Immunosenescence is a complex biological process characterized by progressive remodeling of immune function during aging, leading to increased susceptibility to infections, chronic inflammatory diseases, cancer, and reduced vaccine efficacy. While cumulative antigen exposure, infections, lifestyle factors, environmental exposures, and epigenetic influences contribute to immune decline, a growing body of evidence suggests that genetic predisposition also plays an important modulatory role in shaping the molecular trajectory of immune aging. This review discusses how inherited variation in cytok

Lumbar spinal stenosis (LSS) affects over 100 million people globally, with an increasing incidence due to an ageing population. While LSS is known to be heritable, its genetic basis remains poorly understood. We conduct a genome-wide meta-analysis of LSS in 40,303 cases and 741,469 controls. We identify 73 previously unreported loci in addition to 15 known loci, and highlight spinal degeneration as a key pathogenic mechanism. In 12,784 surgically treated cases, we discover five loci specifically associated with severe disease. Age-of-onset analyses show that most variants influence risk after

Despite the high public health burden of diabetes in Africa, research into its genetic aetiology has been slow, limiting the continent's ability to benefit from an emerging era of diabetes precision medicine. Some progress is evident. In monogenic diabetes, where a molecular diagnosis enables tailored treatment, two cases from Africa successfully illustrate this approach despite the absence of routine affordable genetic testing, and control data from diverse African populations. Although limited, genome-wide association studies from African populations have discovered novel African-specific ty

SUMMARYViral myocarditis is an inflammatory disease of the heart muscle caused by diverse viral pathogens and shaped by complex interactions between viruses and host responses, ultimately leading to myocardial inflammation and dysfunction. While environmental factors contribute to disease onset, growing evidence highlights a central role for host genetic determinants in influencing susceptibility, severity, and clinical outcomes. In addition, sex-specific differences have emerged as important modifiers of immune responses and disease progression. This review examines the genetic architecture o

Type 1 diabetes mellitus (T1DM) is a multifactorial disease wherein a genetic predisposition, upon exposure to environmental factors, triggers seroconversion and subsequent beta-cell destruction. The global incidence of T1DM has risen in recent decades, underscoring the significant role environmental factors play in actuating inherent genetic risk. However, the absence of an identifiable unique triggering factor complicates the identification of risk groups. Advances in bioinformatics and epigenetic research are opening new opportunities for early diagnosis, prevention, and novel therapies for

Ménière's disease is an idiopathic disorder of the inner ear whose causes and pathogenetic mechanisms remain insufficiently elucidated. Advances in genetic analysis technologies have shifted attention toward the role of hereditary components in Ménière's disease, generating a considerable number of studies investigating the relationship between human genetic variations and disease onset. The results of these investigations highlight the complex and heterogeneous nature of pathogenesis, involving both genetic and epigenetic alterations. Studies have identified multiple candidate genes involved

Prion diseases are transmissible neurodegenerative diseases caused by misfolding of prion protein (PrP). All inherited prion diseases, including genetic Creutzfeldt-Jakob disease, Gerstmann-Sträussler-Scheinker disease, and fatal familial insomnia, are caused by pathogenic mutations in the prion protein gene (PRNP). The mutation partly dictates age at disease onset and clinical phenotype, presumably by its effects on the misfolding of PrP, but the clinical features can be highly variable even within the same pedigree. In sporadic prion diseases, genetic factors also play pivotal roles, as poly

Advances in 'omics' technologies have uncovered new insights into photoreceptor development. Dynamic expression of and interaction among transcription factors in post-mitotic photoreceptor precursors guide cone and rod cell fate and maturation, as well as homeostasis. Together with genetic and epigenetic modifications, we are now putting together the pieces of this complex puzzle to elucidate the molecular mechanisms underlying photoreceptor differentiation. In this review, we discuss our current understanding of developmental pathways that result in the generation of cones and rods from a com

PURPOSE: To investigate the heritability of genetic influence on macular choroidal vascularity index (CVI).
METHODS: Total choroidal area (TCA), luminal area (LA), and CVI was measured using horizontal scan of spectral-domain optical coherence tomography with enhanced depth imaging in the 373 healthy twin participants. Characteristics of the participants were investigated, including age, sex, axial length, hypertension, diabetes, drinking habits, and smoking status. Univariate and subsequent multivariate regression analyses were performed to evaluate the associations of these factors with TCA

