Neurovascular coupling to D2/D3 dopamine receptor occupancy using simultaneous PET/functional MRI.

This study employed simultaneous neuroimaging with positron emission tomography (PET) and functional magnetic resonance imaging (fMRI) to demonstrate the relationship between changes in receptor occupancy measured by PET and changes in brain activity inferred by fMRI. By administering the D2/D3 dopamine receptor antagonist [(11)C]raclopride at varying specific activities to anesthetized nonhuman primates, we mapped associations between changes in receptor occupancy and hemodynamics [cerebral blood volume (CBV)] in the domains of space, time, and dose. Mass doses of raclopride above tracer leve

Model-based analysis and design of nerve cuff electrodes for restoring bladder function by selective stimulation of the pudendal nerve.

OBJECTIVE: Electrical stimulation of the pudendal nerve (PN) is being developed as a means to restore bladder function in persons with spinal cord injury. A single nerve cuff electrode placed on the proximal PN trunk may enable selective stimulation of distinct fascicles to maintain continence or evoke micturition. The objective of this study was to design a nerve cuff that enabled selective stimulation of the PN. APPROACH: We evaluated the performance of both flat interface nerve electrode (FINE) cuff and round cuff designs, with a range of FINE cuff heights and number of contacts, as well a

Differential recruitment of the sensorimotor putamen and frontoparietal cortex during motor chunking in humans.

Motor chunking facilitates movement production by combining motor elements into integrated units of behavior. Previous research suggests that chunking involves two processes: concatenation, aimed at the formation of motor-motor associations between elements or sets of elements, and segmentation, aimed at the parsing of multiple contiguous elements into shorter action sets. We used fMRI to measure the trial-wise recruitment of brain regions associated with these chunking processes as healthy subjects performed a cued-sequence production task. A dynamic network analysis identified chunking struc

Accelerated aging of the GABAergic septohippocampal pathway and decreased hippocampal rhythms in a mouse model of Alzheimer's disease.

Patients with Alzheimer's disease (AD) display altered functioning of cortical networks, including altered patterns of synchronous activity and a serious deficit in cholinergic septohippocampal (SH) innervation. However, the mechanisms underlying these alterations and the implication of the GABAergic SH component in AD are largely unknown. In addition, the GABAergic septohippocampal pathway (SHP) is believed to regulate synchronous hippocampal activity by controlling the activity of interneurons. Here we show, using well-characterized pathway tracing experiments, that innervation of the GABAer

Neurologist-associated reduction in PD-related hospitalizations and health care expenditures.

OBJECTIVE: To investigate the impact of neurologist care on Parkinson disease (PD)-related hospitalizations. Recent data indicate that neurologist treatment in PD may be associated with improved survival, yet is underutilized. Factors contributing to this improved survival remain unknown, but may be due in part to optimal disease treatment or avoidance of disease-related complications. METHODS: This was a retrospective cohort study of Medicare beneficiaries diagnosed with PD in 2002 and still living in 2006. Hospitalization for PD-related (neurodegenerative disease, psychosis, depression, uri

Resumption of ambulatory status after surgery for nonambulatory patients with epidural spinal metastasis.

BACKGROUND CONTEXT: Improving the surgical outcome of nonambulatory patients with metastatic epidural spinal compression has been of great interest lately. Although there have been many reports regarding the surgical outcome of spinal metastasis, the surgical outcome in terms of the probability of operative success for nonambulatory patients has not been thoroughly described. If the probability of ambulatory recovery is known, the optimal surgical indications can be determined and implemented. PURPOSE: To predict the surgical outcome and probability of ambulatory resumption for nonambulatory

Predicted effects of sensorineural hearing loss on across-fiber envelope coding in the auditory nerve.

Cross-channel envelope correlations are hypothesized to influence speech intelligibility, particularly in adverse conditions. Acoustic analyses suggest speech envelope correlations differ for syllabic and phonemic ranges of modulation frequency. The influence of cochlear filtering was examined here by predicting cross-channel envelope correlations in different speech modulation ranges for normal and impaired auditory-nerve (AN) responses. Neural cross-correlation coefficients quantified across-fiber envelope coding in syllabic (0-5 Hz), phonemic (5-64 Hz), and periodicity (64-300 Hz) modulatio

Non-motor basal ganglia functions: a review and proposal for a model of sensory predictability in auditory language perception.

While the primary function of the basal ganglia (BG) is linked to motor behaviour, several investigations of non-motor behaviour allocate cognitive and language-specific functions to the BG. What may such seemingly discrepant functions have in common? Some neurophysiologic theories of motor behaviour assign temporal sequencing, others the sequencing of general cognitive patterns to the BG. Turning to auditory language perception and syntax in particular, one may consider syntactic processing as a hierarchical sequencing phenomenon. Furthermore, previous data have shown that if events are predi

Lack of caspase-dependent apoptosis in spinal motor neurons of the wobbler mouse.

