NRF2 coordinates ferroptosis and disulfidptosis in dermal papilla cells via redox metabolic reprogramming in androgenetic alopecia.

Dysfunction of dermal papilla cells (DPCs) is a central feature of androgenetic alopecia (AGA), in which oxidative imbalance plays a critical role. Nuclear factor erythroid 2-related factor 2 (NRF2) is a master regulator of cellular redox homeostasis; however, its role in AGA remains unclear. Here, using primary DPCs, hair follicle organoids, and dihydrotestosterone (DHT)-induced mouse models, we found that NRF2 was markedly downregulated under AGA-associated conditions. Mechanistically, our data suggest that reduced NRF2 activity is associated with two distinct regulated cell death-related pr

Integrin Signaling Imbalance in Periodontitis: A Stage-Dependent Link Between Inflammation, Bone Resorption and Regenerative Failure.

Periodontitis is a chronic inflammatory disease driven largely by dysregulated host responses that lead to destruction of periodontal tissues. Integrins are heterodimeric transmembrane receptors that regulate cell adhesion and bidirectional signaling in epithelial cells, immune cells, periodontal ligament fibroblasts, and osteoclasts. During disease progression, integrin-related responses may shift across overlapping molecular phases. Epithelial integrins such as α3β1 and α6β4 support barrier integrity, whereas α5β1 may facilitate microbial interaction and inflammatory signaling. β2 integrins

Plasma Copper and Zinc Levels in Ataxia-Telangiectasia.

Ataxia-Telangiectasia (A-T) is a rare neurodegenerative multisystem disease caused by mutations in the A-T Mutated (ATM) gene resulting in cerebellar ataxia, immunodeficiency and an increased cancer risk. Copper and zinc play important roles in similar cellular processes as the ATM protein, such as cell growth, apoptosis, and oxidative stress. This study aimed to explore copper and zinc levels in individuals with A-T since imbalances in these trace elements may contribute to the clinical features commonly seen in A-T and therefore be a target for novel therapies; furthermore we aimed to assess

Toxoplasma gondii SIR2A Promotes Tachyzoite Invasion via Lysine Deacetylase Activity.

Accumulating evidence indicates that reversible acetylation modifications are critically involved in the regulation of gene expression, metabolic pathways, and the tachyzoite transformation in Toxoplasma gondii. Silent information regulator 2 (SIR2), an NAD+-dependent lysine deacetylase, contributes to the maintenance of specific silent chromatin domains in mammals. However, its exact functions in T. gondii are unclear. In this study, we systematically analyzed two SIR2 homologs, TgSIR2A, and TgSIR2B, in T. gondii. Our findings demonstrate that TgSIR2A exerts a more pronounced effect on tachyz

The nuclear basket nucleoporin MLP1 is required to maintain nuclear integrity, and mitotic fidelity in Trypanosoma brucei.

Trypanosoma brucei, a divergent eukaryote parasite, is responsible for neglected tropical diseases in humans and animals, specifically sleeping sickness or human African trypanosomiasis and nagana. Beyond its scientific significance, a comprehensive understanding of its biology has substantial medical and economical implications. Nuclear pore complexes (NPCs) are large multiprotein channels embedded in the nuclear envelope that regulate nucleocytoplasmic transport. In addition to this critical function, NPCs are involved in essential nuclear processes such as chromosome segregation, transcript

Atherosclerosis: Molecular Pathogenesis and Therapeutic Interventions.

Atherosclerosis is a multifactor condition and driving force behind many cardiovascular diseases. Moreover, it remains a leading cause of mortality worldwide. The current therapies, including statins and fibrates, focus mainly on lipid lowering. However, this condition is characterized by various biological pathways, particularly the oxidized LDL uptake by macrophages, chronic inflammation, or vascular smooth muscle cells proliferation, migration, and phenotypic switching, which forces the researchers to explore the numerous molecular pathways standing behind those processes. Despite a lot res

Multiscale modeling of host-pathogen interactions and mucociliary clearance during non-tuberculous mycobacterial pulmonary infection.

