Modeling the cell biology of PEX11β deficiency during human neurogenesis.

Peroxisomes are highly specialized organelles that are important for various metabolic functions, including β-oxidation of very-long-chain fatty acids and the synthesis of plasmalogens. Mutations in peroxisomal biogenesis proteins cause Zellweger spectrum disorders (ZSD), rare multisystem disorders often associated with neurological phenotypes. Unlike other peroxisome biogenesis proteins, PEX11β regulates peroxisomal fission, and PEX11β mutations result in milder metabolic phenotypes but persistent neurodevelopmental abnormalities, suggesting a role for PEX11β in neurodevelopment. To model PEX

Revision of Energy Metabolism Adaptations in High-Level Athletes: From Physical Performance Enhancement to Potential Therapeutic Targets in Mental Disorders.

High-level athletic performance requires the implementation of personalized strategies based on the analysis of metabolic pathways involved in energy production: phosphagen, glycolytic, and oxidative pathways. In this context, mitochondria play an essential role as the central regulator of energy production, being closely linked to these three pathways. Exercise boosts cellular respiration, which can also be optimized by nutritional interventions and targeted supplementation, promoting mitochondrial biogenesis, reducing oxidative stress and increasing ATP production. These metabolic adaptation

Human metabolic heat production under varying atmospheric and oxygen pressures during submaximal cycling.

Accurate thermal management in hypoxic and altered-pressure environments is essential, as variations in ambient pressure and oxygen availability can modify human metabolic heat production. This study investigated human metabolic responses under seven atmospheric conditions (normobaric, hypobaric, and micro-hyperbaric) with varying oxygen concentrations. Nine healthy male participants performed submaximal cycling while minute ventilation, respiration rate, respiratory quotient, body temperature, heart rate, and blood lactate were measured, along with subjective thermal sensation and fatigue. Feedback loops and circular causality: a grammar to restore harmony to physiological systems.

Causal inference and its graphical representation using directed acyclic graphs provide invaluable tools for calculating effects and handling confounding in medicine. However, since biological systems involve complex dynamics characterized by numerous feedback loops, practitioners need to abstract out the inner workings of cycles in order to reason with a causal framework. This text presents a step beyond linear causality, toward a language that can adequately capture the circular complexities underlying human health. The most fundamental homeostatic mechanisms are inherently cyclic, including

Cysteine excess triggers a mitochondrial iron-dependent cell death.

Thiol-containing metabolites are central to cellular redox homeostasis1. Among these, cysteine functions as a proteogenic amino acid, supports redox balance and iron-sulfur cluster biogenesis, and, when depleted, triggers ferroptosis2. Cells nevertheless maintain cysteine at low levels, reflecting its intrinsic toxicity, but the mechanisms by which excess cysteine causes cell death remain unclear3. Here we performed a genome-wide CRISPR screen and identified mitochondrial iron transporters as essential mediators of cysteine toxicity. Limiting mitochondrial iron availability suppresses cysteine

Trajectories of short-term operator adaptation in cobot-assisted disassembly: a within-subject multimodal analysis.

Collaborative robots are increasingly deployed in Industry 5.0 disassembly cells, yet how operators adapt across repeated cobot interactions is rarely characterised multimodally. The Day-2 axis of the MultiPhysio-HRC corpus was analysed: forty-two participants performed up to five repetitions each of a Fanuc CRX-20 cobot-assisted disassembly and a matched manual control, with 12-channel dry EEG, ECG, EDA, EMG and respiration recorded throughout; primary inference was based on the paired-analysis cohort (n = 39, ≥ 3 repetitions per context). Mixed-effects models and per-subject physiological re

Glutamine addiction is a therapeutic target to block emergency myelopoiesis.

Inflammation-driven emergency myelopoiesis (EM) contributes to the progression of many solid cancers and inflammatory diseases, yet therapeutic strategies to selectively suppress EM without compromising hematopoiesis remain lacking. Here, we use functional and single-cell transcriptomic analyses to determine metabolic programs organizing the hematopoietic hierarchy, myeloid lineage commitment, and myeloid differentiation. We identify de novo glutamine biosynthesis as a stem cell-specific survival mechanism allowing independence from exogenous glutamine. We show that myeloid differentiation is

Edible Fungi Are a Hidden Source of Carbon Monoxide and Carbon Dioxide.

