Mechanism of quercetin in the treatment of endometriosis based on network pharmacology and transcriptome sequencing.

This study explores how quercetin may treat endometriosis (EMs) by combining network pharmacology and transcriptome sequencing approaches. Through network pharmacology, 132 shared targets between quercetin and EMs were identified, with KEGG pathway analysis suggesting that the MAPK signaling pathway could be a significant therapeutic target. Transcriptome sequencing revealed that PDGFRB was highly expressed in ectopic endometrial tissue, a finding confirmed by immunohistochemistry (IHC) showing elevated levels of PDGFRB, RAS, RAF1, and ERK1/2 in ectopic lesions. In an EMs mouse model, querceti

Alcohol attenuates CRF-induced excitatory effects from the extended amygdala to dorsostriatal cholinergic interneurons.

Alcohol relapse is associated with corticotropin-releasing factor (CRF) signaling and altered reward pathway function, though the precise mechanisms remain unclear. Here, using both mice and rats, we investigated how CRF modulates cholinergic interneurons (CINs) in the dorsal striatum, a region critical in mediating cognitive flexibility and action selection. Using monosynaptic and retrograde circuit tracing, we identified direct inputs from CRF-expressing (CRF+) neurons in the central amygdala (CeA) and bed nucleus of the stria terminalis (BNST) to dorsal striatal CINs. We showed that CINs ex

De novo L-(+)-tartaric acid biosynthesis in multi-modular engineered yeasts.

L-(+)-tartaric acid (L-TA) is a high-value chiral organic acid essential for food and pharmaceuticals. Despite its industrial importance, sustainable green production is constrained by the lack of a fully defined biosynthetic pathway. Here, we report the de novo biosynthesis of L-TA in Saccharomyces cerevisiae through reaction-guided enzyme mining, experimental validation, and Enzyme Commission-specific Catalytic Hybrid Optimizer (ECHO)-assisted enzyme prioritization. We first elucidate the elusive two-step conversion from precursor 5-keto-D-gluconic acid (5-KGA) to L-TA, catalyzed by transket

ZNF711 promotes enzalutamide resistance through transcriptional and epigenetic modification of the androgen receptor signaling pathway.

Although androgen receptor (AR) inhibitors such as enzalutamide are initially effective in castration resistant prostate cancer through suppression of AR signaling pathway, acquired resistance invariably develops, presenting a significant therapeutic challenge. Understanding the mechanisms of enzalutamide resistance (ENZR) is essential for developing improved therapeutic strategies. Here, we demonstrated that ZNF711 was significantly overexpressed in ENZR, and high ZNF711 levels correlated with poor clinical outcomes. Functionally, ZNF711 promoted ENZR progression both in vitro and in vivo. Me

BmSerpin-7 negatively regulates prophenoloxidase activation and antimicrobial peptide synthesis in Bombyx mori.

The prophenoloxidase (PPO) activation pathway and the Toll-mediated antimicrobial peptide (AMP) synthesis pathway constitute critical immune responses within the insect immune system. The activation of these pathways is driven by extracellular serine protease cascades, a process counterbalanced by the negative regulation of serpins. In this study, we identified a typical serpin, BmSerpin-7, and characterized its function in the silkworm, Bombyx mori. BmSerpin-7 expression was up-regulated in the hemocytes but down-regulated in the fat body after Micrococcus luteus and Yersinia pseudotuberculos

Mycobacterium tuberculosis Rv3875 upregulates TRIM21 expression through the type I interferon/STAT1 pathway and promotes intracellular growth.

Mycobacterium tuberculosis (Mtb) has evolved multiple strategies to acquire iron within macrophages, promoting its intracellular growth. We previously demonstrated that Mtb upregulates tripartite motif-containing protein 21 (TRIM21), an E3 ubiquitin ligase to initiate NCOA4-mediated ferritinophagy, enhancing iron bioavailability for bacterial survival. However, the specific virulence factor and underlying signaling pathway involved remain unclear. In this study, we show that TRIM21 upregulation requires stimulation by virulence factors derived from live Mtb. Through screening of Mtb secretory

MMT: A Potential Mechanism of Myocardial Fibrosis - Regulation of the TGF-β1/Smad3 Pathway and Prospects for Targeted Therapy.