Non-tuberculous mycobacterial (NTM) infections are a clinical challenge in cystic fibrosis (CF), where impaired mucociliary clearance and altered mucus rheology promote bacterial colonization despite host immune responses. Understanding how bacterial growth, immune cell dynamics, and mucus transport regulate infection progression is difficult because these processes interact across spatial and temporal scales. We develop a computational framework bridging a mechanistic agent-based model (ABM) of NTM infection with a spatially resolved partial differential equation (PDE) model. The PDE model co

Human brain organoids record the passage of time over multiple years.

The human brain develops and matures over an exceptionally prolonged period of time that spans nearly two decades of life. Processes that govern species-specific aspects of human postnatal brain development are difficult to study in animal models1. While human brain organoids offer a promising in vitro model, they have thus far been shown to largely mimic early stages of brain development. Here we develop human brain organoids for 5 years in culture, optimizing growth conditions to extend excitatory neuron viability beyond previous limits. Using maturation-associated modules derived from endog

Dose-dependent duality: Extracellular vesicles shift from metabolic facilitators to cytotoxic effectors during anaerobic denitrification.

Extracellular vesicles (EVs) mediate critical intercellular interactions, yet their special functions in anaerobic denitrification process remain poorly understood. Utilizing the model denitrifier Pseudomonas aeruginosa PAO1, we investigated how environmental oxygen and nitrate gradients reprogram vesicular cargo and functionality. Proteomic profiling revealed a clear condition-specific cargo: aerobically generated EVs selectively packaged virulence factors and nutrient acquisition proteins. In contrast, imposing severe bioenergetic stress via anaerobic denitrification forced a radical composi

Copper-free clickable alginate microcapsules promote early osteogenic differentiation by in situ conjugation of RGD-DWIVA branched peptides.

Extracellular environments regulate cell proliferation, differentiation, and function through dynamic interactions between cells and the extracellular matrix (ECM), as well as through soluble biochemical cues such as growth factors, all of which are governed by coupled biochemical reactions and mass transport processes. To engineer such environments in a controllable manner, we developed azide-cyclooctyne clickable alginate microcapsules that allow bioactive peptides to be introduced by in situ conjugation via strain-promoted azide-alkyne cycloaddition (SPAAC). Mitochondrial metabolite transporters at the crossroads of metabolic reprogramming, epigenetic regulation and therapeutic vulnerabilities in colorectal cancer.

Metabolic reprogramming is a defining hallmark of CRC. The Warburg effect is the principal metabolic feature of CRC cells, wherein glucose is preferentially catabolized into lactate to sustain accelerated proliferation. In parallel, CRC cells exhibit strong glutamine reliance to replenish tricarboxylic acid (TCA) cycle intermediates required for adenosine triphosphate (ATP) production, lipid biosynthesis and redox homeostasis. Consequently, mitochondria play a central role in supporting the augmented biosynthetic and energetic demands beyond basal energy homeostasis. In this regard, the mitoch

Transcriptional reprogramming in oral squamous cell carcinoma.

Oral squamous cell carcinoma (OSCC) is a prevalent form of cancer globally. This disease is characterized by its complex genetic underpinnings, involving the intricate regulation of multiple genes. Genetic factors influence cellular processes such as growth, differentiation, and apoptosis of oral mucosal cells, thereby promoting or inhibiting tumor formation and progression. Furthermore, environmental factors-including smoking, alcohol consumption, and human papillomavirus (HPV) infection-can significantly increase the risk of developing OSCC. These external influences can impact the disease i

Functional redundancy between penicillin-binding proteins during asymmetric cell division in Clostridioides difficile.

Peptidoglycan synthesis is an essential driver of bacterial growth and division. The final steps of this crucial process involve the activity of the SEDS family glycosyltransferases that polymerize glycan strands and the class B penicillin-binding protein (bPBP) transpeptidases that cross-link them. While many bacteria encode multiple bPBPs to perform specialized roles during specific cellular processes, some bPBPs can play redundant roles that are important for resistance against certain cell wall stresses. Our understanding of these compensatory mechanisms, however, remains incomplete. Endos

Miltefosine and amphoterin B induce membrane rigidity in Leishmania amazonensis and Leishmania-infected macrophages.

