Fatty acid vesicles (FAVs) are closed lipid bilayer nanostructures self-assembled from fatty acids and their ionised derivatives, featuring facile preparation, low cost and excellent biocompatibility, which endows them promising prospects in drug delivery. This study systematically compares FAVs with conventional phospholipid liposomes, highlighting the core limitations of FAVs including high pH sensitivity and poor structural stability in physiological environments. Common stabilisation strategies are summarised, mainly covering chemical modification and amphiphilic additive incorporation. Cl

Breast cancer constitutes a metabolically heterogeneous disorder wherein enzymes associated with lipid metabolism are instrumental in tumor progression, adaptation to the microenvironment, and therapeutic response. Beyond their catalytic roles in fatty acid, cholesterol, and phospholipid metabolism, these enzymes may function as regulatory nodes linking lipid flux to membrane remodeling, lipid peroxide detoxification, autophagy-dependent recycling, epithelial-mesenchymal plasticity, tumor-macrophage crosstalk, epigenetic stabilization, and multidrug resistance (MDR). This review adopts a subty

As a major source of essential polyunsaturated fatty acids, fish oil faces increasing sustainability constraints, underscoring the need for viable lipid alternatives. Volatile organic compounds and lipidomic transformations of fish oil (FO) and algal oil (AO) were characterized. In both oils, aldehydes, ketones, and alcohols constituted the predominant classes, and their relative proportions were broadly comparable. As oxidation temperature increased, FO and AO displayed similar trends in flavor development and lipid compositional changes. FO preferentially developed fatty-green and roasted no

Renal tubular epithelial cells (RTECs) are highly metabolically active and vulnerable to pathological stress. Calcium oxalate (CaOx) crystals disrupt lipid metabolism and damage RTECs, thereby promoting nephrolithiasis. We found that SIRT5, a highly expressed desuccinylase in RTECs, was markedly reduced in kidneys from patients with CaOx stones and in mouse models. Multi-omics analyses integrating succinylomics, lipidomics, and proteomics showed that SIRT5 maintains lipid metabolic homeostasis by desuccinylating key fatty acid oxidation (FAO) enzymes. SIRT5 deficiency impaired FAO, causing pol

PEGylated liposomes are widely used as drug delivery carriers due to their prolonged circulation and enhanced accumulation at pathological sites. However, repeated administration can trigger the accelerated blood clearance (ABC) phenomenon, reducing delivery efficacy. Herein, we report a zwitterionic PEGylation strategy by grafting glutamic acid-lysine (EK) peptides onto PEGylated phospholipid derivatives to assemble liposome (Lip)-based drug delivery systems. Small-angle neutron scattering analysis confirmed that EK modification significantly enhanced Lip hydration, leading to a 40-fold reduc

Alzheimer's disease (AD), a progressive neurodegenerative disorder with a rising global prevalence, is pathologically characterised by the presence of amyloid-β (Aβ) plaques and neurofibrillary tangles (NFTs). These lesions lead to synaptic damage, neuronal loss, and cognitive impairment. Despite the recent approval of immunotherapies for AD treatment, their limited efficacy highlights the urgent need for exploring novel disease mechanisms and developing targeted therapeutic strategies. Annexin A2 (ANXA2), a calcium-dependent phospholipid-binding protein, participates in diverse physiological

The peripheral myelin protein P2 (also known as PMP2/FABP8) has two basic functions in vivo: the transport of fatty acids within the cell to the target compartments and the adhesion of lipid bilayers in the peripheral nervous system myelin to each other. In this work, we focus on the membrane-binding properties of P2 and its Charcot-Marie-Tooth disease variants. Experiments were carried out on lipid monolayers at the air-water interface as a membrane model system, with the lipid composition of the cytoplasmic leaflet of myelin. Our study provides quantitative data on the membrane affinity of P

Lipid droplets (LDs) are intracellular organelles that dynamically regulate lipid and energy homeostasis, mediate hormone production, produce inflammation signaling, while also participating in numerous biological processes and activities. Dysregulation of LD homeostasis is linked to various diseases, such as lipodystrophy, obesity, type 2 diabetes (T2D), cardiovascular diseases (CVD), metabolic dysfunction-associated steatotic liver disease (MASLD), neuronal diseases, and among others. The core of LDs consists of neutral lipids, including triacylglycerol (TAG), cholesteryl esters (CE), and re

The brief lifetime of reduced O2 levels and oxygen species during ferroptosis in cancer cell lines undermines the ferroptosis efficacy. Enhancing the effectiveness of real-time created active oxygen species is crucial for pancreatic adenocarcinoma. The Evofosfamide (EFA) encapsulated polydopamine (PD)-iron (Fe) nanoparticles (Fe/PD-EFA) were modified with unsaturated liposomes (Lipo) to address the limitations of conventional ferroptosis therapy. The Lipo@Fe/PD-EFA can interact with H2O2 to generate •OH, which subsequently targets Lipo, resulting in the formation of a phospholipid peroxide rad