Macrophage-Myofibroblast Transition (MMT) is a process in which macrophages, under the influence of specific factors such as transforming growth factor-beta 1 (TGF-β1) within a fibrotic microenvironment, gradually lose their original immune phenotype and acquire the biological characteristics of myofibroblasts. This transition involves changes in cell morphology, protein expression, and function, representing an important mechanism in the development and progression of tissue fibrosis. The TGF-β1/Smad3 signaling pathway, as a central regulator of fibrosis, plays a key role in MMT by modulating

Tryptophan's Journey Through Pregnancy: The Role of Serotonin.

Tryptophan is an essential amino acid required for protein synthesis and a precursor of key bioactive metabolites produced through the kynurenine, indole and serotonin pathways. Beyond its nutritional role, tryptophan metabolism critically regulates the nervous, immune and endocrine systems, coordinates circadian rhythms via melatonin, and exerts antioxidant effects. These processes are profoundly remodeled during pregnancy. Pregnancy represents a unique physiological state characterized by systemic adaptations, including changes in immunity and erythropoiesis, while the placenta emerges as a

The impact of maternal vitamin D deficiency during pregnancy and lactation on autism-like behavior in offspring.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social impairments and stereotyped behaviors. Many epidemiological studies have found a potential relationship between vitamin D deficiency (VDD) and ASD. However, the mechanism remains unclear. In this study, a VDD model was established by feeding a vitamin D-depleted diet to 5-week-old female mice, from their adulthood through pregnancy to end of lactation. Social deficits, repetitive stereotyped behaviors and anxiety-like behaviors were evaluated in the offspring. The results showed the number of buried marbles

Cystathionine β-synthase is inhibited by epinephrine and norepinephrine over-secretion via NF-κB activation in stress-induced hyperhomocysteinemia.

Stress can elevate plasma homocysteine (Hcy) levels, a key factor in the development of various diseases, leading to hyperhomocysteinemia (HHcy). The enzyme Cystathionine β-synthase (CBS) functions in the trans-sulfuration pathway, which links the methionine cycle to glutathione synthesis. This pathway is essential for the metabolism of homocysteine. However, the precise mechanism by which stress regulates hepatic CBS expression remains unclear. The present study aimed to elucidate the molecular mechanism by which CBS expression is regulated in the livers of restraint-stressed rats. Our result

The Viral Immunoshadow: Early Adenovirus Strategies for Cloaking Innate Immunity with E1A, E4orf1, and Beyond.

Human adenovirus (HAdV), a double-stranded DNA virus, targets terminally differentiated cells in the upper respiratory tract. As a key platform for gene therapy vectors, elucidating HAdV's virulence factors is vital for optimizing therapeutic applications and mitigating risks. To achieve productive replication, HAdV strategically neutralizes host immune defenses and induces S-phase pathways essential for viral propagation. This review synthesizes the latest insights into the key pathways through which HAdVs harness these early proteins to enhance virulence, skilfully evading and counteracting

UDP-glycosyltransferases-mediated vitellogenin protein biogenesis reveals juvenile hormone I-specific dominance in Spodoptera frugiperda reproductive programming.

Juvenile hormone (JH) critically regulates reproduction in Spodoptera frugiperda (FAW) via its signaling cascade. We dissected isoform-specific roles of JH I, II, and III by using hormone injection, RNAi, and RNA-seq. Systematic modulation of JH variants revealed distinct roles in ovarian development, egg production, and vitellogenin (Vg) dynamics. Transcriptomic profiling identified JH I as the most potent inducer of reproductive pathways, specifically enriching the steroid hormone biosynthesis pathway. Within this pathway, two UDP-glycosyltransferases (UGT), SfUGT2 and SfUGT2-like, were iden

Identification and differential expression analysis of microRNAs in the liver and spleen tissues of Yunnan Zebu and Holstein cattle.

