Endogenous viral elements and PIWI gene evolution in Spodoptera: diversity, conservation, and piRNA production.

Endogenous viral elements (EVEs) are viral sequences that have integrated into host genomes. While these elements can be considered molecular fossils of past infections, in some cases, they are co-opted and contribute to host fitness. For instance, in arthropods, some EVEs derived from non-retro-transcribing RNA viruses (nrEVEs) generate PIWI-interacting RNAs (piRNAs), and in Aedes mosquitoes at least, they act as immune effectors. Here, we investigated the diversity and function of nrEVEs across four Spodoptera species (Lepidoptera: Noctuidae), which are notorious agricultural pests. Comparat

A nonlinear time-domain finite-element model of the human middle ear subjected to intense sound waves.

Computational models of the middle ear are typically developed within the linear range of the middle ear's response, which corresponds to typical sounds in daily life. Understanding middle-ear biomechanics under high-intensity sounds is also important for assessing potential auditory damage under adverse conditions. In this study, a finite-element (FE) model of the human middle ear plus the external ear canal is presented, employing a first-order Ogden hyperelastic material formulation and time-domain nonlinear analysis to capture nonlinear deformation of soft tissues and the resulting middle-

Unparalleled nanofibril hydrogel actuators by mimicking nature's design.

Soft actuators aim to bridge the gap between rigid machines and soft matter by mimicking the flexibility and compliance of natural muscles and tissues. Stimuli-responsive hydrogels can fit this purpose through their softness and ability for large reversible shape morphing. However, the performance of hydrogel actuators is restricted by their diffusion-limited water transport, which leads to slow responses, and uniform volumetric changes that generate limited actuation forces and strains. Thus, there is a need to develop faster hydrogel actuators that can efficiently convert swelling into actua

Unraveling the regulatory role of intercellular communication in intestinal immune cells mediated by H₂ in sepsis recovery through single-cell RNA sequencing.

BACKGROUND: Sepsis is a life-threatening condition with dysregulated immune responses, where the intestinal immune system plays a pivotal role. While hydrogen gas (H₂) has shown therapeutic potential, its precise impact on intestinal immune cell communication remains elusive. METHODS: We employed a murine model of sepsis induced by cecal ligation and puncture (CLP). Mice were divided into control, sepsis, and H₂-treated sepsis groups. A comprehensive approach combining single-cell RNA sequencing (scRNA-seq) of intestinal tissues with histological, biochemical, and functional analyses was used

Three Unrelated Children With Childhood Apraxia of Speech: Exome Sequencing and Functional Gene Analysis Imply a Role of Laminin-511 in Early Neurodevelopment.

Childhood apraxia of speech (CAS) is characterized by motor discoordination in the speech domain and also in fine and gross motor systems, implicating the early developing cerebellum. Comorbidity with autism spectrum disorder (ASD) and other neurodevelopmental conditions has been observed. The genetic etiology is highly heterogeneous. Here, we present three unrelated individuals with CAS and concomitant fine and gross motor involvement but different genetic variants of interest. The DNA of the cases and their parents underwent exome sequencing and variant filtering. Using publicly available da

Validation of Cardiorespiratory-Gated Proton Beam Delivery for Stereotactic Arrhythmia Radioablation.

AIMS: Stereotactic arrhythmia radioablation (STAR) is a promising noninvasive therapy for refractory ventricular tachycardia. Using scanned proton beams for STAR enables precisely focused, high doses of ionizing radiation. Effective mitigation of target motion is critical for safety but remains challenging due to the complex interplay between beam scanning and cardiorespiratory motion. We evaluated the safety and feasibility of accurately delivering gated scanned proton beams to a ventricular myocardial target and compared lesion outcomes of respiratory (single)- and cardiorespiratory (dual)-g

Integrating Single-Cell Transcriptomics and Mendelian Randomization to Identify RAC1 as a Causal Metabolic Driver of Pericyte Dysfunction in Systemic Sclerosis.

BACKGROUND: Systemic sclerosis (SSc) is an autoimmune disease characterized by vascular injury and progressive fibrosis. Although microvascular injury is an inciting event and pericytes are recognized as a major source of myofibroblasts, the precise phenotypic heterogeneity of pericytes in the SSc microenvironment and the genetic mechanisms driving their pathological transition remain elusive. METHODS: This study systematically explored the cellular and genetic basis of pericyte dysfunction by integrating single-cell RNA sequencing (scRNA-seq) data from SSc patients with genome-wide associati

Epithelial FOXP3 Orchestrates O-Glycosylated IL6 Secretion to Drive Pancreatic Fibrocarcinogenesis.

BACKGROUND & AIMS: Pancreatic fibrosis characterizes pancreatic ductal adenocarcinoma, driving therapeutic resistance and organ failure. However, the mechanisms initiating fibrosis during early preneoplastic stages remain unclear. We investigated the role of the transcription factor FOXP3, unexpectedly identified within the epithelial cells of precancerous lesions. METHODS: Using human tissues and genetically engineered mouse models, we analyzed FOXP3 expression in premalignant lesions. We used epithelial-specific FOXP3 knockout and knockin strategies to determine its functional impact on fib

High-Intensity Interval Training Attenuates Hepatic Fibrosis by Remodeling Lactate Metabolism in MASLD.

Background: Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as a global metabolic disorder. As a non-pharmacological intervention, the effects of high-intensity interval training (HIIT) on MASLD and its molecular mechanisms remain poorly understood. This study aimed to investigate whether HIIT could ameliorate high-fat diet (HFD)-induced liver fibrosis by recalibrating the intrahepatic lactate metabolic axis. Methods: An HFD-induced murine MASLD model combined with HIIT intervention was utilized to evaluate the therapeutic efficacy and underlying mechanisms. Hepato

The role of monoclonal antibodies against IL-6 or IL-6R in the treatment of thyroid eye disease.