This study provides the first clear evidence that edible mushrooms, such as Lentinula edodes (shiitake), Pleurotus ostreatus and Pleurotus eryngii, can generate carbon monoxide (CO) as part of their metabolic activity-independent of bacteria, illumination or oxygen limitation. Systematic measurements of CO and CO2 emissions were performed over 60 days using multiple fungal species, substrates and growth conditions. Microscopy observations (light, scanning and transmission electron microscopy) confirmed no extracellular and intracellular bacterial endosymbionts involved, supporting a fungal gen

Myoglobin in disease: the good and the bad.

Myoglobin is a member of the globin family, which was the first protein to have its crystal structure resolved, and is predominantly located in the skeletal and cardiac muscles. Its function is integral to various physiological processes, including oxygen storage and transport, nitric oxide (NO) metabolism, lipid metabolism, and mitochondrial respiration. Myoglobin has a dual role in various diseases, serving both as a pathogenic factor in acute kidney injury (AKI), diabetes and its complications, myoglobinopathies, and other conditions, and as a therapeutic factor in cancer, obesity, liver fi

Mitochondrial complex I activity promotes antigen cross-presentation in dendritic cells.

Mitochondrial metabolism modulates immune cell signaling, yet how individual electron transport chain complexes fine-tune dendritic cell (DC) function remains unclear. Here, we identify mitochondrial complex I (CI) as a critical metabolic checkpoint controlling antigen cross-presentation by DCs in mice. Deficiency of the CI subunit NDUFS4 in DCs led to the formation of a nonfunctional CI subcomplex, resulting in mildly impaired mitochondrial respiration without triggering a compensatory glycolytic shift. NDUFS4 deficiency limited endosomal escape of internalized antigens, thereby impairing ant

A gas-permeable edible coating alleviates CO2-induced off-flavor formation and reduces weight loss in postharvest citrus fruit.

Coating can reduce fruit weight loss and delay wrinkling, but often induces off-flavor formation. This study aims to develop coatings for reducing off-flavor formation in citrus fruits. Initially, nine single-component edible coatings were screened, identifying lipid-based carnauba (CNW) and candelilla wax (CW) as the most effective in reducing off-flavor and weight loss, respectively. Twenty CW-CNW composite coatings (ECs) were then formulated, with EC14 (2% CNW + 8% CW) showing optimal performance. The ethanol, acetaldehyde, and weight loss in EC14-coated Satsuma mandarin were 20.25%, 28.78%

Identification of carbon dioxide flux drivers in a High Arctic tundra environment.

We analysed the carbon dioxide fluxes from vegetation, measured by portable flux chambers, and several meteo-climatic and environmental variables obtained during six summer campaigns at two different sites in the High-Arctic tundra in the Bayelva basin near Ny Ålesund, Svalbard, during the period 2019-2025. The two sites have similar climatic conditions and vegetation cover, in terms of species distribution and abundance, but different distance from the glaciers. The measured fluxes provided information on Net Ecosystem Exchange (NEE), Ecosystem Respiration (ER) and Gross Primary Production (G

Low-oxygen storage modulates fruit flavor formation: Metabolic and regulatory insights.

Controlled atmosphere prolongs fruit storage period by reducing oxygen availability, thereby suppressing respiration, ethylene production, softening, and physiological disorders. However, low oxygen can compromise flavor, a critical attribute for consumer acceptance. This review summarizes current knowledge of how hypoxic storage influences sugar and acid metabolism, volatile biosynthesis, and associated molecular mechanisms. Low oxygen promotes sucrose breakdown and increases glucose and fructose accumulation, while suppressing acid degradation. However, volatile production is reduced through

SARS-CoV-2 ORF7a drives mitochondrial dysfunction via PDK4 activation and complex I inhibition.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection reprograms host metabolism to favor viral replication and immune evasion, yet the contribution of accessory proteins remains poorly defined. Here, we characterize the metabolic effects of the SARS-CoV-2 accessory protein ORF7a. Lentiviral expression of ORF7a in human lung epithelial (A549) and monocytic (THP1) cells, combined with integrated transcriptomic, proteomic, and metabolomic analyses, revealed marked dysregulation of glucose and lipid metabolism. ORF7a impaired mitochondrial oxidative phosphorylation, reducing basa

Selectively encapsulated and dry-fabricated sandwich-type patch: CO2/ClO2 differentiated dual-gas release for multi-dimensional preservation of purple cabbage.