Statin exposure has been associated with a broad spectrum of muscle toxicity, ranging from asymptomatic creatine kinase (CK) elevation to immune-mediated necrotizing myopathy (IMNM) with anti-3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) antibodies. The mechanisms underlying these adverse effects are not fully understood, and genetic predisposition may play a role. This observational study evaluated the association of HLA-DRB1*11 and SLCO1B1 rs4149056 variants with statin-induced muscle toxicity. A total of 62 statin-exposed patients treated at a single tertiary center were included

Peritoneal surface malignancies (PSMs) represent a biologically diverse group of cancers that range from primary peritoneal mesothelioma to metastatic gastrointestinal cancers. Because of the heterogenous nature of PSM, there is a large gap in molecular characterization of these cancers. This article reviews the underlying molecular and genetic mechanisms for PSM.

Metabolic and alcohol-related liver disease (MetALD) is a subcategory of steatotic liver disease (SLD) characterized by the coexistence of cardiometabolic risk factors and elevated alcohol intake. The global prevalence of MetALD is estimated to be 2 to 5%, but this is likely underestimated due to self-reporting biases. In real-world settings, fluctuations in alcohol intake mean that many patients with SLD may be classified as having MetALD at some point during their disease. Although MetALD is relatively common, only a minority of patients with the disorder progress to advanced chronic liver d

Muscular dystrophies are a group of heterogenic disorders characterized by progressive muscle weakness, the most common of them being Duchenne muscular dystrophy (DMD). Muscular dystrophies are caused by mutations in over 50 distinct genes, and many of them are caused by different genetic mechanisms. Currently, none of these diseases have a cure. However, in recent years, significant progress has been made to correct the underlying genetic cause. The clinical development of adeno-associated viral vector-based therapies has simultaneously produced excitement and disappointment in the research c

The conversion of proteins into insoluble fibrillar aggregates known as amyloid occurs in a wide variety of diseases, e.g., Alzheimer's disease, amyotrophic lateral sclerosis, systemic transthyretin amyloidosis, and multisystem atrophy. There are more than 60 disease-associated amyloid-forming proteins, and amyloid formation can occur sporadically or can be induced or accelerated by genetic mutations. This Review discusses structural mechanisms by which genetic changes promote amyloid formation and thereby influence disease outcomes. By dividing amyloid-forming proteins into six types accordin

BACKGROUND: Rare bleeding disorders (RBDs) account for almost 3% to 5% of bleeding disorders. Because of their rarity, phenotype and genotype analyses have been conducted in only a limited number of cases.
OBJECTIVES: To characterize the laboratory phenotype and genetic spectrum of a large international cohort of RBDs.
METHODS: A total of 807 individuals suspected of having RBDs were collected (2001-2020). The severity of RBDs was assessed based on plasma clotting factor activity levels. Gene sequencing was performed to confirm the diagnosis. Novel variants were assessed using in silico pred

Klebsiella pneumoniae is a clinically important pathogenic bacteria that poses a serious threat to human health. In particular, the emergence of hypervirulent and multidrug-resistant K. pneumoniae has posed great challenges in clinical anti-infective therapy. In the K. pneumoniae genome, mobile genetic elements (MGEs), such as plasmids, prophages, transposons, and insertion sequences, enhance bacterial viability and adaptation by mediating the horizontal transfer of virulence genes, antibiotic resistance genes, and other adaptive genes. This paper reviews the types and characteristics of the m

Most anxiety disorders 'run within families': people suffering from an anxiety disorder often have family members who are highly anxious as well. In this chapter, we explore recent work devoted to unraveling the complex interplay between genes and environment in the development of anxiety. We review studies focusing on the genetic vulnerability to develop social anxiety disorder (SAD), as SAD is one of the most prevalent anxiety disorders, with an early onset, a chronic course, and associated with significant life-long impairments. More insight into the development of SAD is thus of uttermost