Thyroid eye disease (TED) is a chronic inflammatory autoimmune disorder characterized by expansion and fibrosis of orbital tissues, leading to eyelid retraction, proptosis, periorbital edema, restricted ocular motility, corneal ulceration, and (rarely) compressive optic neuropathy. The relationship between TED pathogenesis and interleukin 6 (IL-6)-mediated signaling has created considerable interest in the therapeutic potential of monoclonal antibodies (mAbs) against interleukin 6 (IL-6) or its receptor (IL-6R). A randomized controlled trial (RCT) and several non-RCTs have reported clinical be

The molecular landscape of hypertrophic cardiomyopathy across disease stages and genotypes.

Hypertrophic cardiomyopathy (HCM) is marked by asymmetric cardiac wall thickening, hypercontractility, diastolic dysfunction, and fibrosis. Pathogenic sarcomere gene variants cause HCM, but comparable abnormalities occur in patients with unexplained disease, albeit with fewer adverse events. To investigate stage- and genotype-specific disease mechanisms, we performed single-nucleus RNA sequencing of cardiac tissues from 47 patients with HCM, spanning obstructive HCM with preserved systolic function and end-stage HCM, and compared them with nonfailing donor and dilated cardiomyopathy hearts. We

Therapeutic Immune Reprogramming by Rapamycin Attenuates Plaque Inflammation and Lymphoid Immune Responses in Aged Atherosclerotic Mice.

Aging is a major risk factor for atherosclerosis and is accompanied by profound changes in the immune system, including effector T cell expansion, senescent cell accumulation, and increased pro-inflammatory signaling. Rapamycin, a promising rejuvenation therapeutic that inhibits mTORC1 and modulates immune aging, was investigated for its potential to attenuate pro-atherogenic immune responses in aged atherosclerotic mice. 80- to 90-week-old male Ldlr-/- mice with established atherosclerotic lesions were treated intraperitoneally with rapamycin or control triweekly for 8 weeks. SlpA-driven enhancement of immunogenicity and protection against Trypanosoma cruzi using a non-recombinant Lactobacillus acidophilus platform.

The rapid development of effective vaccines, particularly for diseases lacking prophylactic options, remains a critical global health priority. Among these, Chagas disease, caused by the intracellular parasite Trypanosoma cruzi, still has no approved vaccine despite decades of research. Conventional platforms often face limitations in terms of scalability, safety, or immunogenicity, especially when targeting complex parasitic infections. Although Lactobacillus acidophilus has been explored as a probiotic vector, its use in customizable vaccine platforms remains underdeveloped. Here, we evaluat

Hologenomic rewiring facilitates dietary adaptation to chitin-rich marine resources in the crab-eating frog.

BACKGROUND: Secondary adaptation of amphibians to marine environments is exceptionally rare. The crab-eating frog, Fejervarya cancrivora, is the only known amphibian capable of completing its life cycle in intertidal zones, where it faces dual challenges: high salinity stress and a diet rich in chitinous crab exoskeletons. While osmoregulatory adaptations have been well documented, the synergistic roles of the host's digestive system and its gut microbiota in this dietary specialization remain unclear. RESULTS: Here, we integrated histological analysis, comparative transcriptomics, chitinase

Comparative Impacts of Oral Amoxicillin, Azithromycin, and Clindamycin on Gut Microbiota and Intestinal Homeostasis.

Background: Amoxicillin, clindamycin and azithromycin are the most frequently prescribed antibiotics for odontogenic infections, but their comparative effects on gut microbiota and intestinal homeostasis remain insufficiently understood. Disruption of gut microbiota, short-chain fatty acid (SCFA) production, and mucosal barrier integrity may contribute to gastrointestinal symptoms. We aimed to compare the impacts of these antibiotics on gut microbiota, SCFA levels, and colonic goblet cells. Methods: C57BL/6N mice were treated with oral amoxicillin, clindamycin, or azithromycin at clinically re

Phase I Study of ROR1-Specific CAR-T Cells in Advanced Hematopoietic and Epithelial Malignancies.

PURPOSE: The receptor tyrosine kinase-like orphan receptor 1 (ROR1) is expressed in hematopoietic and epithelial cancers but has limited expression on normal adult tissues. This phase I study evaluated the safety of targeting ROR1 with autologous T lymphocytes engineered to express a ROR1 chimeric antigen receptor (CAR). Secondary objectives evaluated the persistence, trafficking, and antitumor activity of CAR-T cells. PATIENTS AND METHODS: Twenty-one patients with ROR1+ tumors received CAR-T cells at one of four dose levels: 3.3 × 105, 1 × 106, 3.3 × 106, and 1 × 107 cells/kg body weight, ad

β-Catenin interacts with canonical RBPs including MSI2 to associate with a Wnt signalling mRNA network in myeloid leukaemia cells.

Wnt/β-catenin signalling is important for normal hematopoietic stem/progenitor cell (HSPC) biology and heavily implicated in acute and chronic myeloid leukaemia (AML and CML). The central mediator β-catenin is an attractive therapeutic target in myeloid neoplasms however its targeting has been hampered by a poor characterisation of its molecular interactions in haematopoietic cells, which will differ from its network in solid tissues. Our previous β-catenin interactome study identified the significant enrichment of RNA-binding proteins (RBP) implying post-transcriptional roles for β-catenin in
