As a major cause of cancer-related morbidity in women, cervical cancer is strongly related to chronic infection by high-risk human papillomavirus (HPV). This viral persistence also underpins the pathogenesis of a broad spectrum of cancers. In this review, we summarize the research progress of prophylactic and therapeutic HPV vaccines in clinical trials. Furthermore, we present a critical analysis of future challenges, specifically discussing the type replacement phenomenon and its potential implications for long-term vaccine efficacy. We also expand the discourse to a pan-gender and pan-cancer

Functional integration of transplanted cells with host tissue remains a major challenge in cell-based therapies for tissue damage in organs with complex structures, such as exocrine glands. In this study, we investigated whether salivary gland organoids derived from human induced pluripotent stem cells (hiPSCs) could be integrated into injured salivary glands using cell sheet engineering. Cell sheet engineering has demonstrated therapeutic potential in a range of organs, including the heart, retina, and lungs. We found that hiPSC-derived salivary gland organoids contain long-term maintainable

Immune effector cell-associated neurotoxicity syndrome (ICANS) is a serious complication of chimeric antigen receptor T-cell (CAR-T) therapy, associated with significant morbidity and mortality. While corticosteroids and anakinra are cornerstones of treatment, a subset of patients develop severe, steroid-refractory ICANS, highlighting a critical need for more effective therapies. We present the case of a 51-year-old male with relapsed/refractory Philadelphia chromosome-positive (Ph+) B-cell acute lymphoblastic leukemia (B-ALL) who developed grade 4 ICANS following brexucabtagene autoleucel CAR

Antiphospholipid antibody syndrome (APS) is a vasculopathy with recurrent thrombosis and/or miscarriages mediated by autoantibodies against PL-binding proteins (aPL), mainly beta2glycoprotein I (β2GPI). While clotting is the key in vascular APS, thrombosis is not critical for placenta pathology. Despite that, anticoagulant/antiplatelet drugs are the leading treatments, but this is not a 'one-size-fits-all' therapy, and recurrences are reported. Additional therapies (e.g. antimalarials, statins) and a better characterization of the individual risk profile may improve the outcome. Nevertheless,

The protein tyrosine kinase-7 (PTK7) is an evolutionarily conserved transmembrane receptor that has emerged as a potential therapeutic target for human tumors. PTK7 is a pseudokinase that is involved in the modulation of the Wnt signaling pathway through interactions with other receptors. These interactions result in targeted gene activation that regulates cell polarity, migration, and proliferation during embryogenesis. Aside of this role during development, PTK7 has been shown as overexpressed in numerous cancers including colon carcinoma, leukemia, neuroblastoma, hepatoma, and ovarian cance

Transcription factors (TFs) play a central role in gene regulation by binding to specific DNA sequences and orchestrating the transcriptional machinery. A majority of eukaryotic TFs have a block copolymer architecture, with at least one block being a folded DNA interaction domain, and another block being highly enriched in intrinsic disorder. In this study, we focus on NUP98-HOXA9 (NHA9), a chimeric TF implicated in leukemogenesis. By integrating experiments and simulations, we examine the structural dynamics of NHA9's FG domain across assembly states. We find that the FG domain has different

Cancer immunotherapy has witnessed remarkable advancements, particularly in the development of chimeric antigen receptor (CAR) T cell therapy. Here, we integrated single chain variable fragment (scFv) development with CAR T cell generation based on a newly developed scFv phagemid library. High-throughput long-read PacBio sequencing identified 4.5 × 107 unique full-length scFv proteins within the generated library. As a proof of principle, we screened for scFvs targeting C-type lectin-like molecule-1 (CLL1) with subsequent cloning into a third generation retroviral CAR backbone. Functional assa

Multiple myeloma (MM) remains incurable and patients eventually face the relapse/refractory dilemma. B cell maturation antigen (BCMA)-targeted immunotherapeutic approaches have shown great effectiveness in patients with relapsed/refractory MM, mainly including chimeric antigen receptor T cells (CAR-T), bispecific T cell engagers (TCEs), and antibody-drug conjugates (ADCs). However, their impact on long-term survival remains to be determined. Nonetheless, resistance to these novel therapies is still inevitable, raising a challenge that we have never met in both laboratory research and clinical

Pathogen evolution and narrow vaccine coverage urgently demand broad-spectrum vaccines. This review explores two pivotal technological fronts: structural biology- and immunoinformatics-guided antigen design, and utilizing nanoparticle-based delivery systems to induce broad immune responses. We critically analyze four antigen optimization strategies: (i) structure-based antigen design, (ii) conserved epitope targeting, (iii) consensus sequence-based antigen engineering, and (iv) chimeric immunogen design. Additionally, the common types and characteristics of nanoparticles are described briefly.