The mechanism of motor neuron degeneration in amyotrophic lateral sclerosis (ALS) is still unclear and the post-mortem analysis of samples from ALS patients does not permit a clarification of the early events of cell death occurring in ALS. Animal models of motor neuron degeneration represent a reliable tool to investigate the type of cell death. Attention was focused on the possible role of apoptosis in a spontaneous model of cervical spinal cord motor neuron degeneration, the wobbler mouse. Firstly, the rate of motor neuron loss occurring in the cervical spinal cord region of wobbler mice du

Nicotine-induced dystonic arousal complex in a mouse line harboring a human autosomal-dominant nocturnal frontal lobe epilepsy mutation.

We generated a mouse line harboring an autosomal-dominant nocturnal frontal lobe epilepsy (ADNFLE) mutation: the alpha4 nicotinic receptor S248F knock-in strain. In this mouse, modest nicotine doses (1-2 mg/kg) elicit a novel behavior termed the dystonic arousal complex (DAC). The DAC includes stereotypical head movements, body jerking, and forelimb dystonia; these behaviors resemble some core features of ADNFLE. A marked Straub tail is an additional component of the DAC. Similar to attacks in ADNFLE, the DAC can be partially suppressed by the sodium channel blocker carbamazepine or by pre-exp

Cellular therapies in motor neuron diseases.

Amyotrophic lateral sclerosis (ALS) and spinal muscular atrophy (SMA) are prototypical motor neuron diseases that result in progressive weakness as a result of motor neuron dysfunction and death. Though much work has been done in both diseases to identify the cellular mechanisms of motor neuron dysfunction, once motor neurons have died, one of potential therapies to restore function would be through the use of cellular transplantation. In this review, we discuss potential strategies whereby cellular therapies, including the use of stem cells, neural progenitors and cells engineered to secrete

Sensory gating deficits, pattern completion, and disturbed fronto-limbic balance, a model for description of hallucinations and delusions in schizophrenia.

Schizophrenia, if not the most difficult, is one of the most difficult mysterious puzzles for psychiatrists, psychologists, and neuroscientists to solve. In this paper, based on the previously known pathologies of schizophrenia, a new model is proposed for explanation of the formation of positive psychotic symptoms of hallucinations and delusions. This model can be used for understanding psychotic or psychotic-like positive symptoms of bipolar mood disorder, posttraumatic stress disorder, obsessive compulsive, and amphetamine and drug-induced psychotic disorders. Based on the postulated model,

Stimulation parameter optimization for functional electrical stimulation assisted gait in human spinal cord injury using response surface methodology.

BACKGROUND: The aims of this study were to identify the reflex moment induced by flexion withdrawal reflex and to optimize stimulation parameters for restoring swing motion with respect to initial kinematic conditions in human with spinal cord injury. METHODS: The influence of hip position and passive movement in the reflex moment were tested in six subjects with chronic spinal cord injury. The two-dimensional dynamic models consisted of thigh, shank and foot segments were developed to compute the swing-phase response and the response surface method was also used to optimize stimulation param

How position, velocity, and temporal information combine in the prospective control of catching: data and model.

The cerebral cortex contains circuitry for continuously computing properties of the environment and one's body, as well as relations among those properties. The success of complex perceptuomotor performances requires integrated, simultaneous use of such relational information. Ball catching is a good example as it involves reaching and grasping of visually pursued objects that move relative to the catcher. Although integrated neural control of catching has received sparse attention in the neuroscience literature, behavioral observations have led to the identification of control principles that

Empirical validation of the triple-code model of numerical processing for complex math operations using functional MRI and group Independent Component Analysis of the mental addition and subtraction of fractions.

The suitability of a previously hypothesized triple-code model of numerical processing, involving analog magnitude, auditory verbal, and visual Arabic codes of representation, was investigated for the complex mathematical task of the mental addition and subtraction of fractions. Functional magnetic resonance imaging (fMRI) data from 15 normal adult subjects were processed using exploratory group Independent Component Analysis (ICA). Separate task-related components were found with activation in bilateral inferior parietal, left perisylvian, and ventral occipitotemporal areas. These results sup

Left hemisphere specialization for the control of voluntary movement rate.

Although persuasive behavioral evidence demonstrates the superior dexterity of the right hand in most people under a variety of conditions, little is known about the neural mechanisms responsible for this phenomenon. As this lateralized superiority is most evident during the performance of repetitive, speeded movement, we used parametric rate variations to compare visually paced movement of the right and left hands. Twelve strongly right-handed subjects participated in a functional magnetic resonance imaging (fMRI) experiment involving variable rate thumb movements. For movements of the right