Miltefosine (MTF) and amphotericin B (AmB), drugs approved for leishmaniasis treatment, induce membrane rigidity in Leishmania amazonensis at concentrations that inhibit parasite growth, as demonstrated through spin-probe electron paramagnetic resonance (EPR) spectroscopy. Notably, the rigidity induced by MTF is not due to its direct interaction with the membrane, as shorter incubation periods instead increase fluidity. However, measurements taken following short-term drug exposure reflect conditions before possible oxidative stress has fully developed. AmB causes membrane rigidity, but only a

Limited Proteolysis as a Regulator of Lymphatic Vessel Function and Architecture.

Recent advances have highlighted the multifaceted roles of the lymphatic vasculature in immune cell trafficking, immunomodulation, nutrient transport, and fluid homeostasis. Beyond these physiological functions, lymphatic vessels are critically involved in pathologies such as cancer metastasis and lymphedema, rendering their structural and functional regulation of major interest. Emerging evidence suggests that limited proteolysis is a key regulatory mechanism for lymphatic vascular function. In dyslipidemic conditions, dysregulated calpain activity impairs lymphatic trafficking and destabiliz

Approach to the Child and Adolescent With Adrenal Insufficiency.

The management of adrenal insufficiency (AI) is challenging, and the overall goals of treatment are to prevent life-threatening adrenal crises, to optimize linear growth, to control androgen levels without overdosing in patients with congenital adrenal hyperplasia (CAH), and to improve quality of life in affected individuals. Standard glucocorticoid formulations fail to replicate the circadian rhythm of cortisol and control the adrenal androgen production driven by adrenocorticotropin. To personalize and tailor glucocorticoid therapy and to improve patient outcomes, new pharmacological strateg

Histopathological analysis of Artemia franciscana nauplii under different forms of mercury stress.

Inorganic mercury (Hg) and methylmercury (MeHg) have emerged as global pollutants owing to their long-term environmental stability and bioaccumulation. These heavy metals enter aquatic systems via industrial emissions, coal combustion, and natural processes, posing a serious threat to ecosystems and human health. This study assesses the impact of Hg2+ and MeHg on the growth and development of brine shrimp (Artemia franciscana) nauplii by analyzing the histopathological effects on their tissues. Brine shrimp nauplii from the Bohai Bay in China were selected as the study subjects and exposed to

TGFBI Facilitates Myogenesis and Limits Fibrosis in Mouse Skeletal Muscle Regeneration.

Skeletal muscles are essential for movement and support but are vulnerable to injury. Muscle regeneration relies on the extracellular matrix (ECM), which regulates key cellular processes. Transforming growth factor β-induced (TGFBI), an ECM component involved in cell adhesion, migration, and tissue development, has not been investigated in skeletal muscle regeneration. Here, we examined the role of TGFBI using Tgfbi knockout (KO) mice and C2C12 myoblasts. In vitro, C2C12 cells were treated with recombinant TGFBI following snake venom (SV)-induced injury, and myogenic differentiation and fusion

MiR-378 mediates the ovariectomy induced bone loss via exaggerating osteoclastogenesis and transforming growth factor beta impaired osteogenesis.

Osteoporosis (OP) is a disease characterized by decreased bone mass and damaged architectures. The promising treatment strategy for OP is to inhibit bone resorption while promoting bone formation. MicroRNAs (miRNAs) have been shown to be associated with osteoclastogenesis and osteogenesis processes in OP. In our previous study, we discovered that miR-378 inhibits bone marrow mesenchymal stem cell (BMSC) osteogenesis and bone formation during fracture healing. However, the role of miR-378 during OP progression is not validated. In this study, we found that miR-378 transgenic (Tg) mice exhibited
