Pulsed field ablation (PFA) is emerging as a major advance in cardiac ablation because it induces irreversible electroporation rather than relying primarily on thermal injury. This article provides an overview of the core biophysical principles underlying PFA, focusing on concepts broadly relevant across systems and clinical applications. Externally applied pulsed electric fields interact with biological tissues and affect their transmembrane potential. When a critical threshold is exceeded, nanoscale aqueous pores form in the lipid bilayer, initiating electroporation. Depending on pulse ampli

Retroviruses comprise an ancient and varied group of viruses with the unique ability to integrate DNA from an RNA transcript into the genome, a subset of which are able to integrate in humans. The timing of these integrations during human history has dictated whether these viruses have remained exogenous and given rise to various human diseases or have become inseparable from the host genome (endogenous retroviruses). Given the ability of retroviruses to integrate into the host and subsequently co-opt host cellular process for viral propagation, retroviruses have been shown to be closely assoc

Pediatric burns remain a common injury after which many patients develop a severe form of scarring known as hypertrophic scarring. The formation of the hypertrophic scar arises from excessive production of collagen during wound healing. Wound repair and regeneration represents a complex process that is accomplished through many biological processes involving various cell types, extracellular matrix proteins, cytokines, and other mediators. One important cell type is the monocyte, which displays an altered profile in many wound models. These profile changes may function as biomarkers, reflectin

Virus-like particles are useful materials for studying virus-host interactions in a safe manner. However, the standard production of pseudovirus based on the vesicular stomatitis virus (VSV) backbone is an intricate procedure that requires trained laboratory personnel. In this work, a new strategy for creating virus-like proteoliposomes (VLPLs) and virus-like supported bilayers (VLSBs) is presented. This strategy uses a cell blebbing technique to induce the formation of nanoscale vesicles from the plasma membrane of BHK cells expressing the hemagglutinin (HA) fusion protein of influenza X-31.

Multiple myeloma is a malignant disease characterized by accumulation of clonal plasma cells in the bone marrow. Uncoupling of bone formation and resorption by myeloma cells leads to osteolytic lesions. These are prone to fracture and represent a possible survival space for myeloma cells under treatment causing disease relapse. Here we report on a novel approach suitable for local treatment of multiple myeloma based on hyaluronic acid (HA) hydrogels mimicking the physical properties of the bone marrow. The HA hydrogels are complexed with heparin to achieve sustained presentation and controlled

Benign and malignant thyroid nodules are more prevalent in females than in males. Experimental data suggest that the proliferative effect of oestrogen rather than polymorphisms is responsible for this gender difference. This study analysed whether both differentiated thyroid cells and thyroid stem and progenitor cells are targets of oestrogen action. In thyroid stem/progenitor cells derived from nodular goitres, the ability of 17β-oestradiol (E₂) to induce the formation of thyrospheres and the expression of oestrogen receptors (ERs) and the effect of E₂ on the growth and expression of markers

The avian pancreas has three or four lobes and develops from a dorsal and two ventral buds. The cells that will contribute to formation of the dorsal bud are at first located in the mid-dorsal endoderm, those of the ventral buds in the floor of the foregut. The determination of endoderm to form dorsal and ventral bud derivatives occurs before formation of the buds. The highest concentration of endocrine tissue is in the splenic lobe. The lobes contain A and B islets in which glucagon and insulin cells, respectively, predominate. Islets contain somatostatin and pancreatic polypeptide (PP) cells

The placenta is the critical interface between mother and fetus, and consequently, placental dysfunction underlies many pregnancy complications. Placental formation requires an adequate expansion of trophoblast stem and progenitor cells followed by finely tuned lineage specification events. Here, using single-cell RNA sequencing of mouse trophoblast stem cells during the earliest phases of differentiation, we identify gatekeepers of the stem cell state, notably Nicol1, and uncover unsuspected trajectories of cell lineage diversification as well as regulators of lineage entry points. We show th

While cancer immunotherapy has achieved groundbreaking clinical success, its efficacy is frequently compromised by insufficient T-cell activation, the immunosuppressive tumor microenvironment (TME), and off-target toxicity. Pyroptosis, a highly immunogenic form of programmed cell death characterized by gasdermin-mediated pore formation, massive cytokine release (e.g., IL-1β and IL-18), and robust dendritic cell activation, offers a compelling strategy to overcome these limitations. This review critically examines how nanotechnology-enabled pyroptosis induction can potentiate immunotherapy by (

