The nature of the spatial deficit in young females with Fragile-X syndrome: a neuropsychological and molecular perspective.

Spatial performance in a group of young Fragile-X syndrome females with FMR-1 full mutation was compared to two control groups of mainstream schoolchildren. Performance was assessed across a wide range of spatial tasks including visuo-spatial, visuo-construction, visuo-motor, visuo-perception and spatial-memory. A spatial deficit emerged only on those tasks which comprised a visuo-constructive component, with the Fragile-X group performing worse overall. All other tasks were performed at a comparable level across the three groups. Molecular analysis of the lymphocyte DNA found minimal evidence

Parental expectations and perceptions of augmentative and alternative communication: A Sri Lankan perspective.

From a critical disability rights lens, everyone has a right to communicate using any means available, which includes augmentative and alternative communication. The uptake of augmentative and alternative communication beyond the therapy room may be influenced by awareness, perceptions and acceptance of augmentative and alternative communication by caregivers, family members and the wider society. This study aimed to uncover parental perceptions of augmentative and alternative communication in a Global South context. Ten mothers and six fathers (16 participants) of children with complex commun

Not all roads lead to the immune system: the genetic basis of multiple sclerosis severity.

Multiple sclerosis is a leading cause of neurological disability in adults. Heterogeneity in multiple sclerosis clinical presentation has posed a major challenge for identifying genetic variants associated with disease outcomes. To overcome this challenge, we used prospectively ascertained clinical outcomes data from the largest international multiple sclerosis registry, MSBase. We assembled a cohort of deeply phenotyped individuals of European ancestry with relapse-onset multiple sclerosis. We used unbiased genome-wide association study and machine learning approaches to assess the genetic co

Neonatal exposure to BDE 209 impaired learning and memory, decreased expression of hippocampal core SNAREs and synaptophysin in adult rats.

Polybrominated diphenyl ethers (PBDEs) are a class of flame retardants. While the mechanism remains unknown, the potential neurotoxic effects of PBDEs remain a relevant issue. In the present study, neonatal Sprague-Dawley rats of both sexes were administered BDE 209 (1, 10, or 20mg/kg body weight) or peanut oil once daily from postnatal day (PND) 5 to PND 10. We examined the spatial learning and memory by Morris water maze and the working and reference memory by eight-arm radial maze in the stage of adulthood. Compared with controls, significantly longer escape latencies and fewer platform-cro

Early Detection of Learning Difficulties when Confronted with Novel Information in Preclinical Alzheimer's Disease Stage 1.

We employed a highly demanding experimental associative learning test (the AFE-T) to explore memory functioning in Preclinical Alzheimer's Disease stage 1 (PreAD-1) and stage 2 (PreAD-2). The task consisted in the learning of unknown object/name pairs and our comprehensive setup allowed the analysis of learning curves, immediate recall, long-term forgetting rates at one week, three months, and six months, and relearning curves. Forty-nine cognitively healthy subjects were included and classified according to the presence or absence of abnormal CSF biomarkers (Control, n = 31; PreAD-1, n = 14;

A selective allosteric potentiator of the M1 muscarinic acetylcholine receptor increases activity of medial prefrontal cortical neurons and restores impairments in reversal learning.

M(1) muscarinic acetylcholine receptors (mAChRs) may represent a viable target for treatment of disorders involving impaired cognitive function. However, a major limitation to testing this hypothesis has been a lack of highly selective ligands for individual mAChR subtypes. We now report the rigorous molecular characterization of a novel compound, benzylquinolone carboxylic acid (BQCA), which acts as a potent, highly selective positive allosteric modulator (PAM) of the rat M(1) receptor. This compound does not directly activate the receptor, but acts at an allosteric site to increase functiona

Auditory processing and learning/memory following erythropoietin administration in neonatally hypoxic-ischemic injured rats.

BACKGROUND: Hypoxia-ischemia (HI) is a common injury arising from prematurity/complications at birth and is associated with later language, auditory, and learning impairments. OBJECTIVE: To investigate the efficacy of two doses (300 or 1000 U/kg) of Erythropoietin (Epo) in protecting against neuropathological and behavioral impairments associated with HI injury in rats. METHODS: HI injury (right carotid artery cauterization and 120 min of 8% O(2)) was induced on postnatal day 7 (P7) and Epo or saline was administered i.p. immediately following the procedure. Auditory processing and learning/

Enhanced observation for management of agitated, confused or distressed behaviours in acute general hospitals: systematic review of quality and safety considerations.

BACKGROUND: People who are agitated, confused or distressed seldom receive holistic care within acute general hospitals. Unmet needs relating to conditions such as mental illnesses, learning disabilities or dementia can contribute to patient behaviours which staff find challenging to manage (e.g. self-harm). Enhanced observation, involving one-to-one care and monitoring, is a restrictive intervention often used to manage these presentations. The characteristics and outcomes of high-quality enhanced observation in this setting remain ill-defined, compromising quality and safety. AIMS: To under

Psychosis in a 47,XXX Adolescent: Clinical Management and Multidisciplinary Follow-up.

Triple X (47,XXX) syndrome is a sex chromosome aneuploidy associated with learning disabilities, attentional deficits, and social impairments. Its psychiatric manifestations, particularly psychosis, remain underrecognized. This case report describes a 13-year-old girl with 47,XXX syndrome who developed two recurrent psychotic episodes during adolescence and ultimately achieved sustained remission under clozapine, illustrating the need for individualized, genetically informed care. The patient initially presented with persecutory delusions, auditory hallucinations, behavioral changes, and funct

Performance of teenaged action video game players on the Developmental Eye Movement and King-Devick tests.

Action video game (AVG) players have been reported to demonstrate improved perceptual, cognitive, and motor skills, as well as enhanced eye movements such as saccade latency and accuracy. This study examined if these improvements are also observed in two common clinical eye movement assessments, the Developmental Eye Movement (DEM) and King-Devick tests. Ninth and tenth graders (15-16 years old) without learning disabilities or oculomotor dysfunctions, and who do not regularly play ball sports were tested in the DEM and King-Devick tests in random order. Those playing AVG ≥ 5 hours per week we

Health justice in fragile and shock-prone settings: from theory to practice towards building resilient health systems.

Health justice is an emerging imperative in global health and health policy and systems research, particularly in fragile and shock-prone settings where inequities are deepened by political instability, conflict and structural violence. This practice paper explores how the ReBUILD for Resilience consortium has sought to operationalise health justice as a guiding principle and embedded practice in four diverse contexts: Myanmar, Nepal, Lebanon and Sierra Leone. Drawing from political philosophy, public health ethics and the capability approach, we outline a framework that positions health justi

Thriving Beyond Adversity: A Prospective Longitudinal Cohort Study Using a Strength-Based Approach Depicts Indigenous Adolescents with Less Adverse Childhood Experiences (ACEs) Had Fewer Neurodevelopmental Disorders (NDDs).

Improving social and emotional well-being (SEWB) among Indigenous adolescents is crucial. Since neurodevelopmental disorders (NDDs) are common in Indigenous people and adverse childhood experiences (ACEs) are important contributors to negative health outcomes throughout the lifespan, we investigated whether limited ACE exposure is associated with reduced risk of NDDs in Australian Indigenous teens using the data from multiple waves (Wave 1 to Wave 9, and Wave 11) of the Longitudinal Study of Indigenous Children (LSIC). We also examined the role of other protective factors, such as Indigenous c
