Retinoic acids regulate apoptosis of T lymphocytes through an interplay between RAR and RXR receptors.

Vitamin A deficiency has been known for a long time to be accompanied with immune deficiency and susceptibility to a wide range of infectious diseases. Increasing evidence suggests that retinoic acids derived from vitamin A are involved in the functional regulation of the immune system. Of the two groups of retinoid receptors, retinoic acid receptors (RARs) and retinoid X receptors (RXRs) all-trans and 9-cis retinoic acids are high affinity ligands for RARs and 9-cis retinoic acid additionally binds to RXRs. In cells, at high concentrations, all-trans retinoic acid can be converted to 9-cis re

Multisite phosphorylation and the integration of stress signals at p53.

The p53 tumour suppressor protein is a potent transcription factor that plays a major role in the defence against tumour development. p53 exists in a latent form that can be activated by a range of stresses including DNA damage, hypoxia, cytokines, metabolic changes, viral infection, and activated oncogenes. Activation of p53 can lead to cellular growth arrest prior to entry into either S phase or mitosis or can trigger cell death through apoptosis. The modification of p53 by multisite phosphorylation provides a potential link between stress signalling and the regulation of p53 activity, and t

Light control of the duration of the daily melatonin signal under long and short days in the Soay ram. Role of inhibition and entrainment.

Light acts in two ways to control the duration of the nocturnal melatonin rhythm. It inhibits the production of melatonin from the pineal gland and it entrains the underlying circadian rhythm generators located in the suprachiasmatic nuclei. To investigate the role of these two mechanisms under long and short days, four experiments were carried out using groups of adult Soay rams (n = 6-8). The animals were housed in individual pens in light-controlled rooms and entrained to long (LD 16:8) or short (LD 8:16) days for at least 8 wk. The treatments were as follows: (i) dark period extended by 4

Eyespot development on butterfly wings: the focal signal.

The eyespot colour pattern on butterfly wings is specified in the early pupal epidermis by signals from a central "focus." In Bicyclus anynana we show that a small square of focal epidermis, grafted to a range of distal wing sites, induces eyespot formation in surrounding host tissue. Signaling is limited to the focus, and even an adjacent (parafocal) graft does not maintain its normal fate (of contributing to the eyespot) and does not influence its surroundings. Along the wing, there is an abrupt change in the epidermis, as a focus grafted to a proximal site provokes no host response. The res

N-Myc and STAT interactor as a context-dependent switch in innate antiviral immunity.

N-Myc and STAT interactor (NMI), initially identified as a Myc- and STAT-associated protein, is increasingly recognized as a regulatory node in innate antiviral immunity. Current evidence indicates that NMI does not exert a fixed antiviral or proviral effect. Instead, its functional output is shaped by interacting partners, signaling pathway context, subcellular localization, and stage of infection. In selected acute RNA virus models, NMI suppresses IRF7-dependent type I interferon (IFN-I) signaling. In the IAV model, this involves an NMI-IFP35 complex coupled to the TRIM21-IRF7 axis that prom

Network Topology and Interactomic Analysis Reveal the Regulatory Framework of the Humanin Protein Family (MTRNR2Lx Class).

This study presents an in-depth analysis of an interactome comprising approximately 1033 nodes, focusing on its topology, reliability, and functional implications, with particular attention to the small mitochondrial proteins of the Humanin family and their nuclear-encoded MTRNR2Lx paralogs. The analysis, conducted through stringent high-reliability filters and experimentally supported interaction data, produced a curated network model in which approximately 70% of the retained interactions were supported by experimental evidence, providing a solid basis for network-based functional interpreta

The emerging role of the Hippo signaling pathway in interorgan crosstalk.

Interorgan communication has emerged as a fundamental mechanism for maintaining systemic homeostasis, and its disruption contributes to the development of metabolic diseases, chronic inflammation, and cancer. The Hippo signaling pathway, traditionally known for controlling organ size, has recently been redefined as a context-dependent coordinator of systemic cues. By translating hormonal, metabolic, and microbial signals into coordinated cellular responses, Hippo signaling serves as a bidirectional communication hub that not only interprets systemic inputs but also generates "outputs" that con

SLAMF1 at the crossroads of immunity and disease: Biology, pathology, and therapeutic opportunities.

Signaling lymphocytic activation molecule family member 1 (SLAMF1/CD150) is a multifunctional receptor that regulates both innate and adaptive immunity. Through homophilic interactions and SAP-dependent signaling, SLAMF1 supports invariant natural killer T (iNKT) cell selection, γδ T cell polarization, and natural killer (NK) cell education. It also modulates germinal center dynamics and humoral responses, often in cooperation with other SLAM receptors. Beyond lymphoid compartments, SLAMF1 functions as a microbial sensor in macrophages, promotes phagosome maturation, reactive oxygen species pr

Progranulin as a context-dependent regulator of innate immunity in bacterial infections.