To enable fast and sensitive fusion detection critical for clinical oncology testing, we developed Fuzzion2, a pattern-matching program for detecting targeted gene fusions that employs an index of frequency minimizers and fuzzy matching to accommodate sequence variations. Running against 21,736 reference patterns representing chimeric fusions or internal tandem duplications, Fuzzion2 can analyze an unmapped RNA sequencing (RNA-seq) sample in minutes, at a sensitivity exceeding state-of-the art de novo fusion detection methods as demonstrated by dilution experiments. A comprehensive analysis on

Social determinants of health (SDOH) are an increasingly recognized prognostic factor in patients undergoing hematopoietic cell transplantation (HCT) and chimeric antigen receptor-T cell (CAR-T) therapy. However, awareness among providers of their importance and appropriate evaluation of SDOH factors in transplant candidates is insufficient in standard clinical practice and in research. As part of a coordinated effort by 3 American Society for Transplantation and Cellular Therapy (ASTCT) Special Interest Groups, we have developed a general overview of SDOH in transplantation and cellular thera

Therapeutic antibodies have revolutionized hematology, offering targeted and effective treatments for both malignant and non-malignant diseases. In hematologic malignancies, anti-CD20, anti-CD19, anti-CD38, and anti-B-cell maturation antigen (BCMA) antibodies have markedly improved survival outcomes, whereas antibody-drug conjugates and bispecific antibodies continue to expand therapeutic possibilities. Besides cancer, complement inhibitors such as eculizumab, ravulizumab, and the recently approved crovalimab have redefined paroxysmal nocturnal hemoglobinuria and atypical hemolytic uremic synd

Cytokine release syndrome (CRS) is a rare systemic inflammatory response that can be triggered by certain drugs and infections, commonly diagnosed at a disseminated stage, leading to poor prognosis. This has been well described following chimeric antigen receptor T-cell (CAR-T) therapy but has rarely been reported following antiprogrammed death ligand-1 (PDL-1) therapy. We present the case of an 86-year-old male with metastatic anal melanoma who developed CRS after his 4th cycle of pembrolizumab. His initial presentation was thought to be related to sepsis given his high fevers and hypotension

In rheumatoid arthritis (RA), the impaired function of regulatory T cells (Tregs) induces chronic inflammatory responses in the synovium. Cytotoxic T Lymphocyte Antigen 4 (CTLA4) is crucial for Sustaining Treg function. Uncoupling Protein 2 (UCP2) plays a key role in linking the glycolysis and fatty acid oxidation (FAO) pathways. Here we observed that Tregs in RA were predominantly characterized by a CD25intCTLA4intFOXP3intCD4high Phenotype. Mechanistically, elevated expression of UCP2 in RA Tregs disrupted metabolic homeostasis by downregulating Carnitine Palmitoyltransferase 2 (CPT2), a rate

Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen found on B lymphocytes. It is widely used in the treatment of haematological malignancies and, more recently, autoimmune diseases such as rheumatoid arthritis and scleroderma. While its safety profile is generally considered to be acceptable, a range of gastrointestinal adverse effects have been reported, including colitis. While this event is uncommon, there is a documented association with the development of de novo inflammatory bowel disease, particularly in patients with no prior history of digestive pathology. The c

Idiopathic inflammatory myopathies (IIMs), or myositis, are rare diseases marked by immune-driven muscle damage and complications like skin lesions and interstitial lung disease (ILD). Despite advances, challenges in diagnosis and treatment persist, particularly in inclusion body myositis (IBM), where no effective therapy exists. Recent breakthroughs, including transcriptomics and insights into antibody-mediated immunity and interferon (IFN) signaling, have clarified IIM pathophysiology and spurred the development of new therapies, such as chimeric antigen receptor (CAR) T cells and Janus kina

Mantle cell lymphoma (MCL) is a subtype of non-Hodgkin lymphoma, often characterized by a pattern of continued relapse after front-line chemoimmunotherapy. Although patients are usually able to regain durable disease control with covalent Bruton's tyrosine kinase inhibitors (cBTKi) at first relapse, it is now appreciated that such responses are often not sustained and the management of such patients represents a significant area of unmet need. There is an imperative to better understand resistance mechanisms and identify high-risk subsets of patients for whom cBTKi responses may be particularl

Cellular therapies have been cornerstones of the treatment of mantle cell lymphoma (MCL) for decades and have helped to improve the outcome of this formerly very unfavourable B-cell lymphoma considerably. Current established roles of cellular therapies include autologous hematopoietic cell transplantation (HCT) as part of first-line therapy, chimeric antigen receptor-engineered T-cells (CART) for relapsed/refractory MCL, and allogeneic HCT for settings in which CARTs have failed or are unavailable. Therapeutic innovations have recently entered the MCL treatment landscape and are moving upstrea

Follicular lymphoma (FL) is an indolent disease characterized by multiple relapses and eventual refractoriness to therapy. Despite advancements in therapeutic approaches, FL treatment algorithm and management remain not well-established, necessitating both ongoing research into novel therapeutic strategies and in stating patient journey. We propose a comprehensive overview of current standard treatments for relapsed or refractory (R/R) FL, including chemoimmunotherapy and stem cell transplantation, and insights into emerging therapies such as chimeric antigen receptor (CAR) T-cell therapy and

Osteoradionecrosis (ORN) of the maxilla is relatively uncommon. To the authors' knowledge, no previous reports have described reconstruction of malar and maxillary ORN using a thoracodorsal artery perforator flap combined with scapular bone. This flap has been used in oncologic resection, craniofacial dysplasia, and trauma, and it is also suitable for ORN reconstruction. The authors report a case involving a 60-year-old woman who developed malar and maxillary ORN 3 years after chemoradiotherapy for maxillary sinus carcinoma. The defect was reconstructed using this chimeric free flap, with addi
