Membrane-reinforced three-dimensional electrospun silk fibroin scaffolds for bone tissue engineering.

Electrospun silk fibroin (SF) scaffolds have drawn much attention because of their resemblance to natural tissue architecture such as extracellular matrix, and the biocompatibility of SF as a candidate material to replace collagen. However, electrospun scaffolds lack the physical integrity of bone tissue scaffolds, which require resistance to mechanical loadings. In this work, we propose membrane-reinforced electrospun SF scaffolds by a serial process of electrospinning and freeze-drying of SF solutions in two different solvents: formic acid and water, respectively. After wet electrospinning f

Osteoblast behaviour on in situ photopolymerizable three-dimensional scaffolds based on D, L-lactide, epsilon-caprolactone and trimethylene carbonate.

Polymer networks formed by photocrosslinking of multifunctional oligomers have great potential as injectable and in situ forming materials for bone tissue engineering. Porous scaffolds varying in polyester type and crosslinking density were prepared from methacrylate-endcapped oligomers based on D,L-lactide, epsilon -caprolactone and trimethylene carbonate: LA/CL-hexanediol, LA/CL-dipentaerythritol and LA/TMC-HXD. The biocompatibility and bone formation were related with the degradation time and mechanical properties. The viability of fibroblasts was evaluated after incubation with extraction

Integration of nanozymes and metastructures on implants for confined sono-enhanced catalytic therapy.

Nanocatalytic medicine offers an innovative paradigm for disease treatment, yet systemic administration of nanomaterials raises critical biosafety concerns that restrict its application potential. While implant catalytic medicine represents a promising alternative, insufficient surface-active sites weaken its catalytic therapeutic performance. Here, we propose a universal confined sono-enhanced catalytic strategy realized by integrating nanozymes and metastructures into implants. The porous acoustic metastructure afforded abundant nanozyme immobilization sites and captured ultrasound to genera

Customized ultra-flexible and adhesive spinal cord stimulation electrode for chronic pain management.

Epidural spinal cord stimulation (SCS) has emerged as an effective therapy for chronic pain. However, its long-term efficacy is limited by the poor biocompatibility and instability at the electrode-tissue interface. Herein, we developed an innovative electrode fabrication strategy to address these limitations and validated it in mouse chronic pain models. We designed and synthesized a modified polydimethylsiloxane substrate carrying hydroxyl groups and sulfur anchor points. The hydroxyl groups enhance interfacial adhesion with surrounding tissue, improving mechanical stability and preventing e

Engineering Robust Supramolecular Nanoassemblies from Amino-Acid Functionalized Stiff-Stilbene Amphiphiles.

The unique functional properties and remarkable structural stability of proteins under extreme environments and conditions are largely predetermined by their constituent amino acid groups. However, the specific influence of molecular interactions arising from individual amino acids on overall structural stability of the resulting nanostructures remains insufficiently explored. Herein, we report a newly designed stiff-stilbene amphiphile functionalized with alanine amino acid groups (SA Ala ). The molecular design of SA Ala incorporates advanced π-π interactions between stiff-stilbene cores and

Development of Nigella sativa essential oil loaded keratin/gelatin cryogel with antimicrobial and antioxidant properties for wound dressing.

The effective management of infected and exudative wounds remains a persistent and critical challenge in clinical practice. Hydrogel-based dressings have emerged as a promising solution for wound healing due to their ability to mimic the native extracellular matrix. In this study, we developed an antimicrobial cryogel wound dressing composed of keratin, gelatin (Kr/Gel), and Nigella sativa essential oil (Kr/Gel@NS-EO). The formulation was engineered via a cryogelation process and strengthened through ionic crosslinking of sodium alginate. The physicochemical characterization using scanning ele

Synthesis of BSA-Coated Iron Oxide Nanoparticles with Size Control for High-Performance T1 Contrast Agents in Magnetic Resonance Imaging.

The excellent biocompatibility and favorable physicochemical properties of iron oxide nanoparticles have made them attractive candidates for magnetic resonance imaging. However, it remains challenging to synthesize high-performance T1 contrast agents with controlled sizes and biocompatible coating materials. In this study, we demonstrate a simple and environmentally friendly approach for synthesizing ultra-small iron oxide nanoparticles using bovine serum albumin (BSA) as a template. Following synthesis, the iron oxide nanoparticles (Fe3O4) were oxidized to Fe2O3 via the addition of hydrogen p

3D Bioprinting of Giant Salamander Mucin-Based Bioink for Enhanced Bone Regeneration.