The outcome of bacterial infections depends on a delicate balance between effective pathogen clearance and the containment of immune-mediated damage. Disruption of this equilibrium promotes both microbial persistence and immunopathology, a phenomenon particularly relevant in severe respiratory infections and chronic diseases such as tuberculosis. In this context, progranulin (PGRN) has emerged as a key regulator of the innate immune response. PGRN is a secreted glycoprotein with pleiotropic functions in inflammation, tissue repair, and cell survival, exerting its effects through modulation of

Beyond skin deep: exploring the complex molecular mechanisms and holistic management strategies of vitiligo.

Vitiligo is a multifactorial skin disorder characterized by the progressive loss of pigmentation due to the destruction of melanocytes, affecting 0.5-2% of the global population. This condition not only impacts physical appearance but also has profound psychosocial effects on patients. In this review, various aspects of vitiligo are explored, including its clinical forms, epidemiology, and underlying mechanisms. Advances in research have identified key molecular pathways, such as Wnt/β-Catenin, JAK-STAT, and AhR signaling, which are essential for melanocyte survival and immune regulation. Thes

Hippo signaling in mammalian reproduction.

ABSTRACT: Chronic endometritis, defined by chronic inflammation of the endometrium, remains a clinical and biologic challenge even using hysteroscopy allowing a direct vision of the uterine cavity without anesthesia, and conventional histology using Hematoxylin and Eosin staining. Our primary objectives were to evaluate the relevance of hysteroscopy and conventional histology compared to immunohistochemical expression of syndecan-1 (CD138, a marker of plasma cells), which is a heparan sulfate proteoglycan involved in inflammation and enables diagnosis of chronic endometritis. The second object

The role of suppressor of cytokine signaling 3 in inflammatory bowel disease and its associated colorectal cancer.

Inflammatory bowel disease (IBD) and colorectal cancer (CRC), as two of the major human intestinal diseases, provide challenges for the medical field. Suppressor of cytokine signaling 3 (SOCS3), a protein molecule that negatively regulates cytokine signaling through multiple pathways, is involved in the regulation of various inflammatory diseases and tumors. In IBD, SOCS3 acts on a variety of cells to repair mucosal damage and balance the immune response, including epithelial cells, macrophages, dendritic cells, neutrophils, and T cells. In CRC, SOCS3 is inextricably linked to tumor cell proli

Pax6 affects Ras-Raf-ERK1/2 in mouse aging brain.

Pax6, a transcription factor and multifunctional protein, changes during aging. It also interacts with regulator proteins involved in cell metabolism and survival signalling pathways including Ras-GAP. Many forms of Ras, Raf and ERK1/2 are known but information on their region-specific expression patterns are unavailable from brain during aging. Therefore, it has been intended to evaluate expressions of Pax6 and forms of Ras, Raf, ERK1/2 in hippocampus, caudate nucleus, amygdale, cerebral cortex, cerebellum and olfactory lobe. Association of Pax6 with Ras, Raf and ERK1/2 was evaluated in co-cu

Molecular Clusters and Tumor-Immune Drivers of IgM Monoclonal Gammopathies.

PURPOSE: IgM monoclonal gammopathy of undetermined significance (MGUS) and Waldenström macroglobulinemia (WM) represent a disease spectrum with highly varied therapeutic management, ranging from observation to chemoimmunotherapy. The current classification relies solely on clinical features and does not explain the heterogeneity that exists within each of these conditions. Further investigation is warranted to shed light on the biology that may account for the clinical differences. EXPERIMENTAL DESIGN: We used bone marrow (BM) clonal CD19+ and/or CD138+ sorted cells, matched BM supernatant, a

Not your Mother's MAPKs: Apicomplexan MAPK function in daughter cell budding.

Reversible phosphorylation by protein kinases is one of the core mechanisms by which biological signals are propagated and processed. Mitogen-activated protein kinases, or MAPKs, are conserved throughout eukaryotes where they regulate cell cycle, development, and stress response. Here, we review advances in our understanding of the function and biochemistry of MAPK signaling in apicomplexan parasites. As expected for well-conserved signaling modules, MAPKs have been found to have multiple essential roles regulating both Toxoplasma tachyzoite replication and sexual differentiation in Plasmodium

The role of the Hippo pathway in heart disease.

Heart disease, including coronary artery disease, myocardial infarction, heart failure, cardiac hypertrophy, and cardiomyopathies, is the leading causes of death worldwide. The Hippo pathway is a central controller for organ size and tissue growth, which plays a pivotal role in determining cardiomyocytes and nonmyocytes proliferation, regeneration, differentiation, and apoptosis. In this review, we summarize the effects of the Hippo pathway on heart disease and propose potential intervention targets. Especially, we discuss the molecular mechanisms of the Hippo pathway involved in maintaining c

The role of humanin in the regulation of reproduction.

Humanin, a mitochondria-derived peptide, has been found to exert variously protective function in many tissues, especially in the nervous tissues. However, relatively limited studies have focused on the role of humanin in the regulation of reproduction. Current observations indicate that humanin plays an important role in regulating the response of the cell to oxidative stress and apoptosis in ovaries and testes via the modulation of several signaling pathways, especially when the body is in an abnormal state. Even so, the detailed mechanism of humanin function needs to be explored urgently. I