To promote infections, pathogens exploit host cell machineries such as structural elements of the plasma membrane. Studying these interactions and identifying molecular players are ideal for gaining insights into the fundamental biology of the host cell. Here, we used the anthrax toxin to screen a library of 1,500 regulatory, cell-surface, and membrane trafficking genes for their involvement in the intoxication process. We found that endoplasmic reticulum (ER)-Golgi-localized proteins TMED2 and TMED10 are required for toxin oligomerization at the plasma membrane of human cells, an essential st

Periplocoside A (PSA) has been extracted from the Chinese herbal medicine Periploca sepium Bge to treat rheumatoid arthritis (RA) via immune regulation. We previously found that PSA exhibits immunosuppressive activity both in vitro and in vivo. Balanced regulation of helper T 17 (Th17)/regulatory T (Treg) cells is the current therapeutic direction for the treatment of RA. The present study investigated the mechanism of PSA in treating collagen-induced arthritis (CIA). The therapeutic effects and potential pharmacological mechanisms of PSA were specifically clarified by examining its effects on

The presence of at least three different types of somatostatin receptors in the thymus has been demonstrated using molecular biology techniques or radioimmunological methods. In view of the chemotactical activity shown by somatostatin in other lymphopoietic regions like bone marrow, it was hypothesised that this neuropeptide might play a role in regulating cell migration in the thymus. The selective expression of somatostatin receptors on specific immunocompetent cells, both circulating and in lymphatic organs, allows the neuropeptide to have a modulating action also on the immune system. Cell suspensions from the lymph nodes of small lymphocytic lymphoma (n = 94) and nodular and diffuse follicular center cell lymphomas (n = 330) were analyzed to evaluate the diagnostic significance of the surface immunoglobulin (SIg) intensity and mouse rosette assay (MR). In small lymphocytic lymphoma, SIg was monoclonal in 65 cases (69.15%), with weak fluorescence in 59 (90.77%). It was not detected in 29 cases (30.85%). The MR findings were positive in 68 cases (72.34%) and negative in 26 (27.66%). The combined results of these two assays showed the following: weak SIg/MR+, 35 (37.23%); wea

The tumorigenicity of adenovirus type 12 (Ad12)-transformed cells has been attributed to the low levels of class I major histocompatibility complex (MHC) protein expression by these cells. These levels of class I proteins are thought to be below the threshold critical for cytotoxic T-lymphocyte recognition, a process that may be involved in tumor cell immunosurveillance. We have used gene transfer experiments to investigate the role played by class I protein expression in the tumorigenicity of Ad12-transformed BALB/c mouse cells in naive, syngeneic adult mice. Our Ad12-transformed mouse cells

By means of a modified immune adherence (IA) technique, sera from melanoma patients were tested for the presence of antimelanoma antibodies. In total 13/73 sera tested showed a positive IA reaction of which 4/6 sera showed a positive reaction in the autologous situation. Sera from 33 patients with other tumors, 7 patients with non-neoplastic diseases and 50 healthy individuals did not show any IA reactivity towards melanoma cells. The reaction seemed to be selectively directed against tumor-associated antigens (TAA) on melanoma cells. No correlation with the stage of the disease could be found

The structure of the spleens of 12 patients with idiopathic thrombocytopenic purpura (ITP) who underwent splenectomy was studied. The white pulp was characterized by the presence of large numbers of lymphatic nodules containing highly reactive germinal centers. The marginal zone contained large numbers of vessels surrounded by plasma cells. These findings imply active antibody production and are consistent with the concept that the spleen is a major source of antiplatelet antibody production in ITP. Large numbers of platelets in various stages of degradation were seen in cords, particularly in

UNLABELLED: TNFα is the primary age-related cytokine responsible for hyperreactive platelet formation. In mouse models, increased TNFα induced abnormal megakaryocyte development and platelet activity. Here, we extend these findings to demonstrate that TNFα drives this thrombotic phenotype through expansion of the hematopoietic stem and progenitor cell (HSPCs) compartment. Using HSPC-specific TNFα receptor labeling and single cell RNA sequencing, we found that TNFα receptors are absent from megakaryocytes and their progenitors (MkPs) indicating that TNFα does not directly act on these cells.