There is an urgent need to overcome the limitations associated with limited autologous bone availability, allogeneic bone rejection, and the insufficient bioactivity of traditional materials for bone-defect repair. In this study, skin secretions from Andrias davidianus (SSAD) were compounded with methacrylated gelatin (GelMA) to develop a novel bioink and construct bone-regeneration scaffolds loaded with osteoblasts via extrusion-based 3D bioprinting. The optimized GelMA/SSAD bioink (50% v/v SSAD, 6% w/v GelMA) demonstrated excellent printability characterized by shear-thinning behavior and hi

Ultrathin and ultrastrong hydrogel bioelectronic membranes.

Hydrogels are promising materials for constructing next-generation bioelectronics because of their excellent biocompatibility and mechanical compliance. Yet, creating robust and multifunctional hydrogel devices that conform to the surface of 3D organs remains challenging. Here, we report a biomimetic strategy for engineering ultrathin and ultrastrong hydrogel membranes as an advanced platform for organ-conformal bioelectronics. In these hydrogels, self-organized nanofiber networks confer strain-stiffening characteristics with a phenomenal combination of high mechanical strength (∼13.65 MPa), f

Osteoinductive and Biocompatibility Assessment of a 3D-Printed Polymeric-Hydroxyapatite Composite Interference Screw.

Anterior cruciate ligament reconstruction relies on interference screw fixation, yet insufficient graft osseointegration remains a critical clinical challenge. This study aimed to develop and characterize a 3D-printed polymeric-hydroxyapatite composite interference screw with an osteoinductive surface to enhance localized osteogenic responses. Screws were designed, modeled, and fabricated using fused deposition modeling 3D printing with a polycaprolactone-poly(lactic-co-glycolic acid)-hydroxyapatite composite. Physico-chemical characterization was performed using scanning electron microscopy.

Dual-Oxidation Bacterial Cellulose Sponges with Rapid Shape Recovery for Crosslinker-Free Noncompressible Hemostasis and Tissue Regeneration.

Non-compressible hemorrhage (NCH) caused by deep tissue injury remains a major cause of trauma-related death. Developing hemostatic materials with rapid expansion, effective coagulation promotion, good biocompatibility, and tissue regeneration is still urgently needed. This study reports a simple regioselective dual-oxidation (RDO) strategy for fabricating crosslinker-free hemostatic sponges from bacterial cellulose (BC) with high porosity. Carboxyl and aldehyde groups were both introduced into the BC scaffold after TEMPO-mediated oxidation and sodium periodate oxidation. The dual-oxidation BC

Polydopamine as a biocompatible and precise mitochondrial targeted therapeutic platform for reversing myocardial ischemia-reperfusion injury.

Targeting mitochondria offers a compelling strategy for treating a broad spectrum of major diseases. However, the development of specific and biocompatible mitochondrial delivery vectors remains a key obstacle. In this study, we identified polydopamine (PDA)-a highly biocompatible material with inherent reactive oxygen species (ROS)-scavenging capabilities-as a naturally mitochondria-targeting biomaterial. PDA exhibits strong binding affinity to several critical outer mitochondrial membrane proteins, including the voltage-dependent anion channel and translocases of the outer membrane, conferri

The development of bone repair materials has become a critical focus in addressing bone defects. Existing methods, such as autografts and allografts, are limited by issues like immune rejection and insufficient local anti-infection. Despite the promise of biocompatible scaffolds in drug and cell delivery, challenges persist in creating uniform, stable and injectable scaffolds with multifunctional properties for effective bone repair. Here, we present a customized coaxial microfluidic system designed for the high-throughput fabrication of PDTH porous microspheres (PDTH PMs). This system ensures

Digital Light Processing 4D Printing of Poloxamer Micelles for Facile Fabrication of Multifunctional Biocompatible Hydrogels as Tailored Wearable Sensors.

The lack of both digital light processing (DLP) compatible and biocompatible photopolymers, along with inappropriate material properties required for wearable sensor applications, substantially hinders the employment of DLP 3D printing in the fabrication of multifunctional hydrogels. Herein, we discovered and implemented a photoreactive poloxamer derivative, Pluronic F-127 diacrylate, which overcomes these limitations and is optimized to achieve DLP 3D printed micelle-based hydrogels with high structural complexity, resolution, and precision. In addition, the dehydrated hydrogels exhibit a sha

Intrafibrillar mineralization of type I collagen with calcium carbonate and strontium carbonate induced by polyelectrolyte-cation complexes.

Calcium carbonate (CaCO3), possessing excellent biocompatibility, bioactivity, osteoconductivity and superior biodegradability, may serve as an alternative to hydroxyapatite (HAp), the natural inorganic component of bone and dentin. Intrafibrillar mineralization of collagen with CaCO3 was achieved through the polymer-induced liquid precursor (PILP) process for at least 2 days. This study aims to propose a novel pathway for rapid intrafibrillar mineralization with CaCO3 by sequential application of the carbonate-bicarbonate buffer and polyaspartic acid (pAsp)-Ca suspension.
