ventriculostomy (ETV) is becoming an increasingly widespread treatment for hydrocephalus, but research is primarily based on paediatric populations. In 2009, Kulkarni et al created the ETV Success score to predict the outcome of ETV in children. The purpose of this study is to create a prognostic model to predict the success of ETV for adult patients with hydrocephalus. The ability to predict who will benefit from an ETV will allow better primary patient selection both for ETV and shunting. This would reduce additional second procedures due to primary treatment f

Prediction of postoperative pulmonary complications on the basis of preoperative risk factors in patients who had undergone coronary artery bypass graft surgery.

BACKGROUND AND PURPOSE: Pulmonary complications are among the most frequently reported complications after coronary artery bypass graft (CABG) surgery. However, the risks of postoperative pulmonary complications (PPCs) are not equal for all patients. The aim of this study was to develop a model, based on preoperative factors, for classifying patients with high and low risks for PPCs in order to implement tailored interventions. SUBJECTS AND METHODS: Postoperative pulmonary complications were examined in 117 adult patients who had undergone elective CABG surgery at the University Medical Centr