Extensive research has shown that miRNAs play a crucial role in regulating biological processes within organisms. This study analyzed miRNAs differentially expressed and potentially associated with immune function and tissue-specific expression in Yunnan Zebu and Holstein cattle. To achieve this, 20 liver and spleen tissue samples from the two cattle breeds were collected for high-throughput miRNA sequencing, with the liver tissue as a reference. The findings revealed that bta-miR-122 and bta-miR-143 are the most abundantly expressed known miRNAs in the liver and spleen tissues of Yunnan Zebu

MEHP impairs male reproductive function through the lipid metabolism pathway: Evidence from in vitro human immature testis culture.

Di-(2-ethylhexyl) phthalate (DEHP) is a widely used industrial plasticizer whose main metabolite in the body is mono-(2-ethylhexyl) phthalate (MEHP). The reproductive toxicity of DEHP and MEHP has been documented in some studies. However, precise mechanisms remain unclear, and most research has focused on animal models. In this study, an in vitro culture system of immature human testicular tissue was employed to explore the etiological link between MEHP and developmental toxicity in immature human testes. The examination of micro-morphology, subcellular structure and testosterone secretion dem

SMARCA5 is required for the development of granule cell neuron precursors and Sonic Hedgehog Medulloblastoma growth.

Medulloblastoma constitutes a molecularly diverse group of malignant embryonal brain tumors. Sonic hedgehog molecular group of medulloblastoma (SHH-MB) is a highly heterogeneous tumor entity, characterized by constitutive activation of the SHH signaling pathway. Due to lack of suitable cell line models, little is known about genetic dependencies in SHH-MB outside of the SHH pathway. By performing a CRISPR-Cas9 dropout screen in SMB21 cells derived from SHH-MB in Ptch+/- mice, we aimed to identify genetic vulnerabilities in SHH-MB. Among the top scored gene hits, members of the SNF2-family of A

The Role of Prion Protein in Reelin/Dab1 Signaling: Implications for Neurodegeneration.

The cellular prion protein (PrPC) is studied in prion diseases, where its misfolded isoform (PrPSc) leads to neurodegeneration. PrPC has also been implicated in several physiological functions. The protein is abundant in the nervous system, and it is critical for cell signaling in cellular communication, where it acts as a scaffold for various signaling molecules. The Reelin signaling pathway, implicated both in Alzheimer's and prion diseases, engages Dab1, an adaptor protein influencing APP processing and amyloid beta deposition. Here, we show, using Prnp knockout models (Prnp0/0), that PrPC

METTL14-mediated depression of NEIL1 aggravates oxidative damage and mitochondrial dysfunction of lens epithelial cells through regulating KEAP1/NRF2 pathways.

Abnormal base excision repair (BER) pathway and N6-methyladenosine (m6A) of RNA have been proved to be significantly related to age-related cataract (ARC) pathogenesis. However, the relationship between the Nei Endonuclease VIII-Like1 (NEIL1) gene (a representative DNA glycosylase of BER pathway) and its m6A modification remains unclear. Here, we showed that the expression of NEIL1 was decreased in the ARC anterior lens capsules and H2O2-stimulated SRA01/04 cells. Our findings demonstrated that ectopic expression of NEIL1 alleviated DNA oxidative damage, apoptosis and mitochondrial dysfunction

Designing drugs against leishmaniasis: is targeting the sterol biosynthesis pathway the answer?

INTRODUCTION: Leishmaniasis is a devastating and complex parasitic disease caused by different species of protozoan members of the genus Leishmania. Unfortunately, available drugs are far from ideal and no vaccines are available. Under these circumstances, new effective antileishmanial drugs with reduced host toxicity and improved dosing protocols are urgently needed. The sterol biosynthesis pathway (SBP) is a promising focus for combating Leishmania infections. Thus, various strategies have been documented, such as drug repurposing, combined therapy, rational drug design, and the use of syner