Premature CO₂/ClO₂ loss during wet-casting fabrication bottlenecks moisture-responsive packaging scale-up. To address this, this study prepared polyacrylate nanoparticles loaded with 85.8% sodium chlorite (200-400 nm) and alginate-gelatin films (tensile strength 9.7 MPa, water vapor permeability 19.9 g·mm/m2·day·kPa). Dry nanoparticles, sodium bicarbonate, and citric acid were then loaded into alginate-gelatin pouches to create a sandwich patch, minimizing fabrication-stage gas loss. In humid environments, the polyacrylate core absorbed threefold more water than the film, driving unidirectiona

Lifelong restriction of dietary valine has sex-specific benefits for health and lifespan in mice.

Dietary protein is a key regulator of metabolic health in humans and rodents. Many of the benefits of protein restriction are mediated by reduced intake of dietary branched-chain amino acids (leucine, valine and isoleucine) and restriction of the branched-chain amino acids is sufficient to extend healthspan and lifespan in mice. Here we find that valine restriction (Val-R) improves metabolic health in C57BL/6J mice, promotes leanness and glycemic control across ages, and reduces frailty, cancer prevalence and senescent cell burden in both sexes while increasing median male lifespan by 23%. Oxygen Sensing in Plants.

Hypoxia in plants is detrimental to plant growth and crop productivity. It typically arises from adverse environmental conditions that limit oxygen availability, thereby restricting mitochondrial respiration. However, hypoxia also occurs in specific plant tissues where oxygen levels are low due to intense respiratory activity or limited oxygen diffusion. Under these conditions, the existence of a molecular oxygen-sensing mechanism directs plant metabolism and developmental pathways, matching plant fitness to the environment. Oxygen sensing also allows hypoxia to act as a developmental cue, gui

Faster-and-tighter nitrogen cycle supports mature forest productivity under elevated CO2.

Rising atmospheric carbon dioxide (CO2) concentration could enhance forest carbon (C) uptake, but nitrogen (N) is thought to progressively constrain this effect. Yet, significant biomass gains were demonstrated in a mature oak forest after 6 years of Free Air CO2 Enrichment. This study investigates whether changes in soil N fluxes explain these gains. We show that enhanced N mineralization and ecosystem N conservation support increased productivity under elevated CO2 (eCO2). In situ net and gross N ammonification was enhanced by ∼30%, with a pronounced effect at oak budburst. Higher N ammonifi

Emerging role of mitoNEET as mitochondrial sensor of hypoxia.

Mitochondria are organelles whose function and dynamics, that is fission, fusion, transport and selective removal (mitophagy), are fundamental for eukaryotic cells. Their dysfunction is associated with age-related neurodegeneration and the aging immune system. MitoNEET, an iron-sulfur (Fe-S) cluster-containing protein localised at the outer membrane of mitochondria, has emerged as a key regulator of mitochondrial respiration and linked to fission, fusion, and mitophagy. In this perspective article, we discuss recent emerging evidence describing the mitoNEET [2Fe-2S] cluster role as a sensor of

Young Regulatory T Cell-Derived Extracellular Vesicles Improve Mitochondrial Function and Angiogenesis and Suppress Inflammation in Senescent Brain and Heart.

The core mechanisms underlying aging involve genomic instability, cellular senescence, mitochondrial dysfunction, and chronic inflammation, necessitating multi-dimensional therapeutic interventions. Treg-derived extracellular vesicles (Treg-EVs) therapy, which circumvents the safety risks associated with live cell therapies, exhibits the potential to modulate metabolic and immune functions, offering promise for healthy aging. Here, we isolated Tregs from young male C57BL/6 mice and collected Treg-EVs. In vitro experiments demonstrated that Treg-EVs significantly attenuated cellular senescence,