Alzheimer's dementia (AD) is a disease of the ageing brain. It begins in the hippocampal region with the epicentre in the entorhinal cortex, then gradually extends into adjacent brain areas involved in memory and cognition. The events which initiate the damage are unknown and under intense investigation. Localization to the hippocampus can now be explained by anatomical features of the blood vessels supplying this region. Blood supply and hence oxygen delivery to the area are jeopardized by poor flow through narrowed arteries. In genomic and metabolomic studies, the respiratory chain and mitoc

Myofiber regeneration and membrane repair play crucial roles in maintaining the continuous physiological functioning of the neuromuscular system. A swift and efficient repair mechanism enables the rapid restoration of sarcolemmal integrity following cellular impairment in damaged skeletal muscles. Members of the annexin family of proteins, which are characterized by the peripheral binding to acidic phospholipid membranes, are intrinsically involved in this myofiber repair process. The biochemical and proteomic profiling of dystrophinopathy, a severe and highly progressive neuromuscular disorde

Despite the expanding clinical application of second-generation anti-androgens like enzalutamide (ENZ) in hormone-sensitive prostate cancer (HSPC), therapeutic resistance culminating in castration-resistant prostate cancer (CRPC) persists as an unresolved clinical crisis. Through comprehensive single-cell transcriptomic profiling of ENZ-naïve and ENZ-treated tumors, an expansion of ENZ-resistant myofibroblastic cancer-associated fibroblast (designated STEAP4+ myoCAF) is identified that correlates with adverse clinical outcomes. Strikingly, STEAP4+ myoCAF demonstrated intrinsic ENZ resistance t

The hypothalamus monitors blood glucose levels and regulates glucose production in the liver. In response to hypoglycemia, glucose-inhibited (GI) neurons trigger counterregulatory responses (CRRs), which stimulate the release of glucagon, epinephrine, and cortisol to elevate blood glucose. Recurrent hypoglycemia (RH), however, reduces the effectiveness of these CRRs. This study examined the role of hypothalamic prostaglandins in glucose recovery during acute hypoglycemia and RH. Imaging mass spectrometry and liquid chromatography/mass spectrometry showed phospholipid and prostaglandin levels i

Low-density lipoprotein receptor-related protein 8 (LRP8) is a crucial regulator of lipid metabolism and is implicated in the development and treatment of various cancers. However, its role in bladder cancer (BCa) remains unknown. We analyzed LRP8 expression in BCa using the TCGA database and clinical samples. We manipulated LRP8 expression in tumor cell lines using siRNA or overexpression plasmid transfection. Cell proliferation, migration, invasion, apoptosis, and drug resistance were assessed through CCK-8, transwell, flow cytometry, and IC50 assays. Additionally, a rescue experiment confir

Wounding triggers complex secondary metabolic pathways in plants, including benzylisoquinoline alkaloid (BIA) biosynthesis in opium poppy (Papaver somniferum L.). This study explores transcriptional and metabolic responses to wounding and methyl jasmonate (MeJA) treatment, focusing on BIA biosynthesis and regulatory mechanisms. Real-time expression analysis revealed significant up-regulation of transcripts in the (S)-reticuline and papaverine biosynthetic pathway, while the noscapine pathway was suppressed. The morphinan pathway also showed transcriptional activation, except in the case of cod

Radiolabeled nanocarriers that can simultaneously deliver therapeutic and diagnostic agents are promising for targeted cancer therapy. In this study, we employed molecular dynamics and metadynamics simulations to investigate how the chemical identity of drug molecules influences the behavior of technetium-99 m (⁹⁹ᵐTc)-loaded carbon nanotube (CNT) nanocarriers at the atomic scale. Four clinically relevant drugs-methotrexate, diclofenac, ketotifen, and piroxicam-were conjugated to a single-walled CNT (SWCNT) via a bifunctional chelating group. Our results revealed that drug structure significant

Immunotherapies such as immune checkpoint inhibitors are effective in treating several advanced cancers, but these treatments have had limited success in metastatic ovarian cancer. Here we engineered liposomal nanoparticles carrying a poly-ʟ-arginine/poly-ʟ-glutamate coating that promotes their binding and retention on the surface of ovarian cancer cells. Covalent anchoring of the potent immunostimulatory cytokine interleukin-12 (IL-12) to phospholipid headgroups of the liposome core enabled the polymer-coated particles to concentrate IL-12 in disseminated ovarian cancer tumours following intr
